    Description: The proximate composition and amino acids assay of smoked Clarias gariepinus and Oreochromis niloticus were determined using standard procedures. The mean proximate composition of Oreochromis niloticus are 8.9% moisture, 16.00% lipids, 5.6% ash, 44.5% crude protein, 0.42% crude fiber and 24.9% NFE while Clarias gariepinus has 8.19% moisture, 15.55% lipids, 4.725 ash, 40.46% crude protein, 0.50% crude fiber and 30.58% NFE. There is therefore a nutrient variation between the 2 species of fish Oreochromis niloticus has higher crude protein, lipids, moisture and ash content than Clarias gariepinus.The amino acids profile shows that both fishes have 17 amino acids. Apart from Tryptophan, the following essential amino acids Histidine, Methionine, Threonine, Isoleucine, Leucine, Lysine, Valine and Phenylalanine are present in both fish. Oreochromis niloticus however has higher quantities of the essential amino acids except for Histidine than Clarias gariepinus

    Description: The evaluation of Cr. Cu, Mn, Fe, Ni, Zn,As and Pb concentrations in Clarias gariepinus and Oreochromis niloticus from Lake Geriyo Yola,Adamawa, Nigeria using X - ray fluorescence (XRF) technique is discussed. The analytical samples are irradiated with high energy electrons of 109Cd to produce fluorescent X-rays was produced which passes to the Silicon-lithium detector through Mo target as a source of monochromatic X-rays. The spectrum of energy generated on the detector was processed by a Multi- Channel Analyzer to obtain analytical data. The intensity of the fluorescent X-rays on the detector is proportional to the concentration of the individual element of interest in the sample. This method can identify up to 30 or more elements at the same time. The results showed varying levels of heavy metals in the fishes. The concentrations of Cr, Mn, Cu, and Ni in the fishes were much higher than WHO and PEPA maximum permissible limits, while the concentrations of Zn and Pb were lower than the standards. The results suggest that the lake is polluted with Cr, Mn, Cu and Ni and the consumption of fishes of the lake is life threatening to man.

    Description: Tilapia guineensis eggs were spawned and hatched in different salinities (17, 12, 7, 5, 2 and 0) ppt. The study was conducted in eight replicates. The aim of the investigation was to ascertain disparity in hatching size of Tilapia guineensis attributable to salinity. Immediately hatching was observed, body weight of five hatchlings in each replicate was individually weighed in milligramme. The individual total length of the five hatchlings was also measured in millimeter. The height of the yolk sac larva was determined with micro-metre screw gauge graduated in millimeter from the top of the yolk sac to the dorsal region of the larva. Data obtained were used to calculate yolk sac volume. Significant differences (P〈0.01) were observed in body weight and total length of larvae hatched in different salinities. The heaviest and longest larvae were hatched in 12 ppt saline water. The yolk sac volume of larvae hatched in lower salinities were significantly higher (P〈0.01) than the yolk sac volume of larvae hatched in higher salinities. The smallest yolk sac volume was recorded in 12ppt saline water. Our findings show that salinity could be manipulated for economic production of heavier seeds of Tilapia.

    Description: The water analysis of five selected ponds in Ado-Ekiti in Ekiti State, was carried out to determine their suitability for fish culture. Pa rameters analyzed were: Temperature, Turbidity, Carbon (IV) oxide, Alkalinity, Hardness, pH and Dissolved oxygen. Values for temperature ranged between 20.0~'C and 29.0~'C, Turbidity values were between 46.0-50.2cm, Carbon (IV) oxide: 11.5-12.2mg/litre; alkalinity: 100-120mg/liter; hardness: 110-200mg/litre; pH: 6.8-7.8 and dissolved oxygen: 5.8-6.8mg/liter. The values obtained were found to be within acceptable range for fish production.

    Description: This study focused on the assessment of Terminalia suparba (bark, leave and root) based diet to improve the ovulation of Clarias gariepinus brood stocks. Ten female and ten male brood stocks were obtained from a reputable fishfarm in Ogun State, Nigeria and were fed with the diets for four weeks before the breeding exercise. The latency period observed in treatments 1 to 4 was 4hours 30minutes while the ovulation period in treatment 5 was 8 hours. There was no significant different (P〉0.05) among the means. The brood stocks in Treatment 2 (Root) has the highest fecundity count (2001) while those in Treatment 5 (control) has the least fecundity count (1496). The phytochemical analysis of the plant materials showed the presence of Alkaloids, Tannin, Phenol, Glycoside, Flavonoids, Steroids, Phlobatannin, Saponin, Anthraquinones. The physical and chemical parameters of water observed showed that dissolved oxygen value ranges from 3.28 -4.60, pH 6.5 - 8.5, temperature 28.0 - 28.8 and conductivity 8.9 - 9.20. It is therefore concluded that the inclusion of Terminalia suparba in the feed of Clarias gariepinus broodstock will not only improve ovulation but also reduce the latency period.

    Description: One of the priority research projects of the Nigerian Institute for Oceanography and Marine Research (NIOMR) is broodstock development and mass propagation of silver catfish, Chrysichthys nigrodigitatus of which the broodstock is naturally abundant in New Gala River. Selected physical characteristics and water physico-chemical parameters of the upper reach of the New Calabar River at Aluu were investigated. The highest depth value of 9.20 m was recorded in the middle of the river. The width of the river measured during spring high tide and low tide were 174 m and 110 m respectively. The cross-sectional area and average flow velocity of the river were 797.13 m2 and 0.374 m/s respectively. The discharge of the river was computed as 298.13m3/s. The river water was acidic most of the period (Hydrogen ion concentration, pH: 5.12 to 7.43), with low total alkalinity values (4.00 to 12.00 mgCaCO3/L) and low total hardness values (11.5 to 51.2 mgCaCO3/L) which indicate that the river water is black and soft. There was significant (p~,0.05) seasonal variation in temperature (23.1 to 31.7~'C\}, Secchi transparency (9.30 to 19.00 cm), total hardness (11.5 to 51.2 mgCaCO3/L), dissolved oxygen (5.20 to 7.50 mg/L) and dissolved oxygen deficit (0.02 to 2.95mg/L). These results are useful for several stake holders who use the river as a sink for trade wastes, as source of water for fish farming, as source of broodstocks of Chrysichthys nigrodigitatus, as raw water for water treatment plants, and as means of transportation of industrial materials.

    Description: Eight month physicochemical study of Dadin Kowa Dam was carried out forthnightly from July, 2011 to february 2012. Air temperature ranged from 18.25~'C ~c 28~'C ~c 0.50 to 28~'C ~c 2.16. Water tmperature ranged from 18.75~'C ~c 2.21. Transparency ranged from 11.00cm ~c 1.83 to 29.63 cm ~c ~c 2.14. Conductivity ranged from 13.00~Ks/cm ~c 13.33 to 41.38~Ks/cm ~c 5.54. Hydrogen ion (pH) ranged from 6.69 ~c 1.03 to 8.39 ~c 1.17. Dissolved oxygen ranged from 6.39mg/l ~c 1.03 to 10.14mg/l ~c 1.17. Total alkalinity ranged from o.69mg/l ~c 0.38 1.34mb/l ~c 0.02. Total ammonia ranged from 0.20mg/l ~c 0.027 to 0.83mg/l ~c 0.05. There were various levels of correlation between these parameters. There was no significant difference between both sites and months (p〉0.05) of all the parameters except total nitrogen which show a significant difference between the sites (p〈0.05) and transparency also shows a significance between the months (p〈0.05). All physicochemical parameters were almost within the tolerable limits for supporting aquatic life.

    Description: This study looked into the physicochemical and bacteriological investigation of fish and water in Aiba Reservoir. Water and fish samples collected were studied to assess their bacteriological and physicochemical characteristics and their suitability for domestic purposes. Structural questionnaires were administered to the fishermen around the reservoir and experiments were carried out in the laboratory to analyze the samples. The results showed that the total viable bacteria count was high(1.02 x 105 cfu/ml) while few Staphylococcus aureus and E. coli occurred insignificantly. The population of S. aureus and E. coli in the fish gills, liver and tissue were insignificant. The results obtained from the physicochemical analysis of the water samples showed that the reservoir has optimum water temperature of 29degreesC ~c 0.25, a pH of 7.70 ~c 0.20, dissolved Oxygen level of 7.20mgO2/1 ~c 0.20 and total hardness of 45.00 mg/ICaC03 ~c 1.05. The microbial load in the fish sample is higher than that in the water sample. Further strict hygienic measures and surveillance is recommended so as to improve the water quality and to avoid deleterious effects on the health of the Iwo inhabitants and its neighboring communities who also benefit from the reservoir.

    Description: Comparative primary production in Little Stream Farms, near Uyo was investigated in relation to physico-chemical parameters of the adjacent perennial stream and the fish ponds. Except for temperature, which showed the same values for both stations, physico-chemical variables generally showed significant difference (P〈O.05) between the pond water and the adjacent stream. Values of silicates, Turbidity (NTU), Secchi Disc Transparency and Total Suspended Solids (TSS) exhibited significantly variable trends (P〈O.05), especially in the stream water, in response to the commercial sand dredging activities upstream from the fann location. Lowest value of stream transparency (O.1M) recorded was during the peak of the sand dredging, whereas the stream water showed absolute transparency in the absence of the dredging. The water quality parameters also exhibited a general trend of significant seasonal variation (P〈O.05). Five phytoplankton families: Bacillariophyceae, Chlorophyceae, Cyanobacteria, Euglenophyceae and Dinophyceae occurred in the ponds, while three families, Bacillariophyceae, Chlorophyceae and Cyanobacteria occurred in the stream. Bacillariophyceae showed the highest relative abundance in both stations A and B.

    Description: The physico-chemical parameters of Lake Alau were monitored for four months to assess the water quality. Parameters such as temperature, transparency, conductivity, dissolved oxygen, biochemical oxygen demand, Ca, P, etc were studied in five stations (A, B, C, D and E). Water samples were collected monthly from 8:00 to l2:00noon, for four months (July - October 2008, wet season) and analysed. The physico-chemical parameters were within the ranges of unpolluted water bodies. Variation occurs in almost all the months of the study. Significant difference (P〈0.05) occurred in pH, transparency and Ca among the stations, and insignificant difference (P〉0.05) in temperature, conductivity, dissolved oxygen, biochemical oxygen demand, Mg and P. These variations may be due to effects of fertilizer application, herbicides and insecticides in irrigated farms around the Lake.

    Description: Study of some Physico-chemical parameters and fish species composition of Doma Darn was carried out between the month of August 2012. Fishing communities along the dam were selected at sampling stations and collection of fish species. The values of physico-parameters obtained for stations were in the range of 28.10-92 m for depth, 27.70-28.48~'for temperature, 7.03-7.64 for PH, 7.23-7.64mg/l for dissolved oxygen (DO), 30.08-31.75 ntu for turbidity, 94.64-105.17mg/l for alkalinity and 79.17-80.58mg/l for total hardness. The result indicated that there was significant difference (p〈0.5)for depth, temperature turbidity and alkalinity, while there was no significant difference (p〈0.05) for DO, pH and water hardness among the stations. The result of these parameters among the duration shows that there was significant different (p〈0.05) in all the parameters except pH. The study revealed that there were twenty-five (25) fish species belonging to 12 families in the dam. The family Cichlidae and Mochokidae and Bagridae which has three each, While the family, Polypteridae, Channidae, Centropomidae, flepsetidae and Protopteridae has the lowest number of one (1) each. The overall result revealed that the Phvsico-chemical parameters were mostly influenced by duration and not much by stations; and parametersoaram measured were within the recommended water quality requirement for growth and survival of aquatic organisms.

    Notes: Abstract  Water deficit during meiosis in microspore mother cells of wheat (Triticum aestivum L.) induces male sterility, which reduces grain yield. In plants stressed during meiosis and then re-watered, division of microspore mother cells seems to proceed normally, but subsequent pollen development is arrested. Stress-affected anthers generally lack starch. We employed light microscopy in conjunction with histochemistry to compare the developmental anatomy of water-stress-affected and normal anthers. The earliest effects of stress, detectable between meiosis and young microspore stages, were the degeneration of meiocytes, loss of orientation of the reproductive cells, and abnormal vacuolization of tapetal cells. Other effects observed during subsequent developmental stages were deposition of starch in the connective tissue where it is normally not present, hypertrophy of the middle layer or endothecial cells, and deposition of sporopollenin-like substances in the anther loculus. The resulting pollen grains lacked both starch and intine. These results suggest that abnormal degeneration of the tapetum in water-stressed anthers coupled with a loss of orientation of the reproductive cells could be part of early events leading to abortion of microspores.

