    Notes: Abstract. When DNA topoisomerase II (topo II) activity is inhibited with a non-DNA-damaging topo II inhibitor (ICRF-193), mammalian cells become checkpoint arrested in G2-phase. In this study, we analyzed chromosome structure in cells that bypassed this checkpoint. We observed a novel type of chromosome aberration, which we call Ω-figures. These are entangled chromosome regions that indicate the persistence of catenations between nonhomologous sequences. The number of Ω- figures per cell increased sharply as cells evaded the transient block imposed by the topo II-dependent checkpoint, and the presence of caffeine (a checkpoint-evading agent) potentiated this increase. Thus, the removal of nonreplicative catenations, a process that promotes chromosome individualization in G2, may be monitored by the topo II-dependent checkpoint in mammals.

    Notes: Summary The regulation of cellular LDL metabolism by hypertriglyceridemic LDL was tested before and after treatment with acipimox, a nicotinic acid derivative, in 11 type IV hyperlipidemic patients. Large, less dense LDL particles were found in plasma after acipimox treatment. HDL subfractions were only slightly modified, with an increase of dense, cholesteryl ester-enriched and triglyceride-poor HDL3 particles. The LDL (B, E) receptor activity in human skin fibroblasts of LDL isolated before and after treatment was also evaluated. Hypertriglyceridemic LDL proved rather inefficient in regulating receptor activity, with a close to 30% lower capacity than normal LDL to inhibit receptor-mediated uptake and degradation of125I-LDL. This abnormality was fully corrected after acipimox. The reported findings indicate that acipimox treatment in type IV patients can normalize the defective interaction of hypertriglyceridemic LDL with the LDL (B, E) receptor.

    Notes: Parametrized field theories, which are generally covariant versions of ordinary field theories, are studied from the point of view of the covariant phase space: the space of solutions of the field equations equipped with a canonical (pre)symplectic structure.     Notes: The degree to which the phase space of a closed bosonic string carries a representation of the two-dimensional diffeomorphism group Diff(M) is investigated. In particular, homomorphic mappings from the associated Lie algebra diff(M) into the Poisson algebra of functions on three natural phase spaces associated with the string are constructed. Two of these spaces are extended phase spaces based on the conformal and harmonic gauges, respectively. The third space is essentially the original phase space; the homomorphism in this case relies on the validity of the light-cone gauge. Homomorphisms from diff(M) into the extended phase spaces of the Batalin–Fradkin–Vilkovisky formalism are also constructed. While the methods used to represent diff(M) cannot be extended to represent all of Diff(M), the phase spaces do carry a representation of the subgroup of conformal isometries. It is argued that this subgroup is sufficiently large to serve as the dynamical group for the string. The implications of this work for a true Dirac quantization of the string via operator representations of diff(M) are discussed.

    Notes: The Einstein equations with a cosmological constant, when restricted to Euclidean space-times with anti-self-dual Weyl tensor, can be replaced by a quadratic condition on the curvature of an SU(2) (spin) connection. As has been shown elsewhere, when the cosmological constant is positive and the space-time is compact, the moduli space of gauge-inequivalent solutions to this equation is discrete, i.e., zero dimensional; when the cosmological constant is negative, the dimension of the moduli space is essentially controlled by the Atiyah–Singer index theorem provided the field equations are linearization stable. It is shown that linearization instability occurs whenever the unperturbed geometry possesses a Killing vector and/or a "harmonic Weyl spinor.'' It is then proven that while there are no Killing vectors on compact conformally anti-self-dual Einstein spaces with a negative cosmological constant, it is possible to have harmonic Weyl spinors. Therefore, the conformally anti-self-dual Einstein equations on a compact Euclidean manifold are linearization stable when the cosmological constant is negative provided the unperturbed geometry admits no harmonic Weyl spinors.

    Notes: A natural generalized symmetry of the Yang–Mills equations is defined as an infinitesimal transformation of the Yang–Mills field, built in a local, gauge invariant, and Poincaré invariant fashion from the Yang–Mills field strength and its derivatives to any order, which maps solutions of the field equations to other solutions. On the jet bundle of Yang–Mills connections a spinorial coordinate system is introduced that is adapted to the solution subspace defined by the Yang–Mills equations. In terms of this coordinate system the complete classification of natural symmetries is carried out in a straightforward manner. It is found that all natural symmetries of the Yang–Mills equations stem from the gauge transformations admitted by the equations. © 1995 American Institute of Physics.
