    Beschreibung: In this paper, we demonstrate a prototype of magnetic tunable bandstop filter by using magnetic multilayers of [FeGaB 25 nm/Al 2 O 3 5 nm] 4 prepared by magnetron sputtering. The multilayer structure exhibits a relatively low coercive field and RF loss compared to the single layer film with the same thickness. By placing a single transmission line on the top of magnetic multilayers, a prototype of tunable bandstop filter was fabricated and operates from 4.5 GHz to 7.08 GHz under very low magnetic bias fields from 100 Oe to 400 Oe. These low loss multilayer films are good candidates for RF/microwave applications.

    Beschreibung: Influenza A virus (IAV) has been raising public health and safety concerns worldwide. Cyanovirin-N (CVN) is a prominent anti-IAV candidate, but both cytotoxicity and immunogenicity have hindered the development of this protein as a viable therapy. In this article, linker-CVN (LCVN) with a flexible and hydrophilic polypeptide at the N-terminus was efficiently produced from the cytoplasm of Escherichia coli at a 〉15-l scale. PEGylation at the N-terminal α-amine of LCVN was also reformed as 20 kDa PEGylated linkered Cyanovirin-N (PEG 20k –LCVN). The 50% effective concentrations of PEG 20k –LCVN were 0.43 ± 0.11 µM for influenza A/HK/8/68 (H3N2) and 0.04 ± 0.02 µM for A/Swan/Hokkaido/51/96 (H5N3), dramatically lower than that of the positive control, Ribavirin (2.88 ± 0.66 x 10 3 µM and 1.79 ± 0.62 x 10 3 µM, respectively). A total of 12.5 µM PEG 20k –LCVN effectively inactivate the propagation of H3N2 in chicken embryos. About 2.0 mg/kg/day PEG 20k –LCVN increased double the survival rate (66.67%, P = 0.0378) of H3N2 infected mice, prolonged the median survival period, downregulated the mRNA level of viral nuclear protein and decreased (attenuated) the pathology lesion in mice lung. A novel PEGylated CVN derivative, PEG 20k –LCVN, exhibited potent and strain-dependent anti-IAV activity in nanomolar concentrations in vitro, as well as in micromolar concentration in vivo .

    Beschreibung: Motivation: Systematically predicting gene (or protein) function based on molecular interaction networks has become an important tool in refining and enhancing the existing annotation catalogs, such as the Gene Ontology (GO) database. However, functional labels with only a few (〈10) annotated genes, which constitute about half of the GO terms in yeast, mouse and human, pose a unique challenge in that any prediction algorithm that independently considers each label faces a paucity of information and thus is prone to capture non-generalizable patterns in the data, resulting in poor predictive performance. There exist a variety of algorithms for function prediction, but none properly address this ‘overfitting’ issue of sparsely annotated functions, or do so in a manner scalable to tens of thousands of functions in the human catalog. Results: We propose a novel function prediction algorithm, clusDCA, which transfers information between similar functional labels to alleviate the overfitting problem for sparsely annotated functions. Our method is scalable to datasets with a large number of annotations. In a cross-validation experiment in yeast, mouse and human, our method greatly outperformed previous state-of-the-art function prediction algorithms in predicting sparsely annotated functions, without sacrificing the performance on labels with sufficient information. Furthermore, we show that our method can accurately predict genes that will be assigned a functional label that has no known annotations, based only on the ontology graph structure and genes associated with other labels, which further suggests that our method effectively utilizes the similarity between gene functions.     Beschreibung: Two kinds of carbon nanotube (CNT)-graphene structures, vertical CNT-graphene and paralleled CNT–graphene, were fabricated to investigate the geometrical effect on the transport properties of the CNT–graphene junctions by using AC impedance spectra. The results demonstrated that the geometrical structure showed obvious impact on the resistance rather than the capacity of the junction. It is proposed that the difference caused by the geometrical structure may be associated with the dangling bonds terminated by –OH or –COOH of the open-ended CNTs. The unsymmetrical chemical bonds will increase the dipole moment in CNTs, which enhance the interaction between vertical CNTs and graphene and reduce the contact resistance.

    Beschreibung: Recurrent apnea with intermittent hypoxia is a major clinical problem in preterm infants. Recent studies, although limited, showed that adults who were born preterm exhibit increased incidence of sleep-disordered breathing and hypertension, suggesting that apnea of prematurity predisposes to autonomic dysfunction in adulthood. Here, we demonstrate that adult rats that were exposed to intermittent hypoxia as neonates exhibit exaggerated responses to hypoxia by the carotid body and adrenal chromaffin cells, which regulate cardio-respiratory function, resulting in irregular breathing with apneas and hypertension. The enhanced hypoxic sensitivity was associated with elevated oxidative stress, decreased expression of genes encoding antioxidant enzymes, and increased expression of pro-oxidant enzymes. Decreased expression of the Sod2 gene, which encodes the antioxidant enzyme superoxide dismutase 2, was associated with DNA hypermethylation of a single CpG dinucleotide close to the transcription start site. Treating neonatal rats with decitabine, an inhibitor of DNA methylation, during intermittent hypoxia exposure prevented oxidative stress, enhanced hypoxic sensitivity, and autonomic dysfunction. These findings implicate a hitherto uncharacterized role for DNA methylation in mediating neonatal programming of hypoxic sensitivity and the ensuing autonomic dysfunction in adulthood.