    Notes: Abstract Direct gene transfer to floral meristems could contribute to cell-fate mapping, to the study of flower-specific genes and promoters, and to the production of transgenic gametes via the transformation of sporogenic tissues. Despite the wide potential of its applications, direct gene transfer to floral meristems has not been achieved so far because of the lack of suitable technology. We show in this paper that ballistic micro-targeting is the technique of choice for this purpose, and in this way, we were able to transfer genes efficiently into excised wheat immature spikes. Particle size was adjusted for optimal penetration into the L1 and L2 cell layers of the spikes with limited cell damage. Spikes at different developmental stages were shot either with a plasmid containing two genes involved in anthocyanin biosynthesis or with a plasmid bearing the uidA (β-glucuronidase) gene. The transient expression of these marker genes was observed in the different developmental stages tested and in cells of both the L1 and the L2 layers. The transient expression of the uidA gene was significantly increased when the sucrose concentration in the culture medium was increased from 0.06 to 0.52 M. At the highest concentration, 100% of the targeted spikes expressed the uidA gene, with an average of 69 blue cells per spike. Twelve days after microtargeting, multicellular sectors showing transgene expression and containing up to 17 cells were found in 85% of the shot immature inflorescences. This indicated that targeted cells survived particle bombardment. Sectors were found in primordia of both vegetative and reproductive organs.

    Notes: Abstract  Colonization of Hordeum vulgare L. cv. Salome (barley)and Triticum aestivum L. cv. Caprimus (wheat) roots by the arbuscular mycorrhizal fungus Glomus intraradices Schenck & Smith leads to de novo synthesis of isoprenoid cyclohexenone derivatives with blumenin [9-O-(2′-O-β-glucuronosyl)-β-glucopyranoside of 6-(3-hydroxybutyl)-1,1,5-trimethyl-4-cyclohexen-3-one] as the major constituent and to transient accumulation of hydroxycinnamate amides (4-coumaroylagmatine and -putrescine). Accumulation of these compounds in mycorrhizal wheat roots started 2 weeks after sowing together with the onset of arbuscule formation and proceeded with mycorrhizal progression. Highest levels were reached in 3- to 4-week-old secondary roots (root branches of first and higher order) characterized by the formation of vesicles. In the final developmental stages, the fungus produced massive amounts of spores, enclosing the stele of older root parts (older than 5 weeks) characterized by cortical death. In these root parts, the secondary compounds were detected in trace amounts only, indicating that they were located in the cortical tissues. Some rhizosphere bacteria tested, i.e. Agrobacterium rhizogenes, Pseudomonas fluorescens, and Rhizobium leguminosarum, markedly stimulated both fungal root colonization and blumenin accumulation, thus, acting as mycorrhiza-helper bacteria (MHB). Application of blumenin itself strongly inhibited fungal colonization and arbuscule formation at early stages of mycorrhiza development. This was associated with a markedly reduced accumulation of the hydroxycinnamate amides 4-coumaroylputrescine and -agmatine. The results suggest that both the isoprenoid and the phenylpropanoid metabolism are closely linked to the developmental stage and the extent of fungal colonization. Their possible involvement in the regulation of mycorrhiza development is discussed.

    Notes: Abstract  Oxygen and CO2 fluxes were measured in hydroponically grown mycorrhizal and non-mycorrhizal Triticum aestivum L. cv. Hano roots. The NO3 – uptake of the plants was used to estimate the amount of root respiration attributable to ion uptake. Plants were grown at 4 mM N and 10 μM P, where a total and viable mycorrhizal root colonisation of 48% and 18%, respectively, by Glomus mosseae (Nicol. and Gerd.) Gerd. and Trappe (BEG 107) was observed. The O2 consumption and NO3 – uptake rates were similar and the CO2 release was higher in mycorrhizal than in non-mycorrhizal wheat. This resulted in a significantly higher respiratory quotient (RQ, mol CO2 mol–1 O2) in mycorrhizal (1.27±0.13) than in non-mycorrhizal (0.79±0.05) wheat. As the biomass and N and P concentrations in mycorrhizal and non-mycorrhizal wheat were the same, the higher RQ resulted from the mycorrhizal colonisation and not differences in nutrition per se.

    Notes: Summary Embryogenic cell suspension cultures were established from Triticum aestivum X Leymus angustus F1 hybrids, using compact nodular calli derived from inflorescence segments. Calli originating from leaf segments did not give rise to stable cell suspensions. Growth measurements of the cell suspensions revealed that they continued rapid growth up to 10 days after subculturing. Flow cytometric studies of the cell cycle over a 7 day culture period showed that the majority of cells were in G1 phase while the rest were either in S or G2. During the 7 days of culture, no significant differences in DNA distribution patterns were observed. The cells from suspension cultures produced somatic embryos when they were transferred to different solid media. The embryos germinated and gave rise to plantlets which were successfully rooted and transferred to soil.

    Notes: Abstract Highly regenerable callus cultures have been obtained from immature embryos of hexaploid wheat cv. Oderzo. Friable fast growing calli were induced at high frequency. Suspensions were initiated from the most friable callus lines: they became established in about two months. Suspensions consisted of cell aggregates of 30 to 1000 um in diameter. Upon plating on MS hormone-free medium, suspensions regenerated green plantlets, and their regenerative capability was maintained for at least 10 months. Protoplasts were isolated from 7–8 day old suspension cultures with a yield of 4–6×106 protoplasts/g fresh weight cells. Protoplast culture was either in liquid medium or in a bead-type system with agarose beads. First divisions were detected at day 5. At day 14 visible colonies were detected and the plating efficiency was evaluated between 2 and 8% over the initial number of protoplasts plated. Protoplast-derived calli were cultured in the presence of 1 mg/l IAA and 0.5 mg/l zeatin and were used for reinitiating new suspension cultures. Upon plating onto MS hormone-free medium, with or without the addition of 0.1 mg/l GA3, calliclones were induced to differentiate. Regeneration of complete plantlets, with shoot and roots took about two months. Plantlets were grown in sterile conditions until 12–15 cm height, and were subsequently transplanted in soil.

    Notes: Abstract Spring wheat (Triticum aestivum L.) zygotic embryos were successfully cryopreserved, without the addition of exogenous cryoprotectants, using only an abscisic acid (ABA) pretreatment. Optimum survival was obtained when embryos were cultured in vitro for 10 days on semisolid Murashige and Skoog (MS) nutrient medium supplemented with 0.5 mg/L (±) ABA prior to cryopreservation. The embryos resumed growth within three days when returned to MS medium devoid of ABA but containing 2mg/L 2,4-dichlorophenoxyacetic acid. The embryogenic calli produced from these embryos exhibited normal plant regeneration on auxin-free media. Changes in dw/fw ratio, as well as the esterified fatty acid and sucrose concentrations correlated positively with the development of tolerance to cryopreservation.

When ovules were cultured on Phytagel-solidified medium, and the pericarp removed exclusively at the micropylar tip and the abaxial side, zygotes from up to 79.2% of the ovules underwent embryogenesis with the same developmental pattern as found in planta. Embryos from more than 50% of the cultured ovules germinated when transferred to regeneration medium. More than 100 plantlets were randomly chosen for transfer to soil, all of which developed to phenotypically normal and fertile plants. With this system, the entire process of zygotic embryogenesis can be studied using living material. Furthermore, the method could be used as an embryo rescue technique for plant breeding purposes.

    Notes: Abstract A cloned cDNA to the wheat (Triticum aestivum) early cysteine-labeled metallothionein has many characteristics of a molecular marker for pollen embryogenesis in this plant. This transcript was not detected in uninucleate microspores at the time of culture or in pollen at any stage during normal ontogeny; its mRNA did begin to increase in embryogenic microspores within 6 h of culture, peaked at around 24 h, declined, then leveled off through the 21-day-old embryoid stage. Additionally, the accumulation of the embryoid-abundant EcMt gene transcript showed a direct and positive correlation with an increase of ABA in embryogenic microspores and developing pollen embryoids. Irradiating cultures with high intensity white light or with far-red, or blue light, suppressed EcMt transcript accumulation and the ability of microspores to form embryoids; however, light did not affect ABA concentrations during the early stages of culture. These results suggest that although a promoter of pollen embryogenesis in bread wheat, ABA alone cannot maintain the sporophytic differentiation of microspores subjected to inhibitory regimes of light in vitro. Whether or not light acts directly or indirectly in suppressing EcMt gene expression and pollen embryogenesis remains unknown.

    Notes: Abstract The accumulation of amyloplast DNA during endosperm development was studied in two cultivars of spring wheat, Triticum aestivum L. ‘Chinese Spring’ (CS) and ‘Spica’, small and relatively larger-grained cultivars, respectively. Endosperms were isolated between 9 and 45 days post anthesis (dpa) and the amyloplast DNA content of endosperm nucleic-acid extracts was measured by quantitative hybridisation with a homologous chloroplast-DNA probe. The endosperm cells of CS and Spica accumulated amyloplast DNA during development in a similar way. In both cultivars there was a large increase in the amount of plastid DNA (ptDNA) per endosperm between 9 and about 15 dpa, after which there was no further increase. Because nuclear DNA continued to accumulate until 24 dpa, the percentage contribution of amyloplast DNA to total DNA fluctuated in both cultivars during development, reaching maxima at 12 dpa of about 1.00% and 0.85%, and dropping to apparently constant levels of 0.60% and 0.52% in CS and Spica, respectively, by 24 dpa. In both cultivars, the average number of ptDNA copies per amyloplast was calculated to increase from about 10 copies at 9 dpa to about 50 copies in the mature amyloplasts at 31 dpa. However, the heavier endosperms of Spica contain more cells than those of CS and the varieties therefore differed in the amount of ptDNA that accumulated per endosperm: Spica endosperms accumulated 110 ng of ptDNA by 15 dpa, compared with only 85 ng in CS. The apparent accumulation of ptDNA copies in wheat amyloplasts during endosperm development contrasts with the decline in chloroplast-DNA copies in wheat chloroplasts during leaf development.

    Notes: Summary Poor adaptability or functional quality of much germplasm used for breeding high-protein hard red winter wheats prompted mutagenesis as an alternative means of increasing grain protein content. Four hard red winter wheat genotypes — KS644 (‘Triumph// Concho/Triumph’), ‘Kaw’, ‘Parker’, and ‘Shawnee’ — were treated with 0.40 M ethyl methanesulfonate (EMS). Advanced lines (M8-M10) were selected that had a 3-year mean grain protein advantage of 0.7% to 2.0% over controls. Increased grain protein content was generally associated with decreased grain yield and kernel weight, but some high-protein mutant lines had yields or kernel weights similar to those of original genotypes. Changes in height and lodging induced by EMS were generally favorable, most mutants being shorter and lodging less than controls, but blooming date was generally delayed, a deleterious change. One line also changed from resistant to segregating for wheat soil-borne mosaic virus. Mutant lines might be utilized in cross-breeding programs, particularly if negative pleiotropic effects and linkages are absent.

A sudden halt in protein accumulation is observed as the RNA starts to decrease. The hypothesis is also advanced that maintenance of the RNA content might, in turn, be dependent on the presence, in the endosperm of developing wheat seed, of a certain level of soluble amino nitrogen which could then play the role of limiting factor for protein synthesis.

Analyses were carried out on different parts of dry kernels (embryo plus scutellum and endosperm) using polyacrylamide and starch gel electrophoresis, different electrophoretic buffer systems and various staining methods. The peroxidase isozymes showed a low substrate-specificity and a high tissue-specificity. The embryo plus scutellum and the endosperm always presented different peroxidase patterns. Endosperm peroxidases were associated with chromosome arms 7DS, 4BL and 7AS; whereas the embryo plus scutellum isozymes were related to chromosome arms 3AL, 3BL and 3DS. The different results obtained using various electrophoretic techniques are due to the buffer system used. All staining procedures employed revealed the same peroxidase isozymes.

    Notes: Summary Genetic variability of endosperm esterase has been studied in 42 cultivars of Triticum aestivum L. 2n=6x=42. Different techniques, including sequential electrophoresis and electrofocusing, have been used with various substrates and esterase inhibitors. The electrophoretic patterns in each cultivar are described. Chromosomal location using the nullitetrasomic and ditelosomic lines of Chinese Spring was carried out in order to relate and/or locate the esterase genes to specific chromosomes. Most of the esterase isozymes located were in the long arm of the chromosomes of the homoeology group 3; but we have found six located in the short arms, five of them in the chromosome 3AS and one in the 3DS. This location increases the number of esterase genes described, because no esterase genes had been described so far in short arms of chromosomes of the homoeology group 3. The genetic control is discussed and, according to our results, between 12 and 15 loci, organized in five “compound loci”, control the endosperm esterases in wheat. Also one “modifier” gene modifying the mobility of two esterase bands and present in all the cultivars studied is postulated.

