    Description: Background: Almond witches’-broom (AlmWB), a devastating disease of almond, peach and nectarine in Lebanon, is associated with ‘Candidatus Phytoplasma phoenicium’. In the present study, we generated a draft genome sequence of ‘Ca. P. phoenicium’ strain SA213, representative of phytoplasma strain populations from different host plants, and determined the genetic diversity among phytoplasma strain populations by phylogenetic analyses of 16S rRNA, groEL, tufB and inmp gene sequences. Results: Sequence-based typing and phylogenetic analysis of the gene inmp, coding an integral membrane protein, distinguished AlmWB-associated phytoplasma strains originating from diverse host plants, whereas their 16S rRNA, tufB and groEL genes shared 100 % sequence identity. Moreover, dN/dS analysis indicated positive selection acting on inmp gene. Additionally, the analysis of ‘Ca. P. phoenicium’ draft genome revealed the presence of integral membrane proteins and effector-like proteins and potential candidates for interaction with hosts. One of the integral membrane proteins was predicted as BI-1, an inhibitor of apoptosis-promoting Bax factor. Bioinformatics analyses revealed the presence of putative BI-1 in draft and complete genomes of other ‘Ca. Phytoplasma’ species. Conclusion: The genetic diversity within ‘Ca. P. phoenicium’ strain populations in Lebanon suggested that AlmWB disease could be associated with phytoplasma strains derived from the adaptation of an original strain to diverse hosts. Moreover, the identification of a putative inhibitor of apoptosis-promoting Bax factor (BI-1) in ‘Ca. P. phoenicium’ draft genome and within genomes of other ‘Ca. Phytoplasma’ species suggested its potential role as a phytoplasma fitness-increasing factor by modification of the host-defense response.

    Description: Species introduction, combined with changing access rules, increasing demand, and new road and dam infrastructure, are contributing to remarkable changes in Bolivian Amazon fisheries. This paper examines community responses to the appearance of a commercially valuable introduced fish species, Arapaima cf. gigas (“paiche”) in the Bolivian Amazon. Until the end of the 20th century, fisheries in this region were relatively low intensity, focused in rivers on a small number of native large-sized species by an urban-based commercial fishing fleet, and in floodplain lakes on a high diversity of native medium-sized species for subsistence by rural indigenous communities. In the seventies, Arapaima cf. gigas was introduced from Peru and has since invaded a significant portion of the Madre de Dios and Beni basins in northern Bolivia. This species now represents up to 80 % of commercial catches for the region. Occupying primarily floodplain lakes, many of which are located within indigenous territories, it has created economic opportunities and stimulated conflicts. The evolution of fisheries in one indigenous Tacana community is described, and the perspectives of local fishers are explored. Results suggest that while the new resource has strengthened incipient community-level organization, the current capture strategies and management mechanisms may not be conducive to sustainability or equitable distribution of returns. Commercial fisheries targeting a set of native species have been replaced by a single-species fishery in this community, raising questions about how the changes both in the resource-base and associated livelihood strategies are impacting system resilience. Ecosystem impacts of the introduction remain unclear. Paiche is viewed both as a potential threat and an opportunity by indigenous fishers. The management of this introduced species for a maximum social benefit and minimal environmental damage are topical concerns for communities and government actors and should be treated carefully considering local and broader, regional-scale implications.

    Description: Almost 50 years ago Robert Dahl maintained “we can […] reasonably hope 1 day to achieve great democratic cities. As the optimum unit for democracy in the 21st century, the city has a greater claim, I think, than any other alternative” (Am Polit Sci Rev 61: 953–970, 1967 ). This article intends to ascertain whether the words of one of the greatest scholars of democracy have had a concrete outcome along the pathways taken by democratic theory and whether, therefore, as was the case in classical theory, real superiority has therefore been restored to the city compared with the other territorial institutions of democracy. In this article we begin with two assumptions, each concerning the theoretical status of democratic theory. The first maintains that a realistic and an idealistic dimension coexist in variable dimensions in theoretical democratic models. According to the second assumption, it can be stated that democratic theory envisages the presence of a local territorial dimension, the importance of which is nevertheless variable in the different theoretical models. The thesis we intend to demonstrate here is that the variable nature of the importance of the local-urban territorial dimension depends on the type of balance created between the idealistic and realistic dimensions of the different models of democracy. Concluding, we aim to theoretically demonstrate why (and at which conditions) the city can become, more and better than any other institutional place, the ultimate arena within which the best results may be achieved for democracy in 21st century.

