    Description / Table of Contents: Résumé Une concentration moyenne de 2.4×10−8 g U/g de cendre a été obtenue à partir de l'os humain normal. La microdistribution de l'uranium dans l'os indique que cet élément est surtout limité à surface de l'endoste et, en particulier, aux surfaces de l'os lamellaire et aux parois des canaux de Havers, ouverts dans l'os corticol. Cette répartition suggère que l'uranium se présente sous une forme chimique impropre à son incorporation dans l'apatite osseux: il ne semble donc pas exister une distribution diffuse significative de l'uranium dans l'os.
    Abstract: Zusammenfassung Eine mittlere Konzentration von 2,4×10−8 g Uran/g Asche wurde in normalen menschlichen Knochen gefunden. Die Feinverteilung von Uran im Knochen zeigt, daß dieses Element hauptsächlich an der endostalen Oberfläche vorkommt, insbesondere an der Oberfläche des trabeculären Knochens und an den Wänden der offenen Haversschen Kanäle im kortikalen Knochen. Diese Verteilung läßt vermuten, daß Uran in einer chemischen Form vorliegt, welche sich für den Einbau in das Knochenapatit nicht eignet. Daraus folgt, daß keine signifikante diffuse Verteilung des Urans innerhalb des Knochens vorliegt.
    Notes: Abstract A mean concentration of 2.4×10−8 g U/g ash has been obtained for normal human bone The microdistribution of uranium in bone indicates that this element is mainly restricted to endosteal surfaces; in particular the surfaces of trabecular bone and the walls of open Haversian canals in cortical bone. This distribution suggests that uranium is present in a chemical form that is not acceptable for incorporation into bone apatite and consequently there does not appear to be a significant diffuse distribution of uranium throughout bone.

    Description / Table of Contents: Résumé Une perte de substance ostéo-cartilagineuse, de taille limitée et identique, est réalisée chez le lapin adulte et la cicatrisation est étudiée histologiquement et par autoradiographie après marquagein vitro au35S-sulfate. Une analyse microchimique est pratiquée pour le contenu et la composition en glycosaminoglycanes. 1. Entre la première semaine et la 4ème et 8ème semaine, un tissu conjonctif non-métachromatique se différencie en un cartilage métachromatique et la quantité de sulfate de chondroitine augmente de façon significative aux dépens des glycoprotéines. 2. Jusqu'à la 4ème semaine, la perte de substance est surtout comblée par de l'os néoformé: après cette période, la région est comblée au delà de la limite de la surface articulaire. 3. Le cartilage hyalin, ressemblant morphologiquement, autoradiographiquement et chimiquement au cartilage articulaire, en ce qui concerne la distribution en glycosaminoglycanes, constitute la surface articulaire de la perte de substance comblée dans un tiers des cas après 8 semaines. Le cartilage hyalin s'observe surtout dans les régions où de l'os néoformé a comblé la cavité médullaire. 4. Dans les deux tiers des cas, après 8 semaines, les surfaces articulaires des zones comblées comportent, non seulement du cartilage, mais aussi du tissu fibreux se formant essentiellement sur les parties latérales et dans les régions, où la cavité médullaire, fliant face, à la surface articulaire, n'a pas été comblée par du tissue osseux. La fraction glycoprotéique augmente par rapport à la fraction chondroitine sulfate. 5. Dans la majorité des cas, après 20 semaines, le cartilage néoformé subit des phénomènes dégénératifs, qui se traduisent par une diminution en chondroitine sulfate.
    Abstract: Zusammenfassung Bei ausgewachsenen Kaninchen wurde ein begrenzter, standardisierter, osteochondraler Defekt hervorgerufen, und das regenerierte Gewebe wurde histologisch und autoradiographisch durch Markierung in vitro mit35S-Sulfat und durch mikrochemische Bestimmung des Gehaltes und der Zusammensetzung der Glykosaminglykane untersucht. Die wichtigsten Befunde waren: 1. Zwischen 1 und 4–8 Wochen veränderte sich nichtmetachromatisches Bindegewebe zu metachromatisch gefärbtem Knorpel, und der Anteil an Chondroitin-Sulfat nahm auf Kosten der Glykoproteine signifikant zu. 2. Bis zu 4 Wochen war der Hauptteil des defekten Gebietes mit neugebildetem Knochen gefüllt; nach dieser Zeit lag dieser Bezirk oberhalb der Verknöcherungsgrenze in Richtung der Gelenkoberfläche. 3. Nach 8 Wochen bestand die Gelenkoberfläche des defekten Gebietes in einem Drittel der Fälle aus hyalinem Knorpel, der morphologisch, autoradiographisch und chemisch dem Gelenkknorpel in Bezug auf die Verteilung von Glykosaminoglykanen glich. Hyaliner Knorpel wurde hauptsächlich an Stellen beobachtet, wo neugebildeter Knochen die Markhöhle geschlossen hatte. 3. Nach 8 Wochen bestand die Gelenkoberfläche des defekten Gebietes in einem Drittel der Fälle aus hyalinem Knorpel, der morphologisch, autoradiographisch und chemisch dem Gelenkknorpel in Bezug auf die Verteilung von Glykosaminoglykanen glich. Nach 8 Wochen bestanden Teile der Gelenkoberfläche des Defektes in zwei Dritteln der Fälle nicht nur aus Knorpel, sondern auch aus fibrösem Gewebe, welches vor allem in den seitlichen Teilen des Defektes und an Stellen vorlag, wo die Markhöhle gegenüber der Gelenkoberfläche nicht mit Knochengewebe verschlossen worden war. Die Glykoproteinfraktion nahm im Vergleich zur Chondroitin-Sulfatfraktion zu. 5. Nach 20 Wochen zeigten sich in den meisten Fällen bei neugebildetem Knorpel degenerative Veränderungen, welche durch eine gewisse Abnahme des Chondroitin-Sulfats wiedergegeben wurden.
    Notes: Abstract A limited, standardized osteochondral defect was created in adult rabbits and the regenerated tissue was examined histologically and autoradiographically after labellingin vitro with35S-sulphate, and microchemically for its content and composition of glycosaminoglycans. The principal findings were: 1. Between 1 week and 4 to 8 weeks, non-metachromatic connective tissue differentiated to metachromatically stained cartilage, and the proportion of the chondroitin sulphate increased significantly at the expense of the glycoproteins. 2. Up to 4 weeks, the major part of the defect area was filled with newly formed bone; after this time, the area lay above the level of the “tidemark”, towards the articular surface. 3. Hyaline cartilage with morphological, autoradiographic and chemical resemblance to the articular cartilage in terms of the distribution of glycosaminoglycans constituted the articular surface of the defect area in one-third of the cases at observation times after 8 weeks. Hyaline cartilage was observed mainly in areas where newly formed bone had closed the medullary cavity. 4. In two-thirds of the cases, after 8 weeks, parts of the articular surface of the defect consisted not only of cartilage but also of fibrous tissue, occurring mainly in the lateral parts of the defect and in areas where the medullary cavity facing the articular surface had not been sealed by bone tissue. The glycoprotein fraction increased relative to the chondroitin sulphate fraction. 5. In most cases after 20 weeks, newly-formed cartilage underwent degenerative changes, which were reflected in some reduction of the chondroitin sulphate.

    Description / Table of Contents: Résumé La cinétique de la formation et de la transformation des précipités de phosphate de calcium, obtenus en mélangeant de volumes égaux de solutions à 6×10−3 M de calcium total et/ou phosphate total est étudiée à 25°C. Les solutions de phosphate sont préajustées à un pH de 7.4. Les changements de pH et de turbidité des solutions sont suivis simultanément en fonction du temps. Les précipités sont isolés à des intervalles de temps variables et caractérisés par diverses méthodes physico-chimiques. Initialement un précipité avec un rapport molaire Ca/P de 1.5, amorphe aux rayons X et en diffraction électronique, est formé. Le spectre IR indique la présence de PO 4 3− et de HPO 4 2− . Après une période métastable, on observe la précipitation d'un matériel cristallin dans ou sur la phase amorphe. Vingt quatre heures après préparation de l'échantillon les précipités présentent surtout les caractères du phosphate octocalcique.
    Abstract: Zusammenfassung Die Kinetik der Bildung und Transformation von Calciumphosphat-Niederschlägen wurde bei 25°C untersucht. Es wurden dazu gleiche Volumen von Lösungen gemischt, bei einer Konzentration von 6×10−3M totales Calcium und/oder totales Phosphat. Die Phosphatlösungen wurden zuerst auf pH 7,4 eingestellt. Veränderungen des pH und Trübung der Lösungen wurden gleichzeitig als eine Funktion der Zeit aufgezeichnet. Niederschläge wurden in verschiedenen Zeitintervallen isoliert und mit verschiedenen physiko-chemischen Methoden charakterisiert. Am Anfang wurde ein Niederschlag mit einem molaren Ca/P-Verhältnis von 1,5, im Röntgenbild und in der Elektronendiffraktion amorph, gebildet. Infrarotspektren deuteten die Anwesenheit von PO 4 3− - und HPO 4 2− -Ionen an. Nach einer metastabilen Periode erfolgte ein Niederschlag aus kristallinem Material innerhalb oder auf der amorphen Substanz. 24 Std nach der Herstellung der Proben zeigten die Niederschläge in der Hauptsache die Charakteristiken von Octocalciumphosphat.
    Notes: Abstract The kinetics of the formation and transformation of calcium phosphate precipitates obtained by mixing equal volumes of solutions, 6×10−3 M in total calcium and/or total phosphate was investigated at 25°. The phosphate solutions were preadjusted to pH 7.4. Changes of the pH and turbidity of the solutions were followed simultaneously as a function of time. Precipitates were isolated at various time intervals and characterized by different physicochemical methods. Initially a precipitate with a molar Ca/P ratio of 1.5, amorphous to X-ray and electron diffraction was formed. IR spectra indicated the presence of PO 4 3− and HPO 4 2− ions. After a period of metastability, precipitation of a crystalline material within or upon the amorphous matter occurred. Twenty four hours after sample preparation the precipitates showed mainly the characteristics of octacalcium phosphate.

