    Description / Table of Contents: Die Dobsonmessungen der Stadt Potsdam und der Ozonsondenmessungen von Lindenberg werden homogenisiert und klimatologisch ausgewertet. Zur Homogenisierung der Dobsonmessungen werden dabei neben den allgemein üblichen Verfahren Regressionsbeziehungen mit aerologischen Temperaturwerten als Prediktoren verwendet, die sowohl als Qualitätstest für die Dobsonmessung als auch als Prognosegleichung zur Vorhersage des Gesamtozongehalts verwendet werden können und wurden. Es wird gezeigt, daß sowohl die Verteilungsmuster des Gesamtozongehalts als auch die der langzeitigen Ozonänderungen wetterlagenabhängig sind, wenn zur Wetterlagenklassifikation ein objektives Klassifikationsschema mittels Clusteranalyse auf der Basis des Bodendruckfeldes und des Höhendruckfeldes der 500 hPa-Fläche verwendet wird. Diskutiert werden die Beziehungen zwischen objektiv klassifizierten Wetterlagen und den weit verbreiteten Wetterlagen nach Hess-Brezowsky. Für alle Wetterlagen wird für das Untersuchungsgebiet i.a. Ozonabnahme für alle Jahreszeiten gefunden. Änderungen in der atmosphärischen Zirkulation modifizieren damit auch die Stärke der Ozonabnahme, können aber eine Umkehr des Trends nicht bewirken.

    Description: Homeostatic control of dendritic cell (DC) survival is crucial for adaptive immunity, but the molecular mechanism is not well defined. Moreover, how DCs influence immune homeostasis under steady state remains unclear. Combining DC-specific and -inducible deletion systems, we report that transforming growth factor beta-activated kinase 1 (TAK1) is an essential regulator of DC survival and immune system homeostasis and function. Deficiency of TAK1 in CD11c+ cells induced markedly elevated apoptosis, leading to the depletion of DC populations, especially the CD8+ and CD103+ DC subsets in lymphoid and nonlymphoid tissues, respectively. TAK1 also contributed to DC development by promoting the generation of DC precursors. Prosurvival signals from Toll-like receptors, CD40 and receptor activator of nuclear factor-κB (RANK) are integrated by TAK1 in DCs, which in turn mediated activation of downstream NF-κB and AKT-Foxo pathways and established a gene-expression program. TAK1 deficiency in DCs caused a myeloid proliferative disorder characterized by expansion of neutrophils and inflammatory monocytes, disrupted T-cell homeostasis, and prevented effective T-cell priming and generation of regulatory T cells. Moreover, TAK1 signaling in DCs was required to prevent myeloid proliferation even in the absence of lymphocytes, indicating a previously unappreciated regulatory mechanism of DC-mediated control of myeloid cell-dependent inflammation. Therefore, TAK1 orchestrates a prosurvival checkpoint in DCs that affects the homeostasis and function of the immune system.

