    Notes: Abstract We have developed a mutation detection strategy that combines single strand conformational polymorphism (SSCP) analysis of one strand of a double-stranded amplification product with direct sequencing of the other. Using this strategy, which we find economical of both time and resources, we have identified a G to A transition, which substitutes a serine for glycine residue at position 862 in the major helix of the α1 chain of Type I collagen. We use this mutation, which causes a lethal form of osteogenesis imperfecta, to illustrate the technique.

    Notes: Abstract The near-surface region of 4145 steel following boron implantation was investigated by x-ray photoelectron spectroscopy (XPS). The steel surface was implanted with11B+ ions to a constant dose of 1.0×1017 ions cm−2 at energies of 30 and 135 keV. The XPS spectrum of the implanted surface showed a shift in the B(1s) level towards the higher binding energy. The observed 188.0 eV binding energy of the B(1s) level was found to be in good agreement with the characteristic binding energy of the B(1s) level corresponding to iron boride (Fe2B). Hence the increase in surface hardness reported previously is related to the formation of an iron boride layer in the near-surface region known for its hardening capabilities.

    Notes: A storage ion source has been built for the production of vibrationally cold H+2 and H+3 ions. Using a gas mixture of H2 and rare gases (Ne and He) in the source at high pressure, higher vibrational states of these ions have been effectively depleted by ion–molecule reactions and collisional deactivation. The internal energy of the cold ions was determined by the threshold measurement of collision-induced dissociation. With a mixture of H2 and Ne (ratio 1 : 5) in the source operated at 80 mTorr, H+2 ions were found to be in v=0 and 1 states only and the H+3 ions were in the ground vibrational state.

    Notes: A mathematical model describing simultaneous water diffusion and gelatinization of starch during rice parboiling is proposed. Soaking experiments were conducted on a short-grain rice variety. A computer-aided nonlinear optimization technique is used to find diffusion coefficients and reaction rate constants in the temperature range 50-120°C. It is concluded that the parboiling process is limited by the reaction of starch with water below 85° C and by diffusion of water above 85° C. The activation energy was found to be about half in case of diffusion limited process than that of reaction limited process. The study showed that it is possible to reduce energy input if rice is dehusked before parboiling. The mathematical model shows good correlation between observed and predicted value of water uptake.

    Notes: The effects of water quality on brine discharged from oil and gas recovery operations are described for surface water and ground water in two small watersheds in eastern Kentucky. The brine, which had salinity that was often several times that of sea water, led to significantly higher concentrations of several minerals in surface water, particularly in the first and second order streams. Concentractions as high as 50,000 mg/I for sodium and 64,000 mg/I for chloride were measured in streams. The. differences in chemical concentrations for various chemicals over the period of the study were ascribed to temporal variability, particularly due to differences between wet and dry seasons, and to spatial variability, particularly due to dilution and other chemical decay processes. Chemical decay coefficients for sodium and chloride were developed as a function of watershed area for possible application to similar watersheds. There was some evidence that the brine was influencing the Licking River, the major stream that drains the eastern part of Kentucky.

    Notes: Li+ and N+ ions were implanted into aliphatic polymethylmethacrylate (PMMA), polyvinylalcohol (PVA), and aromatic polyimide (PI) polycarbonate (PC) polymers in the energy range of 100–130 keV. Planar optical waveguides guiding between one and three modes were formed. For low implantation doses (≤ 1014 ions/cm2), total waveguide loss values at λ=633 nm were found to be less than 2 dB/cm. The changes in the refractive index were found to be very large (Δn≥0.05) in the case of PMMA and PVA. We interpret this change in refractive index as being due to the formation of aromatic compounds in the regions of electronic scattering.

    Notes: Transmission electron microscopy in conjunction with x-ray photoelectron spectroscopy (XPS) was used to study the possible microstructural changes occurring in electron-beam-evaporated arsenic trisulfide thin films, when deposited on a sodium chloride substrate from room temperature to 250 °C. These films were found to be amorphous up to 150 °C, while a transition from the amorphous to the polycrystalline phase was observed to take place at 200 °C. The electron-diffraction results showed that the crystallized sulfur-rich phase was predominantly As2S5, along with As2S3. The XPS spectrum showed shifts in binding energy values for As towards higher energies. A shift of 0.9 eV for the As (3d) spectrum was attributed to the transition from the As2S3 to the As2S5 phase occurring at ∼200 °C.
