    Description: Beryl’s various coloring make it a sought-after and precious gemstone. Knowledge on the physical origin behind the various colorings is essential to differentiate artificial colorings from natural ones. The blue color of the variety aquamarine has been proposed to be due to charge transfer between adjacent Fe 3+ -Fe 2+ pairs on the Al site and its adjacent 6 g interstitial position, respectively. The present work presents the first experimental confirmation of such an arrangement. Single-crystal electron paramagnetic resonance (EPR) spectra of a light blue beryl (Springbok, South Africa), before, and after annealing at 800 °C, have been measured at temperatures from 25 to 295 K and reveal two Fe 3+ -Fe 3+ pairs related to an axial Fe 3+ center at the effective g = ~2.00 as well as a subordinate rhombic Fe 3+ center at g = ~4.28. Powder EPR spectra show that the intensity of the axial Fe 3+ center increases after annealing from 400 to 800 °C but becomes constant after 800 °C, whereas the rhombic Fe 3+ center is not affected by annealing. One Fe 3+ -Fe 3+ pair is the same as the one investigated previously by Edgar and Hutton (1982) and arises from Fe 3+ ions at the two nearest Al sites along the c axis. The best-fit spin Hamiltonian parameters show that the second Fe 3+ -Fe 3+ pair is also oriented parallel to the c axis and has a separation of 2.4 Å, corresponding to Fe 3+ ions at an Al site and its nearest 6 g interstitial position. The increased intensities of both Fe 3+ -Fe 3+ pairs after annealing suggest their formation from Fe 3+ -Fe 2+ and Fe 2+ -Fe 2+ precursors. Modeling of synchrotron Fe K -edge X-ray absorption spectra measured on the sample annealed at 1000 °C also shows occupancies of Fe 3+ at both the Al site and the 6 g interstitial position. The assignment of the rhombic Fe 3+ center to the Be site remains tentative.

    Description: In recent years, some approaches have been presented for the seamless integration of WSNs with the existing, widely deployed SOA technologies such as XML, Web services, and the Business Process Execution Language (BPEL) to build a wireless sensor networks service application. However, there a great challenge on fault tolerant in WSNs. In this paper, we present our framework and approach to enhance the reliability of service composition applications in WSNs through modeling and analyzing a wireless sensor networks service application based on BPEL with exception handler and compensation mechanism. At first, we analyze all possible states during the execution of BPEL instance in WSNs. Then, we present a state framework for modeling execution context in BPEL instance in WSNs. Based on this framework, we analyze state transition and operational semantics in the case of both correct execution and exceptional execution of BPEL instance in WSNs. Furthermore, we propose the state transition models for three types of activities in BPEL instance. In the end, we present a formal approach to model the execution context in BPEL for WSNs. Using this formal model, one can describe and analyze the control flow result from the exception handler and compensation mechanism in BPEL instance for WSNs.

    Description: Guanine-rich, single-stranded, DNAs and RNAs are able to fold to form G-quadruplexes that are held together by guanine base quartets. G-quadruplexes are known to bind ferric heme [Fe(III)-protoporphyrin IX] and to strongly activate such bound hemes toward peroxidase (1-electron oxidation) as well as oxygenase/peroxygenase (2-electron oxidation) activities. However, much remains unknown about how such activation is effected. Herein, we investigated whether G-quadruplexes were strictly required for heme activation or whether related multi-stranded DNA/RNA structures such as isoguanine (iG) quadruplexes and pentaplexes could also bind and activate heme. We found that iG-pentaplexes did indeed bind and activate heme comparably to G-quadruplexes; however, iG-quadruplexes did neither. Earlier structural and computational studies had suggested that while the geometry of backbone-unconstrained iG-quintets templated by cations such as Na + or NH 4 + was planar, that of iG-quartets deviated from planarity. We hypothesize that the binding as well as activation of heme by DNA or RNA is strongly supported by the planarity of the nucleobase quartet or quintet that interacts directly with the heme.

    Description: Suprasubduction zone ophiolites are relics of oceanic upper plate forearcs and are typically preserved as discontinuous belts with discrete massifs along suture zones. Ophiolites usually contain an incomplete condensed section compared to average modern oceanic lithosphere. The incompleteness and discontinuity of ophiolites are frequently attributed to dismemberment, but tectonic causes remain poorly constrained. Here we show new paleomagnetic and field geological evidence for the preservation of extensional detachment faults that thinned and dismembered the south Tibetan ophiolite belt during the Early Cretaceous. Similar to those documented in modern slow- and ultraslow-spreading ridges, these detachments exhumed lithospheric mantle, and subophiolitic mélange, to the seafloor, which became unconformably covered by Asia-derived forearc strata. We call this mechanism forearc hyperextension, whereby widespread detachment faults accommodate upper plate extension above a subduction zone. We propose that hyperextension is the key mechanism responsible for dismemberment of the south Tibetan ophiolitic belt shortly after its magmatic accretion.

