    Notizen: Summary Extracellular matrix vesicles from fracture callus cartilage were isolated by differential centrifugation and resolved by equilibrium centrifugation on a discontinuous sucrose gradient into two bands. The phosphohydrolytic activity towards p-nitrophenyl phosphate, tetrasodium pyrophosphate and adenosine triphosphate was distributed similarly after differential and equilibrium centrifugation suggesting the association of this activity with the matrix vesicles. The two bands isolated by equilibrium centrifugation of the partially purified vesicular preparation demonstrated high levels of alkaline phosphatase activity. Observed with an electron microscope, the 1.07–1.14 g/cm3 band from the gradient was enriched in electron luscent matrix vesicles while the 1.27 g/cm3 band contained electron dense matrix vesicles. Enzymatic analysis of the 1.27 g/cm3 band indicated a slight contamination due to the presence of mitochondria and lysosomes while the 1.07–1.14 g/cm3 band gave no enzymatic indication of subcellular contamination. A phosphohydrolytic enzyme active towards p-nitrophenyl phosphate, tetrasodium pyrophosphate and adenosine triphosphate was purified from the 1.07–1.14 g/cm3 fraction by DEAE-cellulose column chromatography. Electron micrographs of callus cartilage sections demonstrated densification of the plasma membrane and matrix vesicles following substrate incubation withβ-glycerophosphate or tetrasodium pyrophosphate. The histochemical and biochemical data indicate that a phosphatase, with multiple substrate specificity, is a component of fracture callus cartilage matrix vesicles.

    Notizen: Abstract The radiation resistant bacteria Micrococcus radiophilus and M. radioproteolyticus were studied by thin sectioning and freeze-etching techniques and the two species were found to be similar in the fine structure. The only significant difference was in the appearance of the surfaces of the cell walls in freeze-etched preparations. Since the two species, together with M. radiodurans, possess a unique cell wall structure and a cell wall peptidoglycan, which is different from that of other micrococci and Gram-positive cocci, it is recommended that they be reclassified into a new genus.

    Notizen: Abstract [2H2]Gibberellin A24 (GA24) and [2H4]-GA9 were applied to the apices of normal-type cucumber (Cucumis sativus L. cv. Yomaki) seedlings treated with uniconazole, an inhibitor of GA biosynthesis. The metabolites from these feeds were identified by full-scan gas chromatography-mass spectrometry (GC-MS) to confirm the conversions of [2H2]GA24 to [2H2]GA9 and of [2H4]GA9 to [2H4]GA4. The results show that GA4 is biosynthesized from GA24 via GA9. In a cucumber hypocotyl elongation bioassay using cv. Yomaki, prohexadione (DOCHC), an inhibitor of 2-oxoglutaratedependent dioxygenase, inhibited the hypocotyl elongation caused by application of GA9, while DOCHC enhanced the elongation caused by application of GA4. These results indicate that GA4 is a physiologically active GA and that the activity of GA9 is due to its conversion to GA4 in cucumber shoots.

    Notizen: Abstract The reaction of Et3SiD with Na2PdCl4 in CD3OD was carried out under conditions where we monitored for production of neutrons and/or ψ-rays. This reaction produces D2 off the surface of Pd colloid. The formation of high surface area Pd metal was confirmed by transmission electron microscopy /TEM/. The heat from the all protio control reaction and the all protio control reaction and the all deutero reaction was monitored and found to be the same within the limits of these experiments. Within the limits of our neutron and gamma detection, we conclude that no measurable fusion occurred during this reaction.

    Notizen: Abstract The effects of gibberellin (GA) on cold-induced stem elongation and flowering of Japanese radish (Raphanus sativus L.) were investigated using application of GA3 and a GA-biosynthesis inhibitor, uniconazole (UCZ). UCZ very strongly inhibited stem elongation and delayed flowering, and the inhibition and delay were completely reversed by GA3 application. These results suggest that GA is necessary not only in the stem elongation but also in the flowering. When cold treatment (CT) was conducted on the plants whose GA level was lowered by UCZ, GA3 applied after CT completely reversed the delay of flowering. Thus low GA level probably did not retard cold induction. Microscopic observation of apical meristem showed that UCZ delayed flowering by delaying the shift from vegetative to dome-shaped meristem. This result suggested that low GA level delayed floral evocation. Consequently it was suggested that low GA level retarded physiological process involved in long day induction or in floral evocation, resulting in delay of floral evocation.

    Notizen: Abstract Solidification and microsegregation studies were performed on alloy CF-8M weld metal which solidified via the primary austenite/eutectic ferrite mode. All of the major alloying elements (chromium, nickel, molybdenum) were observed to segregate to interdendritic areas upon solidification. Electron microprobe analysis revealed a substantial chromium and molybdenum enrichment of the eutectic ferrite relative to the austenite dendrites even in structures water-quenched from the solidus temperature. Scanning transmission electron microscopy/energy dispersive spectrometry (STEM/EDS) profiles taken within the eutectic ferrite phase revealed a similar pattern of major element distribution as has been observed by other investigators in residual primary delta ferrite dendrites. Within the eutectic ferrite, the highest chromium and molybdenum content and the lowest nickel content was found at the eutectic ferrite/austenite boundary. STEM/EDS analyses of in situ water-quenched weld microstructures revealed that compositional modification of the eutectic ferrite had occurred upon cooling from the solidus. In particular, the chromium concentration of the eutectic-ferrite was observed to increase by approximately 3 wt% in the temperature range ∼ 1300 to ∼ 750°C. In the same temperature range, the nickel content of the eutectic-ferrite decreased by approximately 4 wt % and the molybdenum content increased within the same phase by approximately 1 wt%. The transformation of eutectic ferrite to austenite as the weld metal cools to room temperature is consistent with a volume diffusion-control mechanism.

    Notizen: Abstract The nature of the solid-state reaction between hot-pressed Si3N4 and a model Ni-based superalloy was examined and compared to that previously found with SiC. The Si3N4 reacted with the metal to a significantly lesser extent than SiC, presumably because of the dependence of the Si3N4/metal reaction on the partial pressure of N2. Products of the reaction in both metal and ceramic were AIN, Ni2Si, other Ni and Cr silicides, and probably a chromium nitride.

    Notizen: Abstract The levels of gibberellin A1 (GA1), GA8, GA19, GA20, GA29, and GA44 in the short Pisum sativum L. mutants lk, lka, and lkb, and comparable wild-type plants, were determined by gas chromatography-selected ion monitoring (GC-SIM) using 2H or 13C internal standards. The mutants possessed similar GA1 levels to wild-type plants, consistent with their classification as GA-sensitivity rather than GA-synthesis mutants. However, these mutants differ from certain sensitivity mutants in other species, in which substantial accumulation of GA1 occurs. The results suggest that if the proposed feedback model for the regulation of GA synthesis occurs in peas it is not the reduced growth per se that is the trigger for elevated levels of C19 GAs. The results are also consistent with the hypothesis that in those GA-sensitivity mutants which do not accumulate C19 GAs, the biochemical lesion may be well down the transduction pathway which leads from GA1 reception to stem elongation.