Their spike characteristics were similar to those of common wheat but, unlike their parents, they were long-awned. The average meiotic chromosome pairing at MI of F1 hybrids was: 6.39 I +3.75 rodII+8.64 ringII+0.81 III+0.30 IV+0.04 V, the bivalent and multivalent formation of which was much higher than expected from the genomic formulae. It is especially worthwhile to note that the F1 hybrids were self-fertile, self set being 0.15%, and seeds were easily obtained from the backcross of f1 plants with hexaploid and tetraploid wheats; here the seed set was more than 20.0%. The polyploid taxa and the position of A. Michnoi in Agropyron are discussed.

In a specifically designed medium, some of the pollen grains developed to maturity. These were applied to excised stigmas on agar, where they produced pollen tubes. Application to flowers in vivo led to seed set. Pollen was matured in vitro from a variety that produced a different protein banding pattern on SDS-PAGE as compared to the variety that was pollinated. The protein banding in the produced seeds showed the hybrid pattern, demonstrating that the seeds were not produced by self-pollination in this in-breeding species but by pollination with the in-vitro-matured pollen.

    Notes: Summary A series of hexaploid wheat lines containing zero, two or four doses of rye chromosome arm 1RS was used to investigate the response to changes in dosage by the rye genes when in a wheat background. The quantity of protein produced by the secalin protein genes contained on 1RS was directly related to the number of copies of 1RS present in the line. No response could be detected by representative wheat proteins suggesting that the increase in secalin protein observed was due to an increase in mRNA produced when four copies of the secalin gene was present. These results suggest that increasing the dosage of alien genes introgressed into wheat may be a useful tool for enhancing their expression.     Notes: Summary Intergeneric hybrids between Triticum aestivum cv ‘Chinese Spring’ and Agropyron cristatum 4x (2n= 5x=35, ABDPP genomes) with a high level of homoeologous meiotic pairing between the wheat chromosomes were backcrossed 3 times to wheat. Pollination of the F1 hybrid with ‘Chinese Spring’ resulted in 22 BC1 seeds with an average seed set of 1.52%. Five BC1 plants with 39–41 chromosomes were raised using embryo rescue techniques. Chromosome pairing in the BC1 was characterized by a high frequency of multivalent associations, but in spite of this there was no evidence of homoeologous pairing between chromosomes of wheat and those of Agropyron. All of the plants were self sterile. The embryo rescue technique was again essential to produce 39 BC2 plants with chromosome numbers ranging from 37 to 67. The phenomenon of meiotic non-reduction was also observed in the BC3 progenies. In this generation male and female fertility greatly increased, and meiotic pairing was fairly regular. Some monosomic (2n=43) and double monosomic (2n=44) lines were produced. Analysis of these progenies should permit the extraction of the seven possible wheat-Agropyron disomic addition lines including those with the added chromosomes carrying the genes involved in meiotic non-reduction and in suppression of Ph activity.

Several sets of mRNA were isolated from seedling leaf tissues which had been heat-stressed at 37 °C for various time intervals. These mRNAs were hybridized with HSP cDNA or genomic DNA probes (HSP17, 26, 70, 98, and ubiquitin). Protein profiles were compared using in-vitro translation and 2-D gels. The Northern slot-blot data from the heat-stress treatment provide evidence that the heat-tolerant cv ‘Mustang’ synthesized low molecular weight (LMW) HSP mRNA earlier during exposure to heat shock and at a higher level than did the heat-susceptible cv ‘Sturdy’. This was especially true for the chloroplast-localized HSP. The protein profiles shown by 2-D gel analysis revealed that there were not only quantitative differences of individual HSPs between the two wheat lines, but also some unique HSPs which were only found in the ‘Mustang’ HSP profiles. The high level of RFLP between the two wheat lines was revealed by Southern blot hybridization utilizing a HSP17 probe. These data provide a molecular basis for further genetic analysis of the role of HSP genes in thermal tolerance in wheat.

These stocks carried 42 chromosomes and formed the expected multivalents (frequency of 14–25%) at metaphase 1. They gave floret fertility ranging from 40–60%. These stocks were subsequently used to determine the genetic control of low-molecular-weight (LMW) glutenin subunits in ‘Chinese Spring’ and ‘Gabo’ by means of two-step one-dimensional SDS-PAGE. All of the B subunits and most of the C subunits of glutenin were shown to be controlled by genes on the short arms of group-1 chromosomes in these wheats. The other C subunits were not controlled by group-1 chromosomes. The triple translocation line served as a suitable third parent in producing test-cross seeds for studying the inheritance of the LMW glutenin subunits and gliadins in wheat cultivars, e.g. ‘Chinese Spring’ and ‘Orca’. The segregation patterns of the LMW glutenin subunits in these cultivars revealed that the subunits were inherited in clusters and that their controlling genes (Glu-3) were tightly linked with those controlling gliadins (Gli-1). The LMW glutenin patterns d, d and e in ‘Orca’ segregated as alternatives to the patterns a, a and a in ‘Chinese Spring’ controlled by Glu-A3, Glu-B3 and Glu-D3 loci on chromosome arms 1AS, 1BS and 1DS, respectively, thus indicating that these patterns were controlled by allelic genes at these loci.

    Notes: Abstract To elucidate the phylogenetic relationships and cytoplasmic types, restriction endonuclease fragment patterns of chloroplast (cp) and mitochondrial (mt) DNAs isolated from two different accessions of Dasypyrum villosum (L.) candargy were compared with those of tetraploid wheat (Triticum durum Desf., PI265007), hexaploid wheat (Triticum aestivum L., cv Chinese Spring), Aegilops longissimum (S. and M., in Muschli) Bowden and Hordeum vulgare L. T. aestivum and T. durum had identical restriction patterns for their cp and mtDNAs in digestions with four different enzymes. Likewise, no differences were found between the restriction fragment patterns of two accessions of D. villosum. But, there were distinct differences in chloroplast and mitochondrial DNA restriction fragment patterns between D. villosum and tetraploid and hexaploid wheats. A. longissimum (G609) showed a similar pattern to those wheats for PstI digestion of cpDNA. Organellar DNA from Hordeum vulgare (cv Himalaya) showed a distinctly different restriction pattern from those of wheat and D. villosum. These results suggest that D. villosum is unlikely to be the donor of cytoplasm to wheats, and its cytoplasmic organelles were also different from those of A. longissimum.

    Notes: Abstract The objective of this study was to identify molecular markers linked to genes for resistance to powdery mildew (Pm) in wheat using a series of ‘Chancellor’ near-isogenic-lines (NILs), each having one powdery mildew resistance gene. A total of 210 probes were screened for their ability to detect polymorphism between the NILs and the recurrent parent. One of these restriction fragment length polymorphism (RFLP) markers (Xwhs179) revealed polymorphism not only between the NILs for the Pm3 locus, but also among NILs possessing different alleles of the Pm3 locus. The location of the marker Xwhs179 was confirmed to be on homoeologous chromosome group 1 with the help of nullitetrasomic wheat lines. The linkage relationship between this probe and the Pm3 locus was estimated with double haploid lines derived from a cross between wheat cvs ‘Club’ and ‘Chul’ (Pm3b). The genetic distance was determined to be 3.3±1.9 cM.

    Notes: Abstract Diploid-like chromosome pairing in polyploid wheat is controlled by several Ph (pairing homoeologous) genes with major and minor effects. Homoeologous pairing occurs in either the absence of these genes or their inhibition by genes from other species (Ph I genes). We transferred Ph I genes from Triticum speltoides (syn Aegilops speltoides) to T. aestivum, and on the basis of further analysis it appears that two duplicate and independent Ph I genes were transferred. Since Ph I genes are epistatic to the Ph genes of wheat, homoeologous pairing between the wheat and alien chromosomes occurs in the F1 hybrids. Using the Ph I gene stock, we could demonstrate homoeologous pairing between the wheat and Haynaldia villosa chromosomes. Since homoeologous pairing occurs in F1 hybrids and no cytogenetic manipulation is needed, the Ph I gene stock may be a versatile tool for effecting rapid and efficient alien genetic transfers to wheat.

    Notes: Abstract Deficiency of the wheat waxy (Wx) proteins (Wx-A1, Wx-B1 and Wx-D1) was studied in 1,960 cultivars derived from several countries. Gel electrophoretic analyses revealed that the null allele for the Wx-A1 protein occurred frequently in Korean, Japanese and Turkish wheats but was relatively rare in cultivars from other countries and regions. About 48% of the wheats deficient for the Wx-B1 protein were from Australia and India. One Chinese cultivar lacked the WxD1 protein. While 9 Japanese cultivars were deficient in both the Wx-A1 and Wx-B1 proteins, no cultivars lacked both the Wx-A1 and Wx-D1 proteins, both the Wx-B1 and Wx-D1 proteins or all three Wx proteins. Two-dimensional gel electrophoresis revealed polymorphisms of the three Wx proteins that varied according to isoelectric points or molecular weight. The Wx-A1 gene coding the Wx-A1 protein and the Wx-B1 gene coding the Wx-B1 protein were localized in the distal regions of chromosome arms 7AS and 4AL, respectively, by deletion mapping using the deletion lines developed in the common wheat cultivar ‘Chinese Spring’.

    Notes: Abstract Comparative genetic maps among the Triticeae or Gramineae provide the possibility for combining the genetics, mapping information and molecular-marker resources between different species. Dense genetic linkage maps of wheat and barley, which have a common array of molecular markers, along with deletion-based chromosome maps of Triticum aestivum L. will facilitate the construction of an integrated molecular marker-based map for the Triticeae. A set of 21 cDNA and genomic DNA clones, which had previously been used to map barley chromosome 1 (7H), were used to physically map wheat chromosomes 7A, 7B and 7D. A comparative map was constructed to estimate the degree of linkage conservation and synteny of chromosome segments between the group 7 chromosomes of the two species. The results reveal extensive homoeologies between these chromosomes, and the first evidence for an interstitial inversion on the short arm of a barley chromosome compared to the wheat homoeologue has been obtained. In a cytogenetically-based physical map of group 7 chromosomes that contain restriction-fragment-length polymorphic DNA (RFLP) and random amplified polymorphic DNA (RAPD) markers, the marker density in the most distal third of the chromosome arms was two-times higher than in the proximal region. The recombination rate in the distal third of each arm appears to be 8–15 times greater than in the proximal third of each arm where recombination of wheat chromosomes is suppressed.

    Notes: Abstract Forty-eight bread wheat (Triticum aestivum L.) released cultivars and elite advanced lines were evaluated for their ability to produce embryogenic callus using three different media. Basal N6 medium supplemented with dicamba (E1), MS medium containing 2,4-D (E3) or MS medium containing 2,4-D plus different amino acids (E5) were used for callus initiation and maintenance. Plant regeneration was achieved on basal MS medium with indole-3-acetic acid (IAA) and 6-benzylamino purine (BAP) and rooting on MS with 1-naphthaleneacetic acid (NAA). Percentage regeneration varied widely with both genotype and initiation medium, with values ranging from 2% to 94%. The number of plantlets produced per embryo ranged from 6 to 42. Thirteen genotypes showed at least 50% regeneration after culture on E5 medium; 3 genotypes after culture on E3 initiation medium and 1 after initiation on E1. After four subcultures, over a 16-week period, 41 genotypes (85%) lost their ability to regenerate plants while the remaining 7 lines (15%) retained plant regeneration potential but at reduced levels. E3 medium was found to be the best for maintaining regeneration potential after four subcultures.

    Notes: Abstract   Analyses of RFLPs, isozymes, morphological markers and chromosome pairing were used to isolate 12 Triticum aestivum cv Chinese Spring (genomes A, B, and D)-T. peregrinum (genomes Sv and Uv) disomic chromosome addition lines. The evidence obtained indicates that each of the 12 lines contains an intact pair of T. peregrinum chromosomes. One monosomic addition line, believed to contain an intact 6Sv chromosome, was also isolated. A CS-7Uv chromosome addition line was not obtained. Syntenic relationships in common with the standard Triticeae arrangement were found for five of the seven Sv genome chromosomes. The exceptions were 4Sv and 7Sv. A reciprocal translocation exists between 4Sl and 7Sl in T. longissimum and evidence was obtained that the same translocation exists in T. peregrinum. In contrast, evidence for syntenic relationships in common with the standard Triticeae arrangements were found for only one Uv chromosome of T. peregrinum; namely, chromosome 2Uv. All other Uv genome chromosomes are involved in at least one translocation, and the same translocations were found in the U genome of T. umbellulatum. Evidence was also obtained indicating that the centromeric regions of 4U and 4Uv are homoeologous to the centromeric regions of Triticeae homoeologous group-6 chromosomes, that the centromeric regions of 6U and 6Uv are homoeologous to the centromeric regions of group-4 chromosomes, and that 4U and 4Uv are more closely related overall to Triticeae homoeologous group-6 chromosomes than they are to group-4 chromosomes.

