    Notizen: Financial accounting regulatory arrangements adopted in most Western countries often involve the participation of (and interactions between) public-sector and private-sector regulatory agencies. The political process associated with the development of accounting rules not only involves the efforts of interested parties seeking to secure the content of rules favourable to their interests but also the behaviours of regulatory agencies as they compete to influence or control the regulatory ‘agenda’.Regulatory agencies develop their own agendas, in light of their own perceived priorities and the regulatory initiatives of other agencies. The placing of an accounting issue on the agenda of one agency may be warmly supported by other agencies or, alternatively, be viewed as a threat to the regulatory ambitions of those other agencies.This case study reviews the emergence and eventual resolution of an accounting issue that was initially promoted by a government agency in April 1984 but then ultimately developed as a professional accounting standard in December 1991. The study describes the activities of public and private-sector agencies during this eight-year period as they responded to proposals for the introduction of disclosure rules concerning cash flows. Evidence was obtained from public records, documents provided by interest groups, and structured and semi-structured interviews with key participants in the events described.The analysis is underscored by our understanding of competitive regulatory interactions and formal models for analysing agenda entrance (Cobb et al. 1976). This analysis shows how the profession's standard-setting body was unable to control the global agenda for accounting rule-making as a consequence of the intervention of another body, the Australian Stock Exchange.

    Notizen: Regulatory arrangements adopted in most Western countries involve the participation of both private-sector (professional) bodies and public-sector agencies in the formulation and administration of accounting rules. The political processes associated with the efforts of interest groups to secure favoured outcomes may involve disputes over regulatory arrangements as well as the content of specific rules.This case study reviews inter-organizational conflict during the 1980s between government regulatory bodies and the accounting profession as they handled proposals for the introduction of disclosure rules concerning related party transactions (RPTs).The case study illustrates how debates about the content and drafting of rules on RPTs took place in a complex organizational setting; how the activities of the profession's standard-setting bodies were hardly consistent with the pluralist ideal; but that when considered in the context of complex interactions between different agencies, the notion of ‘pluralism’ is of limited value when describing the underlying political processes involved in the development of accounting rules; and how simple models of‘corporatism’ failed to describe the dynamics of political processes surrounding particular issues. It is suggested that identifying the ‘domains’ of major participants in the standard-setting process, and analysing threats to these domains, may provide richer descriptions of regulatory processes than have been presented previously.

    Notizen: Abstract Cancer was an obvious disease to study by magnetic resonance spectroscopy (MRS); it produces large lesions that give clearly abnormal spectra, all treatment methods leave much to be desired, and radiotherapy, in particular, is limited by tissue hypoxia, a process that can be investigated by MRS.31P MRS has shown that tumor cells are not acidic, as had been thought; instead, the pH gradient across the tumor cell membrane is the reverse of that in a normal cell. This change in hydrogen-ion gradient is accompanied by changes in gradients of many other ions. Tumor oxygenation can be monitored in animal tumor models using the techniques employed for functional magnetic resonance imaging (MRI) of the brain. Large changes in signal are observed when drugs that reduce tumor blood flow are administered. 1H NMR spectra of acid extracts of tumor or normal tissue biopsies contain sufficient information to permit classification (and thus, perhaps, diagnosis) if computer-based pattern recognition techniques are employed. Surprisingly, the same technique gives quite good classification of31P spectra takenin vivo. Can MRS beappliedin cancer therapy? Studies on tumor ion balance will help in the design of anticancer drugs and other therapies. Tumor blood flow studies using MRI could be applied to individual patients to predict the usefulness of radiotherapy and to assist in radiotherapy planning. Pattern recognition methods could automate the screening of biopsies and could also assist in interpretation of human spectra takenin vivo.

