    Description: In vitro selection allows rare functional RNA or DNA molecules to be isolated from pools of over 10(15) different sequences. This approach has been used to identify RNA and DNA ligands for numerous small molecules, and recent three-dimensional structure solutions have revealed the basis for ligand recognition in several cases. By selecting high-affinity and -specificity nucleic acid ligands for proteins, promising new therapeutic and diagnostic reagents have been identified. Selection experiments have also been carried out to identify ribozymes that catalyze a variety of chemical transformations, including RNA cleavage, ligation, and synthesis, as well as alkylation and acyl-transfer reactions and N-glycosidic and peptide bond formation. The existence of such RNA enzymes supports the notion that ribozymes could have directed a primitive metabolism before the evolution of protein synthesis. New in vitro protein selection techniques should allow for a direct comparison of the frequency of ligand binding and catalytic structures in pools of random sequence polynucleotides versus polypeptides.

    Description: We present a method that can significantly increase the complexity of protein libraries used for in vitro or in vivo protein selection experiments. Protein libraries are often encoded by chemically synthesized DNA, in which part of the open reading frame is randomized. There are, however, major obstacles associated with the chemical synthesis of long open reading frames, especially those containing random segments. Insertions and deletions that occur during chemical synthesis cause frameshifts, and stop codons in the random region will cause premature termination. These problems can together greatly reduce the number of full-length synthetic genes in the library. We describe a strategy in which smaller segments of the synthetic open reading frame are selected in vitro using mRNA display for the absence of frameshifts and stop codons. These smaller segments are then ligated together to form combinatorial libraries of long uninterrupted open reading frames. This process can increase the number of full-length open reading frames in libraries by up to two orders of magnitude, resulting in protein libraries with complexities of greater than 10(13). We have used this methodology to generate three types of displayed protein library: a completely random sequence library, a library of concatemerized oligopeptide cassettes with a propensity for forming amphipathic alpha-helical or beta-strand structures, and a library based on one of the most common enzymatic scaffolds, the alpha/beta (TIM) barrel.     Description: Convergence rates between the Juan de Fuca and North American plates are calculated by means of their relative, present-day pole of rotation. A method of calculating the propagation of errors in addition to the instantaneous poles of rotation is also formulated and applied to determine the Euler pole for Pacific-Juan de Fuca. This pole is vectorially added to previously published poles for North America-Pacific and 'hot spot'-Pacific to obtain North America-Juan de Fuca and 'hot spot'-Juan de Fuca, respectively. The errors associated with these resultant poles are determined by propagating the errors of the two summed angular velocity vectors. Under the assumption that hot spots are fixed with respect to a mantle reference frame, the average absolute velocity of the Juan de Puca plate is computed at approximately 15 mm/yr, thereby making it the slowest-moving of the oceanic plates.

    Description: A revised model is presented of the tectonic evolution of the Juan de Fuca ridge by propagating rifting. The new model has three different relative rotation poles, covering the time intervals 17.0-8.5 Ma, 8.5-5.0 Ma, and 5.0 Ma to the present. The rotation pole shifts at 8.5 and 5.0 Ma imply clockwise shifts in the direction of relative motion of 10 deg to 15 deg. At each of these shifts, the pattern of propagation reorganizes, and the new ridges formed by propagation are at an orientation closer to orthogonal to the new direction of motion than the orientation of the preexisting ridges. The model, containing a total of seven propagation sequences, shows excellent agreement with the isochrons inferred from the magnetic anomaly data, except in areas complicated by the separate Explorer and Gorda plates. The agreement between model and data near the Explorer plate breaks down abruptly at an age of about 5 Ma, indicating that the probable cause of the rotation pole shift at that time was the separation of the Explorer plate from the Juan de Fuca plate.

    Description: Tests were conducted on two different types of pumps in order to refine the concept and to finalize design details of a positive displacement internal gear pump and a shroudless centrifugal pump. A concept and a system that could be used with pumps to allow a rapid judgement to be made of the suitability of the pump for futher service is developed. Test results and detailed data analysis are included.

    Description: The laboratory tests are described which were conducted on new and damaged bearings to determine the feasibility of using high-frequency vibration as a diagnostic tool. A high-frequency band pass filter and demodulator was assembled to permit field measurements of the high-frequency vibrations. Field tests were conducted on an actual truck and on an axle assembly run in a grease test rig. These field tests were directed toward demonstration of the suitability and capabilities of the high-frequency technique for field application. Two specific areas of field application were identified as being cost effective for railroad use. One area is the examination of railroad roller bearings at a derailment site, and the second is as a wayside detector to supplement present hot box detectors for defective roller bearings.

    Description: The feasibility and efficiency of applying magnetic bearings to free-piston Stirling-cycle power conversion machinery currently being developed for long-term space missions are assessed. The study was performed for a 50-kWe Reference Stirling Space Power Converter (RSSPC) which currently uses hydrostatic gas bearings to support the reciprocating displacer and power piston assemblies. Active magnetic bearings of the attractive electromagnetic type are feasible for the RSSPC power piston. Magnetic support of the displacer assembly would require unacceptable changes to the design of the current RSSPC. However, magnetic suspension of both displacer and power piston is feasible for a relative-displacer version of the RSSPC. Magnetic suspension of the RSSPC power piston can potentially increase overall efficiency by 0.5 to 1 percent (0.1 to 0.3 efficieny points). Magnetic bearings will also overcome several operational concerns associated with hydrostatic gas bearing systems. These advantages, however, are accompanied by a 5 percent increase in specific mass of the RSSPC.

    Description: At the present, the geochemical influence of the Galápagos hotspot (offshore South America) can be seen only along the Galápagos spreading center, north of the hotspot. It is possible, however, that Galápagos plume material also reached the East Pacific Rise in the past. Detecting such influence would be of particular importance for the interpretation of geochemical data from oceanic crust at Ocean Drilling Program (ODP) Site 1256, which formed ∼15 m.y. ago at the East Pacific Rise during a Miocene period of superfast spreading, and is considered to be a reference site for oceanic crust produced at fast-spreading ridges. Here we present geochemical data from Miocene basaltic crust (23–7 Ma) drilled at several Deep Sea Drilling Project (DSDP), ODP, and Integrated Ocean Drilling Program (IODP) sites that formed along the East Pacific Rise between 3°S and 7°N. Lavas formed between ca. 22.5 and ca. 11 Ma show enriched, Galápagos plume–like Pb and Nd isotope ratios (with a peak in enrichment between ≥18 and 12 Ma) compared to lavas created shortly before or after this time interval. Despite their enriched isotope composition, these samples generally show depletion in more-incompatible, relative to less-incompatible, trace elements. Derivation from an enriched Galápagos plume source that had experienced recent melt extraction before it melted further beneath the East Pacific Rise can explain the combined incompatible-trace-element depletion and isotopic enrichment of the 22.5–11 Ma lavas. The influence of plume material correlates with the interval of superfast spreading along the equatorial East Pacific Rise, suggesting a causal relationship. Enhanced ridge-plume interaction ("ridge suction") due to superfast spreading could have facilitated the flow of Galápagos plume material to the ridge. On the other hand, the arrival of Galápagos-type signatures took place immediately after formation of the Galápagos spreading center, which could have provided a pathway for hot plume material to spread into the main ridge network.
