    Notes: Summary The pharmacokinetics of dexamethasone, given at low dose, were studied in 13 patients with congenital adrenal hyperplasia (CAH) to ascertain whether kinetics differed in this inherited disorder of cortisol metabolism from those seen in healthy individuals. Changes in plasma dexamethasone concentration after intravenous bolus, measured using a simple novel radioimmunoassay, were well described by a two-compartment open model with first-order kinetics. Values for λ2: 0.206 h−1, t1/2: 3.53 h, Vc: 24.41 and f: 0.64 were similar to those previously reported for normal subjects. There were considerable interindividual differences in parameter values and Cmaxp.o. (range 22–67 nmol/l). As suppression of the hypothalamo-pituitary-adrenal axis correlates with plasma dexamethasone levels, this variability may partly explain the differing dose and dose schedule requirements necessary to achieve adequate therapeutic control in the clinical management of CAH.

    Notes: The first excited electronic state (S1) vibrational dynamics of aniline(Ar)1 and aniline(CH4)1 van der Waals (vdW) clusters have been studied using molecular jet and time resolved emission spectroscopic techniques. The rates of intramolecular vibrational energy redistribution (IVR) and vibrational predissociation (VP) as functions of vibrational energy are reported for both clusters. For vibrational energy in excess of the cluster binding energy, both clusters are observed to dissociate. The dispersed emission spectra of these clusters demonstrate that aniline(Ar)1 dissociates to all energetically accessible bare molecule states and that aniline(CH4)1 dissociates selectively to only the bare molecule vibrationless state. The emission kinetics show that in the aniline(Ar)1 case, the initially excited states have nanosecond lifetimes, and intermediate cluster states have very short lifetimes. In contrast, the initially excited aniline(CH4)1 states and other intermediate vibrationally excited cluster states are very short lived (〈100 ps), and the intermediate cluster 00 state is observed. These results can be understood semiquantitatively in terms of an overall serial IVR/VP mechanism which consists of the following: (1) the rates of chromophore to vdW mode IVR are given by Fermi's golden rule, and the density of vdW vibrational states is the most important factor in determining the relative [aniline(Ar)1 vs aniline(CH4)1] rates of IVR; (2) IVR among the vdW modes is rapid; and (3) VP rates can be calculated by a restricted vdW mode phase space Rice–Ramsberger–Kassel–Marcus theory. Since the density of vdW states is three orders of magnitude greater for aniline(CH4)1 than aniline(Ar)1 at 700 cm−1, the model predicts that IVR is slow and rate limiting in aniline(Ar)1, whereas VP is slow and rate limiting in aniline(CH4)1. The agreement of these predictions with the experimental results is very good and is discussed in detail.

    Notes: Abstract Routine screening of indigenous and recombinant plasmids in pink facultative methylotrophic bacteria has been difficult, time-consuming, and yields variable results. We report a modified alkaline hydrolysis method for rapid plasmid isolation from these organisms that reproducibly results in good yields of closed circular plasmid DNA which can be readily digested with restriction enzymes. This method greatly facilitates direct screening of indigenous and introduced recombinant plasmids in the methylotrophic host strain. We have confirmed earlier findings that the original NCIB wild-type strain of Methylobacterium sp. strain AM1 (NCIB 9133) contains three cryptic plasmids. However, sizing of these plasmids by comparison to standards and by restriction fragment analysis suggests that they are larger than previously reported. We have designated these plasmids pAM1-1 (65 kb), pAM1-2 (40 kb) and pAM1-3 (33 kb). We have also shown that a rifamycin-resistant strain of Methylobacterium sp. strain AM1 used routinely in our laboratory lacks pAM1-2, although no phenotype has been associated with its loss. Finally, we have shoen that another pink facultative methylotroph, Methylobacterium isolate (#YK1), contains three cryptic plasmids of approximately 43, 37 and 22 kb, respectively.

    Notes: Plasmid pAMβ1 is transferred at low efficiency in filter matings between Bacillus subtilis and Clostridium acetobutylicum. We have constructed a small non-conjugative plasmid, pOD1 (7.2 kbp; ApR, CmR, EmR) that becomes established in pAMβ1-containing strains of B. subtilis in the form of a cointegrate (pAMβ1::pOD1). The cointegrate plasmid is transferred to, and maintained in C. acetobutylicum, with an efficiency similar to that of pAMβ1. We have also constructed derivatives of plasmid pOD1, with single sites for several commonly used restriction endonucleases. They can be used to transfer foreign genes to C. acetobutylicum via B. subtilis.

    Notes: Abstract A filter mating procedure has been used to transfer the plasmid pAMβ;1 from Streptococcus lactis to Clostridium acetobutylicum at high efficiency. Once established in this latter host the plasmid can be transferred, with lower efficiency to other strains of C. acetobutylicum or back to S. lactis. A procedure has also been developed for the co-cultivation of Bacillus subtilis and C. acetobutylicum and the transfer of pAMβ;1 between these organisms has been demonstrated.

    Notes: An amplified tunable picosecond dye laser has been constructed which is pumped by an active-passive mode-locked Nd:YAG laser. The laser is tunable from 555 nm to 〉700 nm. It provides a maximum of 2-mJ pulses with pulse widths of ∼17 ps (FWHM). The tuning range can be extended by frequency doubling and/or mixing with the residual 1064-nm pulses. The dye laser is very stable and relatively inexpensive to construct.

    Notes: A broadband detection scheme for a time-resolved infrared absorption spectrometer, based on a multielement mercury-cadmium-telluride (MCT) array, has been successfully implemented. The spectrometer achieves a resolution on the 10-ns time scale despite the much larger time constant characteristic of the MCT elements. Our signal-collection circuitry takes advantage of the slow decay by integrating the detector response to pulsed IR radiation. The dynamic range is 100–1 and the resultant noise level is near the detector limit. Data acquisition for the 120 elements is fast enough to allow scan rates of 30–40 Hz. The completed electronics are sufficiently compact to be situated local to the array detector, and the design is relatively inexpensive to construct using commonly found electronic components.