    Description / Table of Contents: Résumé Des diagrammes de phase d'équilibre ont été déterminés pour le système CaO-P2O5-H2O en utilisant des techniques de synthèse hydrothermique au cours de variatio nsde température allant de 300–600° et 2 Kb H2O de pression. De l'hydroxyapatite bien cristallisé a été synthétisé et caractérisé. De faibles variations de paramètres de la maille cristalline, liées à la température de synthèse et composition globale du matériel initial, ont été déterminées. Des conditions chimiques précises sont nécessaires pour obtenir de l'apatite, en tant que seule phase solide en équilibre dans la solution. Les résultats de diagramme de phase d'équilibre sont comparés avec ceux obtenus dans des milieux synthétiques.
    Abstract: Zusammenfassung Es wurden Gleichgewichts-Phasendiagramme für das System CaO-P2O5-H2O bestimmt, indem hydrothermale Synthese-Techniken im Temperaturbereich von 300–600° und bei einem Druck von 2 Kb H2O verwendet wurden. Es wurde gut-kristallisiertes Hydroxyapatit erzeugt und charakterisiert. Es wurden geringe Unterschiede in den Parametern der Zelleinheiten festgestellt, welche von der angewandten Temperatur und der Zusammensetzung des Startmaterials abhingen. Es waren genaue chemische Bedingungen nötig, um Apatit als die einzige feste Phase im Gleichgewicht mit der Lösung zu erhalten. Die Resultate der Gleichgewichts-Phasendiagramme werden mit früheren Untersuchungen mit der Synthesetechnik verglichen.
    Notes: Abstract Equilibrium phase diagrams have been determined for the system CaO-P2O5-H2 using hydrothermal synthesis techniques in the temperature range 300–600° and 2 Kb H2O pressure. Well-crystallized hydroxyapatite has been produced and characterized. Small variations in unit cell parameters dependent on temperature of synthesis and bulk composition of the starting materials have been determined. Precise chemical conditions were required to obtain apatite as the only solid phase in equilibrium with solution. Equilibrium phase diagram results are compared with previous synthetic investigations.

    Description / Table of Contents: Résumé Un échantillon minéral provenant de Burgess, Canada s'est révélé être un speciment exceptionnellement pur de fluoroapatite après analyse chimique et cristallographique. La composition globale de cet échantillon est la suivante: $$(Ca^2 )9.98(Sr^{2 + } ,Na^ + ,K^ + ,Mg^{2 + } )0.02(PO_4^{3 - } )5.98(HCO_3^ - ,CO_3^{2 - } )0.02(F^ - )2$$ . L'axe cristallographique C est de 6.865 A et l'axe a de 9.374 A. Des expériences d'échanges réalisés à l'aide de45Ca,32P et18F indiquent la présence de gros cristallites de surface spécifique de l'ordre de 1 m2/g. Il apparait que l'interprétation physique des processus d'échange ne nécessite pas l'existence de compartiments séparés, avec chacun son propre facteur cinétique, les échanges semblent être simplement liés à un changement exponentiel dans l'énergie libre de la réaction. Pour la réaction suivante: $$(Ca)_5 (PO_4 )_3 OH solide + (F^ - ) \rightleftarrows (Ca)_5 (PO_4 )_3 F solide + (OH^ - )$$ , la constante thermodynamique de 101.26 a été calculée, suggérant que le fluorapatite se forme toujours aux dépens de l'hydroxyapatite dans des conditions physiologiques. Cette transformation se continue en abaissant le pH.

Die Gesamtzusammensetzung entspricht: $$(Ca^{2 + } )_{9,98} (Sr^{2 + } ,Na^ + ,K^ + ,Mg^{2 + } )_{0,02} (PO_4^{3 - } )_{5,98} (HCO_3^ - ,CO_3^{2 - } )_{0,02} (F^ - )_2 $$ . Die kristallographische c-Achse wurde bestimmt und ergab 6,865 Å, und die a-Achse ergab 9,374 Å. Austauschwerte, welche durch Anwendung von45Ca,32P und18F erhalten wurden, deuteten auf große Kristalliten mit einer spezifischen Oberfläche von ca. 1 m2/g. Die Befunde deuten darauf hin, daß für die physikalische Erklärung des Austauschvorganges keine separaten Kompartimente mit eigenen kinetischen Faktoren nötig sind, sondern daß der Austausch mit dem exponentiellen Wechsel in der freien Energie der Reaktion in einfacher Beziehung steht. Für die Reaktion $$(Ca)_5 (PO_4 )_3 OH_{in fester Form} + (F^ - ) \rightleftarrows (Ca)_5 (PO_4 )_3 F_{in fester Form} + (OH^ - )$$ wurde als thermodynamische Konstante 101,26 errechnet, was darauf deutet, daß unter physiologischen Bedingungen immer Fluorapatit auf Kosten von Hydroxyapatit entsteht. Diese Umwandlung wird erhöht, wenn das pH erniedrigt wird.
    Notes: Abstract A mineral specimen from Burgess, Canada, proved upon chemical and crystallographic analyses to be an exceptionally pure sample of fluorapatite. The over-all composition corresponds to $$(Ca^{2 + } )_{9.98} (Sr^{2 + } ,Na^ + ,K^ + ,Mg^{2 + } )_{0.02} (PO_4^{3 - } )_{5.98} (HCO_3^ - ,CO_3^{2 - } )_{0.02} (F^ - )_2 $$ . The crystallographic c-axis was determined to be 6.865 Å, and the a-axis 9.374 A. Exchange data obtained by employing45Ca,32P, and18F indicate the presence of large crystallites with a specific surface of the order of 1 m2/g. It is indicated that the physical interpretation of the exchange process does not require the existence of separate departments, each with its own kinetic factor, but that the exchange may be simply related to the exponential change in the free energy of the reaction. For the reaction $$(Ca)_5 (PO_4 )_3 OH_{solid} + (F^ - ) \rightleftarrows (Ca)_5 (PO_4 )_3 F_{solid} + (OH^ - )$$ the thermodynamic constant has been calculated to be 101.26, implying that fluorapatite always will form at the expense of hydroxyapatite under physiologic conditions. This transformation will be furthered by lowering the pH.

    Description / Table of Contents: Résumé Au cours de la phase inhabituelle de mue d'un isopode d'eau courante,Lirceuts brachyrus, la moitié postérieure de l'exosquelette est éliminée 24 heures avant la moitié antérieure. A ce stade, une reminéralisation se développe dans la partie postérieure alors que la partie antérieure est dans un stade de pré-mue. Le pourcentage de différence en calcium dans les deux moitiés à mi-mue et mue complète est respectivement de 22% (p〈0.01) et 33% (p〈0.01), indiquant une complexation du calcium pendant la mue. La rapidité de la reminéralisation est illustrée par le fait que le contenu minéral total double dans la partie postérieure entre la mi-mue et la mue totale et dans la partie antérieure entre la fin de la mue et un jour après. Le carbonate de calcium, sous forme de calcite, a pu être identifié par diffraction électronique de coupes fines des téguments.
    Abstract: Zusammenfassung Während der ungewöhnlichen Häutungssequenz des Frischwasser-IsopodenLirceus brachyurus (Harger) wird die hintere Hälfte des äußeren Skeletts 24 Std vor der vorderen Hälfte abgestoßen. In der halbgehäuteten Phase erfolgt Remineralisation im hinteren Teil, während der vordere Teil in einem Vorhäutungszustand ist. Der prozentuale Unterschied des Calciums in den zwei Hälften bei Halb- und Vollhäutungszustand ist 22% (p〈0,01) bzw. 33% (p〈0,01), was andeutet, daß Calcium während der Häutung abgesondert wird. Die Geschwindigkeit der Remineralisation erhellt aus der Tatsache, daß sich der Gesamtmineralgehalt im hinteren Teil zwischen Halt- und Vollhäutung, in der vorderen Hälfte jedoch zwischen Endhäutung und einem Tag nach der Häutung verdoppelt. Calciumcarbonat in kristalliner Calcitform wurde mittels Elektronendiffraktion von dünnen Hautschnitten nachgewiesen.
    Notes: Abstract During the unusual molt sequence of the fresh-water isopod,Lirceus brachyrus (Harger), the posterior half of the exoskeleton is shed 24 hours before the anterior half. At the half-molt stage, occurs in the posterior part while the anterior portion is in a pre-molt condition. The percentage difference in calcium in the two halves at half-molt and full-molt is22 (p〈0.01) and33 (p〈0.01) respectively, an indication that calcium is sequestered during The rapidity of remineralization is illustrated by the fact that the total mineral content doubles in the posterior part between half and full molt and in the anterior half between the end of molt and one day after ecdysis. Calcium carbonate in the calcite cystalline form was demonstrated by electron diffraction of thin sections of the integument.

