Bacillus subtilis NRRL B-2612, isolated at this laboratory, was used to produce polyglutamic acid from devitalized wheat gluten. Conditions for production of the polypeptide were investigated. When NRRL B-2612 was grown on wheat gluten in shaken flasks with phsophate buffer and salts at pH 6.5, yields of polymer reflected a 25-40% incorporation of glutamic acid into polyglutamic acid. Repeated alcoholic precipitation yielded peptides containing between 45 and 90% D-glutamic acid. Physical and chemical properties of a typical purified polyglutamic acid indicated that the product was a gamma-linked polypeptide composed of 75% D-glutamic acid and 25% L-glutamic acid with a molecular weight of approximately 200,000.