    Description: Background: Workplace sedentary behaviour is a priority target for health promotion. However, little is known about how to effect change. We aimed to explore desk-based office workers’ perceptions of factors that influenced sedentary behaviour at work and to explore the feasibility of using a novel mobile phone application to track their behaviours. Methods: We invited office employees (n = 12) and managers (n = 2) in a software engineering company to participate in semi-structured interviews to explore perceived barriers and facilitators affecting workplace sedentary behaviour. We assessed participants’ sedentary behaviours using an accelerometer before and after they used a mobile phone application to record their activities at self-selected time intervals daily for 2 weeks. Interviews were analysed using a thematic framework. Results: Software engineers (5 employees; 2 managers) were interviewed; 13 tested the mobile phone application; 8 returned feedback. Major barriers to reducing workplace sedentary behaviour included the pressure of ‘getting the job done’, the nature of their work requiring sitting at a computer, personal preferences for the use of time at and after work, and a lack of facilities, such as a canteen, to encourage moving from their desks. Facilitators for reduced sedentariness included having a definite reason to leave their desks, social interaction and relief of physical and mental symptoms of prolonged sitting. The findings were similar for participants with different levels of overall physical activity. Valid accelerometer data were tracked for four participants: all reduced their sedentary behaviour. Participants stated that recording data using the phone application added to their day’s work but the extent to which individuals perceived this as a burden varied and was counter-balanced by its perceived value in increasing awareness of sedentary behaviour. Individuals expressed a wish for flexibility in its configuration. Conclusions: These findings indicate that employers’ and employees’ perceptions of the cultural context and physical environment of their work, as well as personal factors, must be considered in attempting to effect changes that reduce workplace sedentary behaviour. Further research should investigate appropriate individually tailored approaches to this challenge, using a framework of behaviour change theory which takes account of specific work practices, preferences and settings.

    Description: Background: For the development of a sustainable energy system, the public’s contribution to the sustainment of the system’s current operations will become vital. The public’s role is expected to change from that of passive service abiders to active service providers, as anticipated by visions of prosumers, smart grids, demand side management, virtual power plants, and electric vehicle management. Smart technology and new business models will increase system complexity and the experience of uncertainty and risk for all parties. Actionability of the public’s participation in the opportunities arising from the transformation of energy systems will become a problem. Methods: With a qualitative functionalist method, we analyze the prerequisites of the public’s participation: the capacity of social mechanisms, such as trust, for the solution of social problems. Functional problems affect the continuation of communication, i.e., the possibility of the sustainment of social reality. We isolate three case studies that illustrate concrete effects of trust, distrust, and confidence on three parties, respectively: consumers, investors, and those affected by grid extensions.Results and conclusionsWe argue that the social mechanisms of trust and confidence are more vital for consumers, investors, and those affected than previous research has noticed. These mechanisms determine the achievement of sustainability in energy systems by sustaining actionability as a favorable expectation (trust, confidence), or an unfavorable expectation (lack of trust, distrust). Even lack of trust and distrust can be valuable contributors on the road to sustainability, since they uncover flaws and setbacks of a fledgling smart grid.

    Description: Background: Diets rich in the n-3 fatty acid alpha-linolenic acid (ALA) have been shown to reduce breast tumor growth, enhance the effectiveness of the HER2-targeted drug trastuzumab (TRAS) and reduce HER2 signaling in mouse models. It is unclear whether this is due to direct effects of ALA or due to its long-chain n-3 fatty acids metabolites including docosahexaenoic acid (DHA). Methods: The ability of HER2-overexpressing BT-474 human breast cancer cells to convert ALA to long-chain n-3 fatty acids was determined by measurement of phospholipid fatty acids by gas chromatography following treatment with 100 μM ALA. The effects of 96 h treatment with ALA or DHA, at serum levels seen in mice (50–100 μM), alone and combined with TRAS (10 μg/ml), on BT-474 cell growth measured by trypan blue exclusion, apoptosis measured by flow cytometric analysis of Annexin-V/7-AAD stained cells (ALA and TRAS treatment only) and protein biomarkers HER2 signaling measured by western blot were determined. Results: ALA-treated BT-474 cells had higher phospholipid ALA but no increase in downstream n-3 metabolites including DHA. Both ALA and DHA reduced cell growth with and without TRAS. ALA had no effect on apoptosis. ALA and DHA showed opposite effects on Akt and MAPK phosphorylation; ALA increased and DHA decreased phosphorylation. Conclusions: Together these data suggest that, while both ALA and its DHA metabolite can reduce HER2-overexpressing breast cancer growth with and without TRAS, they demonstrate for the first time that DHA is responsible for the effects of ALA-rich diets on HER2 signaling pathways.

