    Description / Table of Contents: Karosseriewerkstoffe und deren Korrosionsschutz: AluminiumblechDie Aluminiumindustrie liefert heute spezielle, auf das Anforderungsprofil der Karosseriebauteile maßgeschneiderte Karosseriebleche. Wichtigste Legierungstypen sind AlMgSi (fließfigurenfrei, für Außenhaut geeignet) und AlMg (für den Innenbereich). Neben den erreichbaren Gewichtseinsparungen ist es vor allem die sehr gute Langzeitbeständigkeit der lackierten Aluminium-Karosserie, welche diesen Leichtbauwerkstoff als Alternative zu Stahl interessant macht. Voraussetzung ist cine „aluminiumgerechte“ Oberflächenbehandlung, insbesondere eine schichtbildende Phosphatierung vor der Lackierung. Dabei kann Aluminium im Verbund mit Stahl und verzinktem Stahl in vorhandenen Anlagen lackiert werden. Von den „statischen“ Beständigkeitstests eignet sich nur die verschärfte Freibewitterung (VDA 621-414) vorbehaltlos für eine Beurteilung des Korrosionsschutzwertes der fertig lackierten Karosserieoberfläche. Wichtig ist, daß neben der „richtigen“ Legierung und Verarbeitung auch konstruktive Maßnahmen zur Erreichung der geforderten Langzeitbeständigkeit des Gesamtsystems berücksichtigt werden.
    Notes: The aluminium industry supplies various car body sheet qualities tailor-made to meet the requirements of the car producers. The most important alloy types are AlMgSi (free from stretcher-strain markings after forming and hence suited for outer skin) and AlMg (for interior use). In addition to the significant weight savings it is its long service life which makes aluminium an attractive alternative to steel sheets. A prerequisite for good corrosion resistance is a surface treatment optimised for aluminium. This includes the creation of a phosphate surface layer to allow lacquer coating together with steel and galvanized steel on existing lines.The Accelerated Outdoor Exposure Test (VDA 621-414) proved to be the only static corrosion test method suitable for assessing the resistance level of the final lacquered car body. In addition to the proper selection of alloy and surface finishing technique it is important that some aluminium-specific precautions are taken during manufacture if the full long service potential of the material is to be realized.

    Notes: A promoter sequence, Pan', was isolated from Mycobacterium paratuberculosis and characterized. This promoter lies adjacent to, and outside, the 3’end of an IS900 insertion element. IS900 contains an open reading frame, ORF2, on the complementary strand which codes for the putative transposase of this insertion sequence. A DNA fragment containing PAN and part of ORF2 was fused to the lacZ gene and inserted into the replicative shuttle vector pRR3. Mycobacterium smegmatis and Mycobacterium bovis BCG (BCG) transformed with this plasmid exhibited β-galactosidase activity. However, lacZ was only expressed in Escherichia coli under the control of Pan, when ORF2 was deleted. Immunization of mice with the recombinant M. bovis BCG expressing lacZ resulted in the induction of a high humoral and cellular response directed against β-galactosidase. The Pan-ORF2 expression system may prove to be particularly useful for cloning and expression of heterologous genes in the BCG vaccine strain.

    Notes: The gene encoding a class A β-lactamase was cloned from a natural isolate of Mycobacterium fortuitum (blaF) and from a high-level amoxicillin-resistant mutant that produces large amounts of β-lactamase (blaF). The nucleotide sequences of the two genes differ at 11 positions, including two in the region upstream from the coding sequence. Gene fusions to Escherichia coli lacZ and transcription and expression analysis of the cloned genes in Mycobacterium smegmatis indicated that high-level production of the β-lactamase in the mutant is mainly or wholly due to a single base pair difference in the promoter. These analyses also showed that transcription and translation start at the same position. A comparison of the amino acid sequence of BlaF, as predicted from the nucleotide sequence, with the determined N-terminal amino acid sequence indicated the presence of a typical signal peptide. The fusion of blaF (or biaF) to the E. coli gene phoA resulted in the production of BlaF-PhoA hybrid proteins that had alkaline phosphatase activity. These results demonstrate that phoA can be used as a reporter gene for studying protein export in mycobacteria.

