    Notes: Summary Smooth muscle from small segments of the vas deferens, urinary bladder and the ureter of the guinea pig was transplanted into the anterior chamber of the eye. Some muscle cells had a normal appearance, but others showed morphological changes during the first few days following transplantation. Changes of two main kinds were observed: some cells contained aggregations of glycogen granules and lipid droplets; other cells were characterized by either a general increase in electron density of the cell with the presence of very thick filaments (which are, possibly, aggregates of thin filaments), or cells containing an electron-dense nucleus, ruptured cellular membranous systems and degenerating mitochondria. Following revascularisation, which commenced one to two days after transplantation, areas containing cells with an appearance similar to that of embryonic smooth muscle were apparent. Evidence of mitosis of smooth-muscle and other cell types was also noted, the most active period being five to ten days after transplantation. Bundles containing normal muscle cells were apparent by four weeks in these transplants. However, whole undissected transplants contained normal-looking muscle bundles after one week. These findings are discussed in relation to the repair of wounds to smooth muscle.

    Notes: Summary Smooth muscle cells of the guinea-pig sphincter pupillae were fixed in the presence of 3% lanthanum nitrate. Although only small amounts of lanthanum were found in plasmalemmal vesicles, large deposits were present within a membrane system resembling sarcoplasmic reticulum. The significance of these findings is discussed in relation to the use of lanthanum nitrate as an extracellular tracer.

    Notes: Summary This paper discusses the possibility that fenestrated capillaries may be induced by the presence or action of epithelial cells. Fenestrations were shown to exist in a number of capillaries in close proximity to the epithelium in the in situ guinea-pig vas deferens and ureter. These results suggest further roles for the vas deferens and ureter than those previously conceived. Transplants of these tissues in the anterior eye chamber were, by two weeks, highly vascularized. Those transplants containing epithelium showed fenestrated capillaries in close proximity to these cells. However, in transplants which did not contain epithelial cells, no fenestrations were noted in capillaries.

    Notes: Summary A fine structural study of sternothyroid muscle spindles of young adult rat demonstrates the rare occurrence of unique nerve endings. These endings are situated in the juxta-equatorial region of nuclear-bag fibers, adjacent to the annulo-spiral sensory endings. They consist of a bundle of terminal axons less than 0.3 μ in diameter and appear to be disposed nearly longitudinally to the axis of the intrafusal fibers. Whereas, the annulo-spiral endings consist of a single axon, coiling around the intrafusal fibers. Transverse sections of these muscle spindles reveal these unique endings scattered around the muscle fibers, fitting into depressions on their surface. The innermost axons directly face the muscle surface and are separated by a narrow gap less than 200 Å in width. No Schwann cell process appears to be associated with these endings. From their unique multi-axonal composition, these endings are termed “bundled endings”. Investigations of developing muscle spindles show the occasional presence of a similar multi-axonal composition of sensory endings in perinatal rats. It is suggested that “bundled endings” are sensory in nature, carrying an immature feature over to adult life.

    Notes: Summary The termination of intrafusal muscle fibres was studied in muscle spindles of the tenuissimus muscle of the Chinese hamster. Most nuclear chain fibres terminated within the spindle capsule. The nuclear bag- and nuclear chain fibres that extended beyond the limit of the spindle capsule terminated in the endomysial connective tissue of the adjacent extrafusal fibres. Three types of intracapsular terminations were found: (1) termination on the neighbouring nuclear bag fibre, (2) termination in the periaxial space without any obvious attachment site, or (3) termination on the spindle capsule. The intrafusal fibres were tapered and longitudinal sarcolemmal invaginations were poorly developed or were completely lacking. The fibre ends were characterized by their folded basal lamina, which appeared to be mechanically reinforced by associated collagen fibrils and elastic fibres.

    Notes: Summary By removing connective tissue components with enzymatic digestion followed by HCl-hydrolysis, myoepithelial cells (MECs) of the terminal portion in a variety of exocrine glands of the rat were examined with the scanning electron microscope. The profile of MECs varied considerably from gland to gland; MECs in the lactating mammary gland have a few long cytoplasmic processes in close contact with those of adjacent cells forming a continuous network around the terminal portion. Those of the exorbital lacrimal gland are stellate with many thin radiating processes with tapered ends that terminate freely. MECs in the sublingual gland are characterized by a number of broad and extensive cellular processes. MECs in the submandibular gland are similar in appearance to those of the exorbital lacrimal gland, but with more extensive cellular processes that form a more or less continuous network with those of the adjacent cells. No MECs were observed on the terminal portion of the parotid gland where the cells appear to be lodged on the intercalated duct. The relative surface area covered by MECs per terminal portion was also found to vary significantly, being 24% in the lactating mammary, 17% in the exorbital lacrimal, 48% in the sublingual, and 25% in the submandibular glands. The findings are discussed in relation to the physical properties of secretions in different glands.