    Notes: Abstract The effects of disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) on bone and serum chemistry were investigated in adult rabbits. EHDP was administered by subcutaneous injection at doses of 0.25, 2.5 and 10 mg/kg body weight/day for of 28 days. Blood samples were obtained weekly from each rabbit and serum levels of total calcium, ionized calcium, inorganic phosphate and alkaline phosphatase were determined. At the end of the treatment period all rabbits were sacrificed and the tibiae removed for chemical analysis and histological evaluation. The effect of EHDP administration on serum chemistry was both dose- and time-related. The highest of the three doses, 10 mg/kg/day, resulted in a time-related decrease in total serum calcium. This dose also caused a rapid but transient reduction in serum ionized calcium. The effect of EHDP on serum inorganic phosphate was biphasic. Administration of 2.5 mg/kg/day resulted in a time-related elevation in this parameter, whereas the 10 mg/kg/day dose resulted in a time-related hypophosphatemic response. There were no significant drug-related changes in tibial fat-free dry weight, ash weight, total calcium or total phosphorus values. However, administration of 2.5 and 10 mg/kg/day EHDP resulted in increased osteoid tissue as measured histologically. These results are compared with data from other EHDP studies, and discussed in relation to the maturity and growth-state of the experimental animals.

    Description / Table of Contents: Résumé Des composants fluorescents de l'os et la dentine sont séparés des hydrolysats alcalins de leur marice sur des colonnes Sephadex C25 CM d'échange cationique. Les concentrations en fluorescence et le spectre d'excitation (λ max 330 nm) et d'émission (λ max 395 nm) sont les mêmes que ceux observés au niveau des matrices intactes et gélatinisées. Les paramètres de fluorescence ne sont pas altérés par hydrolyse. La filtration sur gel à l'aide de colonnes Sephadex G 10 perment de différencier le matériel isolé en deux composants, ayant la même fluorescence et la même absorption UV. La fluorescence est indépendante de pH de 3.5–9.5. Des études de dialyse et de filtration sur gel de matrices gélatinisées indiquent une association étroite du matériel fluorescent avec les chaines polypeptidiques de collagène.
    Abstract: Zusammenfassung Fluorescierende Bestandteile aus Knochen und Dentin wurden in Sephadex C25 CM Kationen-Austauschersäulen von alkalischen Hydrolysaten ihrer Matrices getrennt. Die Fluorescenzintensitäten sowie die Erregungs- (λ max 330 nm) und Emissions- (λ max 395 nm) Spektren waren dieselben wie bei intakten und gelatinisierten Matrices. Die Fluorescenzparameter wurden durch die Hydrolyse nicht verändert. Eine Gelfiltration über Sephadex-G10-Säulen trennte das isolierte Material in 2 Komponenten auf, welche gleiche Fluorescenz- und UV-Absorptionseigenschaften zeigten. Im pH-Bereich zwischen 3,5 und 9,5 war die Fluorescenz unabhängig vom pH. Dialysierversuche sowie Gelfiltrationsexperimente mitden gelatinisierten Matrices zeigten eine starkgefügte Bindung des fluorescierenden Materials mit den Polypeptidketten des Kollagens.
    Notes: Abstract Fluorescent components in bone and dentine were separated from alkaline hydrolysates of their matrices on Sephadex C25 CM cationic exchange columns. The fluorescence levels, and the excitation (λ max 330 nm) and emission (λ max 395 nm) spectra, were the same as those observed in the intact and gelatinised matrices. The fluorescence parameters were unaltered by the hydrolysis procedure. Gel filtration on Sephadex G. 10 columns further resolved the isolated material into two components with the same fluorescence and UV absorption properties. The fluorescence was independent of pH over the range 3.5–9.5. Dialysis and gel filtration studies on the gelatinised matrices indicated a firmly-bonded association of the fluorescent material with the collagen polypeptide chains.

A pH 7.40, de l'ACP frais a un rapport molaire Ca/P de 1.36±0.02 et contient 22.8 (±2.2)% HPO 4 2− . Les résultats obtenus avec du précipité non lavé ne peuvent s'expliquer par du Ca2+ emprisonné et de l'HPO 4 2− ou du Na+, Cl− et CO 3 2− exogènes. Les phosphates de calcium amorphes constituent une classe de sels ayant des caractères chimiques variables et des propriétés physiques identiques, comparables au verre. Le CaHPO4·xH2O non cristallin peut être un ACP, surtout au cours des phases précoces de formation. A des pH physiologiques, l'ACP se transforme en petits cristaux applatis contenant de fortes quantités de phosphate acide facilement remplaçable. Le fait de laver la couche de surface produit un changement chimique dans les nouveaux cristaux: des cristaux non lavés donnent des diagrammes de diffraction d'apatite peu cristallins, ainsi que des spectres infra-rouges peu nets, intermédiaires entre des apatites et du phosphate octocalcique. Des explications structurales sont proposées et les compositions minérales amorphe/cristalline de l'os et du cartilage sont recalculées.
    Abstract: Zusammenfassung Ungewaschene Proben von amorphem Calciumphosphat (ACP) enthalten eine unersetzliche labile Fraktion, welche reich an saurem Phosphat ist und ein niederes Ca/P-Verhältnis hat und welche während des Waschprozesses unwiderruflich verloren geht. Natives ACP, welches im pH-Bereich 6,6–10,6 ausgefällt wurde, variierte im molaren Ca/P-Verhältnis zwischen 1,18 und 1,50 und in HPO 4 2− /totales P zwischen 33,0 und 10,1%. Bei pH 7,40 hatte natives ACP ein molares Ca/P-Verhältnis von 1,36±0,02 und enthielt 22,8 (±2,2)% HPO 4 2− . Die Werte beim ungewaschenen Niederschlag rühren weder von aus dem Überstand aufgenommenem Ca2+ und HPO2−, noch von außen kommendem Na+, Cl− und CO 3 2− her. Die amorphen Calciumphosphate werden als eine Klasse von Salzen erkannt, welche veränderliche chemische, aber identische glasartige physicochemische Eigenschaften haben. Nicht kristallines CaHPO4·xH2O kann auch ein ACP sein, besonders in den frühen Bildungsstadien. Bei physiologischem pH verwandelt sich ACP in kleine plattenförmige Kristalle, welche große Mengen von leicht ersetzbarem saurem Phosphat enthalten. Das Waschen dieser Oberflächenschicht erzeugte chemische Veränderungen in den resultierenden Kristallen; ungewaschene Kristalle zeigten ein Diffraktionsmuster, das nur schwach demjenigen des kristallinen Aspatites glich, aber ein schlecht aufgelöstes Infrarotspektrum, welches zwischen Apatit und Octocalciumphosphat war. Es werden strukturelle Erklärungen für alle diese Phenomena diskutiert, und revidierte amorph/kristalline Mineralzusammensetzungen von Knochen und Knorpel wurden neu berechnet.
    Notes: Abstract Unwashed samples of amorphous calcium phosphate (ACP) contain an irreplaceable labile fraction, rich in acid phosphate and low in Ca/P ratio, which is irreversibly lost during the washing process. Native ACP precipitated in the pH range 6.6–10.6 varied in Ca/P molar ratio from 1.18 to 1.50 and in HPO 4 2− /total P from 33.0% to 10.1%. At pH 7.40, native ACP had a Ca/P molar ratio of 1.36±0.02 and contained 22.8 (±2.2)% HPO 4 2− . Unwashed precipitate data could not be attributed to either trapped supernatant Ca2+ and HPO 4 2− or extraneous Na+, Cl−, and CO 3 2− . The amorphous calcium phosphates are recognized as a class of salts having variable chemical but identical glass-like, physicochemical properties. Non-crystalline CaHPO4·xH2O may also be an ACP, especially during early formative stages. At physiological pH, ACP transforms to small platy crystals containing large amounts of readily-replaceable acid phosphate. Washing this surface layer produced chemical alterations in the resultant crystals; unwashed crystals had poorly-crystalline apatitic diffraction patterns but exhibited poorly-resolved infrared spectra intermediate between apatite and octacalcium phosphate. Structural explanations for all these phenomena are discussed, and revised bone and cartilage amorphous/crystalline mineral compositions have been re-calculated.