    Notizen: Abstract Intact chloroplasts were isolated from sugarbeet leaves by the mechanical disruption technique normally used for spinach. The chloroplast pellet contained a ring of white irregularly shaped crystals which were identified as calcium oxalate. The chloroplasts were greater than 90% intact yet good rates of CO2 fixation were only obtained when inorganic pyrophosphate or 3-phosphoglycerate were added to the assay medium. Chloroplasts free of calcium oxalate were prepared by purification on a three step Percoll gradient. These purified chloroplasts were highly intact and showed high rates of CO2 fixation without adding inorganic pyrophosphate or 3-phosphoglycerate. With optimal assay conditions (0.2 mM orthophosphate and pH 8.0) rates of 110–130 μ mole per milligram chlorophyll per hour were routinely obtained. It is concluded that intact chloroplasts capable of high rates of CO2 fixation can be prepared from sugarbeet leaves using a simple three step Percoll gradient.

    Beschreibung: We have undertaken the first paleoseismological study on an upper plate fault in Chile. The selected structure was the Mejillones Fault, which is marked by a conspicuous fault-scarp. Using cosmogenic 10Be and OSL dating and detailed sedimentary logging of trenches, we have constrained the abandonment of two alluvial surfaces by fault activity at ca. 35 ka and ca.14 ka. Based on stratigraphic observation we characterized the fault evolution in four intervals over the last ca. 35 ka. During the first three intervals the fault had a steady slip rate of 0.61 ± 0.26 m/ka. The fourth interval is delineated by the last vertical fault slip and the accumulation of un-deformed hillslope deposits after ca. 3.3 ka and has a slip rate of 0.22 ± 0.06 m/ka. The younger surface abandonment was caused by two Mw ∼ 7 paleoearthquakes with a recurrence interval of 5.0 ± 3.5 ka. The third interval is characterized by the interaction of hillslope deposits and aseismic slip and/or centimeter scale seismic slip events. At ca. 3.5 ka, a last large (Mw ∼ 6.6) earthquake took place. The recurrence intervals of large (Mw 〉 8.5) subduction earthquakes do not appear to be the same as the recurrence intervals of the Mw ∼ 7 events on the upper plate Mejillones Fault.

    Beschreibung: We have previously demonstrated an increased DNA copy number and expression of IGF1R to be associated with poor outcome in Wilms tumors. We have now tested whether inhibiting this receptor may be a useful therapeutic strategy by using a panel of Wilms tumor cell lines. Both genetic and pharmacological targeting resulted in inhibition of downstream signaling through PI3 and MAP kinases, G1 cell cycle arrest, and cell death, with drug efficacy dependent on the levels of phosphorylated IGF1R. These effects were further associated with specific gene expression signatures reflecting pathway inhibition, and conferred synergistic chemosensitisation to doxorubicin and topotecan. In the in vivo setting, s.c. xenografts of WiT49 cells resembled malignant rhabdoid tumors rather than Wilms tumors. Treatment with an IGF1R inhibitor (NVP-AEW541) showed no discernable antitumor activity and no downstream pathway inactivation. By contrast, Wilms tumor cells established orthotopically within the kidney were histologically accurate and exhibited significantly elevated insulin-like growth factor–mediated signaling, and growth was significantly reduced on treatment with NVP-AEW541 in parallel with signaling pathway ablation. As a result of the paracrine effects of enhanced IGF2 expression in Wilms tumor, this disease may be acutely dependent on signaling through the IGF1 receptor, and thus treatment strategies aimed at its inhibition may be useful in the clinic. Such efficacy may be missed if only standard ectopic models are considered as a result of an imperfect recapitulation of the specific tumor microenvironment.

    Beschreibung: Patient-derived organoids (PDOs) have recently emerged as robust preclinical models; however, their potential to predict clinical outcomes in patients has remained unclear. We report on a living biobank of PDOs from metastatic, heavily pretreated colorectal and gastroesophageal cancer patients recruited in phase 1/2 clinical trials. Phenotypic and genotypic profiling of PDOs showed a high degree of similarity to the original patient tumors. Molecular profiling of tumor organoids was matched to drug-screening results, suggesting that PDOs could complement existing approaches in defining cancer vulnerabilities and improving treatment responses. We compared responses to anticancer agents ex vivo in organoids and PDO-based orthotopic mouse tumor xenograft models with the responses of the patients in clinical trials. Our data suggest that PDOs can recapitulate patient responses in the clinic and could be implemented in personalized medicine programs.
