    Notes: Reagents that block or cross-link sulfhydryl (-SH) groups and those that reduce disulfide (S-S) bonds have been tested for their effects on mating in Chlamydomonas reinhardii. Wild-type (wt) gametes of mating type + (mt+) and mt-, and a fusion-defective mt- mutant, gam-11, were studied. Differential sensitivities of mt+ vs mt- and of wt mt- vs gam-11 mt- were analyzed. Concentrations of reagents that did not disrupt flagellar agglutination, the first stage of the mating reaction, were generally used. Pretreatment of mt+ gametes with the membrane permeable -SH reducing agent dithiothreitol (DTT) inhibits flagellar sexual signaling at concentrations that do not inhibit any part of the mating reaction of mt- gametes. Wt mt- is more sensitive than wt mt+ to inhibition by low concentrations of p-chloromercuribenzoate sulfonate (pCMBS), an organic mercurial. The membrane-impermeable reducing agent, reduced glutathione (GSH), also preferentially inhibits wt mt-. Gam-11 mt-, a fusion-defective mutant, which has been used to study the sensitivity of the adhesion of the plasma membrane-associated mating structures, is less sensitive to GSH and pCMBS inhibition that is wt mt-. DDT and pCMBS cause an increase in mating structure adhesion in pretreated gam-11. The differential inhibition of pair and group formation during gam-11 × wt mt+ matings has suggested a possible mechanism for mating structure adhesion.

    Description: The formulation of the magnetospheric convection problem involves a coordinate system consisting of the electric potential, the magnetic field intensity, and the modified longitudinal invariant. The mapping is extended to include all particles and not just those mirroring in the magnetic equator. Equations of motion for the particle mirror points in this representation are presented. The analysis results in a natural division of the magnetosphere into accelerator and dynamo regions and use of the coordinate system permits trajectory-dependent effects to be distinguished from source- or sink-dependent effects. The representation is illustrated by application to the analysis of the nose structure protons observed by Smith and Hoffman (1974).

    Notes: Phorbol diesters have been reported to stimulate the Na+/H+ antiport of a variety of cells including sea urchin eggs. Since stimulation of the Na+/H+ antiport is necessary for metabolic derepression during fertilization and protein kinase C is a target of phorbol diesters, enhanced Na+/H+ exchange during fertilization may be a result of protein kinase C activity. Protein kinase C is probably physiologically activated by diacylglycerols, which are derived from hydrolysis of phosphatidylinositol. Treatment of sea urchin eggs with 1,2-diacylglycerols was found to stimulate the Na+/H+ antiport. The 1,3-isomers were without effect. Further, the effects of 1,2-diacylglycerol and phorbol diester are not additive with respect to Na+/H+ exchange. While a direct participation of protein kinase C activity during fertilization remains to be demonstrated, these data support the hypothesis that protein kinase C activity plays a role in fertilization. However, the cytotoxic effect of protein kinase C activators suggests effects associated with their pleiotropic nature.

    Notes: The murine myelomonocytic leukemia cell line WEHI-3B D +, which differentiates in response to granulocyte colony stimulating factor (G-CSF), can also be induced to differentiate into monocyte-macrophages by phorbol myristate acetate (PMA) treatment, whereas the WEHI-3B D- subline, which is unresponsive to G-CSF and PMA, can be induced to differentiate to granulocytes as well as monocytes by, 1, 25-dihydroxycholecalciferol [1, 25-(OH)2 D3], the biologically active metabolite of vitamin D3. A newly developed variant of the WEHI-3B D+ line, named WEHI-3B D + G, which was responsive to G-CSF but not to PMA, was also differentiated to granulocytes by 1,25-(OH)2 D3. Although vitamin D3 has been reported to induce macrophage differentiation in responsive tumor cells, this is the first demonstration that 1,25-(OH)2 D3 can induce granulocyte differentiation. In both differentiation pathways, cessation of. cellular proliferation accompanies changes in morphologic and cytochemical properties of the cells. This suggests that leukemic cell lines unresponsive to differentiation agents acting at the cell surface retain their ability to differentiate in response to agents that do not act via the plasma membrane such as 1,25-(OH)2 D3, which has cytosolic/nuclear receptors. Vitamin D3 could act through different cellular pathways inducing differentiation or by bypassing only the first step of a common differentiation cascade used by agents with cell surface receptors such as CSF. These results suggest that low doses of 1,25-(OH)2 D3 may be useful in combination with hemopoietic growth factors (CSFs) as therapeutic agent to induce leukemic cell differentiation in vivo.

