    Notes: In order to further investigate the connection between transport and growth control, 3T3 cells, SV40 transformed 3T3 cells (SV101), and three revertant cell lines derived from SV101 which have regained certain manifestations of growth control were used. Transport rates of 2-aminoisobutyric acid and 3-0-methyl-D-glucose were measured in sparse, confluent, serum-starved, and serum-stimulated cultures.As shown before, cessation of 3T3 cell growth in G0 under conditions of confluence or serum deprivation was associated with reduced rates of transport for both compounds, whereas the density and serum dependence of growth and transport was largely eliminated in SV101. The density revertant F1SV101, which has regained density regulation of growth similar to 3T3 cells, has also regained density regulation of transport. Neither growth nor transport were serum dependent. The serum revertants AγSV7 and LsSV6 have regained both density and serum regulation of growth, but not according to the original mechanism of 3T3 cells of entry into a G0 state. Transport was high under conditions of confluence or serum deprivation. Thus for these cells rates of transport were not reduced simply as a consequence of slower cell growth nor were low transport rates responsible for growth arrest. The data are consistent with the possibility that growth arrest specifically in the G0 state could shut off a number of cellular activities, including transport.

    Notes: Rat thymocytes can be separated into two subpopulations by centrifugation for 20 minutes at 1,600 g in an 18/26/36% (w/v) discontinuous gradient of bovine serum albumin. Approximately 13% of the cells band at the 18/26% interface (light cells) while the remaining cells band at the 26/36% interface (heavy cells). In vitro and in vivo studies of 3H-thymidine incorporation indicate that the light cells are 2- to 3-fold enriched in the rapidly dividing lymphoblast subpopulation of thymocytes as compared to heavy cells. Light cells transport the non-metabolizable glucose analogue 3-O-methylglucose (3-MeGlc) approximately four times faster than heavy cells. The time course of 3-MeGlc uptake is biphasic for light, heavy and unfractionated thymocytes. While the half-times of the rapid (1 minute) and slow (20-45 minute) phases of uptake are similar for all three types of cells, the contributions of the rapid phase to total uptake are 50% for light cells, 20% for unfractionated thymocytes and 10% for heavy cells. The results show that 3-MeGlc transport activity differs markedly within certain thymocyte subpopulations. The correlation between the contributions of the rapid phase of uptake and the proportion of lymphoblasts in the thymocyte fractions suggests that the lymphoblast and small lymphocyte subpopulations might be responsible for the rapid and slow phase of 3-MeGlc uptake, respectively.

    Notes: Tryptophan transport has been examined in A9 and in mutants resistant to 5-fluorotryptophan (5-FT). Evidence indicates that in A9 cells two systems are present for tryptophan transport, which are analogous to the A and L systems found in Ehrlich ascites cells differing, however, in terms of amino acid specificity. Tryptophan uptake via the L system, a high affinity, low capacity system, is Na+ independent and occurs by a counter transport mechanism, while uptake via the A system, a low affinity, high capacity system, is Na+ dependent. Alanine, arginine, lysine, proline, asparagine, and aspartate (listed in order of decreasing inhibitory effect) inhibit tryptophan uptake via the A system from approximately 80-50% while having no inhibitory effect on the L system. In addition, glutamine which inhibits tryptophan uptake by 80% via the L system only inhibits to the extent of 20% via the A system. Previous kinetic studies of 5FT resistant clone FTr37 indicated system A was altered while the analysis of the effects of the mutation on system L was inconclusive. However, in these studies Na+ independent uptake was not altered in FTr 37 indicating system L was not affected. Amino acid competition studies confirmed this observation and suggested that a change in the specificity of system A had occurred in FTr 37. The amino acid competition studies in FTr 23, indicated that the specificities of both systems differed from A9. The possibility that this change may be due to a single mutational event is discussed.

    Notes: A substance promoting the growth of mammalian cells in vitro has been isolated from Bacto-peptone (Difco Labs., Detroit) and identified as 6,8-dihydroxypurine. This compound, which is isomeric with xanthine(2,6-dihydroxypurine), enhances the growth of one Chinese hamster and some other cell lines; xanthine itself is inactive. The biological action of the compound is discussed.

    Description: A rotor loads computer program was used to define the steady state and cyclic loads acting on 60 ft long metal blades designed for the ERDA/NASA 100 kW wind turbine. Blade load and stress analysis used to support the structural design are presented. For the loading conditions examined, the metal blades are structurally adequate for use, within the normal operating range, as part of the wind turbine system.

    Description: The results of dynamic analyses, weight and balance tests, static stiffness tests, and structural vibration tests on the 60-foot-long metal blades for the ERDA-NASA 100-kW wind turbine are presented. The metal blades are shown to be free from structural or dynamic resonance at the wind turbine design speed. Aeroelastic instabilities are unlikely to occur within the normal operating range of the wind turbine.

    Description: The implementation of a general three dimensional hydroelastic mode analysis capability in NASTRAN is presented. Finite elements with polyhedral shapes define the fluid; existing NASTRAN plate elements define the fluid/structure interface. Efficient solution methods were implemented to allow a separate structural matrix reduction and to allow connection of the fluid mass directly to a small set of grid points or modal coordinates representing the structure. Test case results for the various solution options are presented.

    Description: A biaxial method for performing inplane shear tests of materials using a shear frame is described. Aluminum plate and sandwich specimens were used to characterize the uniformity of shear strain imparted by the biaxial method of loading as opposed to the uniaxial method. The inplane stiffening effect of aluminum honeycomb core was determined. Test results for (+ or - 45) graphite-epoxy laminate are presented. Some theoretical considerations of subjecting an anisotropic material to a uniform shear deformation are discussed.

    Notes: When cervical segments 14 to 15 of the chicken spinal cord are cut transversely and studied by routine histological and histochemical methods, an onion-shaped region, filled with thread-like fibers, if seen to surround the ependymal cells of the central canal and to be bounded laterally by the neural elements of the spinal gray matter. This area is negative for succinic dehydrogenase, beta-hydroxybutyrate dehydrogenase and cholinesterase activity, but very strongly periodic acid-Schiff positive. Diastase controls show the positive material to be glycogen. Parasagittal sections through this cervical region and into the upper thoracic cord, show the glycogen-rich region to extend longitudinally throughout the region. Because of its location and histochemical characterization, which, He similar to that of the ventral portion of the glycogen body, the term brachial glycogen budy is proposed for this structure.
