    Description: Ice cores in Antarctica and Greenland reveal ice-crystal fabrics that can be softer under simple shear compared with isotropic ice. Owing to the sparseness of ice cores in regions away from the ice divide, we currently lack information about the spatial distribution of ice fabrics and its association with ice flow. Radio-wave reflections are influenced by ice-crystal alignments, allowing them to be tracked provided reflections are recorded simultaneously in orthogonal orientations (polarimetric measurements). Here, we image spatial variations in the thickness and extent of ice fabric across Dome A in East Antarctica, by interpreting polarimetric radar data. We identify four prominent fabric units, each several hundred metres thick, extending over hundreds of square kilometres. By tracing internal ice-sheet layering to the Vostok ice core, we are able to determine the approximate depth–age profile at Dome A. The fabric units correlate with glacial–interglacial cycles, most noticeably revealing crystal alignment contrasts between the Eemian and the glacial episodes before and after. The anisotropy within these fabric layers has a spatial pattern determined by ice flow over subglacial topography.

    Description: Motivation: microRNAs (miRNAs) are known to play an essential role in the post-transcriptional gene regulation in plants and animals. Currently, several computational approaches have been developed with a shared aim to elucidate miRNA–mRNA regulatory relationships. Although these existing computational methods discover the statistical relationships, such as correlations and associations between miRNAs and mRNAs at data level, such statistical relationships are not necessarily the real causal regulatory relationships that would ultimately provide useful insights into the causes of gene regulations. The standard method for determining causal relationships is randomized controlled perturbation experiments. In practice, however, such experiments are expensive and time consuming. Our motivation for this study is to discover the miRNA–mRNA causal regulatory relationships from observational data. Results: We present a causality discovery-based method to uncover the causal regulatory relationship between miRNAs and mRNAs, using expression profiles of miRNAs and mRNAs without taking into consideration the previous target information. We apply this method to the epithelial-to-mesenchymal transition (EMT) datasets and validate the computational discoveries by a controlled biological experiment for the miR-200 family. A significant portion of the regulatory relationships discovered in data is consistent with those identified by experiments. In addition, the top genes that are causally regulated by miRNAs are highly relevant to the biological conditions of the datasets. The results indicate that the causal discovery method effectively discovers miRNA regulatory relationships in data. Although computational predictions may not completely replace intervention experiments, the accurate and reliable discoveries in data are cost effective for the design of miRNA experiments and the understanding of miRNA–mRNA regulatory relationships.     Description: 〈p〉Cas9 is an RNA-guided endonuclease that targets complementary DNA for cleavage and has been repurposed for many biological usages. Cas9 activities are governed by its direct interactions with DNA. However, information about this interplay and the mechanism involved in its direction of Cas9 activity remain obscure. Using a single-molecule approach, we probed Cas9/sgRNA/DNA interactions along the DNA sequence and found two stable interactions flanking the protospacer adjacent motif (PAM). Unexpectedly, one of them is located approximately 14 base pairs downstream of the PAM (post-PAM interaction), which is beyond the apparent footprint of Cas9 on DNA. Loss or occupation of this interaction site on DNA impairs Cas9 binding and cleavage. Consistently, a downstream helicase could readily displace DNA-bound Cas9 by disrupting this relatively weak post-PAM interaction. Our work identifies a critical interaction of Cas9 with DNA that dictates its binding and dissociation, which may suggest distinct strategies to modulate Cas9 activity.〈/p〉

    Notes: Residence time distribution (RTD) of fluid and particles was measured in the holding tube of an aseptic processing system. A unique injector device instantaneously injected a large number of particles at one time without interruption of flow. A fraction collector device sampled exiting fluid at 1-sec intervals. Exiting particles at specific lapsed times from injection, were counted from those collected. RTD of fluid was determined by dye injection. RTD of fluid and particles under turbulent flow fitted a normal distribution function. Mean residence time of fluid was 0.975 times that calculated from volumetric flow rate with standard deviation ± 4.16%. Mean residence time of particles was 1.032 times that of the fluid, standard deviation was ± 4.62%.

