Notes: Summary The use of 1,1′-[ethenylidenebis(sulfonyl)]bis-benzene as a pre-chromatographic reagent for LC analysis of thiol drugs is proposed. The reagent reacts rapidly (2 min) under mild conditions (pH 7.5, ambient) with thiol drugs and the resulting adducts can be chromatographed under reversed-phase conditions (C-18 column). Excess reagent can be removed from the reaction mixture by simple liquid-liquid extraction with chloroform. The HPLC method using UV detection was successfully applied to the analysis of reduced glutathione (GSH), N-acetylcysteine, N-2-mercaptopropionylglycine and captopril in their commercial formulations.

Notes: Summary A previously described method for determining more than one serine proteinase simultaneously by hydrophobic interaction chromatography of their complexes with aprotinin is inapplicable when other UV-absorbing species are co-eluted. The suitability of125I-labelled aprotinin has therefore been tested as a reagent in the analysis of mixtures containing trypsin, α-chymotrypsin and kallikrein taken as models, in the presence of ribonuclease and lysozyme. A new procedure is described which, without introducing changes in the chromatographic separation, allows direct determination of serine proteinases in terms of molarity. Results obtained in experiments with solutions containing from 0.20 to 30.00 nmol of each serine proteinase are reported.

Notes: Summary The use of 2-bromoacetyl-6-methoxynaphthalene as a fluorogenic labelling reagent in pre-column derivatization for the HPLC separation of biologically active carboxylic acids (fatty acids and bile acids) has been investigated. The compound reacts (30 min. at 70°C) with carboxylic acids to give fluorescent esters that can be separated by reversedphase HPLC and detected at λ ex. 300 nm, λ em. 460 nm. The experimental conditions for the derivatization and chromatographic separation are discussed. Applications to the determination of valproic acid and chenodeoxycholic acid in pharmaceutical formulations are described.

Notes: Summary A compound which becomes a liquid crystal when bonded to a polysiloxane was reacted with dimethylchlorosilane to produce a reagent suitable for bonding to a silica surface. After the silanization reaction was complete, the product was characterized by elemental analysis, diffuse reflectance Fourier transform Infrared Spectroscopy (DRIFT), CP/MAS carbon-13 NMR and differential scanning calorimetry (DSC). Preliminary chromatographic testing was done by retention index measurements using the alkylarylketone homologous series and by measuring the separation factor, α, for anthracene/phenanthrene at two different mobile phase compositions.

Notes: Summary The chromatographic technique as a means for experimental studies of diffusion in porous solids has been demonstrated. In this paper we report the effective diffusion coefficients which we have chromatographically obtained for two dextran samples and three low molecular weight solutes. The chromatographic method of measuring diffusional resistances in porous solids has the advantages of rapidity and simplicity. This method also provides information on the axial dispersion and distribution of a solute between the moving phase and the solvent held stationary in the pore. The elution curves of a packed column in pulse response experiments were analyzed by the method of moments to give the parameters of interest. The results indicate that pore diffusion of the solutes was appreciably restricted in comparison with diffusion in bulk solution. The degree of hindrance depended on the size of the diffusing substances.

Notes: Summary Reversed-phase high performance liquid chromatography with diode array detection was used to analyze phenolic compounds contained in bark extracts of two different poplar clones known to have different resistance toDiscosporium populeum infection. Statistical treatments both monovariate and multivariate were applied to the results in order to identify regularities useful to discriminate the polar resistance and to check the importance of seasonal and cultivation site related factors. For the two clones examined phenolic compounds showed great discrimination ability not influenced by either sample collection period or cultivation site.

Notes: Summary The pore structures of zirconium oxide particles prepared by two different methods (PICA and Oil Emulsion processes) are compared. Nitrogen sorptometry and size exclusion chromatography are used to characterize the two different types of particles. Significant and unexpected differences were found in the accessible pore volumes and the bed packing densities of the two materials. The PICA material was found to have a higher totally included and totally excluded volume than would be normally expected. The chromatographic method provides results which are more useful for the application of these materials to separation sciences.

Notes: Summary The energy of interaction (∈0) between the methylene group and the surface of a sorbent is proposed for use in the quantitative evaluation of the hydrophobicity of a sorbent. A method was developed to determine the ∈0 parameter from chromatographic measurements using the investigated sorbent as the stationary phase and linear methylene-containing homologous series, e.g., alkanols, amino-alkanes, etc., as the test compounds. The effectiveness of this method is demonstrated by using, as examples, a series of bonded stationary phases and a series of cross-linked copolymeric sorbents differing in the concentration of the hydrophobic groups. The proposed method is compared with the Martin rule used extensively in the chromatography of oligomers.