    Description: Significant areas of the southern USA periodically experience intense drought that can lead to episodic tree mortality events. Because drought tolerance varies among species and size of trees, such events can alter the structure and function of terrestrial ecosystem in ways that are difficult to detect with local data sets or solely with remote-sensing platforms. We investigated a widespread tree mortality event that resulted from the worst one-year drought on record for the state of Texas, USA. The drought affected ecoregions spanning mesic to semiarid climate zones and provided a unique opportunity to test hypotheses related to how trees of varying genus and size were affected. The study was based on an extensive set of 599 distributed plots, each 0.16 ha, surveyed in the summer following the drought. In each plot, dead trees larger than 12.7 cm in diameter were counted, sized, and identified to the genus level. Estimates of total mortality were obtained for each of 10 regions using a combination of design-based estimators and calibrated remote sensing using MODIS 1-yr change in normalized difference vegetation index products developed by the U.S. Forest Service. As compared with most of the publicized extreme die-off events, this study documents relatively low rates of mortality occurring over a very large area. However, statewide, regional tree mortality was massive, with an estimated 6.2% of the live trees perishing, nearly nine times greater than normal annual mortality. Dead tree diameters averaged larger than the live trees for most ecoregions, and this trend was most pronounced in the wetter climate zones, suggesting a potential re-ordering of species dominance and downward trend in tree size that was specific to climatic regions. The net effect on carbon storage was estimated to be a redistribution of 24–30 Tg C from the live tree to dead tree carbon pool. The dead tree survey documented drought mortality in more than 29 genera across all regions, and surprisingly, drought resistant and sensitive species fared similarly in some regions. Both angiosperms and gymnosperms were affected. These results highlight that drought-driven mortality alters forest structure differently across climatic regions and genera.     Description: This study aims to compare and validate two soil-vegetation-atmosphere-transfer (SVAT) schemes: TERRA-ML and the Community Land Model (CLM). Both SVAT schemes are run in standalone mode (decoupled from an atmospheric model) and forced with meteorological in-situ measurements obtained at several tropical African sites. Model performance is quantified by comparing simulated sensible and latent heat fluxes with eddy-covariance measurements. Our analysis indicates that the Community Land Model corresponds more closely to the micrometeorological observations, reflecting the advantages of the higher model complexity and physical realism. Deficiencies in TERRA-ML are addressed and its performance is improved: (1) adjusting input data (root depth) to region-specific values (tropical evergreen forest) resolves dry-season underestimation of evapotranspiration; (2) adjusting the leaf area index and albedo (depending on hard-coded model constants) resolves overestimations of both latent and sensible heat fluxes; and (3) an unrealistic flux partitioning caused by overestimated superficial water contents is reduced by adjusting the hydraulic conductivity parameterization. CLM is by default more versatile in its global application on different vegetation types and climates. On the other hand, with its lower degree of complexity, TERRA-ML is much less computationally demanding, which leads to faster calculation times in a coupled climate simulation.

    Description: Host Cell Factor 1 (HCF-1) plays critical roles in regulating gene expression in a plethora of physiological processes. HCF-1 is first synthesized as a precursor, and subsequently specifically proteolytically cleaved within a large middle region termed the proteolytic processing domain (PPD). Although the underlying mechanism remains enigmatic, proteolysis of HCF-1 regulates its transcriptional activity and is important for cell cycle progression. Here we report that HCF-1 proteolysis is a regulated process. We demonstrate that a large proportion of the signaling enzyme O-linked-N-acetylglucosaminyl transferase (OGT) is complexed with HCF-1 and this interaction is essential for HCF-1 cleavage. Moreover, HCF-1 is, in turn, required for stabilizing OGT in the nucleus. We provide evidence indicating that OGT regulates HCF-1 cleavage via interaction with and O-GlcNAcylation of the HCF-1 PPD. In contrast, although OGT also interacts with the basic domain in the HCF-1 amino-terminal subunit, neither the interaction nor the O-GlcNAcylation of this region are required for proteolysis. Moreover, we show that OGT-mediated modulation of HCF-1 impacts the expression of the herpes simplex virus immediate-early genes, targets of HCF-1 during the initiation of viral infection. Together the data indicate that O-GlcNAcylation of HCF-1 is a signal for its proteolytic processing and reveal a unique crosstalk between these posttranslational modifications. Additionally, interactions of OGT with multiple HCF-1 domains may indicate that OGT has several functions in association with HCF-1.

    Description: Canadian Journal of Forest Research, e-First Articles. We investigated the effects of a 5 °C soil + air experimental heating on root and microbial respiration in a boreal black spruce (Picea mariana (Mill.) B.S.P.) forest in northern Manitoba, Canada, that was warmed between 2004 and 2007. In 2007, the 14C/12C signatures of soil CO2 efflux and root and soil microbial respiration were used in a two-pool mixing model to estimate their proportional contributions to soil CO2 efflux and to examine how each changed in response to the warming treatments. In laboratory incubations, we examined whether warming had altered microbial respiration rates or microbial temperature sensitivity. The 14C/12C signature of soil CO2 efflux and microbial respiration in the heating treatments were both significantly (p 〈 0.05) enriched relative to the control treatment, suggesting that C deposited nearer the atmospheric bomb peak in 1963 contributed more to microbial respiration in heated than control treatments. Soil CO2 efflux was significantly greater in the heated than control treatments, suggesting the acclimation to temperature of either root or microbial respiration was not occurring in 2007. Microbial respiration in laboratory incubations was similar in heated and control soils. This study shows that microbial respiration rates still responded to temperature even after 4 years of warming, highlighting that ecosystem warming can cause a prolonged release of soil organic matter from these soils.
