    Description: Nano/micro-size particles are widely applied in various fields. Among the various particles, silver particles are considered among the most prominent nanomaterials in the biomedical and industrial sectors because of their favorable physical, chemical, and biological characteristics. Thus, numerous studies have been conducted to evaluate their properties and utilize them in various applications, such as diagnostics, anti-bacterial and anti-cancer therapeutics, and optoelectronics. The properties of silver particles are strongly influenced by their size, morphological shape, and surface characteristics, which can be modified by diverse synthetic methods, reducing agents, and stabilizers. This Special Issue provides a range of original contributions detailing the synthesis, modification, properties, and applications of silver materials. Nine outstanding papers describing examples of the most recent advances in silver nano/microparticles are included. Silver nano/micro-size particles have many potential advantages as next-generation materials in various areas, including nanomedicine.     Description: Tissue engineering and regenerative medicine is a rapidly evolving research field which effectively combines stem cells and biologic scaffolds in order to replace damaged tissues. Biologic scaffolds can be produced through the removal of resident cellular populations using several tissue engineering approaches, such as the decellularization method. Indeed, the decellularization method aims to develop a cell-free biologic scaffold while keeping the extracellular matrix (ECM) intact. Furthermore, biologic scaffolds have been investigated for their in vitro potential for whole organ development. Currently, clinical products composed of decellularized matrices, such as pericardium, urinary bladder, small intestine, heart valves, nerve conduits, trachea, and vessels, are being evaluated for use in human clinical trials. Tissue engineering strategies require the interaction of biologic scaffolds with cellular populations. Among them, stem cells are characterized by unlimited cell division, self-renewal, and differentiation potential, distinguishing themselves as a frontline source for the repopulation of decellularized matrices and scaffolds. Under this scheme, stem cells can be isolated from patients, expanded under good manufacturing practices (GMPs), used for the repopulation of biologic scaffolds and, finally, returned to the patient. The interaction between scaffolds and stem cells is thought to be crucial for their infiltration, adhesion, and differentiation into specific cell types. In addition, biomedical devices such as bioreactors contribute to the uniform repopulation of scaffolds. Until now, remarkable efforts have been made by the scientific society in order to establish the proper repopulation conditions of decellularized matrices and scaffolds. However, parameters such as stem cell number, in vitro cultivation conditions, and specific growth media composition need further evaluation. The ultimate goal is the development of “artificial” tissues similar to native ones, which is achieved by properly combining stem cells and biologic scaffolds and thus bringing them one step closer to personalized medicine. The original research articles and comprehensive reviews in this Special Issue deal with the use of stem cells and biologic scaffolds that utilize state-of-the-art tissue engineering and regenerative medicine approaches.

    Description: Nowadays, polymer self-assembly has become extremely attractive for both biological (drug delivery, tissue engineering, scaffolds) and non-biological (packaging, semiconductors) applications. In nature, a number of key biological processes are driven by polymer self-assembly, for instance protein folding.     Description: Dear Colleagues, Fused filament fabrication, also known as 3D printing, is extensively used to produce prototypes for applications in, e.g., the aerospace, medical, and automotive industries. In this process, a thermoplastic polymer is fed into a liquefier that extrudes a filament while moving in successive X–Y planes along the Z direction to fabricate a 3D part in a layer-by-layer process. Due to the progressive advances of this process in industry, the application of polymeric (or even composite) materials have received much attention. Researchers and industries now engage in 3D printing by implementing numerous polymeric materials in their domain. In this Special Issue, we will present a collection of recent and novel works regarding the application of polymers in 3D printing.

    Description: Tumor cells commonly exhibit dependence on a single activated oncogenic pathway or protein to maintain their malignant proliferation and survival, a phenomenon called “oncogene addiction”. According to this concept, protein kinases have been identified as promising molecular targets for cancer therapy. There are several possibilities for targeting these proteins in cancer, including monoclonal antibodies, compounds able to favor the proteolytic degradation of the kinase, small-molecule protein kinase inhibitors (PKIs). Moreover, new anticancer treatments have increasingly been developed focusing on tumor suppressor genes and RNA interference. Despite promising results in cancer treatment with targeted cancer drugs, clinical experience has shown that only a fraction of patients respond to targeted therapies, even if their tumor expresses the altered target. This is known as primary resistance. Otherwise, secondary or acquired resistance to the treatment arises, almost invariably, when tumors are treated with cancer drugs. We set out to select some studies containing emerging developments on the subject. In essence, this collection aims to highlight some recent findings regarding resistance mechanisms and reviews of molecular targeting and resistance with 14 contributions, including 10 original research papers and 4 reviews. Aspects relating to solid cancers, such as breast, ovary, colon, and blood cancers such as leukemia, and the identification of resistance mechanisms and new molecular targets, help to create the basis for the preclinical and clinical development of more effective next-generation drugs.

    Description: Acceptance or rejection of implanted biomaterials is strongly dependent on an appropriate bio-interface between the biomaterial and its surrounding tissue. Given the fact that most bulk materials only provide mechanical stability for the implant and may not interact with tissues and fluids in vivo, surface modification and engineering of biomaterials plays a significant role towards addressing major clinical unmet challenges. Increasing data showed that altering surface properties including physiochemical, topographical, and mechanical characteristics, is a promising approach to tackle these problems. Surface engineering of biomaterials could influence the subsequent tissue and cellular events such as protein adsorption, cellular recolonization, adhesion, proliferation, migration, and the inflammatory response. Moreover, it could be based on mimicking the complex cell structure and environment or hierarchical nature of the bone. In this case, the design of nano/micrometer patterns and morphologies with control over their properties has been receiving the attention of biomaterial scientists due to the promising results for the relevant biomedical applications. This Special Issue presents original research papers that report on the current state-of-the-art in surface engineering of biomaterials, particularly implants and biomedical devices.

