    Description: Polycomb group proteins have an essential role in the epigenetic maintenance of repressive chromatin states. The gene-silencing activity of the Polycomb repressive complex 2 (PRC2) depends on its ability to trimethylate lysine 27 of histone H3 (H3K27) by the catalytic SET domain of the EZH2 subunit, and at least two other subunits of the complex: SUZ12 and EED. Here we show that the carboxy-terminal domain of EED specifically binds to histone tails carrying trimethyl-lysine residues associated with repressive chromatin marks, and that this leads to the allosteric activation of the methyltransferase activity of PRC2. Mutations in EED that prevent it from recognizing repressive trimethyl-lysine marks abolish the activation of PRC2 in vitro and, in Drosophila, reduce global methylation and disrupt development.     Description: Histone variants have been proposed to act as determinants for posttranslational modifications with widespread regulatory functions. We identify a histone-modifying enzyme that selectively methylates the replication-dependent histone H3 variant H3.1. The crystal structure of the SET domain of the histone H3 lysine-27 (H3K27) methyltransferase ARABIDOPSIS TRITHORAX-RELATED PROTEIN 5 (ATXR5) in complex with a H3.1 peptide shows that ATXR5 contains a bipartite catalytic domain that specifically "reads" alanine-31 of H3.1. Variation at position 31 between H3.1 and replication-independent H3.3 is conserved in plants and animals, and threonine-31 in H3.3 is responsible for inhibiting the activity of ATXR5 and its paralog, ATXR6.     Notes: A simple method for the measurement of the refractive indices of parallel plate samples is described that uses the shift of the interference pattern when rotating the sample in one arm of a Mach–Zehnder interferometer. The theoretical fringe pattern shift is calculated and the accuracy of the method is derived. A typical accuracy of about 10−4 can be achieved at low refractive indices. The method is especially useful when—because of other reasons—the samples are provided as plates or when prisms are difficult to fabricate.

    Notes: The thermoreflectance technique is applied for imaging electric current distributions and thermal transfer in a temperature reference resistor heated by an alternating current. High-frequency scans (30 kHz) allow imaging of the current density distribution in conducting strips of the resistor while scans of amplitude and phase of the surface temperature variation at lower frequencies reveal plane, cylindrical, and spherical thermal waves. We investigate wave dimensionality as a function of heating geometry and thermal length, and present a simple method allowing a quantitative thermal analysis by exploiting the phase profile of cylindrical thermal waves. © 1996 American Institute of Physics.

    Notes: Noncritical phase-matched second harmonic generation was measured in optically biaxial KNb1−xTaxO3 mixed crystals in the orthorhombic phase. For Nd:YAG-(1064 nm) the coefficient d31 of the tensor of the nonlinear susceptibility was applied, while for the GaAs-laser (905 nm) the coefficient d32 was used. For both laser wavelengths the phase-matching temperature decreases with increasing Ta-concentration. Noncritical phase-matching at room temperature can be reached with the GaAs-laser for a Ta concentration of ≈9%. The corresponding value for the Nd:YAG laser is ≈14%.

    Notes: A photothermal microscope that provides micrometer lateral and submicrometer depth resolution was designed. Thermal conductivity measurements with modulation frequencies up to 12 MHz on single grains in polycrystalline diamond demonstrate its lateral resolution power even for a highly conducting material. Measured conductivities strongly depend on the averaged volume and values up to 2200 W/mK are found in the high frequency limit where the properties inside a grain are sampled. The capability of the instrument to measure thermal parameters on thin films is demonstrated for gold films evaporated on quartz with a thickness ranging from 20 to 1500 nm. Measurements reveal a strong thickness dependence for both thin film conductivity and the contact resistance between film and substrate. Thermal conductivity decreases monotonically from 230 to 30 W/mK whereas the contact resistance rises from 2×10−7 to 8×10−6 m2K/W with decreasing film thickness. © 1997 American Institute of Physics.

    Notes: Abstract We investigated the anisotropic thermal conductivity in pyrolytic graphite by thermoreflectance. A laser-heated circular spot on a surface perpendicular to the planes developed into an elliptical temperature distribution which was recorded by a raster scanning technique at modulation frequencies ranging from 600 Hz to 100 kHz. The ratio of in-plane and perpendicular thermal conductivity was determined by fitting the phase of the temperature data with an analytical model, and was found to decrease with increasing modulation frequency. Highest conductivity values were considerably smaller than previously published data based on steady-state measurements. The frequency dependence and additional features in the phase profiles at high frequencies are discussed in view of sample surface preparation and the local nature of the thermoreflectance measurement.

    Description / Table of Contents: Corrosion Resistant Coating of Thin sheet Metal on Steel and ConcreteInformations on the development and large scale tests of a new corrosion-resistant coating method for coating of equipment, vats, tanks made of concrete, steel or masonry. The new method employs high alloyed thin sheet metal of 0.6 mm thickness, 1000 mm breadth and 3000 mm length the constructive conditions of this coating method are explained and the measures for the fixing of the sheets on the area to be coated are described. They refer to the preparation of the wall to be coated, the selection of a suitable welding process and welding additives, the preparation of the welding seam, the evasion of thermal shrinkings. Directions on the arrangement of the coating sheets and welding seams are given. These fundamental particulars are completed by specifications on the stability and tightness of the welding seams as well as their metallurgical structure, their stability against granular desintegration and their stress-cracking-safety. This method supplies a want within the insufficient or complicated coating methods made of plastics, ceramic masonry and the not always necessary total or rolled plate constructions of high alloyed steels. The new method guarantees industrial efficiency.
    Notes: Berichterstattung über Entwicklung und großtechnische Erprobung eines neuen, korrosionsfesten Verfahrens zum Auskleiden von Apparaten, Bottichen und Tanks aus Stahl, Beton oder Mauerwerk. Die neue Arbeitstechnik verwendet hochlegierte Dünnbleche von 0,6 mm dicke, 1000 mm Breite und 3000 mm Länge. Die konstruktiven Voraussetzungen diese Auskleidetechnik werden erläutert; darüber hinaus werden die Maß-nahmen zur Befestigung der Dünnbleche auf der auszukleidenden Fläche beschrieben. Sie erstrecken sich auf:Vorbereiten der zu schützenden Flächen, Wahl des Schweißverfahrens, des Schweißzusatzwerkstoffes, der Schweißnahtvorbereitung und Vermeidung von Wärmeschrumpfungen.Hinweise für die Aufteilung der Auskleidungsbleche mit Anordnung der Schweißnähte werden gegeben. Diese grundlegenden Angaben werden ergänzt durch Ergebnisse über das Festigkeitsverhalten und die Dichtigkeit der Schweißnähte sowie dem metallurgischen Aufbau der Schweißnähte, ihre Prüfung auf Kornzerfallbeständigkeit und Sicherheit gegenüber Spannungsrissen.Mit diesem neuen Arbeitsverfahren wird eine bestehende Lücke geschlossen, die zwischen unzureichenden oder komplizierten Auskleidungen mit Kunststoffen, keramischen Ausmauerungen oder den nicht immer erforderlichen Voll- oder walzplattierten Konstruktionen aus hochlegierten Stählen liegen. Das neue Verfahren gewährleistet eine wirtschaftliche Basis.
