    Notes: Summary A mathematical model was developed that prediets fluoride accumulation and clearance from the skeleton based upon fluoride bioavailability, bone remodeling rate, and the fluoride binding characteristics of bone. It was assumed that fluoride binds to bone in a nonlinear fashion such that a smaller percentage of fluoride is bound to bone if fluoride intake is increased to high levels. Bone resorption rate was assumed to be proportional to the solubility of hydroxyfluorapatite which is inversely related to bone fluoride content. The predictions made by the model compared favorably with experimental results from fluoride uptake and clearance studies. Parametric studies done using the model showed the following: (1) fluoride can be cleared from the skeleton by bone remodeling, but fluoride clearance takes over four times longer than does fluoride uptake; and (2) fluoride uptake by the skeleton was positively associated with bone remodeling rate. However, the concentration of fluoride in newly formed bone does not decrease with reduced remodeling rates and surpasses 10,000 ppm for intakes of fluoride greater than 9 mg/day. For osteoporosis, daily dose and duration of fluoride treatment should be selected to avoid reaching a toxic cumulative bone fluoride content.

    Notes: Abstract The elastic properties of an anisotropic porous material can be represented as functions of the material's solid volume fraction (or porosity) and the principal diameters of the material's fabric ellipsoid. The fabric ellipsoid is a measure of the anisotropy of the microstructure of a material. The definitions and measurement techniques for fabric ellipsoids in granular materials, foams, cancellous bone, and rocks are discussed. The principal results presented in this work are algebraic expressions for the dependence of the orthotropic elastic constants upon both solid volume fraction and the fabric ellipsoid.

    Notes: The skeleton has the ability to alter its mass, geometry, and strength in response to mechanical stress. In order to elucidate the molecular mechanisms underlying this phenomenon, differential display reverse transcriptase-polymerase chain reaction (DDRT-PCR) was used to analyze gene expression in endocortical bone of mature female rats. Female Sprague-Dawley rats, approximately 8 months old, received either a sham or bending load using a four-point loading apparatus on the right tibia. RNA was collected at 1 h and 24 h after load was applied, reverse-transcribed into cDNA, and used in DDRT-PCR. Parallel display of samples from sham and loaded bones on a sequencing gel showed several regulated bands. Further analysis of seven of these bands allowed us to isolate two genes that are regulated in response to a loading stimulus. Nucleotide analysis showed that one of the differentially expressed bands shares 99% sequence identity with rat osteopontin (OPN), a noncollagenous bone matrix protein. Northern blot analysis confirms that OPN mRNA expression is increased by nearly 4-fold, at 6 h and 24 h after loading. The second band shares 90% homology with mouse myeloperoxidase (MPO), a bactericidal enzyme found primarily in neutrophils and monocytes. Semiquantitative PCR confirms that MPO expression is decreased 4- to 10-fold, at 1 h and 24 h after loading. Tissue distribution analysis confirmed MPO expression in bone but not in other tissues examined. In vitro analysis showed that MPO expression was not detectable in total RNA from UMR 106 osteoblastic cells or in confluent primary cultures of osteoblasts derived from either rat primary spongiosa or diaphyseal marrow. Database analysis suggests that MPO is expressed by osteocytes. These findings reinforce the association of OPN expression to bone turnover and describes for the first time, decreased expression of MPO during load-induced bone formation.     Description: Arc front stratovolcanoes have global chemical systematics that constrain processes at convergent margins. Positive correlations exist for arc averages among “fluid mobile,” “high field strength,” and “large ion lithophile” elements. 143 Nd/ 144 Nd and 87 Sr/ 86 Sr from rear-arc lavas lacking subduction signature align with the oceanic “mantle array,” and correlate with arc front 143 Nd/ 144 Nd. Most chemical parameters (but not isotopes) also correlate well with crustal thickness and slightly less well with the slab thermal parameter, but not with the depth of the slab or model slab surface temperatures. Successful models of arc volcanism should account for these global regularities. Two distinct models can quantitatively account for the observations—different extents of melting of the mantle wedge caused by variations in wedge thermal structure, or varying contributions from the subducting slab owing to variations in the slab thermal structure. The wedge melting model has constant contributions from ocean crust, sediment and mantle wedge to lavas globally, while the slab model varies slab contributions with slab temperature. The wedge melting model fit improves by incorporating convergence rate and slab dip, which should affect the wedge thermal structure; the slab model is not supported by a similar analysis. The wedge model also more easily accommodates the isotope data. The two models predict different primary H 2 O contents, with large variations in H 2 O for the wedge model, and relatively constant H 2 O for the slab model. An evaluation of the effects of varying sediment compositions on arc lavas will benefit from considering the very different consequences of the two models. This article is protected by copyright. Dungan & Stéphane Escrig The processes that lead to the fourfold variation in arc-averaged compositions of mafic arc lavas remain controversial. Control by the mantle-wedge thermal structure is supported by chemical correlations with the thickness of the underlying arc crust, which affects the thermal state of the wedge. Control by down-going slab temperature is supported by correlations with the slab thermal parameter. The Chilean Southern Volcanic Zone provides a test of these hypotheses. Here we use chemical data to demonstrate that the Southern Volcanic Zone and global arc averages define the same chemical trends, both among elements and between elements and crustal thickness. But in contrast to the global arc system, the Southern Volcanic Zone is built on crust of variable thickness with a constant slab thermal parameter. This natural experiment, along with a set of numerical simulations, shows that global arc compositional variability is dominated by different extents of melting that are controlled by the thermal structure of the mantle wedge. Slab temperatures play a subordinate role. Variations in the subducting slab’s fluid flux and sediment compositions, as well as mantle-wedge heterogeneities, produce second-order effects that are manifested as distinctive trace element and isotopic signatures; these can be more clearly elucidated once the importance of wedge thermal structure is recognized.