    Beschreibung: In order to enhance the uniformity of the radial solids distribution and thereby the performance of industrial circulating fluidized-bed (CFB) risers, an approach by using the air jet from the riser circumference is proposed. The Eulerian-Eulerian computational fluid dynamics (CFD) model with the kinetic theory of granular flow is adopted to simulate the gas-solids two-phase flow in a CFB riser with fluid catalytic cracking (FCC) particles. The numerical results indicate that by employing the circumferential air jet approach under appropriate jet velocities, the maximum solids concentration in the near-wall region can be greatly reduced, the entrance region can be shortened, and the uniformity of the flow structure can be significantly improved. A circumferential air jets approach is proposed to improve the uniformity of lateral or radial solids concentration distributions mainly in the fully developed region. A comprehensive numerical study on the effects of this approach on the flow structure of gas-solids two-phase flows in a circulating fluidized-bed (CFB) riser is conducted. The results could permit optimal designs and operations of CFBs.

    Beschreibung: Neurodegeneration is a devastating manifestation in the majority of 〉50 lysosomal storage disorders (LSDs). Neuronal ceroid lipofuscinoses (NCLs) are the most common childhood neurodegenerative LSDs. Mutations in 13 different genes (called CLN s) underlie various types of NCLs, of which the infantile NCL (INCL) and congenital NCL (CNCL) are the most lethal. Although inactivating mutations in the CLN1 gene encoding palmitoyl-protein thioesterase-1 (PPT1) cause INCL, those in the CLN10 gene encoding cathepsin D (CD) underlie CNCL. PPT1 is a lysosomal thioesterase that cleaves the thioester linkage in S -acylated proteins required for their degradation by lysosomal hydrolases like CD. Thus, PPT1 deficiency causes lysosomal accumulation of these lipidated proteins (major constituents of ceroid) leading to INCL. We sought to determine whether there is a common pathogenic link between INCL and CNCL. Using biochemical, histological and confocal microscopic analyses of brain tissues and cells from Cln1 –/– mice that mimic INCL, we uncovered that Cln10 /CD is overexpressed. Although synthesized in the endoplasmic reticulum, the CD-precursor protein (pro-CD) is transported through endosome to the lysosome where it is proteolytically processed to enzymatically active-CD. We found that despite Cln10 overexpression, the maturation of pro-CD to enzymatically active-CD in lysosome was disrupted. This defect impaired lysosomal degradative function causing accumulation of undegraded cargo in lysosome leading to INCL. Notably, treatment of intact Cln1 –/– mice as well as cultured brain cells derived from these animals with a thioesterase-mimetic small molecule, N - tert -butyl-hydroxylamine, ameliorated the CD-processing defect. Our findings are significant in that they define a pathway in which Cln1 mutations disrupt the maturation of a major degradative enzyme in lysosome contributing to neuropathology in INCL and suggest that lysosomal CD deficiency is a common pathogenic link between INCL and CNCL.

    Beschreibung: The Levant constitutes an important region for assessing linkages between climate and societal changes throughout the course of human history. However, large uncertainties remain in our understanding of the region's hydroclimate variability under varying boundary conditions. Here we present a new high-resolution, precisely-dated speleothem oxygen-carbon isotope and Sr/Ca records, spanning the last 20ka from Jeita Cave, northern Levant. Our record reveals a higher (lower) precipitation-evaporation (P-E) balance during the Last Glacial Maximum and Bølling interstadial (Heinrich stadial 1). The early-mid Holocene is characterized by a trend towards higher P-E state, culminating between ~7 and 6 ka BP. The mid-late Holocene is characterized by two millennial-length drier periods during 5.3–4.2 and 2.8–1.4ka BP. On sub-millennial scale, the northern Levant climate variability is dominated by 500-year periodicity. Comparisons with the regional proxy records suggest persistent out-of-phase climate variability between the northern and southern Levant on a wide-range of timescales.

    Beschreibung: We have simulated the invasion of a polyelectrolyte complex made of a polycation chain and a polyanion chain, by another longer polyanion chain, using the coarse-grained united atom model for the chains and the Langevin dynamics methodology. Our simulations reveal many intricate details of the substitution reaction in terms of conformational changes of the chains and competition between the invading chain and the chain being displaced for the common complementary chain. We show that the invading chain is required to be sufficiently longer than the chain being displaced for effecting the substitution. Yet, having the invading chain to be longer than a certain threshold value does not reduce the substitution time much further. While most of the simulations were carried out in salt-free conditions, we show that presence of salt facilitates the substitution reaction and reduces the substitution time. Analysis of our data shows that the dominant driving force for the substitution process involving polyelectrolytes lies in the release of counterions during the substitution.
