    Description: The Convention on Biological Diversity (CBD)'s strategic plan advocates the use of environmental surrogates, such as ecosystems, as a basis for planning where new protected areas should be placed. However, the efficiency and effectiveness of this ecosystem-based planning approach to adequately capture threatened species in protected area networks is unknown. We tested the application of this approach in Australia according to the nation's CBD-inspired goals for expansion of the national protected area system. We set targets for ecosystems (10% of the extent of each ecosystem) and threatened species (variable extents based on persistence requirements for each species) and then measured the total land area required and opportunity cost of meeting those targets independently, sequentially and simultaneously. We discover that an ecosystem-based approach will not ensure the adequate representation of threatened species in protected areas. Planning simultaneously for species and ecosystem targets delivered the most efficient outcomes for both sets of targets, while planning first for ecosystems and then filling the gaps to meet species targets was the most inefficient conservation strategy. Our analysis highlights the pitfalls of pursuing goals for species and ecosystems non-cooperatively and has significant implications for nations aiming to meet their CBD mandated protected area obligations.

    Description: The sustainability of Short Sea Shipping (SSS) is central to a clean, safe, and efficient European Union (EU) transport system. We report on key challenges for advancing reliability, quality, and safety, and removing unnecessary costs and delays at SSS hubs, with a particular focus on Cyprus and the Eastern Mediterranean. Specifically, we consider the effect of port-2-port (P2P) communication on port efficiency by investigating the factors influencing the various waiting times at the Port of Limassol, both from a qualitative and a quantitative perspective. The qualitative results are based on the views of key stakeholders involved in the port call process. The quantitative analysis relies on data from over 8000 port calls during 2017–2018, which are analyzed with respect to ship type, port of origin, and shipping agent. The calculated Key Performance Indicators (KPIs) include arrival punctuality, berth waiting, and berth utilization. The analysis clearly reveals considerable variation in agent performance regarding the KPIs, suggesting a lack of attention to the social aspect of a port’s socio-technical system. We propose measures for improving agent performance based on the principles of Port Collaborative Decision Making (PortCDM), including P2P communication, data sharing and transparency among all involved in a port call process including the agents, and open dissemination of agent-specific KPIs.

    Description: Geologic carbon capture and storage (CCS) is an option for reducing CO 2 emissions, but leakage to the surface is a risk factor. Natural CO 2 reservoirs that erupt from abandoned oil and gas holes leak to the surface as spectacular cold geysers in the Colorado Plateau, United States. A better understanding of the mechanisms of CO 2 -driven cold-water geysers will provide valuable insight about the potential modes of leakage from engineered CCS sites. A notable example of a CO 2 -driven cold-water geyser is Crystal Geyser in central Utah. We investigated the fluid mechanics of this regularly erupting geyser by instrumenting its conduit with sensors and measuring pressure and temperature every 20 sec over a period of 17 days. Analyses of these measurements suggest that the timescale of a single-eruption cycle is composed of four successive eruption types with two recharge periods ranging from 30 to 40 h. Current eruption patterns exhibit a bimodal distribution, but these patterns evolved during past 80 years. The field observation suggests that the geyser's eruptions are regular and predictable and reflect pressure and temperature changes resulting from Joule–Thomson cooling and endothermic CO 2 exsolution. The eruption interval between multiple small-scale eruptions is a direct indicator of the subsequent large-scale eruption.

    Description: Bears produce the most altricial neonates of any placental mammal. We hypothesized that the transition from colostrum to mature milk in bears reflects a temporal and biochemical adaptation for altricial development and immune protection. Comparison of bear milks with milks of other eutherians yielded distinctive protein profiles. Proteomic and metabolomic analysis of serial milk samples collected from six giant pandas showed a prolonged transition from colostrum to main-phase lactation over approximately 30 days. Particularly striking are the persistence or sequential appearance of adaptive and innate immune factors. The endurance of immunoglobulin G suggests an unusual duration of trans-intestinal absorption of maternal antibodies, and is potentially relevant to the underdeveloped lymphoid system of giant panda neonates. Levels of certain milk oligosaccharides known to exert anti-microbial activities and/or that are conducive to the development of neonatal gut microbiomes underwent an almost complete changeover around days 20–30 postpartum, coincident with the maturation of the protein profile. A potential metabolic marker of starvation was detected, the prominence of which may reflect the natural postpartum period of anorexia in giant panda mothers. Early lactation in giant pandas, and possibly in other ursids, appears to be adapted for the unique requirements of unusually altricial eutherian neonates.

    Description: We accurately determine the fundamental system parameters of the neutron star X-ray transient Cen X–4 solely using phase-resolved high-resolution UV–Visual Echelle Spectrograph spectroscopy. We first determine the radial-velocity curve of the secondary star and then model the shape of the phase-resolved absorption line profiles using an X-ray binary model. The model computes the exact rotationally broadened, phase-resolved spectrum and does not depend on assumptions about the rotation profile, limb-darkening coefficients and the effects of contamination from an accretion disc. We determine the secondary star-to-neutron star binary mass ratio to be 0.1755 ± 0.0025, which is an order of magnitude more accurate than previous estimates. We also constrain the inclination angle to be 32 ${^{\circ }} ^{+8^{\circ }}_{-2^{\circ }}$ . Combining these values with the results of the radial-velocity study gives a neutron star mass of 1.94 $^{+0.37}_{-0.85}$ M consistent with previous estimates. Finally, we perform the first Roche tomography reconstruction of the secondary star in an X-ray binary. The tomogram reveals surface inhomogeneities that are due to the presence of cool starspots. A large cool polar spot, similar to that seen in Doppler images of rapidly rotating isolated stars, is present on the Northern hemisphere of the K7 secondary star and we estimate that ~4 per cent of the total surface area of the donor star is covered with spots. This evidence for starspots supports the idea that magnetic braking plays an important role in the evolution of low-mass X-ray binaries.

    Description: Multiple sclerosis (MS) is a complex disease with underlying genetic and environmental factors. Although the contribution of alleles within the major histocompatibility complex (MHC) are known to exert strong effects on MS risk, much remains to be learned about the contributions of loci with more modest effects identified by genome-wide association studies (GWASs), as well as loci that remain undiscovered. We use a recently developed method to estimate the proportion of variance in disease liability explained by 475,806 single nucleotide polymorphisms (SNPs) genotyped in 1,854 MS cases and 5,164 controls. We reveal that ~30% of MS genetic liability is explained by SNPs in this dataset, the majority of which is accounted for by common variants. These results suggest that the unaccounted for proportion could be explained by variants that are in imperfect linkage disequilibrium with common GWAS SNPs, highlighting the potential importance of rare variants in the susceptibility to MS.     Description: Protein-chromophore interactions are a central component of a wide variety of critical biological processes such as color vision and photosynthesis. To understand the fundamental elements that contribute to spectral tuning of a chromophore inside the protein cavity, we redesigned human cellular retinol binding protein II (hCRBPII) to fully encapsulate all-trans-retinal and form a covalent bond as a protonated Schiff base. This system, using rational mutagenesis designed to alter the electrostatic environment within the binding pocket of the host protein, enabled regulation of the absorption maximum of the pigment in the range of 425 to 644 nanometers.