Three wheat germplasm lines possessing resistance to WSMV, derived from Triticum aestivum×Th. intermedium crosses, are analyzed by C-banding and genomic in situ hybridization (GISH) to determine the amount and location of alien chromatin in the transfer lines. Line CI15092 was confirmed as a disomic substitution line in which wheat chromosome 4A was replaced by Th. intermedium chromosome 4Ai?2. The other two lines, CI17766 and A29-13-3, carry an identical Robertsonian translocation chromosome in which the complete short arm of chromosome 4Ai?2 was transferred to the long arm of wheat chromosome 4A. Fluorescence in situ hybridization (FISH) using ABD genomic DNA from wheat as a probe and S genomic DNA from Pseudoroegneria stipifolia as the blocker, and vice versa, revealed that the entire short arm of the translocation was derived from the short arm of chromosome 4Ai?2 and the breakpoint was located at the centromere. Chromosomal arm ratios (L/S) of 2.12 in CI17766 and 2.15 in A29-13-3 showed that the translocated chromosome is submetacentric. This translocated chromosome is designated as T4AL ⋅ 4Ai?2S as suggested by Friebe et al. (1991).

Twenty out of eighty eight wheat microsatellites (WMS) tested were able to distinguish Triticum aestivum-Ae. markgrafii addition lines. Six, three, three, one and six of 18 WMS can be used as markers for single Ae. markgrafii chromosomes B, C, D, F and G, respectively. Addition line A is not available but additional bands, appearing only in Ae. markgrafii and the T. aestivum-Ae. markgrafii amphiploid and not in any of the available addition lines, indicate that three WMS detect markers for Ae. markgrafii chromosomes A. Addition line E could not be detected by any of the WMS markers applied, although the 20 WMS represented all the homologous groups of wheat. All three WMS located on the short arm of group-2 chromosomes were located on Ae. markgrafii chromosome B; three of four WMS, located on the long arm of wheat group-2 chromosomes, were specific to Ae. markgrafii chromosome G and three of four WMS, specific to group-5 chromosomes, were markers for Ae. markgrafii chromosome C, indicating the homoeology of these wheat chromosome arms with the respective Ae. markgrafii chromosomes.

    Notes: Abstract  Aegilops tauschii is the diploid D-genome progenitor of bread wheat (Triticum aestivum L. em Thell, 2n=6x=42, AABBDD). A genetic linkage map of the Ae. tauschii genome was constructed, composed of 546 loci. One hundred and thirty two loci (24%) gave distorted segregation ratios. Sixty nine probes (13%) detected multiple copies in the genome. One hundred and twenty three of the 157 markers shared between the Ae. tauschii genetic and T. aestivum physical maps were colinear. The discrepancy in the order of five markers on the Ae. tauschii 3DS genetic map versus the T. aestivum 3D physical map indicated a possible inversion. Further work is needed to verify the discrepancies in the order of markers on the 4D, 5D and 7D Ae. tauschii genetic maps versus the physical and genetic maps of T. aestivum. Using common markers, 164 agronomically important genes were assigned to specific regions on Ae. tauschii linkage, and T. aestivum physical, maps. This information may be useful for map-based cloning and marker-assisted plant breeding.

    Notes: Abstract  A new Hessian fly (Mayetiola destructor) resistance gene from Aegilops ventricosa and its transfer to hexaploid wheat is described. The 4D(4Mv) substitution line H-93-33 derived from the cross [(Triticum turgidum H-1-1×Aegilops ventricosa no. 11)×Triticum aestivum H-10-15] was highly resistant to the Spanish population tested. Resistance seemed to be inherited as a single dominant factor in the F2 generation resulting from a cross of H-93-33 with its susceptible parent (H-10-15). Resistance in Ae. venticosa no. 10 was located on chromosome 4Mv using Mv wheat/Ae. ventricosa addition lines. The resistance gene transferred from Ae. ventricosa no. 11 to H-93-33 (H27) is allelic with respect to that of Ae. ventricosa no. 10 and is non-allelic with respect to the genes H3 and H6 from Monon and Caldwell respectively. The assignment of H27 gene to chromosome 4Mv is further supported by its linkage to a gene encoding isozyme Acph-Mv1, previously located on chromosome 4Mv in the line H-93-33. A new marker from homoeologous chromosome group 4 (Amp-Mv2) present in H-93-33 and the 4Mv addition line is described.

    Notes: Abstract  Aegilops markgrafii contains resistance genes to powdery mildew, leaf rust and stripe rust, and also has high crude protein and lysine contents, which can be useful for wheat improvement. These important traits are localized on different chromosomes. Disomic Triticum aestivum-Ae. markgrafii addition lines and euploid introgression lines showing leaf-rust and powdery mildew resistance were screened with RAPDs to detect chromosome-specific markers which can accelerate the breeding process. RAPD markers for all six available disomic addition lines were obtained. The additional chromosomes B, C, D, E, F and G were identified by three, three, three, two, one and seven primers, respectively. All three chromosome-B-specific RAPD markers demonstrated the presence of alien chromatin in the leaf-rust-resistant 42-chromosome introgression lines as well as in the segregating progeny. The three chromosome-C-identifying primers also demonstrated the presence of that chromosome in powdery mildew-resistant euploid introgression lines. The substitution lines (5A)5C and (5D)5C with different genetic backgrounds for both parents, in comparison to the lines mentioned above, showed the chromosome C-specific band with only two of the three primers. The chromosome F-specific primer and a primer evident on all the Ae. markgrafii chromosomes analysed did not generate the expected fragments on the chromosome Fdel addition line, indicating that the markers are located on the deleted part of chromosome F.

    Notes: Abstract  The cereal cyst nematode (Heterodera avenae) is an important root parasite of common wheat. A high level of resistance was transferred to wheat from Aegilops triuncialis (TR lines) using the cross [(T. turgidum×Ae. triuncialis)×T. aestivum]. Low fertility (3–5 viable kernels per plant) was observed during the process but the surviving hybrid plants were highly vigorous. To obtain stable resistant lines further crosses to T. aestivum were performed. The resistance in TR lines seems to be transferred from the C genome of Ae. triuncialis (genomes CCUU). Ae. triuncialis was highly resistant to the two Spanish populations of H. avenae tested, as well as to four French races and two Swedish populations. The histological analysis showed a hypersensitive reaction in the roots of a resistant TR line inoculated with the Ha71 pathotype of H. avenae, whereas well-formed syncytia were observed in the roots of the susceptible control. Resistance to the H. avenae Ha71 pathotype seemed to be inherited as determined by a single dominant factor in the crosses between resistant TR lines and susceptible cultivars.

    Notes: Abstract Using recently developed solution culture techniques, the effect of a non-resource abiotic stress, nickel toxicity, was tested on intraspecific nutrient competition among wheat. The choice of an appropriate statistical model was of paramount importance in interpreting these effects. We argue that a multiplicative model is more appropriate for experiments on interactions of competition and abiotic stress. By such an analysis, nickel had no relative effect on the ability of competition to reduce plant size in two experiments, and caused a small reduction in competition in another. These results are contrary to other reports of the effect of a non-resource abiotic stress on competition and appear to be due to an increased demand for nutrients in the presence of toxic levels of nickel. The effects of an abiotic stress on competition may thus be specitic to the stress and not generalized across all abiotic stresses.

    Notes: Summary The study of interstellar molecules broadly includes two areas of interest. One area uses the unique ability of molecules to act as probes of the physical conditions in the cold, dense, visually opaque component of the interstellar medium. The physical properties of this and other components of the interstellar medium are summarized. The other area deals with the chemistry of interstellar molecules, recent aspects of which are emphasized in this review. Gas-phase chemistry, shock chemistry, and grain surface chemistry are discussed in the context of recent observations. No present observations suggest that surface reactions are relevant, but neither can they be ruled out. Ion-molecule reactions are clearly operative, at least for the simpler species. Chemical isotope fractionation is reviewed, andd it is concluded that the complexities of the chemistry allow no cosmological conclusions to be drawn from observations of deuterium in interstellar molecules, while the presence of13C in interstellar molecules permits an estimate of the12C/13C ratio which is consistent with the current concepts of the nucleosynthesis history of the Galaxy. Possible connections between interstellar molecules and the early molecular history of the solar system are discussed.

    Notes: Summary The direct and indirect effects of plant genetic diversity on epidemics and the influence of disease on plant competition were investigated using the wheat (Triticum aestivum)/stripe rust (Puccinia striiformis) system. Replacement series consisting of a susceptible and a resistant wheat genotype or two wheat genotypes susceptible to different races of stripe rust were grown in the presence and absence of the pathogen. Stripe rust severity, number of seed heads, seed yield, and seed weight were determined separately for each wheat genotype in the mixtures and the pure stands. The frequency of susceptible genotypes in a mixture explained up to 67% of the variation in disease severity. However, competitive interactions among plant genotypes sometimes appeared to alter susceptibility and obscured the relationship. In pure stands of single genotypes, disease severity explained between 52 and 58% of the variation in seed yield. In mixtures, coefficients of determination were only 10 and 31%, suggesting a strong influence of plant-plant interactions on seed yield. These results suggest that host-parasite coevolutionary models need to account for the strong effect that specific plant genotype combinations may have on disease severity and plant reproduction.

    Notes: Summary Competition for light among species in a mixed canopy can be assessed quantitatively by a simulation model which evaluates the importance of different morphological and photosynthetic characteristics of each species. A model was developed that simulates how the foliage of all species attenuate radiation in the canopy and how much radiation is received by foliage of each species. The model can account for different kinds of foliage (leaf blades, stems, etc.) for each species. The photosynthesis and transpiration for sunlit and shaded foliage of each species is also computed for different layers in the canopy. The model is an extension of previously described single-species canopy photosynthesis simulation models. Model predictions of the fraction of foliage sunlit and interception of light by sunlit and shaded foliage for monoculture and mixed canopies of wheat (Triticum aestivum) and wild oat (Avena fatua) in the field compared very well with measured values. The model was used to calculate light interception and canopy photosynthesis for both species of wheat/wild oat mixtures grown under normal solar and enhanced ultraviolet-B (290–320 nm) radiation (UV-B) in a glasshouse experiment with no root competition. In these experiments, measurements showed that the mixtures receiving enhanced UV-B radiation had a greater proportion of the total foliage area composed of wheat compared to mixtures in the control treatments. The difference in species foliage area and its position in the canopy resulted in a calculated increase in the portion of total canopy radiation interception and photosynthesis by wheat. This, in turn, is consistent with greater canopy biomass of wheat reported in canopies irradiated with supplemental UV-B.

    Notes: Abstract  Molecular markers were identified in common wheat for the Pm24 locus conferring resistance to different isolates of the powdery mildew pathogen, Erysiphe graminis DM f. sp. tritici (Em. Marchal). Bulked segregant analysis was used to identify amplified fragment length polymorphism (AFLP) markers and microsatellite markers linked to the gene Pm24 in an F2 progeny from the cross Chinese Spring (susceptible)× Chiyacao (resistant). Two AFLP markers XACA/CTA-407 and XACA/CCG-420, and three microsatellite markers Xgwm106, Xgwm337 and Xgwm458, were mapped in coupling phase to the Pm24 locus. The AFLP marker locus XACA/CTA-407 co-segregated with the Pm24 gene, and XACA/CCG-420 mapped 4.5 cM from this gene. Another AFLP marker locus XAAT/CCA-346 co- segregated in repulsion phase with the Pm24 locus. Pm24 was mapped close to the centromere on the short arm of chromosome 1D, contrary to the previously reported location on chromosome 6D. Pm24 segregated independently of gene Pm22, also located on chromosome 1D. An allele of microsatellite locus Xgwm337 located 2.4±1.2 cM from Pm24 was shown to be diagnostic and therefore potentially useful for pyramiding two or more genes for powdery mildew resistance in a single genotype.

    Notes: Summary Cyanogen bromide (CNBr) peptides were prepared of the insoluble collagen of bovine dental cementum. Following chromatographic separation, the peptides were identified by their amino-acid composition. Type I collagen ([α1(I)]2α2) accounted for more than 90% of the organic matrix, while Type III collagen ([α1(III)]3) was present at a level of approximately 5%. Amino-acid analyses revealed that the CNBr peptides from α1(I) and α2 chains of cementum closely resembled the corresponding peptides from calf skin. The only systematic difference was a higher level of hydroxylation of prolyl and lysyl residues of the cementum peptides.

La microdistribution de l'uranium dans l'os indique que cet élément est surtout limité à surface de l'endoste et, en particulier, aux surfaces de l'os lamellaire et aux parois des canaux de Havers, ouverts dans l'os corticol. Cette répartition suggère que l'uranium se présente sous une forme chimique impropre à son incorporation dans l'apatite osseux: il ne semble donc pas exister une distribution diffuse significative de l'uranium dans l'os.
    Abstract: Zusammenfassung Eine mittlere Konzentration von 2,4×10−8 g Uran/g Asche wurde in normalen menschlichen Knochen gefunden. Die Feinverteilung von Uran im Knochen zeigt, daß dieses Element hauptsächlich an der endostalen Oberfläche vorkommt, insbesondere an der Oberfläche des trabeculären Knochens und an den Wänden der offenen Haversschen Kanäle im kortikalen Knochen. Diese Verteilung läßt vermuten, daß Uran in einer chemischen Form vorliegt, welche sich für den Einbau in das Knochenapatit nicht eignet. Daraus folgt, daß keine signifikante diffuse Verteilung des Urans innerhalb des Knochens vorliegt.

