    Description / Table of Contents: The superposition of stratified rocks is an unmistakable manifestation of the history of sedimentary processes through deep time. However, the relationship between the preserved strata of the rock record and the passage of geological time, indisputable in principle, is unknowable in detail; incompleteness is an essential property of the record. That gaps exist at all scales in sedimentary successions is easily demonstrated from consideration of sediment accumulation rates, and expectations of continuity and completeness at any scale are correspondingly inadvisable. Locating and quantifying the gaps in the record is, however, very much less straightforward. Predictive modelling of strata – essential for their practical exploitation – requires such geohistorical understanding, yet over-simplified assumptions about how time is represented in rock can still lead to inadequate or even false conclusions. The contributions to this volume describe a range of practical studies, theoretical investigations, and numerical experiments in which the nature of the strata–time relationship is explored.

    Notes: Abstract  Retinoic acid (RA), a derivative of vitamin A, plays a critical role as a signaling molecule in axial patterning of vertebrates. Here we report that RA exposure of zebrafish (Danio rerio) and mummichog (Fundulus heteroclitus) embryos during gastrulation results in homeotic duplications of the pectoral fins in up to 94% of fish. We have observed three to four pairs of fins in an individual fish. Although some duplications are partial, many represent complete axial duplications of the pectoral girdle and fin and include coracoscapulae, proximal radials, and dermal fin elements. Fin duplications are observed only at a defined dose of RA. Inhibition of RA synthesis by exposure to citral during a narrow developmental window leads to fish which lack pectoral fins but can be rescued by addition of exogenous RA, suggesting that RA signaling is critical to fin specification during early development. The ability to consistently induce multiple fins in a large number of vertebrate embryos should contribute to the understanding of genetic regulation of the normal positioning of limbs during embryogenesis.

    Notes: Summary The growth of Rhodotorula glutinis is inhibited by both D-threo chloramphenicol and an L-threo isomer of chloramphenicol (lacking the dichloroacetyl group), causing an increase in the mean generation time, in a variety of media, approximately proportional to the concentration of antibiotic. The antibiotic is not removed from the growth medium in any quantity during this inhibition of growth. The oxygen uptakes of normal and chloramphenicol-grown cells of R. glutinis are similar when expressed on a dry weight basis. The oxygen uptake of normal and L-threo isomer-grown cells is strongly inhibited by antimycin A, whereas D-threo chloramphenicol-grown cells are unaffected. There was no evidence to suggest that any uncoupling of phosphorylation occurred with either isomer. Pythium ultimum mycelium also showed similar oxygen uptakes per unit dry weight whether grown in the presence or absence of D-threo chloramphenicol. The D-threo chloramphenicol-grown mycelium was also insensitive to antimycin A in contrast to the normal mycelium which was strongly inhibited. P. ultimum grows slowly in the presence of 100 μg/ml D-threo chloramphenicol in a glucose salts medium, but is completely inhibited by a similar concentration in a glycerol salts medium. The L-threo isomer does not inhibit the growth of P. ultimum. The mitochondria of Rhodotorula glutinis show a progressive disorganization when grown in the presence of increasing concentrations of D-threo chloramphenicol up to 1000 μg/ml. There is an associated over synthesis of cell wall material in the higher concentrations of the antibiotic. The L-threo isomer produces no obvious fine structural abnormalities even at concentrations of 1000 μg/ml.

    Notes: Summary Microtubular structures, apparently continuous with the plasmalemma, have been observed in thin sections of two strains of group D streptococcal L-forms. The tubules had an external diameter of about 250 Å and a hollow core 100–150 Å in diameter. The tubules were found protruding either into or out of the L-form cells and were only found in cultures growing in the presence of penicillin.

    Notes: Summary Spherical or elongated phospholipid inclusions have been observed in vacuoles of Saccharomyces cerevisiae. The periodicity of these lipid inclusions is 56 Å and the material of which they are composed appears to be derived from spherosomes. The spherosomes arise in the cytoplasm, penetrate the tonoplast, and discharge their contents into the vacuole. It is postulated that the phospholipid matrix of the spherosome then aggregates to produce the myelin-like inclusions. Tween 80 and ergosterol together in the growth medium do not prevent the formation of the lipid inclusions.

    Notes: Summary L-form colonies derived from a Group D streptococcus showed alterations in fine structure during propagation. For the first 6 months after preparation the L-forms were capable of reversion and fibrillar and lamellate inclusions were found in some cells. Fibrillar bundles, composed of fibrils with an external diameter of 12.5 nm were found attached to the cytoplasmic membrane of the large 20 μm surface cells. Small, 1 μm cells within the agar contained lamellate inclusions with a periodicity of 14 nm, also attached to the cytoplasmic membrane. In empty cells both inclusions appeared as hollow fibrils. The composition of these structures is not known. Long filamentous evaginations of the cytoplasmic membrane were also found in a small proportion of cells. After 15 months' propagation, when the L-form had become stable, the inclusions and filaments were no longer visible.

    Notes: Summary Apical growth of hyphae of Fusarium culmorum was demonstrated using an immunofluorescent labelling technique. An antiserum prepared against hyphal tips contained a series of antibodies, detected by immunodiffusion, not present in antisera against mature hyphae or conidia. Absorption of the tip antiserum with hyphae allowed a specific immunofluorescence reaction with hyphal tips only. The antiserum against mature hyphae gave non-fluorescent tips to the hyphae.

    Notes: Summary The first stage in the formation of a bud in Rhodotorula glutinis is the production of a tapered plate of new wall material between the existing wall and the plasmalemma. The parent cell wall is lysed, allowing the bud to emerge enveloped in this new wall. Mucilage is synthesised to surround the developing bud. As the bud grows a septum forms centripetally dividing the two cells. When the daughter cell reaches maximum size the septum cleaves along its axis, producing the bud scar on the parent cell and the birth scar on the daughter cell. The birth scar is obliterated later as the wall of the young cell grows. A system of endoplasmic reticulum and vesicles is found in young buds and is thought to be responsible for the transport of wall material precursors.