    Description: In a low-pressure radio-frequency (13.56 MHz), inductively coupled argon plasma generated by a normal cylindrical rf coil, electric field, current density, and absorbed power density is calculated from magnetic field measured with a phase-resolved magnetic probe. The anomalous skin effect (ASE) for the cylindrical rf coil is compared to those previously reported for the planar and re-entrant cylindrical rf coils. Physical reasons for our observed characteristics of ASE are presented. With the increasing discharge power, the size and the number of negative and positive power absorption regions evolve into several distinct patterns. For the low discharge power (at 156.9 W), there is one area of positive and one area of negative power absorption in the radial direction. For the medium discharge power (279 W–683.5 W), there are two areas of negative and two areas of positive power absorption. For the even higher discharge power (above 803.5 W), the number of areas is the same as that of the medium discharge power, but the size of the inner positive and negative power absorption areas is approximately doubled and halved, respectively, while the outer positive and negative power absorption areas slightly shrinks. The evolution of positive and negative power absorption regions is explained as a result of electron thermal diffusion and the energy conversion between rf current and electric field. The spatial decays of electric field and current density are also elucidated by linking them with the positive and negative power absorption pattern.

    Description: A model is presented for describing the cylindrical implosion of a shock wave driven by an accelerated piston. It is based in the identification of the acceleration of the shocked mass with the acceleration of the piston. The model yields the separate paths of the piston and the shock. In addition, by considering that the shocked region evolves isentropically, the approximate profiles of all the magnitudes in the shocked region are obtained. The application to the dynamic Z-pinch is presented and the results are compared with the well known snowplow and slug models which are also derived as limiting cases of the present model. The snowplow model is seen to yield a trajectory in between those of the shock and the piston. Instead, the neglect of the inertial effects in the slug model is seen to produce a too fast implosion, and the pressure uniformity is shown to lead to an unphysical instantaneous piston stopping when the shock arrives to the axis.

    Description: Hydrogen production from ethanol solution (ethanol/water) by pulsed spark discharge was optimized by varying the volume of ethanol solution (liquid volume). Hydrogen yield was initially increased and then decreased with the increase in solution volume, which achieved 1.5 l/min with a solution volume of 500 ml. The characteristics of pulsed spark discharge were studied in this work; the results showed that the intensity of peak current, the rate of current rise, and energy efficiency of hydrogen production can be changed by varying the volume of ethanol solution. Meanwhile, the mechanism analysis of hydrogen production was accomplished by monitoring the process of hydrogen production and the state of free radicals. The analysis showed that decreasing the retention time of gas production and properly increasing the volume of ethanol solution can enhance the hydrogen yield. Through this research, a high-yield and large-scale method of hydrogen production can be achieved, which is more suitable for industrial application.

    Description: A new conodont biostratigraphic study is presented from a key section on the flanks of the Permian-Triassic Great Bank of Guizhou, an isolated carbonate platform from South China, that has recently provided much key data for understanding the nature of this mass extinction interval. Detailed investigation at Bianyang (Guizhou Province) has revealed ten conodont zones, in ascending order: Clarkina yini Zone; Hindeodus changxingensis Zone; Hindeodus parvus Zone; Sweetospathodus kummeli Zone; Neospathodus dieneri Zone; Neospathodus cristagalli Zone; Discretella discreta Zone; Pachycladina-Parachirognathus assemblage zone; Icriospathodus collinsoni Zone; Triassospathodus homeri Zone. This allowed the Permian-Triassic boundary (PTB) to be defined in the section based on the first occurrence of Hindeodus parvus . Furthermore, it is proposed that the first occurrence of Discretella discreta can be used as an auxiliary reference for defining the Induan-Olenekian boundary when Novispathodus waageni is absent both at Bianyang and elsewhere.