This distribution suggests that uranium is present in a chemical form that is not acceptable for incorporation into bone apatite and consequently there does not appear to be a significant diffuse distribution of uranium throughout bone.

Une analyse microchimique est pratiquée pour le contenu et la composition en glycosaminoglycanes. 1. Entre la première semaine et la 4ème et 8ème semaine, un tissu conjonctif non-métachromatique se différencie en un cartilage métachromatique et la quantité de sulfate de chondroitine augmente de façon significative aux dépens des glycoprotéines. 2. Jusqu'à la 4ème semaine, la perte de substance est surtout comblée par de l'os néoformé: après cette période, la région est comblée au delà de la limite de la surface articulaire. 3. Le cartilage hyalin, ressemblant morphologiquement, autoradiographiquement et chimiquement au cartilage articulaire, en ce qui concerne la distribution en glycosaminoglycanes, constitute la surface articulaire de la perte de substance comblée dans un tiers des cas après 8 semaines. Le cartilage hyalin s'observe surtout dans les régions où de l'os néoformé a comblé la cavité médullaire. 4. Dans les deux tiers des cas, après 8 semaines, les surfaces articulaires des zones comblées comportent, non seulement du cartilage, mais aussi du tissu fibreux se formant essentiellement sur les parties latérales et dans les régions, où la cavité médullaire, fliant face, à la surface articulaire, n'a pas été comblée par du tissue osseux. La fraction glycoprotéique augmente par rapport à la fraction chondroitine sulfate. 5. Dans la majorité des cas, après 20 semaines, le cartilage néoformé subit des phénomènes dégénératifs, qui se traduisent par une diminution en chondroitine sulfate.
    Abstract: Zusammenfassung Bei ausgewachsenen Kaninchen wurde ein begrenzter, standardisierter, osteochondraler Defekt hervorgerufen, und das regenerierte Gewebe wurde histologisch und autoradiographisch durch Markierung in vitro mit35S-Sulfat und durch mikrochemische Bestimmung des Gehaltes und der Zusammensetzung der Glykosaminglykane untersucht. Die wichtigsten Befunde waren: 1. Zwischen 1 und 4–8 Wochen veränderte sich nichtmetachromatisches Bindegewebe zu metachromatisch gefärbtem Knorpel, und der Anteil an Chondroitin-Sulfat nahm auf Kosten der Glykoproteine signifikant zu. 2. Bis zu 4 Wochen war der Hauptteil des defekten Gebietes mit neugebildetem Knochen gefüllt; nach dieser Zeit lag dieser Bezirk oberhalb der Verknöcherungsgrenze in Richtung der Gelenkoberfläche. 3. Nach 8 Wochen bestand die Gelenkoberfläche des defekten Gebietes in einem Drittel der Fälle aus hyalinem Knorpel, der morphologisch, autoradiographisch und chemisch dem Gelenkknorpel in Bezug auf die Verteilung von Glykosaminoglykanen glich. Hyaliner Knorpel wurde hauptsächlich an Stellen beobachtet, wo neugebildeter Knochen die Markhöhle geschlossen hatte. 3. Nach 8 Wochen bestand die Gelenkoberfläche des defekten Gebietes in einem Drittel der Fälle aus hyalinem Knorpel, der morphologisch, autoradiographisch und chemisch dem Gelenkknorpel in Bezug auf die Verteilung von Glykosaminoglykanen glich. Nach 20 Wochen zeigten sich in den meisten Fällen bei neugebildetem Knorpel degenerative Veränderungen, welche durch eine gewisse Abnahme des Chondroitin-Sulfats wiedergegeben wurden.
    Notes: Abstract A limited, standardized osteochondral defect was created in adult rabbits and the regenerated tissue was examined histologically and autoradiographically after labellingin vitro with35S-sulphate, and microchemically for its content and composition of glycosaminoglycans. The principal findings were: 1. Between 1 week and 4 to 8 weeks, non-metachromatic connective tissue differentiated to metachromatically stained cartilage, and the proportion of the chondroitin sulphate increased significantly at the expense of the glycoproteins. 2. Up to 4 weeks, the major part of the defect area was filled with newly formed bone; after this time, the area lay above the level of the “tidemark”, towards the articular surface. 3. Hyaline cartilage with morphological, autoradiographic and chemical resemblance to the articular cartilage in terms of the distribution of glycosaminoglycans constituted the articular surface of the defect area in one-third of the cases at observation times after 8 weeks. Hyaline cartilage was observed mainly in areas where newly formed bone had closed the medullary cavity. 4. In two-thirds of the cases, after 8 weeks, parts of the articular surface of the defect consisted not only of cartilage but also of fibrous tissue, occurring mainly in the lateral parts of the defect and in areas where the medullary cavity facing the articular surface had not been sealed by bone tissue. The glycoprotein fraction increased relative to the chondroitin sulphate fraction. 5. In most cases after 20 weeks, newly-formed cartilage underwent degenerative changes, which were reflected in some reduction of the chondroitin sulphate.

    Description / Table of Contents: Résumé La cinétique de la formation et de la transformation des précipités de phosphate de calcium, obtenus en mélangeant de volumes égaux de solutions à 6×10−3 M de calcium total et/ou phosphate total est étudiée à 25°C. Les solutions de phosphate sont préajustées à un pH de 7.4. Les changements de pH et de turbidité des solutions sont suivis simultanément en fonction du temps. Les précipités sont isolés à des intervalles de temps variables et caractérisés par diverses méthodes physico-chimiques. Initialement un précipité avec un rapport molaire Ca/P de 1.5, amorphe aux rayons X et en diffraction électronique, est formé. Le spectre IR indique la présence de PO 4 3− et de HPO 4 2− . Après une période métastable, on observe la précipitation d'un matériel cristallin dans ou sur la phase amorphe. Vingt quatre heures après préparation de l'échantillon les précipités présentent surtout les caractères du phosphate octocalcique.
    Abstract: Zusammenfassung Die Kinetik der Bildung und Transformation von Calciumphosphat-Niederschlägen wurde bei 25°C untersucht. Es wurden dazu gleiche Volumen von Lösungen gemischt, bei einer Konzentration von 6×10−3M totales Calcium und/oder totales Phosphat. Die Phosphatlösungen wurden zuerst auf pH 7,4 eingestellt. Veränderungen des pH und Trübung der Lösungen wurden gleichzeitig als eine Funktion der Zeit aufgezeichnet. Niederschläge wurden in verschiedenen Zeitintervallen isoliert und mit verschiedenen physiko-chemischen Methoden charakterisiert. Am Anfang wurde ein Niederschlag mit einem molaren Ca/P-Verhältnis von 1,5, im Röntgenbild und in der Elektronendiffraktion amorph, gebildet. Infrarotspektren deuteten die Anwesenheit von PO 4 3− - und HPO 4 2− -Ionen an. Nach einer metastabilen Periode erfolgte ein Niederschlag aus kristallinem Material innerhalb oder auf der amorphen Substanz. 24 Std nach der Herstellung der Proben zeigten die Niederschläge in der Hauptsache die Charakteristiken von Octocalciumphosphat.
    Notes: Abstract The kinetics of the formation and transformation of calcium phosphate precipitates obtained by mixing equal volumes of solutions, 6×10−3 M in total calcium and/or total phosphate was investigated at 25°. The phosphate solutions were preadjusted to pH 7.4. Changes of the pH and turbidity of the solutions were followed simultaneously as a function of time. Precipitates were isolated at various time intervals and characterized by different physicochemical methods. Initially a precipitate with a molar Ca/P ratio of 1.5, amorphous to X-ray and electron diffraction was formed. IR spectra indicated the presence of PO 4 3− and HPO 4 2− ions. After a period of metastability, precipitation of a crystalline material within or upon the amorphous matter occurred. Twenty four hours after sample preparation the precipitates showed mainly the characteristics of octacalcium phosphate.

    Description / Table of Contents: Résumé Des diagrammes de phase d'équilibre ont été déterminés pour le système CaO-P2O5-H2O en utilisant des techniques de synthèse hydrothermique au cours de variatio nsde température allant de 300–600° et 2 Kb H2O de pression. De l'hydroxyapatite bien cristallisé a été synthétisé et caractérisé. De faibles variations de paramètres de la maille cristalline, liées à la température de synthèse et composition globale du matériel initial, ont été déterminées. Des conditions chimiques précises sont nécessaires pour obtenir de l'apatite, en tant que seule phase solide en équilibre dans la solution. Les résultats de diagramme de phase d'équilibre sont comparés avec ceux obtenus dans des milieux synthétiques.
    Abstract: Zusammenfassung Es wurden Gleichgewichts-Phasendiagramme für das System CaO-P2O5-H2O bestimmt, indem hydrothermale Synthese-Techniken im Temperaturbereich von 300–600° und bei einem Druck von 2 Kb H2O verwendet wurden. Es wurde gut-kristallisiertes Hydroxyapatit erzeugt und charakterisiert. Es wurden geringe Unterschiede in den Parametern der Zelleinheiten festgestellt, welche von der angewandten Temperatur und der Zusammensetzung des Startmaterials abhingen. Es waren genaue chemische Bedingungen nötig, um Apatit als die einzige feste Phase im Gleichgewicht mit der Lösung zu erhalten. Die Resultate der Gleichgewichts-Phasendiagramme werden mit früheren Untersuchungen mit der Synthesetechnik verglichen.
    Notes: Abstract Equilibrium phase diagrams have been determined for the system CaO-P2O5-H2 using hydrothermal synthesis techniques in the temperature range 300–600° and 2 Kb H2O pressure. Well-crystallized hydroxyapatite has been produced and characterized. Small variations in unit cell parameters dependent on temperature of synthesis and bulk composition of the starting materials have been determined. Precise chemical conditions were required to obtain apatite as the only solid phase in equilibrium with solution. Equilibrium phase diagram results are compared with previous synthetic investigations.

L'axe cristallographique C est de 6.865 A et l'axe a de 9.374 A. Des expériences d'échanges réalisés à l'aide de45Ca,32P et18F indiquent la présence de gros cristallites de surface spécifique de l'ordre de 1 m2/g. Il apparait que l'interprétation physique des processus d'échange ne nécessite pas l'existence de compartiments séparés, avec chacun son propre facteur cinétique, les échanges semblent être simplement liés à un changement exponentiel dans l'énergie libre de la réaction. Pour la réaction suivante: $$(Ca)_5 (PO_4 )_3 OH solide + (F^ - ) \rightleftarrows (Ca)_5 (PO_4 )_3 F solide + (OH^ - )$$ , la constante thermodynamique de 101.26 a été calculée, suggérant que le fluorapatite se forme toujours aux dépens de l'hydroxyapatite dans des conditions physiologiques. Cette transformation se continue en abaissant le pH.

Diese Umwandlung wird erhöht, wenn das pH erniedrigt wird.

The over-all composition corresponds to $$(Ca^{2 + } )_{9.98} (Sr^{2 + } ,Na^ + ,K^ + ,Mg^{2 + } )_{0.02} (PO_4^{3 - } )_{5.98} (HCO_3^ - ,CO_3^{2 - } )_{0.02} (F^ - )_2 $$ . The crystallographic c-axis was determined to be 6.865 Å, and the a-axis 9.374 A. Exchange data obtained by employing45Ca,32P, and18F indicate the presence of large crystallites with a specific surface of the order of 1 m2/g. It is indicated that the physical interpretation of the exchange process does not require the existence of separate departments, each with its own kinetic factor, but that the exchange may be simply related to the exponential change in the free energy of the reaction. For the reaction $$(Ca)_5 (PO_4 )_3 OH_{solid} + (F^ - ) \rightleftarrows (Ca)_5 (PO_4 )_3 F_{solid} + (OH^ - )$$ the thermodynamic constant has been calculated to be 101.26, implying that fluorapatite always will form at the expense of hydroxyapatite under physiologic conditions.     Notes: Summary The calcium homeostatic mechanism was challenged in adult female rats by feeding them a calcium-deficient diet containing oxalate, and by subjecting them to pregnancy and lactation. The regimen caused a substantial weight loss, especially in those animals which reared their young well. Severe hypocalcaemia was observed in the lactating rats. Serum-P was slightly elevated. The content of hydroxyproline in serum was considerably elevated, reflecting the degree of calcium deprivation. Serum proteins were least influenced. The calcium depriving regimen reduced the growth of long bones, but did not stop it. The ash content of the bones was considerably reduced, the degree of reduction depended on the degree of calcium deprivation. Ash as percentage of total bone organ was reduced, but not to the same extent as total ash. Analyses of different parts of femur showed that the proximal and distal parts had lost more bone mineral than the diaphyseal shaft. The ash content of cortical bone tissue from the femur was estimated by a volumetric method. No differences were observed between test groups and controls, indicating that no measurable amounts of bone mineral had been removed from the walls of the vascular canals or by osteocytic osteolysis. Planimetric determinations on cross sections from femora disclosed that a great amount of bone had been removed from the endosteal surface of the diaphysis, while the periosteal surface demonstrated reduced bone apposition.

