Notes: A new method for determining diffusion coefficients of point defects using synchrotron radiation topography (SRT) is described. Since the method is based on dynamical observations of dislocation motion in a thick specimen with a low dislocation density, advantage of the SRT is fully utilized. It is shown by typical observations on ice crystals that an absolute value of the diffusion coefficient is determined by a simple analysis of the climb motion of the dislocations.

Notes: srnB is an F-plasmid encoded gene, otherwise silent, whose expression is induced by added rifampicin, leading to the release of cellular Mg2+ and degradation of stable RNA. In the absence of rifampicin, transcripts from the srnB gene were relatively short. S1 nuclease mapping revealed that the short mRNA species terminated within the leader, at the 3’end of a potential stem-and-loop structure. A deletion in the stem-loop resulted in constitutive synthesis of the mRNA that extended beyond the termination site into the structural gene. Even with the wild-type gene, transcription continued beyond the terminator sequence in the presence of added rifampicin. Most of the transcripts synthesized in the presence of rifampicin were long enough to encode the srnB protein. We hypothesize from these results that RNA polymerase associated with rifampicin can read through the terminator to induce srnB expression.

Notes: Summary The inducing effect of feprazone, a pyrazolone anti-inflammatory agent, on hepatic drug-metabolizing enzymes has been studied in healthy volunteers. The ratio of 6β-hydroxycortisol (6β-OHF) to 17-hydroxycorticosteroids (17-OHCS) in urine, used as an indicator of oxidative drug-metabolizing enzyme activity, was increased up to 1.6-times the original level after 5 days of oral treatment with feprazone 300 mg/day. This indicates that feprazone induces hepatic drug-metabolising enzymes in man as does phenylbutazone.

Notes: Summary In order to obtain distinct and reliable information concerning the localization of ionized anionic groups in tissues, fine-granular cationic ferric hydroxide colloid solution (Fe-Cac-f) was newly devised. This can be obtained by boiling a mixture of ferric chloride and ammonium cacodylate solutions. the colloid particles of Fe-Cac-f are about 1.0 nm in size, i.e., one-fifth of the size of ferric cacodylate colloid (Fe-Cac; Seno et al. 1983a). As with Fe-Cac, Fe-Cac-f particles in the pH range of 1.6–7.6 carry a positive electric charge, but the latter show a better permeation of tissues. Using the Prussian blue reaction, Fe-Cac-f gives a distinct deep-blue color and can be used for the detection of anionic groups of acid mucopolysaccharides and proteins by light microscopy. It is also useful for detecting the exact sites of ionized anionic groups in deep tissue areas using electron microscopy.

Notes: Summary The immunocytochemical localization of fatty acid binding protein (FABP) of liver type was studied at light and electron microscopic levels by the peroxidase-antiperoxidase (PAP) method using a specific polyclonal antibody against FABP in the liver of fed and fasted rats. In the liver of rats fed ad libitum, the intense immunoreactivity was confined to portions of the liver cell cytoplasm adjacent to the glycogen area. After 2-days' fasting, such a focal intracellular localization of the immunoreactivity was abolished, in association with the disappearance of the glycogen area, and was replaced by a diffuse distribution of the immunoreactivity throughout the cytoplasm, with higher intensity at the periphery of the cells. In liver cells exhibiting an overall hypertrophy of smooth endoplasmic reticulum (SER) induced by the treatment of fasted rats with phenobarbital, the peripheral localization of FABP immunoreactivity ramained unchanged compared with that obtained in the case of fasting alone, and the immunoreactivity did not occur in association with the proliferated SER in the central cytoplasm. These results suggest that FABP, although cytosolic in nature, changes its localization within the liver cells in response to the general metabolic alterations caused by the starvation, inferring that FABP is intimately involved in the intracellular transport and metabolism of free fatty acids.

Notes: Abstract Ultrafine powders of the ZrO2-SiO2 system prepared by spraying aqueous mixed solutions of ZrO(NO3)2·2H2O and silicone oil into an ultrahigh temperature inductively coupled plasma (the spray-ICP technique) have been investigated in connection with particle growth and phase change caused by heat treatment. The prepared powders were composed of roundish ultrafine particles 10 to 20 nm in size, and their components were tetragonal ZrO2 (t-ZrO2) and amorphous SiO2 (a-SiO2).a-SiO2 alone did not crystallize below 1400° C, though the particles grew above 900° C.t-ZrO2 converted to monoclinic ZrO2 (m-ZrO2) above 400° C, also followed by particle growth. On the other hand composite powders oft-ZrO2 anda-SiO2 exhibited no particle growth below 1200° C. Above 1200° Ct-ZrO2 converted tom-ZrO2, and its amount decreased with an increase in SiO2 content. At 1400° C particle growth occurred, and the round particles of ZrO2 were dispersed in ana-SiO2 glass medium. These results indicate that ZrO2 and SiO2 mutually block particle growth, and that SiO2 contributes to the quenching oft-ZrO2 by keeping the particles of ZrO2 within the critical particle size oft-ZrO2.

Notes: Abstract Two series of alumina-zirconia composites, i.e. alumina-unstabilized zirconia and alumina-partially stabilized zirconia with 3 mol % Y2O3, with different zirconia content were slip casted and fired at 1550°C for 3 h. Elastic constant, bending strength and fracture toughness were measured. Internal friction was determined to follow the formation of cracks, nondestructively, which could be one of the fracture origins. The crack length of the fracture origin and the fracture surface energy were calculated by applying Griffith's fracture theory. Microstructures of the fracture surfaces were observed using a scanning electron microscope. For the unstabilized zirconia system, the increase in the internal friction of the order from 10−4 to 10−3 was a guide to find the formation of cracks which lead to the fracture. The increase in the cracks becoming a fracture origin lead to the increase inK lc and also to the apparent increase in the fracture surface energy. For the partially stabilized zirconia system, the increase in the fracture surface energy with an increase in zirconia content, keeping low internal frictions of the order of 10−4, indicates the intrinsic strengthening of the grain boundaries in comparison to the unstabilized zirconia system. Internal friction is the most suitable nondestructive physical quantity to find the microcracks which leads to the fracture.