    Description / Table of Contents: Korrosion und Kriechverhalten von Alloy 800 H unter simulierten KohlevergasungsbedingungenBei der Prüfung der FeNiCr-Legierung Alloy 800 in einem Gemisch aus H2S, CO, H2O (sowie in einem CO-freien Gas dieser Zusammensetzung bei 800°C und einem Gesamtdruck von 1,5 bar) kommt es sowohl zu innerer als auch zu äußerer Korrosion unter Bildung von Sulfiden, Oxiden und Carbiden. Die Zeitstandfestigkeit wird bei hoher Belastung der Proben durch die Korrosion deutlich verringert, während sie bei mittleren und niederen Belastungen praktisch unverändert bleibt oder sogar besser wird. Unerwarteterweise entsteht bei der Einwirkung der genannten Gasgemische kein Nickelsulfid, was dem Umstand zugeschrieben wird, daß die Aktivität des Nickels in der Legierung verhältnismäßig niedrig ist, so daß erst bei höheren Schwefelpartialdrucken, als hier verwendet, mit Sulfidbildung zu rechnen wäre. Die unter Belastung auftretende Beschleunigung der Rißbildung läßt sich mit der Annahme erklären, daß die auf dem Metall vorhandene oxidische Deckschicht dann aufreißt, so daß das aggressive Gas freien Zutritt zur Metalloberfläche hat. Durch die Versuche mit dem CO-freien Gas konnte gezeigt werden, daß die Entfestigung vor allem durch die Sulfidierung und Oxidation bedingt ist, während die Aufkohlung, die zu einer Verfestigung führt, die Gesamtentfestigung etwas verlangsamt.
    Notes: When the FeNiCr alloy Alloy 800 is exposed to a mixture of H2S, CO and H2O (as well as to a CO free gas of this composition at 800°C and total pressure of 1.5 bar), internal as well as external corrosion takes place and sulfides, oxides and carbides are formed. Corrosion results in a decrease of creep strength under high load while it remains practically unchanged or is even improved under medium and low load. The unexpected absence of nickel sulfide formation in the above gas mixture is attributed to the comparatively low nickel activity in the alloy so that sulfide formation would occur only at sulphur partial pressures exceeding the pressures used in this investigation. The increase rate of crack propagation under load can be interpreted in terms of the cracking of the oxide surface layer on the metal, the cracks allowing the aggressive gas to freely penetrate to the metal surface. The tests with the CO free gas demonstrate that the strength loss is principally caused by sulfidation and oxidation while carburisation which leeds to a strengthening results in somewhat slower destrengthening rate.

    Notes: Summary All emissions from incomplete combustion contain polycyclic aromatic hydrocarbons (PAH) which are a well-known class of carcinogens. The question whether additional carcinogenic compounds do exist in these emissions can be answered by separating the emission condensate into a PAH-containing and a PAH-free part. These parts have been tested in an animal experiment by means of a carcinogen-specific test system such as topical application onto the skin of mice or injection into the lung of rats. The investigation on the contribution of PAH-fractions and of benzo(a)pyrene to the carcinogenic potential of emission condensate from gasoline driven engines, Diesel engines, coal combustion in domestic furnaces and sidestream smoke of cigarettes shows that the carcinogenic effect of the particle phase of these pyrolytic condensates is predominantly caused by polycyclic aromatic compounds such as PAH. In all cases investigated the PAH-fraction containing 4 and more rings accounted for more than 75% of the total carcinogenic effect resulting from the implantation into the lung of rats or from the topical application onto mouse skin. The contribution of benzo(a)pyrene to the carcinogenic potency of various condensates, however, is minor in all cases investigated and accounts for only 0.17% to 4% of the total carcinogenicity as evaluated from implantation into the lung of rats (Table 1).