    Description: The innate immune system is the first line of defense against bacterial and viral infections and sterile inflammation through the recognition of pathogen-associated molecular patterns (PAMPs) by pathogen recognition receptors (PRRs) resulting in the production of proinflammatory and antiviral cytokines and chemokines. Several damage-associated molecular patterns (DAMPs), which were released by passive or active mechanisms under sterile conditions, are additionally recognized by PRRs and can cause or even aggravate the inflammatory response. In this special issue many aspects of innate immunity are summarized. Mechanisms of different DAMPs to induce pro- and anti-inflammatory activities, functions of different immune cells, as well as the crosstalk between coagulation and innate immunity were described. Furthermore, aspects of autoinflammatory diseases, types of programmed cell death pathways, and insect immunity are covered. Finally, therapeutic options for the treatment of diseases related to autoimmunity or infections are suggested. Overall, this special issue presents a broad overview of activities related to sterile inflammation and defense mechanisms of innate immunity.

    Description: This Special Issue gathers original studies (eight articles and one review) that aim to improve our knowledge concerning mycotoxin toxicity. The most recent research concerning the impact of zearalenone (ZEA) and the immune response is presented in a review that aims to explore the immunotoxicity produced by ZEA on different types of immune cells (phagocytes related to innate immunity and lymphocytes related to acquired immunity) as well as on immune organs, and also to identify the mechanism of action used by ZEA to modulate the immune response. The role of zearalenone in macrophage-mediated innate immunity after bacterial lipopolysaccharide (LPS) stimulation was the subject of another study, suggesting that the intake of ZEA-contaminated food might result in decreasing innate immunity. Another important study related to the co-exposure to mycotoxins and other food/feed contaminants showed that combining patulin with cadmium induces enhanced hepatotoxicity and nephrotoxicity both in vitro and in vivo, while the exposure to both deoxynivalenol and porcine circovirus has an additive effect on inflammatory cytokines by inducing the mRNA expression of interleukine-1 beta (IL-1β) and interleukine-6 (IL-6). The use of feed compounds rich in bioactive constituents for mitigating the mycotoxins effects in swine was the subject of other three studies herein, which have shown the efficacy of antioxidants from dietary grapeseed and sea buckthorn meal waste to decrease the toxicity of a diet contaminated with aflatoxin B1 or both aflatoxin B1 and ochratoxin A at the level of organs involved in the metabolism and excretion (liver and kidney), as well as at the level of secondary lymphoid organs (mesenteric lymph nodes). The use of different models (cell co-culture model, rat model) are recommended by another two studies in order to evaluate the ochratoxin A toxicity or for better prediction of the mycotoxins risk for human cancer.

    Description: Biopharmaceutical and pharmaceutical manufacturing are strongly influenced by the process analytical technology initiative (PAT) and quality by design (QbD) methodologies, which are designed to enhance the understanding of more integrated processes. The major aim of this effort can be summarized as developing a mechanistic understanding of a wide range of process steps, including the development of technologies to perform online measurements and real-time control and optimization. Furthermore, minimization of the number of empirical experiments and the model-assisted exploration of the process design space are targeted. Even if tremendous progress has been achieved so far, there is still work to be carried out in order to realize the full potential of the process systems engineering toolbox. Within this reprint, an overview of cutting-edge developments of process systems engineering for biopharmaceutical and pharmaceutical manufacturing processes is given, including model-based process design, Digital Twins, computer-aided process understanding, process development and optimization, and monitoring and control of bioprocesses. The biopharmaceutical processes addressed focus on the manufacturing of biopharmaceuticals, mainly by Chinese hamster ovary (CHO) cells, as well as adeno-associated virus production and generation of cell spheroids for cell therapies.

    Description: Mitochondria are the powerhouses of cells; however, mitochondrial dysfunction causes energy depletion and cell death in a variety of diseases. Altered oxidative phosphorylation and ion homeostasis are associated with ROS production resulting from the disassembly of respiratory supercomplexes and the disruption of electron transfer chains. In pathological conditions, the dysregulation of mitochondrial homeostasis promotes Ca2+ overload in the matrix and ROS accumulation, which induces the mitochondrial permeability transition pore formation responsible for mitochondrial morphological changes linked to membrane dynamics, and ultimately, cell death. Finally, studies on the impaired mitochondrial bioenergetics in pathology could provide molecular tools to counteract diseases associated with mitochondrial dysfunction.

    Description: Carotenoids are a group of approximately 600 compounds synthesized by photosynthetic organisms. These pigments are abundant in fruits and vegetables, as well as in certain animal products such as eggs and salmon, being responsible for their colorful appearance. The bioactive properties of certain carotenoids in human health are clear, as some of these compounds have antioxidant properties and serve as the only precursors of vitamin A in nature. The aim of this Special Issue entitled “Carotenoids and Human Health” is to provide the scientific community with an updated perspective of this exciting and growing research area.     Description: Tor mahseer (Tor tor), possess high commercial and recreational value as they are potential game as well as food fish of India. Two cell culture systems were developed from fin and heart of T. tor (Hamilton-Buchanan). The explants excised aseptically from fingerling of T. tor were cultured in Leibovitz-15 (L-15) medium with 20% fetal bovine serum (FBS). Radiation of cells started after 72 hours and 48 hours of explant attachment from caudal fin and heart respectively. Confluent monolayer of cells with heterogeneous morphology around fin explants was observed after 7-10 days, where as a homogenous confluent layer of fibroblastic cells from heart explant was observed after 12-13 days. The establishment of cell culture systems from different organs and tissues of commercial important species would facilitates in vitro research.
