    Notes: Summary CAROLINE I represents a DDC software package utilizing to full advantage the power of modern microcomputer systems in a multitasking environment. The software in no way imposes restrictions on the number of configurable elements as I/O channels and function calls. More than 100 functions allow a rigorous process control concerning events depending on time and logic. The user interface is adaptable to any process setup by configurable menus whose number is limited only by the memory capacity of the computer. The menu configuration includes graphic layouts and the actions performed in response to operator input. Special data structures allow the integration of data acquired by analytical instruments such as mass spectrometers, IR-analyzers etc. The software is written in ‘C’ and easily portable to any multitasking operating system like OS9, real time UNIX or QNX. The tests were run using OS9 on VME computer systems.

    Notes: Abstract We examined how hand-trajectory smoothness changed during the practice of a motor task where smoothness was quantified by jerk-cost. Four human subjects each moved his nondominant arm between an upper target and a lower target, while circumnavigating a barrier that extended outward from the vertical plane of the targets. The two targets and the barrier placed boundary constraints on hand trajectories, but the motion was not restrained in any other way. Arm movements were recorded on high-speed ciné film, and linear and angular kinematical data were obtained for all arm segments. In each of 100 practice trials, subjects attempted to minimize movement time. After the practice trials, subjects repeated the same motor task but at movement times corresponding to the slowest, mid-range and fastest motion that had occurred during practice. Thus, jerk-cost could be compared for movements of different speeds during practice and after practice. Because the movement task contained several changes in hand-path direction, the changes in the vector characteristics of the hand accelerations were expected to be important for explaining the modulations in jerk-cost with practice. Total jerk-cost, therefore, was calculated as well as the separate magnitudinal and directional jerk-cost components. During practice, total movement time decreased, hand paths became more parabolic in shape, and significant changes occurred in hand acceleration magnitude, direction, and timing. Total jerk-cost and the magnitudinal and directional jerk-cost components were significantly less when slowest hand movements were compared after practice versus during practice. The decrease in jerk-cost indicated an increased smoothness of the practiced movements.

    Notes: Summary A new hydrogen bacterium has been isolated by enrichment culture on hydrogen, carbon dioxide, and oxygen. It is a strictly aerobic, Gram-positive, immotile rod. Irregular cell shapes, the snapping type of cell division and the partial fragmentation of the rods into coccoid cells in older heterotrophic cultures indicate that strain 14 g is a coryneform bacterium. Strain 14 g grows well in a mineral salts medium under a gas atmosphere of 80% hydrogen, 10% carbon dioxide and 10% oxygen. With increasing oxygen partial pressures, the growth rate decreases. While growth is good or excellent on organic acids, carbohydrates are utilized not at all. Autotrophically grown cells are able to oxidize organic substrates after a relatively short lag-phase. Growth is retarded when hydrogen is present in the gas atmosphere. Citrate utilization is even inhibited by hydrogen. In gluconate grown cells gluconokinase, the enzymes of the Entner-Doudoroff system and a NAD-dependent 6-phosphogluconate dehydrogenase are present. All enzymes are inducible. The uptake rate of the oxygen-hydrogen mixture by resting cells is very high and amounts to 2500 μl gas/mg protein·h. Carbon dioxide does not stimulate the oxyhydrogen-reaction. The hydrogenase activity of intact cells measured with methylene blue as a hydrogen acceptor is limited by the penetration of methylene blue into the cell. In cell-free extracts the hydrogen uptake rate amounts to 2260 μl H2/mg protein·h with methylene blue as H-acceptor. The hydrogenase of strain 14 g is a strictly inducible enzyme. It is completely part culate and does not reduce pyridime nucleotides.

    Notes: Abstract Six new strains of Alcaligenes enriched for and isolated as nickel-resistant bacteria resemble Alcaligenes eutrophus H16 and contain both an NAD-reducing, tetrameric soluble hydrogenase and a membrane-bound hydrogenase. None of the soluble hydrogenases share with the Rhodococcus opacus MR11 enzyme tetramer the property of being cleaved easily into two dimeric moieties [a hydrogenase (βδ) and an NADH:acceptor oxidoreductase (αγ)], in the absence of nickel or at low ionic strength. The soluble hydrogenase of the newly isolated strain MR22 of R. opacus equalled that of strain MR11. The absence of a membrane-bound hydrogenase in Alcaligenes denitrificans strain 4a-2 and in Alcaligenes ruhlandii was confirmed.

    Notes: Abstract A Gram-negative bacterial strain, identified as Acidovorax facilis strain 72W, has been isolated from soil by enrichment using 2-ethylsuccinonitrile as the sole nitrogen source. This strain grows on a variety of aliphatic mono- and dinitriles. Experiments using various heating regimes indicate that nitrile hydratase, amidase and nitrilase activities are present. The nitrilase is efficient at hydrolyzing aliphatic dinitriles to cyanoacid intermediates. It has a strong bias for C3–C6 dinitriles over mononitriles of the same chain length. Whole, resting cell hydrolysis of 2-methylglutaronitrile results in 4-cyanopentanoic acid and 2-methylglutaric acid as the major products. Heating, at least 20 min at 50 °C, eliminates nitrile hydratase and amidase activities, resulting in greater than 97% selectivity to 4-cyanopentanoic acid. The nitrilase activity has good heat stability, showing a half-life of 22.7 h at 50 °C and a temperature optimum of at least 65 °C for activity. The strain has been deposited as ATCC 55746.

    Notes: Abstract Here we describe experiments aimed at developing an element-selective ion source for thorium (Th). The technique applied is resonance ionization spectroscopy (RIS) with a thermal atom beam. Ionization schemes for isotopically nonselective ionization of Th as well as for isotopically selective ionization of 230Th are proposed. The RIS-scheme used is two-photon two-colour ionization with excitation in the ultraviolet spectral range between 244 nm and 267 nm or in the visible spectral range between 485 nm and 529 nm. Ionization of the excited atoms is performed either by ultraviolet photons or by visible photons, depending on the energy required for this process.