    Abstract: Zusammenfassung Während der ungewöhnlichen Häutungssequenz des Frischwasser-IsopodenLirceus brachyurus (Harger) wird die hintere Hälfte des äußeren Skeletts 24 Std vor der vorderen Hälfte abgestoßen. In der halbgehäuteten Phase erfolgt Remineralisation im hinteren Teil, während der vordere Teil in einem Vorhäutungszustand ist. Der prozentuale Unterschied des Calciums in den zwei Hälften bei Halb- und Vollhäutungszustand ist 22% (p〈0,01) bzw. 33% (p〈0,01), was andeutet, daß Calcium während der Häutung abgesondert wird. Die Geschwindigkeit der Remineralisation erhellt aus der Tatsache, daß sich der Gesamtmineralgehalt im hinteren Teil zwischen Halt- und Vollhäutung, in der vorderen Hälfte jedoch zwischen Endhäutung und einem Tag nach der Häutung verdoppelt. Calciumcarbonat in kristalliner Calcitform wurde mittels Elektronendiffraktion von dünnen Hautschnitten nachgewiesen.
    Notes: Abstract During the unusual molt sequence of the fresh-water isopod,Lirceus brachyrus (Harger), the posterior half of the exoskeleton is shed 24 hours before the anterior half. At the half-molt stage, occurs in the posterior part while the anterior portion is in a pre-molt condition. The percentage difference in calcium in the two halves at half-molt and full-molt is22 (p〈0.01) and33 (p〈0.01) respectively, an indication that calcium is sequestered during The rapidity of remineralization is illustrated by the fact that the total mineral content doubles in the posterior part between half and full molt and in the anterior half between the end of molt and one day after ecdysis. Calcium carbonate in the calcite cystalline form was demonstrated by electron diffraction of thin sections of the integument.

    Notes: Abstract The effects of disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) on bone and serum chemistry were investigated in adult rabbits. EHDP was administered by subcutaneous injection at doses of 0.25, 2.5 and 10 mg/kg body weight/day for of 28 days. Blood samples were obtained weekly from each rabbit and serum levels of total calcium, ionized calcium, inorganic phosphate and alkaline phosphatase were determined. At the end of the treatment period all rabbits were sacrificed and the tibiae removed for chemical analysis and histological evaluation. The effect of EHDP administration on serum chemistry was both dose- and time-related. The highest of the three doses, 10 mg/kg/day, resulted in a time-related decrease in total serum calcium. This dose also caused a rapid but transient reduction in serum ionized calcium. The effect of EHDP on serum inorganic phosphate was biphasic. Administration of 2.5 mg/kg/day resulted in a time-related elevation in this parameter, whereas the 10 mg/kg/day dose resulted in a time-related hypophosphatemic response. There were no significant drug-related changes in tibial fat-free dry weight, ash weight, total calcium or total phosphorus values. However, administration of 2.5 and 10 mg/kg/day EHDP resulted in increased osteoid tissue as measured histologically. These results are compared with data from other EHDP studies, and discussed in relation to the maturity and growth-state of the experimental animals.

La filtration sur gel à l'aide de colonnes Sephadex G 10 perment de différencier le matériel isolé en deux composants, ayant la même fluorescence et la même absorption UV. La fluorescence est indépendante de pH de 3.5–9.5. Des études de dialyse et de filtration sur gel de matrices gélatinisées indiquent une association étroite du matériel fluorescent avec les chaines polypeptidiques de collagène.
    Abstract: Zusammenfassung Fluorescierende Bestandteile aus Knochen und Dentin wurden in Sephadex C25 CM Kationen-Austauschersäulen von alkalischen Hydrolysaten ihrer Matrices getrennt. Die Fluorescenzintensitäten sowie die Erregungs- (λ max 330 nm) und Emissions- (λ max 395 nm) Spektren waren dieselben wie bei intakten und gelatinisierten Matrices. Die Fluorescenzparameter wurden durch die Hydrolyse nicht verändert. Eine Gelfiltration über Sephadex-G10-Säulen trennte das isolierte Material in 2 Komponenten auf, welche gleiche Fluorescenz- und UV-Absorptionseigenschaften zeigten. Im pH-Bereich zwischen 3,5 und 9,5 war die Fluorescenz unabhängig vom pH. Dialysierversuche sowie Gelfiltrationsexperimente mitden gelatinisierten Matrices zeigten eine starkgefügte Bindung des fluorescierenden Materials mit den Polypeptidketten des Kollagens.
    Notes: Abstract Fluorescent components in bone and dentine were separated from alkaline hydrolysates of their matrices on Sephadex C25 CM cationic exchange columns. The fluorescence levels, and the excitation (λ max 330 nm) and emission (λ max 395 nm) spectra, were the same as those observed in the intact and gelatinised matrices. The fluorescence parameters were unaltered by the hydrolysis procedure. Gel filtration on Sephadex G. 10 columns further resolved the isolated material into two components with the same fluorescence and UV absorption properties. The fluorescence was independent of pH over the range 3.5–9.5. Dialysis and gel filtration studies on the gelatinised matrices indicated a firmly-bonded association of the fluorescent material with the collagen polypeptide chains.

    Notes: Abstract An assessment is made of the relative contribution of certain classes of energetic particle precipitation to the chemical composition of the middle atmosphere with emphasis placed on the production of odd nitrogen and odd hydrogen species and their subsequent role in the catalytic removal of ozone. Galactic cosmic radiation is an important source of odd nitrogen in the lower stratosphere but since the peak energy deposition occurs below the region where catalytic removal of O3 is most effective, it is questionable whether this mechanism is important in the overall terrestrial ozone budget. The precipitation of energetic solar protons can periodically produce dramatic enhancement in upper stratospheric NO. The long residence time of NO in this region of the atmosphere, where catalytic interaction with O3 is also most effective, mandates that this mechanism be included in future modelling of the global distribution of O3. Throughout the mesosphere the precipitation of energetic electrons from the outer radiation belt (60°≲Λ≲70°) can sporadically act as a major local source of odd hydrogen and odd nitrogen leading to observable O3 depletion. Future satellite studies should be directed at simultaneously measuring the precipitation flux and the concomitant atmosphere modification, and these results should be employed to develop more sophisticated models of this important coupling.

A pH 7.40, de l'ACP frais a un rapport molaire Ca/P de 1.36±0.02 et contient 22.8 (±2.2)% HPO 4 2− . Les résultats obtenus avec du précipité non lavé ne peuvent s'expliquer par du Ca2+ emprisonné et de l'HPO 4 2− ou du Na+, Cl− et CO 3 2− exogènes. Les phosphates de calcium amorphes constituent une classe de sels ayant des caractères chimiques variables et des propriétés physiques identiques, comparables au verre. Le CaHPO4·xH2O non cristallin peut être un ACP, surtout au cours des phases précoces de formation. A des pH physiologiques, l'ACP se transforme en petits cristaux applatis contenant de fortes quantités de phosphate acide facilement remplaçable. Le fait de laver la couche de surface produit un changement chimique dans les nouveaux cristaux: des cristaux non lavés donnent des diagrammes de diffraction d'apatite peu cristallins, ainsi que des spectres infra-rouges peu nets, intermédiaires entre des apatites et du phosphate octocalcique. Des explications structurales sont proposées et les compositions minérales amorphe/cristalline de l'os et du cartilage sont recalculées.
    Abstract: Zusammenfassung Ungewaschene Proben von amorphem Calciumphosphat (ACP) enthalten eine unersetzliche labile Fraktion, welche reich an saurem Phosphat ist und ein niederes Ca/P-Verhältnis hat und welche während des Waschprozesses unwiderruflich verloren geht. Natives ACP, welches im pH-Bereich 6,6–10,6 ausgefällt wurde, variierte im molaren Ca/P-Verhältnis zwischen 1,18 und 1,50 und in HPO 4 2− /totales P zwischen 33,0 und 10,1%. Bei pH 7,40 hatte natives ACP ein molares Ca/P-Verhältnis von 1,36±0,02 und enthielt 22,8 (±2,2)% HPO 4 2− . Die Werte beim ungewaschenen Niederschlag rühren weder von aus dem Überstand aufgenommenem Ca2+ und HPO2−, noch von außen kommendem Na+, Cl− und CO 3 2− her. Die amorphen Calciumphosphate werden als eine Klasse von Salzen erkannt, welche veränderliche chemische, aber identische glasartige physicochemische Eigenschaften haben. Nicht kristallines CaHPO4·xH2O kann auch ein ACP sein, besonders in den frühen Bildungsstadien. Bei physiologischem pH verwandelt sich ACP in kleine plattenförmige Kristalle, welche große Mengen von leicht ersetzbarem saurem Phosphat enthalten. Das Waschen dieser Oberflächenschicht erzeugte chemische Veränderungen in den resultierenden Kristallen; ungewaschene Kristalle zeigten ein Diffraktionsmuster, das nur schwach demjenigen des kristallinen Aspatites glich, aber ein schlecht aufgelöstes Infrarotspektrum, welches zwischen Apatit und Octocalciumphosphat war. Es werden strukturelle Erklärungen für alle diese Phenomena diskutiert, und revidierte amorph/kristalline Mineralzusammensetzungen von Knochen und Knorpel wurden neu berechnet.
    Notes: Abstract Unwashed samples of amorphous calcium phosphate (ACP) contain an irreplaceable labile fraction, rich in acid phosphate and low in Ca/P ratio, which is irreversibly lost during the washing process. Native ACP precipitated in the pH range 6.6–10.6 varied in Ca/P molar ratio from 1.18 to 1.50 and in HPO 4 2− /total P from 33.0% to 10.1%. At pH 7.40, native ACP had a Ca/P molar ratio of 1.36±0.02 and contained 22.8 (±2.2)% HPO 4 2− . Unwashed precipitate data could not be attributed to either trapped supernatant Ca2+ and HPO 4 2− or extraneous Na+, Cl−, and CO 3 2− . The amorphous calcium phosphates are recognized as a class of salts having variable chemical but identical glass-like, physicochemical properties. Non-crystalline CaHPO4·xH2O may also be an ACP, especially during early formative stages. At physiological pH, ACP transforms to small platy crystals containing large amounts of readily-replaceable acid phosphate. Washing this surface layer produced chemical alterations in the resultant crystals; unwashed crystals had poorly-crystalline apatitic diffraction patterns but exhibited poorly-resolved infrared spectra intermediate between apatite and octacalcium phosphate. Structural explanations for all these phenomena are discussed, and revised bone and cartilage amorphous/crystalline mineral compositions have been re-calculated.

    Notes: Summary A method is described for the isolation and purification of a sulphated glycopeptide from human supragingival calculus. The compound was isolated after using EDTA treatment, 2 M CaCl2 extraction, proteolytic digestion, ethanol precipitation, and finally purified by DEAE cellulose chromatography. It migrated as a single component on cellulose acetate electrophoresis, and chemical and infrared spectral analysis showed the presence of covalently attached sulphate groups. The sulphated glycopeptide was distinguished from being a sulphated glycosaminoglycan.

    Notes: Summary The bone resorptive factor and amphipathic antigen (AcA) previously identified by us in preparations fromActinomyces viscosus have been partially purified, characterized chemically, and compared. They elute at the same location on chromatography with Ac 22. The fatty acid composition of AcA and the bone resorptive factor is the same. Some differences in carbohydrate composition are observed. TheActinomyces factor does not affect calcium influx or cyclic AMP in isolated bone cells. Therefore it is concluded that AcA stimulates resorption either by gaining entrance into bone cells or by way of a yet undetermined second messenger.