    Notes: Summary This study describes three-dimensional aspects of the development and pseudo-unipolarization of neuroblasts and the maturation of satellite cells in prenatal rat dorsal root ganglia, using scanning electron microscopy, after removal of extracellular connective tissue components by trypsin digestion and HC1 hydrolysis. At 14 days of gestation, the vast majority of neurons are spindle-shaped or bipolar and only 3% are unipolar, while at 16 and 18 days this percentage has increased to 30% and 91%, respectively. The initial portions of the central and peripheral neuronal processes gradually approach each other and form a common initial portion. Finally, the cytoplasm of this common initial portion becomes thinner and elongates to form the stem process of the mature cell. Satellite cells are present from the beginning of the period studied, but intricate networks of branching satellite cell processes only develop after about day 17.

    Notes: Summary The cytoarchitecture of the medial layer of rat thoracic aorta was examined by scanning electron microscopy after removal of the connective tissue. The outermost lamella showed a lattice-like structure of muscle bundles of closely apposed smooth muscle cells (SMCs), whereas the inner lamellae consisted of more-or-less continuous muscle sheets of vaguely defined subgroups of parallel SMCs. Longitudinal rows of ridges ran along the adventitial surface of these muscle bundles and sheets. The SMCs of the outermost lamella, were 5.1 μm wide, and varied in shape, whereas those of the inner lamellae, were 52.7 μm long, 2.6 μm wide and 4.1 μm thick, and were elongated, spindle-shaped cells with serrated outlines. These latter SMCs extended obliquely, and partially overlapped each other. The surface of the SMCs in the outermost lamella exhibited a rugged texture, with nodular protrusions and oblique and longitudinal laminar folds, while the inner lamellar cells showed longitudinal laminar folds and finger-like processes on both sides of the ridges, pointing in opposite directions to the ridges. The angle of deviation from the transverse axis of the vessel, of the muscle bundles and subgroups in the outermost lamella, was 33.6°, in the second and third lamellae, 22.5°, and in the innermost lamellae, 12.8°. The mean angle of the muscle bundle and subgroup arrangement, with respect to the long axis of the vessel, however, was basically 90° in all lamellae.

    Notes: Summary The architecture of the media of arterial vessels in dog brain was investigated using scanning electron microscopy. The arrangement and shape of the circularly-oriented smooth muscle cells varied with vessel diameter: The arteries (〉100 μm in diameter) had 4–10 layers of spindle-shaped smooth muscle cells; the muscular arterioles (30–100 μm), 2–3 layers of spindle-shaped smooth muscle cells; the terminal arterioles (10–30 μm), a compact layer of spindle-shaped smooth muscle cells with more dominant nodular or rod-like processes and thin lateral processes; and the precapillary arterioles (5–15 μm), a less compact layer of branched smooth muscle cells. Longitudinally-oriented muscles were observed in the medio-adventitial border. The distribution and arrangement of these muscles varied with vessel size: in the large arteries (〉 300 μm in diameter), at the branching sites only; in the small arteries (100–300 μm), at both the branching and non-branching sites; in the muscular arterioles, at both the branching and non-branching sites in a reticular arrangement with some muscle cells having an “asteroid” appearance; in the terminal aterioles, only “asteroid-like” muscle cells were found at the branching and non-branching sites.

    Notes: Summary Three-dimensional aspects of smooth muscle cells of the microvas-culature were studied ultrastructurally in laboratory rodents by means of serial thin sections and reconstruction of muscle cell models. It was demonstrated that a muscle cell of an arteriole (luminal diameter (LD) 17 μm) in hamster striated muscle was spindle-shaped, 70 μm long, and wound twice round the vessel axis. The volume of the cell was calculated as 750 μm3 and its surface area as 1330 μm2. A muscle cell in an arteriole (LD 6 μm) in the rat retina was irregular in shape, about 22 μm long, and had branched processes. The cell volume was calculated as 139 μm3 and its surface area as 298 μm2.
