Summary The authors describe a photometric micro determination of the fatty acids. The analytical principle is based on the formation of intensively coloured salts of the colourless carbinol bases of the fuchsone dye stuffs with the fatty acids. As most important supposition for the reliability of the method, a stable solution of the pure carbinol base sensitive to acids has been made. As such one a solution of rosaniline in benzol proved suitable. By using this stable reagent upon fatty acids, the best conditions of the formation of the coloured salts were found out. The red reaction colour was measured with thePulfrich photometer of the house Carl Zeiss. The results were calculated according to the usual principles of the photometry. The reach of the method is limited between 0,05 and 0,5 mgs. of the higher fatty acids, but it can be extended into both directions. The method is distinguished by simplicity and high sensibility. Very probably, by suitable alterations the procedure also can be made useful for the determination of other organic and anorganic acids. The method is especially qualified for the determination of most little amounts of fatty acids in biological material. Only the characteristics of its use for determining the fatty acids in blood could be discussed in this paper.

Résumé (W) On décrit une méthode pour la détermination photomicrométrique des acides gras. Son principe analytique repose sur la formation de sels d'acides gras fortement colorés, obtenus à partir des bases carbinolique incolores des colorants de la fuchsone. La condition principale à l'obtention d'une méthode exacte est la suivante: on fait une solution stable d'une base carbinolique pure et sensible aux acides sous forme d'une solution de rosaniline dans le benzol. Grâce à l'emploi d'un tel réactif d'acide, on réalise les meilleures conditions pour la formation du sel. La mise en évidence de la couleur rouge de la réaction du sel d'acide gras de la rosaniline a été faite par le photomètre dePulfrich de la Maison Carl Zeiss selon les règles habituelles de la photométrie. L'intervalle de détermination de la méthode est placé pour les acides gras supérieurs entre 0,05 et 0,5 mgr. Mais en principe on peut l'agrandir des deux côtés. La méthode se distingue par sa simplicité et sa grande sensibilité. On peut probablement supposer que ces procédés, grâce à des modifications appropriées peuvent être utilisés pour la détermination d'autres acides organiques ou inorganiques. La méthode se prête particulièrement à la détermination quantitative de très petites quantités d'acide gras dans le domaine de la recherche biologique. La mode d'emploi pour la détermination des acides gras dans le sang ne peut être établi que dans les grandes lignes.

Zusammenfassung Es wird eine photometrische Mikrobestimmung der Fettsäuren beschrieben, deren analytisches Prinzip auf der Bildung intensiv gefärbter fettsaurer Salze der farblosen Carbinolbasen der Fuchsonfarbstoffe beruht. Als wichtigste Voraussetzung für die Zuverlässigkeit der Methode wurde eine stabile Lösung der reinen, säureempfindlichen Carbinolbase in Gestalt einer Lösung von Rosanilin in Benzol geschaffen. Unter Verwendung dieses stabilen Fettsäurereagens wurden die optimalen Bedingungen der Farbsalzbildung ermittelt. Die Auswertung der roten Reaktionsfarbe des fettsauren Rosanilins erfolgte mit demPulfrich-Photometer der Firma Carl Zeiß nach den üblichen Grundsätzen der Photometrie. Das Bestimmungsbereich der Methode liegt zwischen 0,05 und 0,5 mg höheren Fettsäuren, läßt sich aber grundsätzlich nach beiden Seiten erweitern. Die Methode zeichnet sich durch Einfachheit und hohe Empfindlichkeit aus. Es besteht die berechtigte Annahme, daß sich das Verfahren durch geeignete Modifikationen auch zur Bestimmung anderer Säuren organischer und anorganischer Natur nutzbar machen läßt. Die Methode eignet sich besonders gut zur quantitativen Erfassung kleinster Fettsäuremengen in biologischem Untersuchungsgut; ihre Anwendung zur Bestimmung der Blutfettsäuren konnte nur in den Grundzügen dargelegt werden.