Des analyses chimiques et par diffraction aux rayons X du follicule ont confirmé ce résultat. — En se basant sur l'augmentation du calcium et du phosphore, les calcifications débutent dans le tissue folliculaire 6–12 h après une blessure d'intensité moyenne de la peau de rats, ayant reçu du dihydrotachysterol (DHT), et 24–48 h après une blessure similaire chez des rats non injectés. Les diagrammes de diffraction aux rayons X sont diffus. Trois heures après la blessure, on note une augmentation du calcium du tissu folliculaire qui ne semble pas en rapport avec le DHT qui traduit probablement une liaison de calcium plutôt qu'un dépot minéral.
    Abstract: Zusammenfassung Frühere histologische Untersuchungen haben gezeigt, daß der Haarfollikel besonders anfällig für Verkalkungen ist, wenn die Haut von hypercalcämischen Ratten verletzt wird. Dieses Resultat wurde nun durch direkte chemische Bestimmungen und Röntgendiffraktions-analysen von Follikelgewebe bestätigt. Aufgrund der erhöhten Calcium- und Phosphatwerte kann gesagt werden, daß nach einer leichten Quetschung der Haut von Ratten, die mit Dihydrotachysterol (DHT) behandelt wurden, im Haarfollikelgewebe nach 6–12 Std Mineral-ablagerungen stattfanden, wogegen Kontrollratten mit der gleichen leichten Hautverletzung diese Ablagerungen erst nach 24–48 Std zeigten. Röntgendiffraktionsanalysen ergaben ein diffuses Apatit-Muster. Innerhalb 3 Std nach der Verletzung wurde ein Anstieg des Calcium-gehaltes im Follikelgewebe beobachtet, der nicht im Zusammenhang mit der DHT-Behandlung stand, also nicht eine Mineralablagerung, sondern eher eine Bindung von Calcium widerspiegelte.
    Notes: Abstract Previous histological investigations have shown that the hair follicle is particularly susceptible to mineralization when the skin of hypercalcaemic rats is injured. Direct chemical and X-ray diffraction analyses of follicle tissue have now confirmed this finding. As judged by increases in both calcium and phosphorus, mineral deposits began to form in hair follicle tissue 6–12 h after a mild crush injury to the skin of rats dosed with dihydrotachysterol (DHT), and 24–48 h after a similar injury to the skin of non-dosed rats. X-ray diffraction gave a diffuse apatite pattern. Within 3 h of injury there was a rise in the calcium content of follicle tissue which was not related to DHT-dosing and which was probably a reflection of calcium binding rather than mineral deposition.

    Description / Table of Contents: Résumé Un programme d'ordinateur a été mis au point pour calculer les activités ioniques du calcium et l'orthophosphate dans un grand nombre de solutions. Dans le cas de solutions synthétiques, les calculs sont vérifiés en comparant les valeurs de pH, obtenues par ordinateur, avec celles observées expérimentalement. Des essais de ce type, avec des solutions possèdant des concentrations de calcium et d'orthophosphate trouvées dans les liquides biologiques et à des valeurs de pH variant de 3.00 à 10.00, indiquent que le programme est adapté pour des applications biologiques. Le programme n'est pas effectif pour des solutions, dans les lesquelles l'ion bromure est la source principale de la force ionique, sans doute, par manque d'équation étendue de Debye-Hückel dans ces circonstances. Aucune formation de complexe de phosphate de sodium n'a été notée à des concentrations biologiques normales.
    Abstract: Zusammenfassung Es wurde ein Computer-Programm ausgearbeitet, um die Ionenaktivitäten von Calcium und Orthophosphat in einer breiten Varietät von Lösungen zu berechnen. Die Berechnungen wurden bei synthetischen Lösungen durch Vergleiche zwischen den auf diese Weise errechneten pH-Werten und den experimentell gefundenen kontrolliert. Diese Art Kontrollen mit Calcium-und Orthophosphatkonzentrationen, wie sie in biologischen Flüssigkeiten gefunden werden, und mit pH-Werten zwischen 3,0 und 10,0 wies darauf hin, daß das Programm für biologische Anwendungen geeignet war. Das Programm konnte nicht benützt werden für solche Lösungen, bei welchen hauptsächlich das Bromidion zur Einstellung der Ionenstärke verwendet wurde, vermutlich weil die erweiterte Debye-Hückel-Gleichung unter diesen Umständen nicht anwendbar ist. Die Bildung eines Natriumphosphat-Komplexes unter normalen biologischen Konzentrationen konnte nicht nachgewiesen werden.
    Notes: Abstract A computer program has been designed to calculate the ionic activities of calcium and orthophosphate in a wide variety of solutions. In the case of synthetic solutions the calculations were checked by comparing the computed pH values with those observed experimentally. Tests of this type with solutions having the concentrations of calcium and orthophosphate found in biological fluids and with pH values ranging from pH 3.0–10.0 indicated that the program was suitable for biological applications. The program was not effective for solutions in which the bromide ion was a principal source of ionic strength probably because of the failure of the extended Debye-Hückel equation under those circumstances. No evidence for the formation of any sodium phosphate complex at normal biological concentrations could be found.

    Notes: Summary Wheat mitochondria) DNA has been isolated from callus cultures initiated from both immature embryos and the corresponding parental cultivar. A Sall restriction pattern study has shown that the organization of callus culture mitochondria) DNA underwent extensive change, characterized by either the disappearance or the decrease in the relative stoichiometry of several restriction bands. Hybridization of labelled mitochondrial fragments obtained from a recombinant cosmid library to Southern blots of callus and parental line restricted mitochondria) DNAs has shown that a fraction of the mitochondria) genome was lost in callus cultures. Data from a Sall + HindIII restriction map of a defined part of the wheat mitochondria) genome concerned with some of these variations strongly suggest that the observed variations correspond to the disappearance of at least one mitochondria) DNA subgenomic molecule in callus cultures.

    Notes: Summary The nucleotide sequence of the wheat mitochondrial 26S ribosomal RNA gene and flanking regions was determined and compared with mitochondrial 26S rRNA genes from maize and Oenothera. When the plant mitochondrial 26S rRNA genes are compared with Escherichia coli 23S rRNA and chloroplast 23S and 4.5S rRNA genes, a third variable region is apparent close to the 3′ end of the gene. The 5′ and 3′ ends of the wheat mitochondrial gene were determined by S1 nuclease mapping. Computer analysis of the wheat mitochondrial gene revealed several small sequences present either in the 5′ region of the 26S rRNA gene or in the 18S rRNA gene.

    Notes: Summary Embryogenic and non-embryogenic long-term callus cultures of hexaploid wheat exhibit differences in the organization of their mitochondrial genome. Embryogenic and non-embryogenic fractions of callus cultures initiated from immature embryos of the wheat cultivar “Chinese Spring” have been isolated and subsequently subcultured. DNA-DNA hybridization experiments using labelled cloned wheat mitochondrial DNA fragments have shown that the mitochondrial DNA organization of embryogenic subcultures derived from embryogenic parts of “Chinese Spring” calli is closely related to that of the initial “Chinese Spring” calli, while non-embryogenic subcultures derived from non-embryogenic fragments of “Chinese Spring” calli exhibit a mitochondrial DNA organization similar to that found in non-embryogenic calli derived from cultivar “Aquila”. In addition, somatic tissue cultures initiated from three other non-embryogenic wheat cultivars (“Talent”, “Thésée” and “Capitole”) display mitochondrial DNA arrangements similar to those found in cultivar “Aquila”. These results strongly suggest that, in wheat callus cultures, a particular mitochondrial genome organization is correlated with the ability of cultured cells to regenerate whole plants.

    Notes: Abstract The mitochondrial genome of the selfed progeny of a plant regenerated from long-term somatic tissue culture displays specific structural rearrangements characterized by the appearance of novel restriction fragments. A mitochondrial DNA library was constructed from this selfed progeny in the SalI site of cosmid pHC79 and the novel fragments were subsequently studied. They were shown to arise from reciprocal recombination events involving DNA sequences present in the parental plant. The regions of recombination were sequenced and the nucleotide sequences were aligned with those of the presumptive parental fragments. We characterized an imperfect short repeated DNA sequence, 242 bp long, within which a 7-bb DNA repeat could act as a region of recombination. The use of PCR technology allowed us to show that these fragments were present in both parental plants and tissue cultures as low-abundance sequence arrangements.

    Notes: Summary The mitochondrial 18S and 5S ribosomal RNA (rRNA) genes of rye, plus a total of about 90 kilobase pairs of flanking DNA, have been cloned and maps of restriction enzyme cleavage sites have been constructed. Like their homologs from hexaploid wheat, the rye genes are closely linked and are part of a three-copy family of recombining repeats (the “18S/5S repeat”). The rye repeat probably also contains a mitochondrial tRNAfMet gene, which the wheat repeat is known to carry. However, despite the overall organizational similarity between the wheat and rye 18S/5S repeats in the immediate vicinity of their coding regions, extensive rearrangement of flanking sequences has taken place during evolutionary divergence of the two species. Our data provide additional support for an emerging picture of plant mitochondrial genomes as evolving much more rapidly in structure than in sequence.

    Notes: Summary An internal part of the chloroplast atpA gene has been identified in the mitochondrial DNA of Triticum aestivum. It is located near the 18S-5S ribosomal genes and partially contained within a repeated sequence. Comparison of the transferred sequence with the original ct sequence reveals several nucleotide changes and shows that neither 5′ nor 3′ ends are present in the mt genome. No transcript of this region could be detected by Northern analysis. This sequence is present in mitochondrial genomes of other tetraploid and diploid species of Triticum, also in the vicinity of the 18S-5S ribosomal genes, suggesting a unique transfer event. The date of this event is discussed.

    Notes: Summary The nucleotide sequence of a region of wheat chloroplast DNA containing the psbB gene for the 47 kDa chlorophyll a-binding protein of photosystem II has been determined. The gene encodes a polypeptide of 508 amino acid residues which is predicted to contain six hydrophobic membrane-spanning regions. The psbB gene is located 562 bp upstream of the psbH gene for the 10 kDa phosphoprotein of photosystem II. A small open reading frame of 38 codons is located between psbB and psbH, and on the opposite strand the psbN gene, encoding a photosystem II polypeptide of 43 amino acid residues, is located between orf38 and psbH. S1 nuclease mapping indicated that the 5′ ends of transcripts were located 371 and 183 bp upstream of the psbB translation initiation codon. Predominant transcripts of 2.1 kb and 1.8 kb for psbB and 0.4 kb for psbH were present in RNA isolated from etiolated and greening wheat seedlings. Immunodecoration of Western blots indicated that the 47 kDa polypeptide was absent, or present in very low amounts, in dark-grown tissue and accumulated on greening, whereas the 10 kDa polypeptide was present in similar amounts in both dark-grown and greening seedlings. The 10 kDa polypeptide was phosphorylated in vitro by incubating wheat etioplast membranes with [γ3 2P] ATP.

    Notes: Summary To identify transcription initiation sites in wheat mitochondria, the nascent 5′-ends of transcripts were specifically labeled by incubation of wheat mitochondrial RNA with [α-32P]GTP in the presence of the enzyme guanylyltransferase. After separation of the resulting capped transcripts by electrophoresis in polyacrylamide gels, individual RNAs were recovered and directly sequenced. Four RNA sequences obtained in this way were localized upstream of the protein-coding genes atpA, coxII, coxIII and orf25. Comparison of mRNA and gene sequences allowed precise positioning of transcription initiation sites for these four genes. Sequence similarities immediately upstream of these sites define a conserved motif that we suggest as a candidate regulatory element in wheat mtDNA. The relationship between this motif and putative mitochondrial promoters in other plant species is discussed.

    Notes: Abstract  We examined the utility of otolith minor and trace element chemistry, assayed with inductively coupled plasma mass spectrometry (ICP-MS), as a means of delineating population structure in the Nassau grouper (Epinephelus striatus). We characterized the elemental composition of otoliths collected in 1993 from three locations in Exuma Sound, Bahamas and from Glover Reef, Belize in 1995. A single location in Exuma Sound was sampled in 1994 to test temporal variability in otolith composition. Five elements (Ca, Zn, Sr, Ba and Pb) were routinely detected, at levels significantly above background, by solution-based ICP-MS. Results from analysis of variance of elemental data, expressed as a ratio to Ca, indicated that there were no significant differences among the Exuma locations for any element, but significant variability was found between Glover Reef and the pooled Exuma localities for Zn/Ca, Sr/Ca and Ba/Ca ratios. Significant inter-annual differences at one Exuma Sound location was restricted to Ba/Ca ratios. Discriminant function analysis correctly classified 86% and 95% of the Belize and pooled Exuma sites, respectively. Otoliths from Belize were characterized by low Zn/Ca and high Ba/Ca and Pb/Ca ratios compared to otoliths from fish collected in Exuma Sound. Although differences in Ba levels may be related to upwelling at Glover Reef, more data are needed to definitely link otolith composition with regional differences in water chemistry.

    Notes: Summary Although the mitochondrial genomes of the Chinese Spring and Aquila varieties of wheat are normaly similar in organization, this is not so in tissue cultures initiated from their immature embryos where the mitochondrial genomes of both are rearranged and in different, characteristic, ways. However, the mitochondrial genomes of tissue cultures of reciprocal F1 crosses between these varieties were almost identical to one another, showing that nuclear genes control the rearrangement processes. These rearrangements are either due to the appearance of new structures or else result from changes in the relative amounts of subgenomic components. The severe reduction in the amount of certain molecular configurations in tissue cultures from reciprocal crosses is probably due to the presence of dominant information in the Aquila nuclear genome. Data obtained from tissue cultures initiated from F2 embryos of the cross Aquila x Chinese Spring suggest that at least two complementary genes are involved in this control. In contrast, the presence of new molecular arrangements appears to be under the control of a dominant allelic form of a Chinese Spring gene or genes. Thus, this study demonstrates that at least two sets of nuclear genes control the reorganization of the mitochondrial genome which occurs when tissue cultures are initiated from the immature embryos of wheat.

