    Notes: Summary High-pressure liquid chromatography (HPLC) was applied for separation and quantification of the concentration of aflatoxin in 500 samples of shelled unroasted whole peanut kernels.     Notes: Summary Phenolic acid derivatives of cinnamic acid are investigated by reversed-phase thin-layer chromatography, using silica gel silanised or impregnated with paraffin oil in saturated and sandwich tanks, as well as by HPLC using LiChrosorb RP-2 as adsorbent. The RM vs. % H2O relationships are linear in a wide concentration range. The results prove the possibility to correlate the results obtained by TLC in sandwich tanks with those obtained by HPLC. The chromatographic parameters of the investigated compounds are interpreted in terms of their chemical structure.

    Notes: Summary The performance of injection valves with both curtain- and single-flow column couplings has been evaluated using reverse-phase octadecylsilane columns and a standard test mixture. The results are compared with those obtained from septum and stopped-flow syringe injections. Stopped-flow valve injection has been examined similarly. Configurations and couplings which give optimum efficiency and reproducibility are recommended.

    Description / Table of Contents: Zusammenfassung Die Möglichkeiten zur Reversed-phase-HPLC von Metall-Dithiocarbamaten wurden systematisch untersucht. Außer den Diäthyldithiocarbamat-Chelaten wurden in die Untersuchungen die Tetramethylendithiocarbamate einbezogen. Als geeignet und voneinander trennbar erwiesen sich die Diäthyldithiocarbamate von Pb, Ni, Co, Cu and Hg sowie die Tetramethylendithiocarbamate von Cd, Pb, Ni, Co, Zn, Cu und Hg. Allen anderen Metallchelate dieser Gruppen zersetzen sich im chromatographischen System, ergeben unsymmetrische oder wegen zu geringer Retention nicht trennbare Banden. Die Ergebnisse aus der Reversed-phase-Chromatographie werden mit den bereits beschriebenen Trennmöglichkeiten mit der Adsorptions-Dünnschicht-Chromatographie und denen der Gas-Chromatographie verglichen und diskutiert.
    Notes: Summary The possibilities for reversed-phase HPLC of metal dithiocarbamates have been systematically examined. Beside the diethyldithiocarbamate chelates examined also included are the tetramethylenedithiocarbamates. The diethyldithiocarbamates of Pb, Ni, Co, Cu and Hg and the tetramethylenedithiocarbamates of Cd, Pb, Ni, Cl, Zn, Cu and Hg are suitable for this technique and have proved separable from one another. All other metal chelates of these groups are decomposed in the chromatographic system, give asymmetrical peaks or because of low retention unresolved bands. The results from the reversed-phase chromatography are discussed and compared with previously described separations using thinlayer adsorption chromatography and gas chromatography.

    Notes: Summary The retention behaviour of reference proteins on commercial siliceous size-exclusion supports was studied. Sorption was observed on both surface modified and unmodified supports. When sodium dodecylsulfate was added to the aqueous mobile phase, normal elution patterns were found. With this system, proteins, such as those isolate from different alfalfa genotypes, may be compared rapidly. Comparisons were facillitated by use of on-line central data processing capability.

    Notes: Summary Chemically bonded stationary phases (CBSP) were prepared by reaction of di- and trialkylchloro- and methoxy-silanes, R1R2SiCl2 and R1R2Si(OMe)2 with varying chain length of the ligands R1 and R2 with Lichrosorb Si 100, 10 μm, Merck. The influence of the structure of the CBSPs on the retention and selectivity in reversed phase chromatography (RPC) was investigated and compared with results obtained with monoalkyl-CPSPs. Plots of log k′ versus the percentage carbon yield straight lines. The slopes and intercepts are dependent on the structure of the ligands and the structure of the solute. An attempt was made to explain how structure of the CBSP influences the selectivity in RPC.

    Description / Table of Contents: Zusammenfassung Es wird der Einsatz von Säulenschaltungen in der Hochleistungs-Flüssigkeits-Chromatographie, speziell auf dem Gebiet der organischen Spurenanalytik, beschrieben. Außerdem wird die Möglichkeit eines einfachen Aufbaues solcher Schaltungen durch Kopplung eines Säulenschaltventils mit einem Mikroprozessor-gesteuerten Chromatographen aufgezeigt. Beispiele sollen die Leistungsfähigkeit dieser Methode zeigen.
    Notes: Summary The application of column-switching-techniques in high-performance liquid chromatography, especially in the field of organic trace analysis is shown. A possible construction of such systems with a high pressure multiport micro valve coupled to a microprocessor controlled chromatograph is demonstrated. With some examples the performance and effort of this method is documented.

    Notes: Summary The use of high-performance liquid chromatography in kinetics and metabolism studies is demonstrated. The following kinetic parameters are ascertained after single oral and intravenous administration of a drug: absorption by areal comparison, elimination constant in the β-phase, and clearance. Concentration levels in serum are measured after repeated oral administration on several days, with pharmacokinetic model fit to estimate limit concentrations in the steady state. Species-dependencies in metabolism are investigated in 24-h urines. The necessary characterization of drug and metabolite peaks is obtained by in-series connection of several detectors. The relative bioavailability of a controlled release form is determined by measuring drug concentrations in the blood.

    Description / Table of Contents: Zusammenfassung Aliphatische Di- und Polysulfide lassen sich im reverse-phase-Verfahren an einer chemisch gebundenen Octadecylphase (Micro-Pak-CH) mit MeOH-H2O als Eluens sauber trennen. Die qualitative Zuordnung der Peaks wird durch folgende Effekte vereinfacht: 1. log k′ nimmt bei konstanter S-Zahl linear mit der C-Zahl zu; 2. log k′ nimmt bei konstantem Alkylrest linear mit der S-Zahl zu; 3. log k′ fällt linear mit zunehmendem Gehalt des Eluens an MeOH. Bei Auswahl eines geeigneten Eluensgradienten können homologe Reihen wie R 2 S 2 mit R = Methyl bis R = Hexyl oder R 2 S x mit x = 2−7 mit einer Auflösung Res ∼ 2 zwischen allen Komponenten getrennt werden. Die Polysulfide werden bei diesen Trennungen nicht verändert.
    Notes: Abstract Aliphatic di- and polysulphides were separated by reverse-phase bonded-phase liquid chromatography. Chemically bonded octadecyl groups formed the stationary phase, mixtures of water and methanol were the mobile phase. For the qualitative analysis of polysulphide mixtures the following findings are important: 1. With constant S-number log k′ increases linearly with the C-number. 2. With constant hydrocarbon group log k′ increases linearly with the S-number. 3. log k′ decreases linearly with increasing methanol percentage in the eluent. With proper choice of gradient homologous series like R 2 S 2 with R = methyl to R = hexyl or R 2Sx with x = 2 ... 7 could be separated with a resolution of about 2 between all components. The polysulphides are not altered during these separations.