    Notes: We have isolated cDNA sequences from TPA-treated primary human fibroblasts, which indicate RNA species that are coordinately regulated after treatment of these cells with either ultraviolet light, mitomycin C, the UV-induced factor EPIF, or TPA. The levels of RNA are elevated in Bloom syndrome (cells of two out of three patients). After transformation with SV40 one of the sequences is overexpressed while another one is reduced. Both genes maintain their inducibility by the agents mentioned.

    Notes: A comparison of the binding and overall processing of epidermal growth factor (EGF) was made in ten mammalian fibroblast-like and three epithelial-like cell lines. EGF stimulated the growth of five fibroblastic cell lines (132 to 224%) after 10 days in the constant presence of EGF and were termed “responsive”. Eight of the lines did not respond or were growth inhibited by EGF (-64 to + 21%) and were listed as “nonresponsive”. Both “responsive” and “nonresponsive” cell lines possessed specific saturable membrane receptors for EGF, but no consistent differences were found between the number of apparent receptors per cell or the concentrations of 125I-EGF required for half maximal binding. However, a consistent difference between the “responsive” and “nonresponsive” cell lines was observed when the amount of cell associated 125I-labeled EGF was measured as function of incubation time at 37°C in the constant presence of the hormone. In every cell line we classified as responsive, the binding of 125I-EGF reached a maximum after 30 minutes incubation at 37°C and the cell associated radiolabel subsequently decreased by 42-68% within 4 hours. In contrast, the “nonresponsive” cell lines required 1-2 h to reach maximal binding and showed a minimal decrease of 1-10% during the 4-h period. These data indicate that a variety of different cell lines can possess receptors with similar binding properties but process EGF in a dissimilar manner. This difference in processing may reflect the specific events which are necessary for the induction of cell growth.

    Description: Fifteenth-order commensurability of the orbit of TETR-3 (1971-83B) is studied. The study is designed to obtain good discrimination of 15th-order resonances through a better range of inclinations. The first low inclination orbit, 33 deg, is used for this purpose; it is very sensitive to the high degree terms which were rather poorly represented by previously analyzed orbits.

    Notes: Recent work has shown that 19-norandrostenedione is a major steroidal component of porcine follicular fluid; however, little is known about its role(s) in the regulation of follicular function. This study was designed to examine the effect of 19-norandrostenedione on porcine oocyte maturation in vitro. Oocyte-cumulus complexes were isolated from medium (3-6-mm diameter)-sized prepubertal pig follicles and incubated for 12 h in medium with or without dibutyryl cyclic AMP ((Bu)2cAMP, 1 mM) with or without testosterone (5 x 10-7 M) or 19-norandrostenedione (5 x 10-7 M). In medium alone, 70.8% of oocytes spontaneously resumed meiosis as evidenced by the occurrence of germinal vesicle breakdown. Oocyte maturation was inhibited by (Bu)2cAMP (44.6% of oocytes matured). Although neither steroid alone affected maturation, both testosterone and 19-norandrostenedione enhanced the effect of (Bu)2cAMP (22.5 and 19.6%, respectively, resumed meiosis).The effects of testosterone and 19-norandrostenedione on (Bu)2cAMP-inhibited oocyte maturation were dose dependent and there was no significant difference between the actions of the steroids. The effect of 19-norandrostenedione was reversible and dependent on the presence of an intact cumulus. Hydroxyflutamide (SCH-16423), a nonsteroidal compound known to block androgen receptors, abolished the effects of both testosterone and 19-norandrostenedione on germinal vesicle breakdown, indicating that the actions of these steroids are truly androgenic.The results of this study suggest that 19-norandrostenedione may be of physiological importance in the regulation of porcine oocyte maturation.

    Description: A recent reanalysis of the International Latitude Serivce (ILS) polar motion data-day has been processed using Kalman filtering techniques to generate the polar motion excitation function over the time-span from 1960 to 1965. The resulting excitation function has been examined for the effects of 1960 Chile in an attempt to determine experimentally how large earthquake affect polar motion. The resulting upper bound of about 75 x 10 to the 22nd N-m for a 10-deg dip (about 36 x 10 to the 22nd N-m for 20-deg dip) is consistent with results obtained from previous seismic studies, including a recent normal mode excitation result. Following future great earthquakes, monitoring of polar motion by space-based techniques such as VLBI should continue at high temporal resolution for several weeks in order to directly measure the rheological parameters of the upper mantle.
