    Beschreibung: Compact two-channel radiometers for solar occultation experiments have been constructed in order to measure stratospheric trace gases. The instruments can be used as filter- or correlation-type radiometers, depending on the trace gas under investigation. Within the LIMS correlative measurement program, balloon flights were performed with a payload of up to four of these two-channel radiometers. From the filter-type measurements, profiles of the trace gases H2O and HNO3 are inferred for the height region between the tropopause and the balloon float level. The data evaluation also includes a comprehensive analysis of the error sources and their effect on the accuracy of the trace gas profiles. The derived H2O and HNO3 profiles are assessed against the observations of other authors and are discussed in the light of the trace gas distributions calcualted from photochemical models.

    Beschreibung: The Limb Infrared Monitor of the Stratosphere (LIMS) instrument on the Nimbus 7 spacecraft sensed limb emission profiles for 7 months, starting in October 1979. Vertical temperature profiles are inferred from measurements in two CO2 channels, and constituent profiles are obtained form the O3, HNO3, NO2, and water vapor channels. The line parameters used to calculate transmittances in the retrieval algorithm are presented. Their adequacy for LIMS and for other applications are assessed, and recommendations for future investigations are outlined.

    Beschreibung: In the LIMS experiment using thermal IR limb scanning to sound the composition and structure of the upper atmosphere, one of the LIMS channels was spectrally centered at 6.9 micrometers to measure the vertical profile and global distribution of stratospheric water vapor. This channel's characteristics, the data from it, and the steps taken to validate results are described. The mean difference between the LIMS measurements and data from 13 balloon underflights is about 0.6 ppmv with LIMS mixing ratios biased high; this difference is of about the same order as estimated LIMS accuracy and less than the sum of the errors for LIMS and the balloon techniques. In-orbit precision is 0.2-0.3 ppmv and accuracy is estimated at 20-30 percent from 50 mbar to the stratopause. An unexplained diurnal variation exists in the vertical profile data which is largest at the 1-mbar level and virtually nonexistent at 10 mbar; day values are higher than night. More confidence is placed in zonal mean distributions averaged over several days than in single profiles. A zonal mean pressure-latitude cross section is described for January 5-9, 1979.

    Beschreibung: The validation of results from the nitrogen dioxide channel and the quality of the data are examined in connection with the LIMS experiment which ran from late October 1978 to late May 1979. Factors studied include: channel characteristics, experiment errors due to instrument and spacecraft effects, predicted and measured precision, predicted accuracy, and comparisons with correlative measurements made in a series of balloon underflights. Features such as profile shape and slope of the mixing ratio altitude distribution are in good agreement. The LIMS data also fall within the range of previous mixing ratio measurements and are consistent with model estimates. The calculated on-orbit precision is about 0.3 ppbv and the estimated accuracy from simulations is about 2 ppbv over the 3-10-mbar range. Accuracy is less at higher and lower pressure levels. These results provide the first day-night set of nitrogen dioxide measurements from space.

    Beschreibung: Total odd nitrogen, NO(y), may be defined as the sum of all active nitrogen species that interchange photochemically with one another on a time scale of the order of weeks or less. As noted, NO + NO2 reactions dominate the processes controlling the ozone balance in the contemporary stratosphere. The observational data from non-satellite platforms are reviewed. The growth in available satellite data in the past four years is considered. Some of the most important scientific issues are discussed, taking into account new results from atmospheric models (mainly 2-D). The model results are compared with the observational data.

    Notizen: Abstract The genome of the slow-growing Bradyrhizobium japonicum (strain 110) was mutagenized with transposon Tn5. A total of 1623 kanamycin/streptomycin resistant derivatives were screened in soybean infection tests for nodulation (Nod) and symbiotic nitrogen fixation (Fix). In this report we describe 14 strains possessing a stable, reproducible Nod+Fix- phenotype. These strains were also grown under microaerobic culture conditions to test them for free-living nitrogen fixation activity (Nif). In addition to strains having reduced Fix and Nif activities, there were also strains that had reduced symbiotic Fix activity but were Nif+ ex planta. Analysis of the genomic structure revealed that the majority of the strains had a single Tn5 insertion without any further apparent physical alteration. A few strains had additional insertions (by Tn5 or IS50), or a deletion, or had cointegrated part of the vector used for Tn5 mutagenesis. One of the insertions was found in a known nif gene (nifD) whereas all other mutations seem to affect different, hitherto unknown genes or operons. Several mutant strains had an altered nodulation phenotype, inducing numerous, small, widely distributed nodules. Light and electron microscopy revealed that most of these mutants were defective in different stages of bacteroid development and/or bacteroid persistence. The protein patterns of the mutants were inspected by two-dimensional gel electrophoresis after labelling microaerobic cultures with l-(35S)methionine. Of particular interest were mutants lacking a group of proteins the synthesis of which was known to be under oxygen control. Such strains can be regarded as potential regulatory mutants.

    Notizen: Abstract At least five highly conserved, but disparately regulated groESL operons are present in Bradyrhizobium japonicum. Expression of groESL 3 is coregulated with symbiotic nitrogen fixation genes, implying a role of GroESL chaperonins in the nitrogen fixation process. Null mutants of individual groEL genes, however, were not impaired in symbiotic nitrogen fixation activity. By contrast, the groEL 3-plus-groEL 4 double mutant strain D4, which is mutated in those groEL genes that contribute most to the GroEL pool under symbiotic conditions, exhibited less than 5% Fix activity as compared to the wild-type. Expression of lacZ fusions made to several representative nif and fix genes was not, or only marginally, reduced in mutant D4, indicating that the requirement of chaperonins for nitrogen fixation does not occur at the level of RegSR-NifA-σ54- or FixLJ-FixK2-dependent gene regulation. Instead, immunoblot analyses revealed that the level of NifH and NifDK nitrogenase proteins was drastically decreased in extracts prepared from D4 bacteroids and from free-living cells grown anaerobically. Transcriptional fusions of the anaerobically induced groESL 3 promoter (P3) to all five B. japonicum groESL operons and also to groESL from Escherichia coli were integrated into the chromosome of mutant D4. Strains harboring P3 fused to groESL 1, groESL 2, groESL 5, or E. coli groESL partially complemented the symbiotic defect of mutant D4, whereas the wild-type phenotype was completely restored in strains complemented with P3 fused to groESL 3 (control) or groESL 4. Likewise, the growth defect of an E. coli groEL mutant could be corrected at least partially by individual B. japonicum groESL operons. In conclusion, both series of complementation analyses were not indicative of a strict substrate specificity of any of the B. japonicum groESL gene products, which is in good agreement with their high degree of sequence conservation.
