    Notizen: Non-aseptic production of biosurfactant from molasses by a mixed culture was investigated in stirred batch reactors. Biosurfactant production was quantified by surface tension reduction, critical micelle dilution (CMD), and emulsification capacity (EC). Biosurfactant production was directly correlated with biomass production, and was improved by pH control and addition of yeast extract. Centrifugation of the whole broth increased emulsifying capacity and reduced surface tension. Acidification of the whole broth increased the emulsification capacity but reduced the apparent biosurfactant concentration (CMD), without affecting the surface tension. The emulsification capacity of the cell-free broth was equivalent to that of a 100 mg/L solution of sodium dodecyl sulfate. The emulsification capacity of the whole broth and cell-free broth were reduced by about 50% at and above NaCl concentrations of 100mM. Preliminary characterization suggests that the biosurfactant activity is primarily associated with one or more protease-sensitive species, released from cells in larger quantities after more vigorous centrifugation. © 1994 John Wiley & Sons, Inc.

    Notizen: The mechanism of extraction of rat cytochrome b5 from water into a sodium dioctylsulfosuccinate (AOT) micellar organic phase was studied using protein engineering of surface charged residues. The extraction behavior of native cytochrome b5 and modified proteins with substitutions of the type glutamic acid → lysine at positions 44 (E44K), 56 (E56K), and 92 (E92K), was studied as a function of pH. The results indicate that an important mechanism of extraction is an electrostatic interaction of this protein with the negatively charged surfactant. We demonstrate that it is possible to improve extraction by engineering the protein surface charge, increasing the driving force responsible for the protein transfer to the micellar phase. © 1994 John Wiley & Sons, Inc.

    Beschreibung: Direct measurements from satellites of the solar 'constant' (the total irradiance at mean sun-earth distance) during more than ten years show variations over time scales from minutes to years and decades. At high frequencies, solar oscillations contribute to the variance. The most important influences are related to solar activity: during the passage of active regions on the solar disk (sunspots and faculae) changes of a few 0.1 percent lasting for several days are observed. The effects of spots can be well reproduced by the projected sunspot index, whereas the influence of faculae have to be modeled from proxy data like the Ca-K plage index or the He I index. Long-term trends are detected which are connected to the 11-yr solar activity cycle.

    Beschreibung: A model of solar Lyman alpha irradiance developed by multiple linear regression analysis, including the daily values and 81-day running means of the full disk equivalent width of the Helium line at 1083 nm, predicts reasonably well both the short- and long-term variations observed in Lyman alpha. In contrast, Lyman alpha models calculated from the 10.7-cm radio flux overestimate the observed variations in the rising portion and maximum period of solar cycle, and underestimates them during solar minimum. Models are shown of Lyman alpha based on the He-line equivalent width and 10.7-cm radio flux for those time intervals when no satellite observations exist, namely back to 1974 and after April 1989, when the measurements of the Solar Mesosphere Satellite were terminated.

    Beschreibung: Solar Lyman alpha irradiance is estimated from various solar indices using linear regression analyses. Models developed with multiple linear regression analysis, including daily values and 81-day running means of solar indices, predict reasonably well both the short- and long-term variations observed in Lyman alpha. It is shown that the full disk equivalent width of the He line at 1083 nm offers the best proxy for Lyman alpha, and that the total irradiance corrected for sunspot effect also has a high correlation with Lyman alpha.