    Description: The CRISPR/Cas has been recently shown to be a powerful genome-editing tool in a variety of organisms. However, these studies are mainly focused on protein-coding genes. The present study aims to determine whether this technology can be applied to non-coding genes. One of the challenges for knockout of non-coding genes is that a small deletion or insertion generated by the standard CRISPR/Cas system may not necessarily lead to functional loss of a given non-coding gene because of lacking an open reading frame, especially in polyploidy human cell lines. To overcome this challenge, we adopt a selection system that allows for marker genes to integrate into the genome through homologous recombination (HR). Moreover, we construct a dual guide RNA vector that can make two cuts simultaneously at designated sites such that a large fragment can be deleted. With these approaches, we are able to successfully generate knockouts for miR-21, miR-29a, lncRNA-21A, UCA1 and AK023948 in various human cell lines. Finally, we show that the HR-mediated targeting efficiency can be further improved by suppression of the non-homologous end joining pathway. Together, these results demonstrate the feasibility of knockout for non-coding genes by the CRISPR/Cas system in human cell lines.

    Description: Elucidating the key signal transduction pathways essential for both antipsychotic efficacy and side-effect profiles is essential for developing safer and more effective therapies. Recent work has highlighted noncanonical modes of dopamine D2 receptor (D2R) signaling via β-arrestins as being important for the therapeutic actions of both antipsychotic and antimanic agents. We thus sought to create unique D2R agonists that display signaling bias via β-arrestin–ergic signaling. Through a robust diversity-oriented modification of the scaffold represented by aripiprazole (1), we discovered UNC9975 (2), UNC0006 (3), and UNC9994 (4) as unprecedented β-arrestin–biased D2R ligands. These compounds also represent unprecedented β-arrestin–biased ligands for a Gi-coupled G protein–coupled receptor (GPCR). Significantly, UNC9975, UNC0006, and UNC9994 are simultaneously antagonists of Gi-regulated cAMP production and partial agonists for D2R/β-arrestin-2 interactions. Importantly, UNC9975 displayed potent antipsychotic-like activity without inducing motoric side effects in inbred C57BL/6 mice in vivo. Genetic deletion of β-arrestin-2 simultaneously attenuated the antipsychotic actions of UNC9975 and transformed it into a typical antipsychotic drug with a high propensity to induce catalepsy. Similarly, the antipsychotic-like activity displayed by UNC9994, an extremely β-arrestin–biased D2R agonist, in wild-type mice was completely abolished in β-arrestin-2 knockout mice. Taken together, our results suggest that β-arrestin signaling and recruitment can be simultaneously a significant contributor to antipsychotic efficacy and protective against motoric side effects. These functionally selective, β-arrestin–biased D2R ligands represent valuable chemical probes for further investigations of D2R signaling in health and disease.

    Description: A conical conformal leaky-wave antenna based on substrate integrated waveguide (SIW) technology is proposed and demonstrated in this paper. This antenna conforms to a conical shape surface with the angle of 40°. It has a narrow beam that scans from 80° to 97° with varying frequency (34 GHz~37 GHz). Both conformal and nonconformal antennas are fabricated through the standard PCB process. Their performances are compared within the desired frequency.

    Description: Neuronal mechanisms of auditory distance perception are poorly understood, largely because contributions of intensity and distance processing are difficult to differentiate. Typically, the received intensity increases when sound sources approach us. However, we can also distinguish between soft-but-nearby and loud-but-distant sounds, indicating that distance processing can also be based on intensity-independent cues. Here, we combined behavioral experiments, fMRI measurements, and computational analyses to identify the neural representation of distance independent of intensity. In a virtual reverberant environment, we simulated sound sources at varying distances (15–100 cm) along the right-side interaural axis. Our acoustic analysis suggested that, of the individual intensity-independent depth cues available for these stimuli, direct-to-reverberant ratio (D/R) is more reliable and robust than interaural level difference (ILD). However, on the basis of our behavioral results, subjects’ discrimination performance was more consistent with complex intensity-independent distance representations, combining both available cues, than with representations on the basis of either D/R or ILD individually. fMRI activations to sounds varying in distance (containing all cues, including intensity), compared with activations to sounds varying in intensity only, were significantly increased in the planum temporale and posterior superior temporal gyrus contralateral to the direction of stimulation. This fMRI result suggests that neurons in posterior nonprimary auditory cortices, in or near the areas processing other auditory spatial features, are sensitive to intensity-independent sound properties relevant for auditory distance perception.
