    Notizen: Continuous cultivation of Plasmodium falciparum presently requires the nutritionally complex medium, RPMI 1640. A basal medium of KCl, NaCl, Na2HPO4, Ca(NO3)2, MgSO4, glucose, reduced glutathione, HEPES buffer, hypoxanthine, phenol red (in RPMI 1640 concentrations), and 10% (v/v) exhaustively dialyzed pooled human serum was used to determine which vitamins and amino acids had to be exogenously supplied for continuous cultivation. Supplementation of basal medium with calcium pantothenate, cystine, glutamate, glutamine, isoleucine, methionine, proline, and tyrosine was necessary for continuous growth. This semi-defined minimal medium supported continuous growth of four isolates of P. falciparum at rates slightly less than those obtained with RPMI 1640. Adding any other vitamin or amino acid did not improve growth. Incorporation of several non-essential amino acids, particularly phenylalanine and leucine, into proteins was markedly enhanced in the minimal medium compared to RPMI 1640.

    Notizen: Eighty subprimals were fabricated from 20 pairs of beef strip loins from lean (fat thickness, 0.2-0.4 cm) US Standard and US Good carcasses. Three vacuum packaged (VP) and one dry aged treatments were randomly assigned to short loin sections of each carcass. Steaks from VP intact and VP not-reassembled subprimals were rated highest in overall appearance and eating quality. Dry aging reduced retail caselife, produced greater shrinkage and trim losses and lower overall satisfaction ratings by panelists. Precutting subprimals into steaks followed by VP aging tends to decrease retail caselife (especially if steaks are reassembled) and decrease juiciness and tenderness.

    Notizen: A gas chromatographic technique was used to determine sulfur dioxide and carbon dioxide in the headspace of packaged apples during storage. The headspace sulfur dioxide so determined decreased more rapidly and correlated better with nonenzymatic browning of the apples than free or total sulfur dioxide. The decrease in headspace sulfur dioxide, which was not accompanied by a comparable decrease in free sulfur dioxide, was attributed to the binding of sulfite to organic components of the apple. Free sulfur dioxide was found to measure not just the inorganic sulfite, which is commonly assumed, but also bound sulfite which was rapidly released under the acid conditions. Neither free nor total sulfur dioxide in a product could be quantitatively determined from headspace gas chromatographic techniques.

    Notizen: Trypsin inhibitors were isolated from seeds of Amaranthus hypochondriacus by extraction at pH 7.5, heat treatment at 70°C for 15 min and affinity chromatography with trypsin-Sepharose 4B. Inhibitors of trypsin and chymotrypsin were identified in polyacrylamide gels following electrophoresis using a stain procedure that employed the chromogenic substrate acetyl-D, L-phenylalanine-2-napthyl ester. Three of the thirteen trypsin inhibitors identified in the electrophoresis gels, also had antichymotryptic activity. The inhibitor preparation was very thermostable retaining 20% of its original activity after 7 hr at 100°C.

    Notizen: Examination of the diet of Lates calcarifer, based on 3684 specimens of 4–1200 mm total length (T.L.), from two localities in north Australia showed it is an opportunistic predator with an ontogenetic progression in its diet from microcrustacea to macrocrustacea to fish. The increasing importance of fish in the diet of L. calcarifer in salt water (up to 87% of food consumed) involved a dietary shift from feeding on bottom-dwelling forms to pelagic forms. There were few differences in the diet of L. calcarifer between the two localities. Cannibalism was rarely encountered in the Gulf of Carpentaria although it was significant (11–4% of the diet of L. calcarifer of 1001–1200 mm T.L.) in Van Diemen Gulf. A significant correlation between predator and prey length was found for Ariidae, Dorosomidae, Engraulidae, L. calcarifer, Mugilidae and Polynemidae. The maximum prey/predator length ratio observed was 0–61 for the dorosomid, Nematolosa erebi.

    Notizen: Metabolic consequences of chronic elevation of cortisol in the diet of yearling channel catfish, Ictalurus punctatus, were studied. Cortisol was incorporated into the diet in concentrations of 1, 10, 50 and 100 μg/g of food. This diet was offered at 3% of the body weight per day for 10 weeks. Fish were individually weighed and measured at 2-week intervals and feeding rates were adjusted. Body weight, liposomatic index and condition factor were significantly lower and food conversion was significantly higher in fish fed 50 or 100 μng cortisol/g of food when compared with controls. The hepatosomatic index of fish fed cortisol at the rate of 100 μg/g of food decreased significantly. Specific activity of hepatic tyrosine aminotransferase was significantly higher at the two highest cortisol doses. Long-term cortisol administration can reduce growth and condition factor by activating gluconeogenic mechanisms in which lipids and amino acids, rather than carbohydrates, are used for energy production. The metabolic effects of exogenous cortisol in this study offer an explanation for the decreased growth of fish under conditions that activate the secretion of endogenous cortisol.

In response to the needs of Corps professionals, the Hydrologic Engineering Center (HEC) has developed and supports a family of computer programs designed to aid them in their work. These programs include catchment, channel, alluvial, and statistical process models, system operation models, plan evaluation models, and data management programs. These models individually and collectively have been used throughout the Corps in a wide range of water resources planning studies.

    Notizen: Abstract Sulphate incorporation into glycopeptides appears to be a key event in the development of a number of organisms. An inhibitor of sulphation, sodium selenate, has been used in this study to examine the possibility that sulphation has a comparable role in the development of Dictyostelium discoideum. At concentrations of 0.1 mM and 1.0 mM, exogenously supplied selenate reversibly arrested the growth of bacterially grown amoebae of D. discoideum. In contrast, the effect of selenate on development was minimal. In the presence of 1.0 mM selenate, aggregation and tip formation were delayed 2–3 h and aggregates were slightly smaller; exogenous 0.1 mM selenate had no visible effect on development. However, the possibility that starved amoebae are impermeable to selenate was not excluded. The vegetative growth and development of an axenic strain in the presence of selenate closely resembled that of the bacterially grown strain. Since an inhibitory effect of 1.0 mM selenate on [35S]sulphate incorporation into acetone precipitable protein was also demonstrated, these results suggest that sulphation is necessary for the growth of D. discoideum.