Les diagrammes de diffraction aux rayons X sont diffus. Trois heures après la blessure, on note une augmentation du calcium du tissu folliculaire qui ne semble pas en rapport avec le DHT qui traduit probablement une liaison de calcium plutôt qu'un dépot minéral.
    Abstract: Zusammenfassung Frühere histologische Untersuchungen haben gezeigt, daß der Haarfollikel besonders anfällig für Verkalkungen ist, wenn die Haut von hypercalcämischen Ratten verletzt wird. Dieses Resultat wurde nun durch direkte chemische Bestimmungen und Röntgendiffraktions-analysen von Follikelgewebe bestätigt. Aufgrund der erhöhten Calcium- und Phosphatwerte kann gesagt werden, daß nach einer leichten Quetschung der Haut von Ratten, die mit Dihydrotachysterol (DHT) behandelt wurden, im Haarfollikelgewebe nach 6–12 Std Mineral-ablagerungen stattfanden, wogegen Kontrollratten mit der gleichen leichten Hautverletzung diese Ablagerungen erst nach 24–48 Std zeigten. Röntgendiffraktionsanalysen ergaben ein diffuses Apatit-Muster. Innerhalb 3 Std nach der Verletzung wurde ein Anstieg des Calcium-gehaltes im Follikelgewebe beobachtet, der nicht im Zusammenhang mit der DHT-Behandlung stand, also nicht eine Mineralablagerung, sondern eher eine Bindung von Calcium widerspiegelte.
    Notes: Abstract Previous histological investigations have shown that the hair follicle is particularly susceptible to mineralization when the skin of hypercalcaemic rats is injured. Direct chemical and X-ray diffraction analyses of follicle tissue have now confirmed this finding. As judged by increases in both calcium and phosphorus, mineral deposits began to form in hair follicle tissue 6–12 h after a mild crush injury to the skin of rats dosed with dihydrotachysterol (DHT), and 24–48 h after a similar injury to the skin of non-dosed rats. X-ray diffraction gave a diffuse apatite pattern. Within 3 h of injury there was a rise in the calcium content of follicle tissue which was not related to DHT-dosing and which was probably a reflection of calcium binding rather than mineral deposition.

Dans le cas de solutions synthétiques, les calculs sont vérifiés en comparant les valeurs de pH, obtenues par ordinateur, avec celles observées expérimentalement. Des essais de ce type, avec des solutions possèdant des concentrations de calcium et d'orthophosphate trouvées dans les liquides biologiques et à des valeurs de pH variant de 3.00 à 10.00, indiquent que le programme est adapté pour des applications biologiques. Le programme n'est pas effectif pour des solutions, dans les lesquelles l'ion bromure est la source principale de la force ionique, sans doute, par manque d'équation étendue de Debye-Hückel dans ces circonstances. Aucune formation de complexe de phosphate de sodium n'a été notée à des concentrations biologiques normales.
    Abstract: Zusammenfassung Es wurde ein Computer-Programm ausgearbeitet, um die Ionenaktivitäten von Calcium und Orthophosphat in einer breiten Varietät von Lösungen zu berechnen. Die Berechnungen wurden bei synthetischen Lösungen durch Vergleiche zwischen den auf diese Weise errechneten pH-Werten und den experimentell gefundenen kontrolliert. Diese Art Kontrollen mit Calcium-und Orthophosphatkonzentrationen, wie sie in biologischen Flüssigkeiten gefunden werden, und mit pH-Werten zwischen 3,0 und 10,0 wies darauf hin, daß das Programm für biologische Anwendungen geeignet war. Das Programm konnte nicht benützt werden für solche Lösungen, bei welchen hauptsächlich das Bromidion zur Einstellung der Ionenstärke verwendet wurde, vermutlich weil die erweiterte Debye-Hückel-Gleichung unter diesen Umständen nicht anwendbar ist. Die Bildung eines Natriumphosphat-Komplexes unter normalen biologischen Konzentrationen konnte nicht nachgewiesen werden.
    Notes: Abstract A computer program has been designed to calculate the ionic activities of calcium and orthophosphate in a wide variety of solutions. In the case of synthetic solutions the calculations were checked by comparing the computed pH values with those observed experimentally. Tests of this type with solutions having the concentrations of calcium and orthophosphate found in biological fluids and with pH values ranging from pH 3.0–10.0 indicated that the program was suitable for biological applications. The program was not effective for solutions in which the bromide ion was a principal source of ionic strength probably because of the failure of the extended Debye-Hückel equation under those circumstances. No evidence for the formation of any sodium phosphate complex at normal biological concentrations could be found.

    Notes: Abstract Alpine lakes in siliceous catchments of Tyrol and Carinthia (Austria) show signs of acidification. About 9% of the studied lakes have no alkalinity, more than 20% are below pH 6. About two thirds of all lakes have acid neutralizing capacities below 100 μeq 1−1. In spite of moderate precipitation acidity, some lakes show considerable concentrations of dissolved reactive aluminum during or shortly after snowmelt. High altitude lakes of the Alps are definitely more acidic than high mountain lakes in remote areas. Large differences in water and soil chemistry of nearby situated lakes were attributed to heterogeneities of bedrock geology. Paleolimnological investigations on former pH values of five lakes, based on diatom assemblages in the sediment, showed different developments: recent and past acidification, stable conditions, and alkalinization.

    Notes: Abstract It is now known that cardiac atria play an important role in blood pressure and volume regulation. Mechanical distension of the atria results in the release of a potent diuretic and natriuretic agent or agents termed the atrial natriuretic factor (ANF). Several structurally related forms of ANF exist in man and it is thought that these represent precursory forms of a single optimally active molecule and/or the presence of more than one form of active ANF. The chemical structure of ANF between different mammalian species is similar. ANF receptors have been identified in kidney, brain, vascular endothelial and smooth muscle cells, tracheal and bronchial smooth muscle and the adrenal glands of many mammalian species, including man. This would suggest that ANF influences blood pressure and volume homoeostasis by affecting any one of a number of biochemical or physiological mechanisms via different target tissues. ANF is now considered a potentially valuable therapeutic agent for the treatment of hypertension and congestive heart failure. Synthesis of potent receptor antagonists could be extremely useful in the treatment of various clinical situations which are produced or complicated by endogenously produced ANF, such as chronic orthostatic hypotension.

    Notes: Amino acids can be identified unambiguously in form of their dansylated methyl esters by combination of thin-layer chromatography on polyamide and subsequent mass spectrometry without prior substance elution from the chromatographic polyamide carrier. The evaporation intensity of the amino acid derivatives is sufficient to get excellent mass spectra with significant structural information also with very small amounts of starting material (picomole level), which guarantee a high separation power in the TLC step. Besides the methylester derivatives, in part, products additionally methyl substituted at the sulfonamide nitrogen could be separated.

    Notes: The simple combination of thin-layer chromatography on polyamide with subsequent mass spectrometry of the separated compounds together with the carrier material has been employed for unambiguous identification of individual dipeptides in a mixture. The dipeptides were analysed in the form of their dansyl methyl ester derivatives. The appearance of molecular ions in the electron impact mass spectra simultaneously with the ions dans-NH=CHR⌝+ indicating the corresponding N-terminal amino acid residue enables a rapid and sensitive structure elucidation of the dipeptides. The chromatography and mass spectrometry can be performed with 0.2-0.3 nmol dipeptide.

    Notes: Analysis of reports in the world's literature suggests that average sperm densities for groups of unselected males were relatively constant at about 108 million cells per ml prior to 1950. Subsequent to that time mean sperm densities appear to have declined. Regression analysis indicates the existence of significant negative correlations between mean sperm densities and production of synthetic organic chemicals among other parameters. Phthalate esters are one class of large volume organic chemicals that are known to disturb testicular function in laboratory animals. These compounds are also the most abundant man-made chemicals in the environment. Plots of the concentration of dibutylphthalate in the cellular fraction of ejaculates against either the sperm density or the total number of sperm for the same ejaculates gave two clusters of points. These clusters suggest the existence of two or more populations vis à vis phthalate metabolism; both of which show a negative correlation between phthalate concentration and sperm production.

    Notes: We report a case of n-nonacosane storage disease, which went undiagnosed until the death of a 55-year-old farmer. Clinical, histological, and biochemical features are discussed. n-Nonacosane storage was identified by gas chromato-graphic-mass spectrometric analysis of different tissue extracts, n-nonacosane concentration reaching 1.2 mg g-1 of lung tissue and 0.32 mg g-1 of liver tissue. It was possible to rule out a work-induced intoxication, and n-nonacosane storage appeared to be accounted for by a lifelong, heavy consumption of unpeeled apples and Brussels sprouts.