    Notes: Summary The mitochondrial genomes of wheat and rye each contain a three-member family of recombining repeat peat sequences (the “18S/5S repeat”) that encode genes for 18S and 5S rRNAs (rrn18 and rrn5) and tRNAfMet (trnfM). Here we present, for wheat and rye, the sequence and boundaries of the “common sequence unit” (CSU) that is shared between all three repeat copies in each species. The wheat CSU is 4,429 base-pairs long and contains (in addition to trnfM, rrn18 and rrn5) a putative promoter, three tRNA-like elements (“t-elements”), and part of a pseudogene (“ψatpA c”) that is homologous to chloroplast atpA, which encodes the α subunit of chloroplast F1 ATPase. The rye CSU is somewhat smaller (2,855 base pairs) but contains much the same genic and other sequence elements as its wheat counterpart, except that two of the three t-elements as well as ψatpA c are found in only one of the three downstream flanks of the 18S/5S repeat, outside the CSU boundaries. In interpreting the seuuence data in terms of the evolutionary history of the 18S/5S-repeat family of wheat and rye, we conclude that (1) the wheat-rye form of the 18S/5S repeat most likely originated between 3 and 14 million years ago, in a lineage that gave rise to wheat and rye but not to barley, oasts, rice or maize; (2) the close linkage (1-bp apart) between trnfM and rrn18 is similarly limited in its taxonomic distribution to the wheat/rye lineage; (3) the trnfM-rrn18 pair arose via a single mutation that inserted a sequence block containing trnfM immediately upstream of rrn18; and (4) the presence of a putative promoter upstream of rrn18 in all wheat and rye repeats is consistent with all three repeat copies being transcriptionally active. We discuss these conclusions in the light of the possible functional significance of recombining-repeats in plant mitochondrial genomes.

Mexi-Pak-65) in a field experiment. The dry matter and N yields were significantly increased with fertilizer N application compared to those from unfertilized soil. The wheat crop used 33.6–51.5 and 30.5–40.9% of the N from ammonium nitrate and urea, respectively. Splitting the fertilizer N application had a significant effect on the uptake of fertilizer N by the wheat. The fertilizer N uptake showed that ammonium nitrate was a more available source of N for wheat than urea. The effective use of fertilizer N (ratio of fertilizer N in grain to fertilizer N in whole plant) was statistically similar for the two N fertilizers. The application of fertilizer N increased the uptake of unlabelled soil N by wheat, a result attributed to a positive added N interaction, which varied according to the fertilizer N split; six split applications gave the highest added N interaction compared to a single application or two split applications for both fertilizers. Ammonium nitrate gave 90.5, 33.5, and 48.5% more added N interaction than urea with one, two, and six split N applications. A values were not significantly correlated with the added N interaction (r=0.557). The observed added N interaction may have been the result of pool substitution, whereby added labelled fertilizer N replaced unlabelled soil N.

    Notes: Summary The effects of Spray Seed (diquat + paraquat), Roundup (glyphosate), Banvel-D (dicamba), Treflan (trifluralin), Glean (chlorsulfuron) and Dacthal (chlorthal dimethyl) at concentrations of 0–500 ppm product on the vegetative growth, vigour and pathogenicity of Gaeumannomyces graminis var. tritici (Ggt) on wheat were examined. All herbicides with the exception of dicamba and chlorsulfuron inhibited fungal growth on potato dextrose agar (PDA) at concentrations 10–500-fold of rates recommended for use in the field. The vegetative growth of the pathogen growing out of straw colonized on PDA supplemented with 100 ppm diquat + paraquat or glyphosate was reduced by 47.4% and 42.4%, respectively. When portions of these colonies were subcultured onto unamended PDA, their growth and the pathogenicity of straw pieces colonized by these subcultures were found to be unaltered. Straw colonized by Ggt on agar amended with concentrations of diquat + paraquat or at all concentrations of glyphosate produced less root disease in wheat seedlings in comparison to those colonized on unamended agar. It is proposed that the reduced pathogenicity of inocula prepared on agar amended with these two herbicides is due to poor colonization by the pathogen of straw on these media, and that a similar effect on saprophytic colonization in the field could lead to a reduction in the field inocula of the pathogen.

    Notes: Summary The use of vertical biopores by wheat (Triticum aestivum) seminal roots for easy access to the subsoil and the consequences for plant water supply and yield has been investigated by computer simulation. Parameters included were: biopore density and diameter, depth of cultivation and strength of the subsoil — all under a wide range of seasonal weather conditions. The model predicts that biopores add significantly to root penetration at depth, even at a density of 0.1% v/v of small, vertical pores, while 1.5% to 2.0% v/v can ensure maximum root penetration. When the growing season is shorter a larger number of biopores is needed to ensure timely root penetration to depth. With shallow tillage, biopores occur closer to the soil surface, and their importance is increased. Deeper root penetration invariably gives greater water uptake and transpiration, but may have a negative effect on grain yield, especially under the driest climatic conditions. An increase in early water use may result in less soil water being available during the grain-filling period. The effect of biopores on plant transpiration varies from year to year, depending on the amount of rain and its distribution in time, and on the amount of soil water stored at time of sowing.

    Notes: Abstract Alpine lakes in siliceous catchments of Tyrol and Carinthia (Austria) show signs of acidification. About 9% of the studied lakes have no alkalinity, more than 20% are below pH 6. About two thirds of all lakes have acid neutralizing capacities below 100 μeq 1−1. In spite of moderate precipitation acidity, some lakes show considerable concentrations of dissolved reactive aluminum during or shortly after snowmelt. High altitude lakes of the Alps are definitely more acidic than high mountain lakes in remote areas. Large differences in water and soil chemistry of nearby situated lakes were attributed to heterogeneities of bedrock geology. Paleolimnological investigations on former pH values of five lakes, based on diatom assemblages in the sediment, showed different developments: recent and past acidification, stable conditions, and alkalinization.

    Notes: Summary Combining ability studies for grain yield and its primary component traits in diallel crosses involving seven diverse wheat cultivars of bread wheat (Triticum aestivum L.) over generations F1-F5 are reported. The general and specific combining ability variances were significant in all generations for all the traits except specific combining ability variance for number of spikes per plant in the F5. The ratio of general to specific combining ability variances was significant for all the traits except grain yield in all the generations. This indicated an equal role of additive and non-additive gene effects in the inheritance of grain yield, and the predominance of the former for its component traits. The presence of significant specific combining ability variances in even the advanced generations may be the result of an additive x additive type of epistasis or evolutionary divergence among progenies in the same parental array. The relative breeding values of the parental varieties, as indicated by their general combining ability effects, did not vary much over the generations. The cheap and reliable procedure observed for making the choice of parents, selecting hybrids and predicting advanced generation (F5) bulk hybrid performance was the determination of breeding values of the parents on the relative performance of their F2 progeny bulks.

    Notes: Summary In this first analysis the protein patterns obtained by two-dimensional gel electrophoresis of 8 day-old leaves from 18 alloplasmic wheat lines are compared. From 440 spots retained on the basis of their reproducibility, 36 proteins were observed to vary in different cytoplasms, allowing us to distinguish the T. aestivum cytoplasm from 5 Aegilops cytoplasms. Twenty-four of the 36 variable proteins could be structurally related to the large subunit of RuBPCase. Nuclear variation between 3 wheat varieties was observed for 14 proteins.

    Notes: Summary Intergeneric hybrids of Triticum aestivum (2n=42,AABBDD) with Agropyron ciliare (2n= 28,SSYY), A. trachycaulum (2n=28,SSHH), A. yezoense (2n=28,SSYY) and A. scirpeum (2n=28) are reported for the first time. F1 hybrids of T. aestivum were also produced with A. intermedium (2n=42,E1E1E2E2Z1Z1) and A. junceum (2n=14,JuJu). All wheat-Agropyron hybrids were obtained by embryo rescue technique. Cultivars and reciprocal crosses differed for seed set, seed development and F1 plant production. The F1 hybrids were sterile. Attempts to obtain amphiploids were unsuccessful. However, backcross derivatives were obtained with wheat as the recurrent parent. The level of chromosome pairing in A. trachycaulum x wheat, A. yezoense x wheat and wheat x A. junceum hybrids provided no evidence of homologous or homoeologous pairing. Mean pairing frequencies in A. ciliare x wheat, wheat x A. scirpeum and wheat x A. intermedium hybrids indicated homoeologous or autosyndetic pairing. Ph gene was more effective in regulating homoeologous pairing in A. yezoense x wheat hybrids than in A. ciliare x wheat hybrid. Chromosome pairing data of BC1 derivatives indicated that either some of the wheat chromosomes were eliminated or Agropyron chromosomes caused reduced pairing of wheat homologues.

    Notes: Summary Each of the three genomes in hexaploid wheat controls the expression of a specific lectin in the embryo. The chromosomes which control their synthesis were determined using nullisomic-tetrasomic and inter-varietal chromosome substitution lines of ‘Chinese Spring’. All three wheat lectins were shown to be controlled by the homoeologous group 1 chromosomes. Using ditelosomic lines of ‘Chinese Spring’ the lectin genes could be localized on the long arms of chromosomes 1A and 1D. Inter-specific addition and substitution lines of Aegilops umbellulata chromosomes to ‘Chinese Spring’ indicated that chromosome 1U, which is homoeologous to the group 1 chromosomes of wheat, controls lectin synthesis.

    Notes: Summary Inheritance of gliadin components in winter wheat has been studied by one-dimensional polyacrylamide gel electrophoresis. Single F2 grains from 36 intervarietal hybrid combinations have been analysed. The genetic analysis has revealed blocks, including 1–6 gliadin components, which are inherited as individual mendelian traits. About 80 variants of blocks have been detected. On the basis of the allelism test they are grouped into 6 series in accordance with the number of known gliadin-coding loci located on chromosomes of the homoeologous groups 1 and 6. Each series includes 8–18 blocks controlled by different alleles of one gliadin-coding locus. Blocks of components have been confirmed to be inherited codominantly in accordance to the gene dose in the triploid endosperm. The highest similarity between members of one series is observed in groups of blocks controlled by chromosomes ID and 6D. On the contrary, many blocks controlled by chromosomes 1A and 1B have no bands in common. The presented catalogue of blocks of components may be used to make up gliadin genetic formulae and to compare electrophoregrams obtained by different authors. Blocks of gliadin components are suitable genetic markers for use in revealing and studying heterogeneity of wheat varieties, in tracing their origin, in identifying recombinations, translocations and substitutions of the genetic material and in solving many other problems of the origin, evolution and selection of hexaploid wheat.

    Notes: Summary Blocks of gliadin components found both in a number of varieties and in single F2 grains of winter wheat intervarietal hybrids have been studied by two-dimensional electrophoresis combining electrophoresis in acidic aluminium-lactate buffer (pH3.1) and SDS-electrophoresis. Gliadin components (spots) have been shown to be inherited as linked groups (blocks), codominantly and in accordance with a gene dosage in triploid endosperm. Blocks include components differing in their electrophoretic mobility and molecular weight. Some allelic variants of blocks differ only in presence of few additional components or in the electrophoretic mobility of components with similar molecular weights; other variants may contain no similar components. Apparently, in the course of evolution, mutations in individual genes of gliadin-coding loci and processes changing the number of expressing genes and the sizes of their structural part occurred.

    Notes: Summary Restriction enzyme digestion of the ribosomal RNA genes of the nucleolar organisers of wheat has revealed fragment length polymorphisms for the nucleolar organiser on chromosome 1B and the nucleolar organiser on 6B. Variation between genotypes for these regions has also been demonstrated. This variation has been exploited to determine the recombination frequency between the physically defined nucleolar organiser on 1B (designatedNor1) and other markers; two loci,Glu-B1 andGli-B1 which code for endosperm storage proteins andRf3, a locus restoring fertility to male sterility conditioned byT. timopheevi cytoplasm.Gli-B1 andRf3 were located on the short-arm satellite but recombine with the nucleolar organiser giving a gene order ofNor1 — Rf3 — Gli-B1. Glu-B1 is located on the long arm of 1B but shows relatively little recombination withNor1, which is, in physical distance, distal on the short arm. This illustrates the discrepancy between map distance and physical distance on wheat chromosomes due to the distal localisation of chiasmata. The recombination betweenNor1 andRf3 indicates that, contrary to previous suggestions, fertility restoration is not a property of the nucleolar organiser but of a separate locus.
