    Notes: Polycrylamide gel electrophoresis of chicken lens proteins showed 17 crystallins, divided over three groups. Within each group physicochemical heterogeneity was combined with (partial) immunological homogeneity. It is assumed that more than one gene is involved in the synthesis of any crystallin species. During development of the chicken embryo, α-crystallin was first demonstrated by immunofluorescence in centrally located lens fibers at 3 days. At 8 days the epithelium became positive and the fibers lost some fluorescence. This continued until in 5-week-old chickens the lens core was negative. Lens placode cells showed immunofluorescence for δ-crystallin at 52 hours, mainly in their basal parts. The reaction gradually spread and at 3 days the entire lens was positive. From 8 days on the epithelium reacted progressively weaker, but the fibers remained positive. Five weeks after hatching, epithelium and cortex were negative, while the center still showed strong fluorescence. The behavior of β-crystallin was intermediate between that of the other two. Immunoelectrophoresis suggested a differential production onset for the components of each single crystallin type. Under normal conditions no crystallins were found outside the lens. Therefore, crystallin synthesis occurs after placode formation has taken place and must be restricted to the lens itself. Autoradiography after 3H-thymidine treatment indicated that all placode cells still replicate, though some already produce crystallins. A generation time of 8 to 10 hours was determined with an M phase of 30 minutes, an S phase of 6 hours, and a G2 of 2 ½ hours. During DNA synthesis the nuclei were located in the basal parts of the cells, and for mitosis they migrated to the lumen. Autoradiography after 3H-glucosamine application suggested that the placode cells take active part in the synthesis of the basement membrane interposed between lens rudiment and optic cup. This membrane later becomes the lens capsule, and in mice with the “shrivelled” gene, abnormal masses of anterior epithelial cells also clearly produce extra capsule material. This results in anterior polar cataracts. Several of the above findings are in disagreement with some of the current theories on the regulation of lens differentiation. No substitutes are presently offered, however.

    Notes: Inhibition of cellular respiration by treatment with the nonionic detergent Triton WR-1339 was found to be related to the cytotoxic response of cell to the surfactant. Respiration of sensitive cell lines (AV-3 and HeLa) markedly inhibited by Triton concentrations as low as 125 μgm/ml. Conditionally sensitive lines (BHK-21 and L-929) were affected by 500 μgm/ml while the respiration of insensitive cultures (primary rat and chick embryo cells) was unaffected by this concentration. Macrocyclon, a cyclic analogue of Triton, failed to alter the respiration rate of any of the above cell cultures.The levels of isocitric and succinic dehydrogenases in sensitive and conditionally sensitive cells were depressed within 2 hours after treatment with 500 μgm/ml of Triton was initiated and by 6 hours the activity was only 25% of the untreated controls. Similar results were obtained with mitochondrial preparations from these cells. Enzyme levels in insensitive cells were unaffected by Triton treatment.Mitochondrial damage was the most striking characteristic noted in treated cells examined by electron microscopy. The mitochondria were quite distorted and had lost most of their cristae formation. This mitochondrial damage was seen in all cell types examined although the rate at which it occurred varied. With sensitive cultures, damage was pronounced within 6 hours after the addition of Triton while mitochondria from conditionally sensitive cells were not grossly affected until 48 hours and they appeared to repair the damage following the removal of Triton.

    Notes: The enigmatic lophophoral organs of phoronids are demonstrated to by accessory sex glands. In the hermaphroditic Phoronis vancouverensis, each mature adult has two pairs of such glands: female nidamental glands that facilitate the temporary brooding of the embryos and male accessory spermatophoral organs that function in spermatophore elaboration. Functional males of Phoronopsis harmeri have complex accessory spermatophoral organs while females of this non-brooding dioecious species lack accessory sex glands.In both species, coelomic spermatozoa are accumulated within the nephridia and passed en masse into the partially enclosed spermatophoral organs. There each sperm mass is molded into a characteristic shape and encased in one or more membranes before release as a spermatophore into the ambient water. The structure of the spermatophore is characteristic for each species and is correlated with the functional morphology of the distinctive spermatophoral organs.The existence of spermatophores, the non-primitive nature of the spermatozoa, and additional features of reproductive biology dictate that fertilization is internal in these species, but attempts to determine the means of sperm transfer proved futile. Reproductive potential and behaviour of male Phoronopsis, means of gamete segregation in Phoronis, and general pattern of reproductive biology for both phoronids were also studied.

    Notes: The morphologic, functional, and biochemical changes produced by hematoporphyrin and light in human platelets have been characterized. by phase microscopy the cells appeared swollen and resembled signet rings; by electron microscopy they showed considerable loss of cytoplasm and their contour was smoother than normal. irradiated platelets were not aggregable by thrombin and calcium chloride, although they contained clottable protein, and were incapable of supporting clot retraction. a linear relationship was demonstrated between the per cent depletion of serotonin from irradiated platelets and the log dose of hematoporphyrin. the depletion of serotonin from these platelets was related lineraly to the log of time of exposure to light during the initial six minutes of exposure; but thereafter continued at a constant rate. the temperature of incubation influenced directly the rate of depletion of serotonin from irradiated platelets but did not influence the movement of serotonin into these platelets. atp was diminished considerably in irradiated platelets. these changes are attributable to damage to the membrane of the platelet by hematoporphyrin and light.These studies provide additional information about the blood platelet in terms of its response to photodynamic action.