    Notes: A new technique known as thermally-assisted fast atom bombardment (TA-FAB) has been applied to the analysis of a series of cyclic tetrapeptide mycotoxins in order to demonstrate the usefulness of the method for structural elucidation. TA-FAB uses saturated aqueous solutions of highly hydroxylated compounds, such as fructose, as alternatives to the usual viscous liquid matrices employed in conventional FAB. During the TA-FAB analysis, the probe tip is resistively heated causing diffeiences to occur in the desorption profiles for the analyte and the matrix ions enabling an optimization for analyte desorption as a function of temperature. In this study, direct comparisons are made between TA-FAB, conventional FAB, and electron impact ionization for the analysis of the Helminthosporium carbonum mycotoxins at the 1.5 μg level. The results demonstrate the superior capacity of TA-FAB to provide both molecular weight confirmation and significant fragmentation to aid in the structural elucidation of these important biomolecules.

    Notes: Three glutathione conjugates of acetaminophen were characterized by fast atom bombardment/mass spectrometry (FAB/MS) and fast atom bombardment/mass spectrometry/mass spectrometry (FAB/MS/MS). The conjugates, 3-(glutathion-S-yl)acetaminophen, 3-(glutathion-S-yl)diacetaminophen and 3-(diglutathion-S-yl)diacetaminophen showed intense [MH]+ ions at m/z 457, 606 and 911, respectively. Only 3-(glutathion-S-yl)acetaminophen showed any fragmentation by FAB/MS. Structurally characteristic fragmentation was observed with all three conjugates when the [MH]+ ions were collisionally activated. The loss of the glycine (GLY) and glutamic acid (GLU) moieties indicated the presence of at least one glutathione (GSH) group. Multiple losses, some of which could only occur via cleavages in both GSH moieties, were observed with the diglutathione conjugate.

    Notes: A GC-MS procedure has been developed for the quantitation in plasma and urine of rimantadine, an antiviral drug effective against type A influenza. The assay utilizes selective ion monitoring, methane negative ion chemical ionization (NCI) and stable isotope dilution. Sensitivity to NCI is effected by derivation of rimantadine with pentafluorobenzoyl chloride. The method has been used to quantitate plasma concentrations of rimantadine over a range from 4.2 ng/ml to 416 ng/ml, and urinary concentrations of rimantadine over a range of 21 ng/ml to 2077 ng/ml.

    Notes: The behaviour of 1,1·,1″-phosphinothioylidinetrisaziridine (thiotepa) in water and aqueous salt solution of various acidity was studied by field ionization mass spectrometry (FI-MS), TLC and potentiometric titration. The rate of thiotepa hydrolysis increased with increasing thiotepa, or sodium chloride concentration, and with decreasing pH. As hydrolysis products of thiotepa, mono- and dichloro derivatives formed in neutral, salt-containing and acidic solution, tepa, and products formed by P—N bond cleavage and release of aziridine have been identified.

    Notes: Methodology for the qualitative and quantitative analysis of the anthelmintic fenbendazole and its metabolites in goat feces using electron impact (EI)/direct exposure probe (DEP)/mass spectrometric (MS) and tandem mass spectrometric (MS/MS) techniques is presented. Analyses were conducted on extracts from spiked feces and feces from animals treated per os with 5 mg fenbendazole/kg, with samples being collected at zero time and at twelve hour intervals for 144 h. The results of the EI/DEP/MS quantitation of these samples are compared to those for the same samples analysed by high pressure liquid chromatography (HPLC). Mass spectral data for fenbendazole and its metabolites are presented and the advantages of the use of EI/DEP/MS and/or DEP/MS/MS over HPLC are discussed. This methodology may be used as a confirmatory method for the HPLC analysis of fenbendazole and its metabolites or may be used as a method in its own right for the rapid qualitative and quantitative analysis of these compounds.

    Notes: Known and possible phenolic and primary and secondary amine metabolites of lidocaine were added to urine from drug-naive rats. These metabolites were derivatized in this aqueous medium by acetylation with acetic anhydride or by pentafluorobenzoylation with pentafluorobenzoyl chloride. The derivatives were simultaneously extracted into an organic solvent. The products were separated by gas chromatography (flame ionization detection for acetates and electron-capture detection for pentafluorobenzoates) and identified by combined gas chromatography/mass spectrometry. Mass spectral fragmentation pathways were readily deduced; diagnostic fragment ions were identified and were valuable for characterization purposes. Structural isomers could be distinguished on the basis of their GC retention times.Extractive derivatization using pentafluorobenzoyl chloride is an attractive analytical procedure for the identification of phenolic and dealkylated metabolites of lidocaine.

    Notes: Thermospray HPLC/MS analysis of the metabolites of arachidonic acid proved to be sensitive and specific. The compounds included were hydroxy-fatty acids (5-HETE, 12-HETE, and 15-HETE) and prostaglandins PGD2, PGE2, PGF2α, PGA2, PGA1, TXB2, and 6-Keto-PGF1α. Thermospray HPLC/MS analysis allows for simultaneous monitoring of each compound without the need for additional sample preparation or derivatization. The thermospray spectra for the metabolites exhibited [M + NH4]+ ions and fragment ions because of sequential loss of equivalents of H2O. HPLC/MS showed detection limits in the 0.5 to 5 ng range when using multiple ion detection for most of the metabolites. Post-column derivatization of these metabolites using trimethylanilinium hydroxide (TMAH) to form the methyl esters is also presented. This derivatization resulted in a gain in ion current by a factor of 3-6 for most compounds while adding potential specificity to the analysis. The thermospray spectra of the derivatives were nearly identical to the spectra of the free acid except the peaks were incremented by 14 daltons due to the methyl ester formation. The derivatization of the carboxylic acid group proved to be complete under thermospray conditions producing the maximum ion current and causing no compromise in operation of the interface.

    Notes: Dinucleoside monophosphates bearing usual protecting groups at the exocyclic amino functions of the heterocyclic bases, the 5′-hydroxy group, the phosphate moiety and the O-6-position of the guanine ring have been analysed by FAB mass specrometry in the negative ion mode. These non-charged key intermediates for oligodeoxynucleotide synthesis exhibit very simple spectra in the mass range of m/z 400-1500 using tetra-ethylene glycol as matrix. Most of the observed ions could easily be assigned. Apart from deprotonated molecular ions sequence specific ions arising from C-O-P bond cleavage and ions derived from loss of protecting groups were observed. The simplicity of spectra interpretation makes FAB mass spectrometry to a rapid and valuable tool for unambiguous identification of dimeric synthons for oligonucleotide synthesis.

    Notes: Mixtures of both synthetic and naturally occurring (human plasma) cholesteryl esters have been examined by capillary gas chromatography/mass spectrometry (GC/MS). A magnetic sector mass spectrometer was used and a variety of ionization modes were assessed with a view to obtaining structural information on intact cholesteryl esters. By employing ammonia as reagent gas, with negative ion scanning, spectra were produced from which the nature of steryl and fatty acyl moieties could be readily deduced. Analyses were performed at an ion source temperature of 300°C in order to maintain the integrity of the gas chromatographic profile. The technique described is of general use for the GC/MS analysis of steryl esters, particularly in conjunction with magnetic sector instruments.

    Notes: Thermospray spectra on a magnetic sector mass spectrometer, by definition measured at high accelerating voltages, are seen to be highly fragmented. This is rationalised in terms of the necessity to accelerate ions through a region of high pressure in the vicinity of the ion-collecting orifice at the spray volume, where ions may be energized by ion molecule collisions. In this region, a high percentage of the parent ion current is lost through the dispersion of the ion beam and by the collisional activation of molecular ions resulting in extensive fragmentation. The investigation of means by which these effects may be minimized has led to the construction of an improved sector thermospray source. Comparative results for gramicidin S demonstrate that its fragmentation is reduced and that the intensity of the protonated molecular ion is greatly enhanced.

    Notes: Acetates of 12 positionally isomeric tetradecenols (Δ2-Δ13) have been prepared and their mass spectra measured without any prior chemical modification. Comparison of spectra of all the isomers has shown the variation in intensities of some predominant fragments with the position of the double bond on the 14 carbons chain. The mass spectra were interpreted in terms of the relative intensity ratio of the fragment ion peaks to locate the double bond positions. A procedure for elucidation of the positional isomerism of tetradecenyl acetates is given.

    Notes: Eight normal subjects ingested 125 mg of p-tyramine-β,β-2H2 hydrochloride and the 3 h and following 21 h urine collections were analysed by mass spectrometry with selected ion monitoring for the deuterated metabolites: free and conjugated p-tyramine-β,β-2H2, free p-octopamine-β-2H1, free and conjugated p-hydroxyphenylacetic acid-α-2H1 and -α,α-2H2, and free p-hydroxymandelic acid-α-2H1. These metabolites accounted for 72% of the ingested label, of which conjugated p-tyramine and free p-hydroxyphenylacetic acid constituted 90%. Approximately 50% of the total deuterated tyramine and 70% of the total deuterated p-hydroxyphenylacetic acid were excreted in the first three hours, although there was considerable variation between individuals. The presence of a small amount of p-hydroxyphenylacetic acid-α-2H1 suggests that some exchange of deuterium occurred at the intermediate p-hydrophenylacetaldehyde stage.These results, based as they are on metabolites labelled with the stable isotope deuterium, ought to be more reliable than earlier studies in which unlabelled tyramine was ingested with the resultant metabolites, indistinguishable from their endogenous metabolic counterparts, being measured by fluorimetry or gas chromatography.

