    Notes: Abstract. Reeve & Crozier (1980) have recently proposed a method of evaluating the accuracy of a plant growth substance analysis by using information theory to estimate whether the analysis would distinguish the growth substance from all other components of the sample. In the present article, it is argued that their method requires knowledge of the relevant analytical characteristics of all potential components of the sample, and hence is not applicable to plant extracts. Instead, the identity of a growth substance in a plant extract can only be conclusively established using techniques which yield information on its chemical structure.

    Notes: Plants adapt to environmental stresses through specific genetic responses. The molecular mechanisms associated with signal transduction, leading to changes in gene expression early in the stress response, are largely unknown. It is clear, however, that gene expression associated with acclimatory responses is sensitive to the redox state of the cell. Of the many components which contribute to the redox balance of the cell, two factors have been shown to be crucial in mediating stress responses. Thiol/disulphide exchange reactions, particularly involving the glutathione pool and the generation of the oxidant H2O2, are central components of signal transduction in both environmental and biotic stresses. These molecules are multifunctional triggers, modulating metabolism and gene expression. Both are able to cross biological membranes and diffuse or be transported long distances from their sites of origin. Glutathione and H2O2 may act alone or in unison, in intracellular and systemic signalling systems, to achieve acclimation and tolerance to biotic and abiotic stresses.

    Notes: Stem elongation growth in a slender line of Pisum sativum L. was much greater than in its two dwarf parental lines due to a greater rate of production of longer internodes. The cellular basis of the greater length of slender internodes was tissue specific. In the epidermis, greater cell length primarily accounted for the longer internodes of the slender plants, but in the outer cortex, greater cell number was the more important factor. The soluble and salt-extractable peroxidase (EC 1.11.1.7) activities of expanding internodes were much lower in slender plants than in dwarf plants. The proportional effects of ethylene treatment on epicotyl length, diameter and orientation were similar in etiolated slender and dwarf seedlings.

    Notes: Abstract The endogenous levels of abscisic acid (ABA) and jasmonic acid (JA) were analyzed in wheat seedlings grown in water, a system which in the past has been used to test the effects of these plant growth inhibitors. The levels in different plant parts and in the medium were measured by gas chromatography-mass spectrometry-selected ion monitoring, using [2H3]ABA and [2H6]JA as internal standards. In every plant part, JA levels were about 2 orders of magnitude greater than those of ABA. The exudation of JA from roots per seedling was about 14,000-fold greater than that of ABA, although the roots contained only about 170 times more JA than ABA. It is suggested that JA is a possible allelopathic compound.

    Notes: Abstract the cytokinins of tobacco crown-gall tissue have been analysed by quantitative mass spectrometry using 2H2-labelled cytokinin riboside 5′-monophosphates and 15N4-labelled cytokinin glycosides as internal standards. The principal endogenous cytokinin of this tissue is zeatin riboside 5′-monophosphate. The biologically inactive 7-glucoside of zeatin is the most abundant basic cytokinin in the tissue. These findings expose the limitations of previously reported analyses of similar tissues, which were restricted to biologically active basic cytokinins. The present study demonstrates that the endogenous cytokinins of tobacco crowngall tissue show a clear correspondence to the range of metabolites formed when exogenous cytokinins are supplied to nontumorous tobacco cells.

    Notes: Abstract The amyloplasts found in the apical hook cells of etiolated pea (Pisum sativum L.) epicotyls were randomly distributed. Sedimentation of endodermal amyloplasts in the direction of gravity became apparent in the transition from the hook to the top of the main axis of the epicotyl. Cortical amyloplasts in this region were not, however, sedimented. These patterns of sedimentation could not be related to changes in amyloplast size, and it is proposed that cytoplasmic properties determine amyloplast behaviour. The differentiation of plastids in the hook differed between the amyloplast-containing endodermal cells and the cortical cells, in which amoeboid plastids predominated over amyloplasts. Amyloplasts disappeared from the cortical cells in the main axis of the epicotyl, but in the endodermal cells sedimented amyloplasts were found throughout the upper epicotyl. Etiolated epicotyls induced to grow horizontally by treatment with ethylene had a normal content of amyloplasts, sedimented in the direction of gravity.

    Notes: Abstract The character of the current controversy over geneticallymodified (GM) agriculture, typified by protesters' use of emotivesymbolism, has been largely inspired by the Green movement'snon-governmental organizations and political parties. This articleexplores the deeper philosophical and spiritual motivations of the Greenmovement, to inquire why it is implacably opposed to GM agriculture. TheGreen movement's anti-capitalism, exemplified by the hate-symbol statusof Monsanto as the company pioneering GM crops, is viewed within thewider context of alienation in the modern era. A complex of meanings isseen in Frankenstein as the focal symbol of GM protests, includingperceptions of risk, fears of the remixing of living identities seen ingenetic engineering, and resentment at the spiritual nihilism of thereduction of life to the digital code of DNA. By contrast, RobertGoodin's Green Theory of Value, which postulates the deep psychologicalimportance of nature in locating the self in a meaningful context largerthan ourselves, can explain the power of the Green symbol of thethreatened environment, Gaia. The advent of GM agriculture seems toimply that capitalism and technology can now enframe nature itself,leaving a world devoid of natural myth or meaning, with no escape fromthe alienation and nihilism of modernity. The central question posed forprotagonists of the GM debate is whether their agenda is based on thesepowerful but mythical conceptions of the environment, or whetherpreservation of the real environment is their primary ethic.

    Notes: A procedure has been developed for the synthesis of 6-chloropurine labelled with four 15N atoms. This compound can be used in the synthesis of a range of labelled cytokinin bases, which can be ribosylated by an enzymatic procedure. 15N labelled zeatin riboside was synthesized and used as an internal standard in the measurement by selected ion monitoring of the levels of this cytokinin in cultured crown gall and normal tissues of Vinca rosea, which were found to contain 402 and 2.5 ng per g fresh weight respectively.