    Notes: Human platelets, exposed to hematoporphyrin and light, were depleted of potassium and acid phosphatase. A linear relationship was demonstrated between the per cent depletion of potassium from irradiated platelets and the log dose of hematoporphyrin. The initial release of potassium and acid phosphatase from irradiated platelets was first order. Rate constants demonstrated that potassium was released five times faster than acid phosphatase. At 4°C, release of both potassium and acid phosphatase was the same as at 37°C. These results are compared to previous data on the release of serotonin from irradiated platelets.

    Notes: Actinomycin D was studied as a photodynamic agent and fluorochrome. With Venezuelan equine encephalitis virus propagated in chick fibroblasts, it was shown that actinomycin D induces both direct and indirect photoactivation of the virus. Some capacity of antibiotic-treated and irradiated virus to produce plaques larger than usual was not retained with passage of virus isolated from large plaques. Cells stained with actinomycin D exhibited a weak brownish fluorescence of nuclear structure.

    Notes: The cytological effects of 2 mM hydroxyurea upon Chinese hamster cells at various phases of the cell cycle were examined. Cells in the G1, G2, or M phases of the generation cycle treated with hydroxyurea showed no chromosomal aberrations. Cell treated in S phase became moribund and eventually lysed. Some of these moribund S cells reached mitosis much later and were found to have chromatid aberrations. Cells in the log phase of growth, surviving exposure to 2 mM hydroxyurea for six hours, also showed no aberrations. Thus, viable (colony-forming) cells, resulting from synchrony procedures with hydroxyurea are free of chromosomal aberrations.

    Notes: The gross anatomy of muscles, the topography of nerve tissues, and the histology of the pregenital abdominal glands of Nomia melanderi Ckll. are reported in detail. The movable and fixed points of muscle attachment were utilized in establishing a system of nomenclature for a typical abdominal segment. Names of nervules correspond to those of the tissues they innervate. The points of attachment of muscles of the fifth abdominal segment are essentially the same in both Nomia and Apis, except for the second tergo-sternal muscle which, in Nomia, has shifted its point of movable attachment to the membranous integument in front of the intersegmental membrane gland where it helps in relasing glandular secretion. The general plan of the nerves in the fifth abdominal segment in Nomia is more diffuse than in Apis, but there is no difficulty in establishing homology between the nervules of the two species. A pair of intersegmental stretch organs was found in abdominal segments 3-6. Glands of the sixth intersegmental membrane possess a reservoir with peripheral pouches both of which are absent in those of the fifth. Both types of glands have neither closing nor opening mechanisms, and neither is innervated. Release of glandular secretion is accomplished by the action of the tergo-sternal muscles.

    Notes: Each muscle of the third metasomal segment of the male of Nomia melanderi Ckll. is described in detail. The points of attachment of each muscle are compared with their homologs in other pregenital segments and with their homologs in the female. The function desgnated for each muscle describes its action alone or in conjunction with other muscle(s). New names are given to genital muscles by referring in the name to their points of attachment. Each intratergal muscle has homologous points of attachment in the pregenital segments of both sexes. The median tergo-dorsoplical muscle of the seventh segment and the oblique tergo-dorsoplical muscle of the eighth segment have changed their points of attachment. The intrasternal muscles are modified to suit the needs of courtship and mating, thus they are different from their homologs in the female. The spiracular muscles are well developed in all segments except the eighth, where the sterno-spiracular muscle is absent. The extrinsic genital muscles are derived from the intrasternal muscles of the eighth and ninth segments. The parameral and volsellar muscles are reduced in number. The aedeagal muscles, except the aedeago-phallic, have retained similar points of attachment to those found in primitive Hymenoptera. The topography of the metasomal nervous system is reported in detail by following each nerve and nervule to its termination. The study shows that (at least in Nomia) the criterion of nerve-concentration should not be used alone to indicate evolutionary levels. To accommodate the morphological changes in the terminal segments the Anterior and Posterior Lateral Nerves have migrated to new locations. The pattern of nerve topography (even at the nervule level) is homologous both in the different pregenital segments and between the sexes. The fact that homology does not exist between the external genitalia of the male and the modified ovipositor of the female supports the thesis that the male genital capsule is of phallic rather than prephallic origin. A pair of intersegmental membrane glands located between the seventh and eighth sterna is described. These glands may be the source of a pheromon responsible for gregariousness among “sleeping” males.

    Notes: The apparently inconsistent reports on flagellar ATPase properties may be resolved by elimination of adenylate kinase from the system. Removal of the adenylate kinase from alkaline M/2 KCl extracts of bull sperm flagella yields a spermosin-ATPase which liberates only the terminal phosphate of ATP. In any case spermosin is preferentially activated by calcium. However, combination of spermosin with flactin (e.g., by addition of digitonin and MgCl2 to the extraction medium) produces an ATPase much more highly activated by magnesium. But flactospermosin has so far resisted purification from its adenylate kinase contaminant. In divalent cation activation, pH optima and nature of ATP hydrolysis, the flagellar contractile protein system closely parallels the muscle system.