    Notes: Human exposure to aromatic amines may be monitored by measuring the amount of sulfinamide adduct bound to hemoglobin. In order to develop a sensitive and selective method for measuring aromatic amines present in low concentrations the mass spectra of the pentafluoropropionamide derivatives of a series of aromatic amines were determined under various ionization conditions. With electron ionization the aromatic amine derivatives typically yield intense molecular ions with fragmentation leading to elimination of C2F5 and COC2F5. Higher molecular weight polycyclic aromatic amines, e.g., 6-aminochrysene, give prominent molecular ions as well as intense ions corresponding to M-174 (loss of CNHCOC2F5). With positive chemical ionization using methane as reagent gas the derivatives give protonated molecular ions as the base peaks, as well as the associated ions at M + 29 and M + 41. Negative ion chemical ionization yields, primarily, peaks corresponding to the loss of HF from the molecular anions. Negative ion chemical ionization with selected ion monitoring is generally more suitable than positive chemical ionization or electron ionization for the quantitative analysis of aromatic amines present in biological samples in the mid femtomolar range. Exposure of human subjects to aromatic amines was determined via basic hydrolysis of the isolated sulfinic acid amide hemoglobin adducts. Analysis of the derivatized extracts by gas chromatography/negative ion chemical ionization/mass spectrometry demonstrated the presence of aniline, ortho-, meta-, and para-toluidine, 2-naphthylamine, and 4-aminobiphenyl. The hemoglobin adduct levels of these amines in both cigarette smokers and nonsmokers were determined. The mean values of the adduct levels of ortho- and para-toluidine, 2-naphthylamine, and 4-aminobiphenyl were higher in smokers than in nonsmokers, while the mean adduct values of aniline and meta-toluidine were similar in both groups. Adducts of 2-, 3-, 4-ethylaniline, 2,3-, 2,4-, 2,5-, 2,6-dimethylaniline, and 3-aminobiphenyl have been detected in smokers' blood. 1-Aminoanthracene, 9-aminophenanthrene, and 6-aminochrysene have not been detected. GC/MS procedures were also used to determine the extent of binding of aniline-2,3,4,5,6-d5 to hemoglobin in rats administered aniline-d5.

    Notes: The single dose pharmacokinetics of deuterium-labelled cannabinol (2H2-CBN) were evaluated in six male cannabis users with different degree of abuse after smoking an average dose of 19 mg and after intravenous administration of 20 mg CBN. Plasma levels were measured for up to 72 h with selected ion monitoring by GC/MS using 2H7-CBN as internal standard. The systemic availability of smoked CBN was found to be 39 ± 26% (min-max 6-65%). The mean plasma clearance was 19.1 ± 2.6 ml min-1 kg-1 and the volume of distribution was determined to 50 ± 23 1 kg-1. The apparent terminal half lives for CBN were 32 ± 17 h and 43 ± 29 h after intravenous administration and smoking, respectively.

    Notes: Derivatization of prostaglandins and thromboxane B2 using diethyl amino ethyl chloride and diethyl amino ethyl bromide improved thermospray HPLC/MS sensitivity. The derivatization was evaluated for prostaglandin A1, A2, D2, E2, F1α, F2α, and thromboxane B2. The derivatization reaction, thermospray operating conditions, and mode of detection were optimized to produce the most intense [M + H]+ or [M - H]- ions for the derivative. Derivatization was better than 99% complete in 1 hour at 75°C. No thermal degradation of the prostaglandins was observed. Positive thermospray ionization proved the mode most sensitive, enabling detection from 10 to 300 pg of each prostaglandin under multiple ion detection. The thermospray spectra exhibited intense [M + H]+ ions for the derivative with a few fragment ions from sequential losses of water from the [M + H]+ ion. Detection of a prostaglandin metabolite in plasma over the concentration range from 3 ppm to 30 ppb was possible using this derivatization.

The sensitivity defined as the quantity that could be passed through the gas chromatographic column.The three derivatives showed a mass spectrometric fragmentation pattern with cleavage of the C12—C13 bond as an important feature. Particularly, the allyldimethylsilyl ether derivative of the three compounds studied exhibited a high tendency for C12—C13 bond cleavage resulting in a fairly intense ion at m/z 435. However, the mass spectra indicated multiple fragmentation pathways due to the presence of double bonds, leading to decreased intensities of the high mass ions. A quantitative analysis by selected ion monitoring of the most intense high mass ions in the respective mass spectrum demonstrated that neither derivative would allow measurements in the low picogram range. Catalytic hydrogenation of the double bonds was performed and the methyl ester trimethylsiyl, methyl ester allyldimethylsilyl and methyl ester tert-butyldimethylsilyl ether derivatives of the reduced compounds were prepared. Saturation of the double bonds increased the gas chromatographic sensitivity for the three derivatives as well as the intensities of the high mass ions in their mass spectra. The high sensitivity that can be obtained by measurement of such high mass ions was demonstrated by quantification of leukotriene B4 in lung tissue samples by selected ion monitoring.

    Notes: In the process of developing a high-sensitivity negative chemical ionization gas chromatographic/mass spectrometric assay for brain and cerebrospinal fluid (CSF) levels of quinolinic acid (QUIN, 2,3-pyridine dicarboxylic acid), (18O4)QUIN was prepared. Its properties as an internal standard were compared with those of the structural isomer 2,4-pyridine decarboxylic acid (2,4-PDC) previously used by others. All oxygen atoms in QUIN were labeled by heating in 3N HCl/(18O)water for 48 h at 80°C. Back-exchange of (18O4)QUIN was prevented during derivatization to an electron-capturing dihexafluoropropanol ester by using trifluoroacetylimidazole as catalyst instead of perfluroacyl anhydrides. When mixtures of QUIN and (18O4)QUIN and/or 2,4-PDC were followed through a procedure to isolate and quantify brain and CSF QUIN, the variability in the ratio of QUIN:2,4-PDC was greater than for QUIN:(18O)QUIN. We conclude that (18O)QUIN is the preferred internal standard in gas chromatographic/mass spectrometric quantification of brain and CSF QUIN, and that (18O)-labeled carboxylic acids may be esterified effectively without back-exchange using acylimidazole reagents.

    Notes: Oxygen-enhanced negative chemical ionization MS is sensitive and specific for the detection of polychlorinated biphenyls (PCBs) without isobaric interferences (qualitative or quantitative) from congeners of higher levels of chlorination. The collision-induced dissociation (CID) spectra of the [M - Cl + O]- ion of 40 PCB congeners have been acquired on a triple quadrupole mass spectrometer.Qualitative relationships between structure of the congener and its CID spectrum suggest that reliable identification of PCB congeners is possible even where capillary column gas chromatographic separation is incomplete. Congeners with hydrogen atoms located para and meta to the biphenyl linkage show most abundant collision-induced loss of HCl, whereas congeners with a penta-chloro phenyl ring show least CID loss of HCl. Congeners IUPAC 105 and 121 have CID spectra suggesting a pathway for sequential loss of HCl and CO neutrals during the collision-induced dissociation of the [M - Cl + O]- ion of these molecules. Unambiguous identification of PCB congeners in natural materials will require a complete set of pure, synthetic PCB congeners: identification problems are illustrated using an extract of Northern Shark (dogfish) liver oil.

    Notes: The 13C enrichment of the carboxyl carbon of leucine was measured by isotope ratio mass spectrometry after conversion to CO2 by reaction with ninhydrin in a Vacutainer and cryogenic purification using the Finnigan MAT Breath Gas Analysis System designed for processing 13C breath test samples. The sources of error which arise with submicromole samples are examined and corrections provided for suboptimal mass spectrometer signals and contamination of the evolved CO2 with CO2 from the reaction medium. The main limitations to the accuracy and precision of the method are not instrumental but arise from the contamination with residual CO2 in the reaction medium, and this sets a lower limit of around 0.25 μmol leucine on the practical sample size. This is an improvement of about five-fold on the previous manual method of CO2 isolation.

    Notes: The fragmentation pathways of methyl abscisate and its 2E isomer in methane positive ion chemical ionization (PICI) and methane negative ion chemical ionization (NICI) mass spectrometry have been elucidated using isotopically labelled analogues. It is shown that in PICI the most abundant ions are formed by the loss of water, methanol, both methanol and water and of methyl formate. The PICI mass spectrum of methyl 2E abscisate differs significantly in that the loss of water is much less important. In NICI, the most abundant ions from methyl abscisate are due to the molecular anion, [M - H2O]- and [M - CH3OH]-. It is shown, however, that the methyl ester methyl is not lost in this last fragmentation. Two other significant ions contain the side chain and the ring of methyl abscisate, respectively. In contrast, the NICI mass spectrum of methyl 2E abscisate differs principally in showing an abundant loss of a hydrogen atom.

