    Notes: Salinity aggravates B toxicity symptoms in several plant species. In the present study the interactive effects of B toxicity and salinity stresses on the subcellular distribution of boron, cations and proteins in basal and apical leaf sections of wheat were investigated. High B supply increased total B concentrations in all leaf parts, but values remained below 25 mg B kg−1 dry weight (DW) in basal sections, whereas they reached more than 600 mg B kg−1 DW in leaf tips. In basal leaf sections intercellular soluble B concentrations closely reflected the external supply, whereas intracellular soluble B concentrations remained lower by a factor of two, indicating some retention of excess B in the apoplast. Combined salinity and B toxicity stresses significantly increased soluble B concentrations in inter- and intracellular compartments of basal leaf sections in comparison with either stress alone, probably related to salinity-induced changes in water status. The combined stresses also induced quantitative and qualitative changes in inter-, but not intracellular protein composition. Most obvious was the induction of a 25 kDa protein and an increase in amount of a 33 kDa protein. It is suggested that these changes might be due to structural modifications of the cell wall. The concentration of soluble boron in cells is proposed to be an indicator of boron toxicity.

    Notes: Abstract  The secretory vesicles of some cells of the islets of Langerhans of the pancreas contain high amounts of immunoreactive tyrosine phosphatase of the PTP1B/TCPTP subfamily. The cells are located in the peripheral parts of the islets and were identified as glucagon- and pancreatic polypeptide-forming cells. The tyrosine phosphatase is also enriched in some of the somatostatin-producing cells but is not elevated either in insulin-producing B-cells or in the exocrine pancreas. Virtually the same patterns were found in pancretic tissues of rats, guinea pigs, pigs, and mice. High levels of detergent-soluble tyrosine phosphatase were measured in the particular fraction of pancreatic islets with a substrate preferred by PTP1B/TCPTP-type protein tyrosine phosphatases.

    Notes: Summary The acinar activity pattern of phosphoenolpyruvate carboxykinase (PEPCK) was investigated in livers of streptozotocin diabetic male and female rats and in addition in livers of diabetic males, which had undergone estrogen treatment. In all diabetic animals blood glucose levels were supranormal and liver PEPCK activity was increased. This increase in activity was greatest in estrogen treated diabetic males and lowest in diabetic females. Plasma insulin levels were reduced after the application of streptozotocin to otherwise normal male and female rats. Yet, in males treated in addition with estrogens the plasma insulin levels reached the normal range again. The PEPCK activity showed a heterotopic distribution along the acinus. The periportal to perivenous gradient was steeper in males compared to females in the untreated as well as in the diabetic state. The application of estrogens to males resulted in a further steepening of the gradient.

    Notes: Summary The distribution patterns of the enzyme activities of phosphoenolpyruvate carboxykinase, pyruvate kinase, glucose-6-phosphate-dehydrogenase and malic enzyme were determined in the liver acini of men and women by microquantitative means. The activity of PEPCK was higher in men compared to the activity in women. In both sexes no heterotopic distribution pattern was observed. PK activity of men was higher, but in both sexes no heterotopic distribution was detectable. G6PDH and ME showed relatively low activity. The distribution of G6PDH and ME activity was to some extent different in men and women. Yet their heterotopic patterns were not particularly distinct.

    Notes: Summary Male and female juvenile as well as adult rats were treated with acetylsalicilic acid in order to examine the effect of the drug on over-all activity and activity distribution of phosphoenolpyruvate carboxykinase (PEPCK) in the liver acinus. Upon administration of acetylsalicilic acid PEPCK activity increased in juvenile males and adult females, but was reduced in juvenile females and adult males. The periportal-perivenous activity gradient along the sinusoidal length, which is flatter in untreated juvenile rats compared to the livers of adult rats, was distinctly steepened by acetylsalicilic acid treatment. Acetylsalicilic acid did not affect the gradient in adult rats.

    Notes: Summary Phosphoenolpyruvate carboxykinase activity in rat liver was shown to be heterotopically distributed within the acinus under varying feeding conditions. Highest values of PEPCK activity were found in the periportal zone of the acinus from where it decreased continuously towards the perivenous zone. 84 h of starvation resulted in an increase of activity, which was most prominent in the perivenous zone, but nevertheless resulted in a steeper gradient. Refeeding of starved rats with a high carbohydrate diet for 6 nights led to a decrease in PEPCK activity which was most prominent in the periportal zone, but almost negligible in the perivenous zone, resulting in a further change in the activity gradient. Sex-dependent differences for total PEPCK activity were found i) in controls, where the activity was lower in females, ii) after starvation, where the induction was much higher in females, and iii) after refeeding of starved rats, where the activity in females remained higher compared to that of the controls. Differences in the intra-acinar localization of the activity in dependence of the sex were registrated in the control group and in starved rats. Livers from female rats contained a higher periportal/perivenous ratio compared to males. In starved and starved and refed animals the periportal/perivenous ratios were almost the same in both sexes.

    Notes: Summary G6PDH and ME activity was determined biochemically in homogenates and demonstrated histochemically in cryostat sections of rat liver. Control animals were sham-operated, the male and female rats of the experimental groups were castrated. After castration groups of rats were treated with daily doses of 3 or 6 μg/estradiolbenzoate for 21 days. The results show that in the controls there is a sex-dependent distribution pattern of the two enzymes; in males the rather low activity is mainly located in the periportal area, in females the higher activity is demonstrable in the perivenous area. After castration G6PDH activity (and to a lesser extent ME activity) increases, mainly in the periportal zone. Estrogen treatment results in the high activity of both enzymes, which are exclusively located in the perivenous zone. In the periportal zone no G6PDH or ME activity is demonstrable histochemically. This zone-typical effect of estrogen is interpreted in terms of the concept of Metabolic Zonation, according to which it is supposed that the NADPH generating enzymes in the perivenous area have a lipogenic function whereas the periportal activity contributes to bile acid production.