    Description: Background: Despite the significant interest in β 2 -Adrenergic receptor (ADRB2) polymorphisms related to asthma, whether ADRB2 genetic variants are similarly associated with acute respiratory tract infections have not been studied. We hypothesized that genetic variants in ADRB2 associated with a response to asthma therapy during an asthma exacerbation were also associated with severity of acute respiratory tract infections. Methods: To test this hypothesis, we genotyped 5 common polymorphisms in the promoter region and coding block of the ADRB2 gene (loci -2387, -2274, -1343, +46, and +79) from 374 Caucasian and African American term infants who were enrolled at the time of acute respiratory illness over four respiratory viral seasons. Severity of respiratory tract infections was measured using a bronchiolitis severity score (BSS; range = 0-12, clinically significant difference = 0.5) with a higher score indicating more severe disease. We assigned the promoter, coding and combined promoter and coding haplotypes to the unphased genotype data. The associations between each of these five single-nucleotide polymorphisms (SNPs) as well as the haplotypes and infant BSS were analyzed using nonparametric univariate analysis and multivariable proportional odds model separately in Caucasians and African Americans. Results: There was no significant association between infant BSS and each of the SNPs in both Caucasians and African Americans. However, promoter haplotype CCA was associated with a decreased BSS in African Americans in a dose dependent manner. The median (interquartile range) BSS of infants with no copies of the CCA haplotype, one copy, and two copies of the CCA haplotype were 5.5 (2.0, 8.0), 4.0 (1.0, 7.5), and 3.0 (1.0, 4.0), respectively. This dose dependent relationship persisted after adjusting for infant age, gender, daycare exposure, secondhand smoke exposure, prior history of breastfeeding, siblings at home, and enrollment season (adjusted odds ratio: 0.59, 95 % confidence interval: 0.36, 0.98). There was no similar protective relationship of haplotype CCA on severity of respiratory tract infections identified in Caucasians. Conclusions: ADRB2 genotype may be predictive of severity of acute respiratory tract infections in African Americans, and potentially identify a subset of infants who may respond to beta-agonist therapy.

    Description: Background: Carbonic anhydrases (CAs, EC 4.2.1.1) are ubiquitous enzymes that catalyze the reversible hydration reaction of carbon dioxide. CAs are present as six structurally divergent enzyme families: α, β, γ, δ, ζ and η. β-CAs have a wide distribution across different species including invertebrates. Previously, we showed that Drosophila melanogaster β-CA is a highly active mitochondrial enzyme. In this study, we investigated the function of Drosophila β-CA by silencing the expression of the β-CA gene using UAS/GAL4-based RNA interference (RNAi) in Drosophila in vivo. Results: Crossing β-CA RNAi lines over ubiquitous Actin driver flies did not produce any viable progeny, indicating that β-CA expression is required for fly development. RNAi silencing of β-CA ubiquitously in adult flies did not affect their survival rate or function of mitochondrial electron transport chain. Importantly, β-CA RNAi led to impaired reproduction. All β-CA knockdown females were sterile, and produced few or no eggs. Whole ovaries of knockdown females looked normal but upon cadherin staining, there was an apparent functional defect in migration of border cells, which are considered essential for normal fertilization. Conclusions: These results indicate that although Drosophila β-CA is dispensable for survival of adult flies, it is essential for female fertility.

    Description: The paired analyses of the Mg/Ca ratio and oxygen isotopic composition (δ 18 O c ) of surface-dwelling planktonic foraminifera have become a widely used method for reconstructing the oxygen isotopic composition of ambient seawater (δ 18 O sw ) as a robust proxy for surface salinity. Globigerinoides ruber (G. ruber) is a mixed-layer dweller, and its fossil shell is an ideal archive for recording past sea surface water conditions, such as those caused by variability in the East Asian summer monsoon (EASM). Here, we investigate the validity of shell-derived δ 18 O sw estimates for G. ruber using core-top sediments from the East China Sea (ECS). First, we determined a local δ 18 O sw –salinity equation for the eastern part of the ECS in July [δ 18 O sw  = −7.74 + 0.23 × salinity]. Then, we calculated δ 18 O sw from core-top δ 18 O c and Mg/Ca values in G. ruber using the δ 18 O c –temperature equation of Bemis et al. (Paleoceanography 13(2):150–160, 1998) and the Mg/Ca–temperature equation of Hastings et al. (EOS 82:PP12B-10, 2001). The core-top δ 18 O sw and salinity were estimated to be in the ranges of −0.2 to +0.39 ‰ and 33.7 to 34.5, respectively, which fall close to the local δ 18 O sw –salinity regression line. The core-top data showed that the Mg/Ca–temperature calibration by Hastings et al. (EOS 82:PP12B-10, 2001) and the δ 18 O c –temperature equation by Bemis et al. (Paleoceanography 13(2):150–160, 1998) are appropriate for calculating δ 18 O sw in the ECS. Furthermore, we measured core-top Ba/Ca ratios of G. ruber (Ba/Ca G. ruber ), which ranged from 0.66 to 2.82 μmol mol −1 . There was not a significant relationship between the salinity and Ba/Ca G. ruber ratios due to the highly variable Ba/Ca G. ruber data. Given the seawater Ba/Ca data and the published partition coefficient for Ba (D Ba  = 0.15–0.22), pristine Ba/Ca G. ruber ratios at northern Okinawa Trough sites should be less than 0.84 μmol mol −1 . Anomalously high Ba/Ca G. ruber ratios (〉0.84 μmol mol −1 ) might be attributable to contamination by sedimentary barite adherent on fossil shells. Therefore, further evaluation of the Ba/Ca G. ruber ratio as a paleo-salinity proxy requires diethylene triamine pentaacetic acid (DTPA)-cleaned Ba/Ca data that can minimize the influence of barite contamination.