    Notes: A moving belt interface was used to identify budesonide metabolites, obtained from rat and mouse liver incubations, by liquid chromatography/mass spectrometry (LC/MS). The metabolites were separated on a small-bore C18 column with an ethanol/water gradient as mobile phase at a flow rate of 0.2 ml min-1. A spray device was used for deposition of the aqueous solvent on to the belt. Chemical ionization mass spectra were obtained with methane as the reagent gas. Deuterium-labelled budesonide, which was used to facilitate metabolite identification by the isotope cluster technqiue, was found to be slightly separated from the unlabelled analogue on the LC column. Incubations were also performed under 18O2 to elucidate the mechanism of a new metabolic pathway (16α, 17α-acetal splitting) and to confirm the oxidative nature of reactions leading to hydroxylated metabolites. The moving belt LC/MS technique afforded higher sensitivity, and gave more abundant MH+ ions of the compounds studied, than previously found by direct probe mass spectrometry. Phthalate ester background, partly from the polymide belt, complicated the identification of minor metabolites.

    Notes: Dehydroepiandrosterone sulphate (DHAS) has been quantified in human blood serum by fast atom bombardment (FAB)/tandem mass spectrometry of immunoadsorption extracts. FAB of DHAS yielded abundant ions corresponding to the intact steroid sulphate; these were selected by a double-focusing mass spectrometer prior to collisionally activated decomposition in a quadrupole collision cell and mass analysis by a quadrupole mass filter. [HSO4]- (m/z 97) was the sole prominent daughter ion. For quantitative analyses the quadrupole mass filter was set to transmit m/z 97 and a narrow-range magnet scan yielded a spectrum of parents, including m/z 367 and 369, corresponding to DHAS and the (2H2)-analogue (used as internal standard), respectively. Serum concentrations by this procedure were in good agreement with data obtained by gas chromatographic/mass spectrometric analyses of DHA heptafluorobutyrate, formed by direct derivatization of the steroid sulphate.

    Notes: A method is described for the quantification of tulobuterol in human plasma, based upon selective extraction and high resolution capillary gas chromatography-mass spectrometry. The mass spectrometric determination is based on selected ion monitoring of two fragment ions derived from the N,O-heptafluorobutyryl derivatives of tulobuterol and its internal standard, deschlorotulobuterol. The plasma work-up procedure consists of extraction of 1.0 ml alkalinised plasma with dichloromethane, back-extraction into an acidified aqueous phase, followed by a final extraction with dichloromethane after realkalinisation. The method gave interference-free and linear results. The between-assay variability is 4.7% CV at the 3.0 μg l-1 plasma concentration level. The assay permitted quantitative measurements down to 170 ng tulobuterol per litre of plasma and hence provided sufficient sensitivity for quantification of plasma levels after therapeutic doses.

    Notes: In phytochemical and chemotaxonomic research work mass spectrometry plays an outstandingly important role. Using gas chromatography/mass spectrometry (GC/MS) we established the chemotaxa of Tanacetum vulgare L. Chemotypes with essential oils countaining 60-90% of artemisia ketone, carveol, dihydrocarvone, myrtenol, umbellulone, terpinen-4-ol, davanone, and Tagetes species containing various essential oils can be clearly distinguished by their spectra; we examined many variations of Tagetes erecta, T. lucida, T. minuta, T. patula and T. tenuifolia. We have identified α-β-pinene-, 1,8-cineol-, linalool-, camphor-, nerol-, geraniol- and γ-gurjonene as components of Achillea distans L. Injecting the essential oil direct from the oil-secreting organs of T. minuta plants we identified using GC/MS 6-10 and 16% eugenol from the involucral bract and hypsophyll, respectively, as well as β-ocimene, dihydrotagetone, tagetone, Z- and E-ocimenones. In the course of studies on essential fatty acids Borago officinalis and Lappula squarrosa were selected from 70 species of the family Boraginaceae to obtain seed oil as a source of γ-linolenic acid, and for the PG synthesis we isolated several grams of γ-linolenic acid, as well as C18:4, i e. octadecatetraenic acid, from L. squarrosa on the basis of the mass spectra. From the seed oil of Aquilegia vulgaris C18:3 (5) from the oil of Limnanthes dougloasii C20:1 (5) and from the seed oils of Delphinium consolida and of Tropaeolum species (T. majus, T. minus, T. peregrinum) C20:1 (11) fatty acids were identified on the basis of spectra.

    Notes: The problem of reproducing electron capture negative ion mass spectra has been addressed by studying parameters that affect negative ion abundance on a Hewlett Packard 5985B mass spectrometer. Parameters affecting negative ion formation in the ion source, such as ion source temperature, pressure, sample concentration and electron energy, were studied in conjunction with the effect of lenses used to extract and transmit ions to the quadruples. From these experiments, it was found that, in addition to ion source temperature, the ion focus potential has the most dramatic effect on the abundance of molecular ions relative to fragment ions like Cl-. With proper control, it was found that the relative abundance of ions from decafluorotriphenylphosphine could be reproduced over a period of one year.

The structure was confirmed by proton nuclear magnetic resonance and direct probe fast atom bombardment mass spectrometry. A mixture of elliptinium and its deuterated analogue was administered intravenously to rats. In urine, after analysis by liquid chromatography/mass spectrometry (LC/MS), unchanged drug and N-acetylcysteinylelliptinium were found.     Notes: Bile from rats treated with acetaminophen was analyzed by direct injection onto a thermospray liquid chromatography/mass spectrometry (LC/MS) system. Two conjugated metabolites of acetaminophen were separated by the high-pressure liquid chromatographic system and analyzed by mass spectrometry. The conjugates were identified as the glutathione-acetaminophen conjugate and the glucuronide-acetaminophen conjugate by comparison of the chromatographic retention times and the mass spectra to that of the synthetic standards. No evidence of acetaminophen metabolites was observed when bile samples were subjected to direct analysis by fast atom bombardment mass spectrometry.

    Notes: The analysis of extracts from the South American plant Justicia pectoralis Jacq. permitted the identification, among other compounds, of coumarin, dihydrocoumarin, umbelliferone and 3-(2-hydroxyphenyl)propionic acid by gas chromatography/mass spectrometry (GC/MS); the acids and phenolic compounds were derivatized with diazomethane. GC/MS of simple coumarins, phenylpropionic acids and their hydroxylated isomers was performed after derivatization through methylation and trimethylsilylation; these results may be useful for the identification and quantification of these compounds in other biological materials.

    Notes: The fast atom bombardment mass spectra of 24 α-amino acids have been studied. These include the mass spectra as well as the metastable ion MI and collisional activation CA spectra of [M + H]+ and [M - H]- ions. The extent of the common neutral losses as NH3, H2O and CO2H2 in the positive ion spectra is governed by the nature of the side chain. The relationship between the fragmentation behaviour of the negative ions and the presence of particular functional groups is less obvious. Mixtures of amino acids in glycerol show pronounced surface effects.

    Notes: A conventional bore liquid chromatograph has been interfaced to quadrupole and magnetic sector mass spectrometers configured for fast atom bombardment ionization via a continuous flow FAB probe. It is shown that post-column addition of FAB matrix and in-line ultraviolet detection facilities do not significantly compromise chromatographic integrity and that high quality mass spectra are obtainable from such FAB LC/MS studies.

    Notes: A method which involves the use of tandem mass spectrometry (MS/MS) for the identification of drug metabolites has been demonstrated with a triple quadrupole mass spectrometer. The method is based on the fact that metabolites usually retain various substructures of the original drug molecule. MS/MS is capable of rapidly identifying molecules with characteristic substructures without prior separation. It is shown that this method makes it possible to postulate possible drug metabolite structures rapidly and systematically without the use of standards. The MS/MS method, as it was applied to the identification of the metabolites of a new antiepileptic drug, zonisamide, is discussed. In this case it was possible to identify isomeric metabolites due to their differences in vaporization times off the probe and their different daughter spectra. The complementary uses of the neutral loss and parent scans for the determination of the site of metabolism is demonstrated. A new figure of merit, the limit of identification, is introduced. The amount of the epoxide metabolite of carbamazepine necessary for its reliable identification in urine was shown to be 0.4 ng/μl. The application of various techniques to confirm preliminary findings with this MS/MS method are described.

