    Notes: An approximate experimental correction of electron diffraction intensities from an epitaxically crystallized phospholipid bilayer for dynamical scattering is described. This correction, which is useful for certain low-angle centrosymmetric data sets, compares intensities recorded at high and low electron-accelerating voltages to ascertain which reflections are most affected by n-beam interactions. When applied to experimental intensity data from 1,2-dihexadecyl-sn-glycerophosphoethanolamine (DHPE), the correction facilitates a direct phase determination based on the probabilistic estimate of three-phase invariants because a more accurate estimate of the hierarchy of |El| values is obtained. When a multisolution technique is used, incorporating algebraic unknowns for certain phase values, the best phase assignment can be assessed by comparison of the single convolution of phased structure factors to the observed structure-factor magnitudes for the low-voltage data. This approach exploits an approximate analogy made earlier by Moodie between the Sayre equation and the phase grating series and is valid as long as the single convolution adequately models experimental low-voltage data (a condition favored by light-atom structures in a low-angle region of reciprocal space). In real space, the correct structure can also be readily identified as the one having the smoothest density profile.

    Notes: Electron diffraction intensity data were collected at 1200 kV from thin epitaxically oriented crystals of copper perbromophthalocyanine (C32Br16CuN8) in a projection down molecular columns. Measured cell constants for the projection with cmm symmetry are d100 = 17.88 (9), b = 26.46 (15) Å. The structure was determined by Fourier refinement after three heavy-atom positions were identified in an initial potential map. In addition to the copper and halogens, all light-atom positions were found. Although the final R value for all data is 0.41, n-beam dynamical calculations for crystal thicknesses corresponding to the estimated sample dimension account for the observed amplitudes that deviate most from their kinematical values.

    Notes: Abstract We describe methods of fine scale chemical and topographical patterning of silicon substrates and the selected attachment and growth of central nervous system cells in culture. We have used lithography and microcontact printing to pattern surfaces with self-assembled monolayers and proteins. Chemical patterns can be created that localize and guide the growth of cells on the surfaces. Self-assembled surface texturing with structures at the tens of nanometers scale and lithographic based methods at the micrometer scale have been used to produce a variety of surface topographical features. These experiments suggest that surface texture at the scale of tens of nanometers to micrometers can influence the attachment of these cells to a surface and can be used as a mechanism of isolating cells to a particular area on a silicon substrate.

    Notes: Application of electron microscopy in a wide variety of fields of investigation has placed ever-expanding demands on the various components of the instrument. In situ specimen manipulation is one such demand and can often be critical to the success of an experiment. Control of specimen orientation is the most common manipulation, but control of a variety of other physical and chemical parameters may also be important. Temperature, gaseous and/or liquid environment, and mechanical operations are examples. Control and variation of these parameters in a small device (occupying a few cm3) operated in a strong magnetic field inside a vacuum system is often a considerable challenge. This must also be done at extreme stability: at least as good as the resolution limit of the microscope. Optimization of stage performance is too often sacrificed for optical performance or vice versa. The next generation of objective lenses and specimen stages are being designed in concert: an approach which should lead to an improved in situ laboratory, observed with optimum optics.

    Notes: Three-dimensional morphology and electrophysiology were correlated from individual neurons in a thick brain slice preparation. The hippocampal formation from immature and adult rats was cut transverse to the longitudinal axis into 500 μ Um-thick slices which were maintained under physiologic conditions. Individual neurons were impaled and physiologically characterized using microelectrodes. Recordings were made from the soma and in some cases from a dendrite. The impaled neurons were filled through the microelectrode with the fluorescent dye lucifer yellow and imaged by confocal scanning laser microscopy using an analog preprocessor. As many as 180 optical sections were recorded as a function of depth through the slices. Images are presented as a series of optical sections, stereo pairs, or three-dimensional reconstructions. Both stereo contouring and volume rendering methods were employed, and the reconstructions were viewed from any arbitrary perspective. Dendritic and axonal fields were separated from each other and displayed separately or as different pseudocolors. The three-dimensional reconstructions provided perspectives that were difficult or impossible to appreciate by viewing the optical sections or conventionally formed stereo pairs.