    Notes: A liquid-solid extraction method suitable for rapid screening of sulfonated azo dyes in municipal wastewater has been developed. The dyes (Color Index Acid Yellow 23, Red 14 and Yellow 49) were separated on a short, reversedphase liquid chromatographic column with a water-methanol gradient containing 2 mM sulfuric acid. The column effluent was directed via a split-valve to an ion spray liquid chromatography/mass spectrometry (LC/MS) interface to an atmospheric pressure ionization mass spectrometer. The ion spray LC/MS system produced abundant [M - Na]- and [M - 2Na]2- ions according to the number of sulfonic acid substituents. Collision-induced dissociation of the parent ions for the dyes studied gave the SO3-. fragment common to sulfonated compounds plus additional daughter ion fragments characteristic of each dye. The dyes were quantified by monitoring from four to six different daughter ions of each dye. The relative abundances and the sum of the daughter ion current counts were used for confirmation and quantification with external standards. Recoveries of the dyes were in the range of 70-122%, and the relative standard deviation of replicate determinations was 5.4-12%. The method detection limit was three times higher for Acid Yellow 23 compared with the other two dyes, which could be detected down to 50 ppb in municipal wastewater.

    Notes: Self-chemical ionization Fourier transform ion cyclotron resonance (FT-ICR) mass spectra are reported for bases, nucleosides, and alkylated and exocyclic adducts of bases and nucleosides. The technique always produces a protonated molecular ion and in the majority of cases this is a single, intense peak. Analysis of a base mixture and a nucleoside mixture demonstrates the technique as an excellent method to identify the constituent compounds qualitatively. The high resolution capabilities and tandem mass spectrometric techniques (msn) in FT-ICR are discussed with respect to developing the technique as a future method to identify and characterize nucleic acid constituents, specifically adducts.

    Notes: Nucleotides of the structure P1,Pn-di(adenosine-5′)-n-phosphate (n = di- through penta-) in the form of salts, and P1,P4-di(guanosine-5′)tetraphosphate sodium salt have been analyzed by fast atom bombardment (FAB) mass spectrometry. A 0.2 molar solution of p-toluenesulfonic acid in glycerol has been evaluated as a matrix. In this matrix, the metal ions of the nucleotide salts are readily exchanged for protons, resulting in a simple spectrum with only one peak in the molecular ion region corresponding to the free phosphoric acid of the nucleotide plus or minus a proton (positive or negative mode), instead of the multiplicity of peaks arising from a series of metal and matrix adduct ions found with glycerol as matrix. The detection limit for analytes using this matrix is improved by a factor of ten compared to glycerol alone. It appears that the high acidity and the surfactant properties of p-toluenesulfonic acid both contribute to this result. Useful spectra are obtained from 250 ng of each of the above mentioned nucleotides, with the detection limit being somewhat lower in the positive mode. However, both positive and negative FAB spectra are useful and the results are complementary.

    Notes: Direct analysis of unpurified urine from patients with β-ketothiolase deficiency and glutaryl-coenzyme A dehydrogenase deficiency was carried out by methylation and fast atom bombardment mass spectrometry. Previously unidentified signals consistent with unusual acylcarnitines were detected. In the former disease, thermospray liquid chromatography/mass spectrometry analysis confirmed the identification of tiglylcarnitine and differentiated it from a biological isomer, 3-methylcrotonylcarnitine. In glutaric aciduria, glutarylcarnitine was confirmed by detection of glutaric acid liberated upon base hydrolysis of a purified acylcarnitine fraction. The discovery of these metabolites suggests that L-carnitine therapy might be beneficial for the enhanced excretion of toxic metabolites that accumulate in patients with these disorders.

Abundant, diagnostic fragment ions arise from cleavage of the bonds at the positions of the original double bonds, combined with loss of a molecule of trimethylsilanol for every two vicinal trimethylsiloxy groups, with the exception of similar ions formed by rupture of an original conjugated double bond and containing the other conjugated double bond, which are weak or absent. The method was applied to several alkadienoic, alkatrienoic and alkatetraenoic acids containing a conjugated diene unit.

    Notes: Long-chain unsaturated fatty acids (UFA) can easily be converted on a microgram scale to the corresponding 2-alkenyl-4,4-dimethyloxazolines by condensation with 2-amino-2-methylpropanol (AMP). These modified molecules with a ‘hidden’ carboxyl group have been proved to be a class of useful derivatives for gas chromatography/mass spectrometry (GC/MS) of UFA mixtures. While possessing very good GC characteristics, the title compounds show regular, well-recognizable diagnostic ion peaks of the double bond position in the chain. Detailed description of the method as well as electron impact (E1) mass spectra of derivatives resulting from mono-, di and polyenoic (maximally containing six double bonds) acids are presented.

    Notes: A stable isotope assay for the quantification of debrisoquine (1) and its major urinary metabolite 4-hydroxydebrisoquine (2) is described. The method consists of extractive derivatization of 1 and 2 by use of 1,3-diketones, chiral derivatization of the 4-hydroxy group of 2, and gas chromatography/negative ion chemical ionization mass spectrometry in the presence of deuterated analogues of 1 and 2. In comparison with synthetic R-(-)-2 and S-(+)-2 it is shown that in vivo benzylic 4-hydroxylation of 1 is highly stereoselective, leading predominantly to S-(+)-4-hydroxydebrisoquine (enantiomeric excess ≥90%).

    Notes: A method for qualitative and quantitative analysis of trace amounts of quaternary organoarsenicals such as arsenocholine and acetylarsenocholine has been developed. The method is based on pyrolysis, gas chromatography/mass spectrometry and use of deuterium-labelled internal standards. Arsenocholine and acetylarsenocholine have been estimated in fish from arsenic-polluted brackish water and compared with the same species of fish from unpolluted water. The investigation also includes some fish and crustacea from marine water. The presence of arsenocholine and acetylarsenocholine in different aquatic organisms indicate the existence of a general metabolic pathway for these compounds in aquatic ecosystems.

Collisional activation of the [M - H]- anions of sulfate conjugates and bile salts predominantly yields fragment ions arising by reactions occurring remote from the charge site. These reactions are sometimes sensitive to differences in stereochemistry at positions remote from the charge site and are useful for positional isomer differentiation. On the other hand, collisional activation of the [M - H]- anions of the glucouronide conjugates leads primarily to charge-driven fragementations.

Collision-induced dissociation mass spectra of the most intense parent ion in the high-mass region were taken. Collisionally activated decomposition mass spectra of the [C(12)-C(20)]+ ion of TxB2 and 2,3-dinor-TxB2 show an intense but not specific daughter ion, whereas the collison-induced dissociation mass spectrum of the [M - (C(16)-C(20)]+ ion of 11-dehydro-TxB2 results in the formation of numerous daughter ions.

    Notes: Steviol (ent-13-hydroxykaur-16-en-19-oic acid), the aglycone of various plant-derived glycoside sweeteners consumed by human populations, is known to be mutagenic toward Salmonella typhimurium strain TM677 when metabolically activated using a 9000 × g supernatant fraction derived from the liver of Aroclor 1254-pretreated rats. Mass spectral analysis of this diterpenoid and some analogs revealed characteristic patterns reflecting differential stereochemistry at the C/D rings and variations in the nature of the substituents present. Such information has been used to help identify several in vitro metabolites of steviol in conditions known to produce a mutagenic response, when analyzed by human populations, is known to be mutagenic toward Salmonella typhimurium strain TM677 when metabolically be allylic oxidation and epoxidation. 15-Oxosteviol, a product of oxidation of the major steviol metabolite, 15α-hydroxysteviol, was found to be a direct-acting mutagen.

Determination of G2-AP and G3-AP produced from G5-AP by the action of α-amylases, based on the abundance of their [M + H]+ ions relative to that of cellobiose reductively aminated with 2-amino-6-methylpyridine as the internal standard, allowed rapid and reproducible assay of these enzymes. It was advantageous for clinical investigation that the proportion of pancreatic and salivary α-amylase activities could be determined.

The method is based on a computerized analysis of data obtained with dual-column gas chromatography/FID (GC/FID) and gas chromatography/mass spectrometry (GC/MS) profiling of urinary organic acids. It involves: (1) isolation of the compounds by solvent or solid-phase extraction; (2) conversion into trimethylsilyl (TMS) or TMS-oxime derivatives; (3) GC/FID analysis on SE-52 and OV-1701 capillary columns; (4) tentative identification by comparing the methylene unit (MU) values on both columns with a user-built library of reference compounds; (5) quantitative evaluation of the excretion profile; and (6) analysis by GC/MS of samples with an abnormal profile using an automated peak identification programme.

    Notes: The electron impact (EI) mass spectra (75 eV) of the new carboxylic acid ionophore griseochelin and some of its derivatives are discussed. The mass spectral fragmentation was studied using exact mass measurements and deuterium labelling. Furthermore, the negative ion mass spectra (2-4 eV) of these compounds are compared with their EI mass spectra.

    Notes: Fast atom bombardment (FAB) combined with tandem mass spectrometry has been used to verify the amino acid sequence in Org 2766 [H-Met(O2)-Glu-His-Phe-D-Lys-(Phe-OH)] and its deuterated analogues. It is shown that FAB mass spectrometry in addition to the [M + H]+ ion yields dominantly N-terminal sequence ions and some C-terminal sequence ions. These results together permit determination of the amount of deuterium incorporated and the position of the denterated amino acids in the peptide.
