    Notes: The prograde amphibole that coexists with chlorite, epidote, muscovite, albite, quartz and hematite in Sanbagawa schists was examined to investigate the relationship between the prograde P-T paths of individual rocks and the metamorphic field gradient in the Sanbagawa metamorphic belt, central Shikoku. The amphibole changes from actinolite, through ferri-winchite and crossite, to barroisite and hornblende with increasing grade along the metamorphic field gradient. However, the sequence of prograde amphibole compositions in each sample varies in different mineral zones. The general scheme can be summarized as: magnesioriebeckite-riebeckite crossite in the upper chlorite zone of lower-grade rocks; crossite or glaucophane barroisite in the garnet zone of medium-grade rocks; and actinolite or winchite barroisite hornblende in the albite-biotite zone of higher-grade rocks. Changes of amphibole composition indicate that the prograde P-T path recorded in the higher-grade rocks was situated on the higher-temperature side of that of the lower-grade rocks and on the lower-pressure side of the metamorphic field gradient. The systematic change of P-T paths implies an increasing dP/dT during continuous subduction. These features can be interpreted as documenting prograde metamorphism within a young subduction zone that has a non-steady-state geotherm.

    Notes: Two nematode-resistant trisomic lines which were derived from interspecific Beta, vulgaris × B. procumbens hybrids were intercrossed or backcrossed with susceptible diploid sugar beer and their progenies were screened for nematode resistance. The transmission rate of resistance varied from 1.5 % to 47.6 % with an average of 20.4 % in the progenies of individual insomics derived from the two trisomic lines. Eleven resistant diploads were selected with a frequency of 0.2 %. These resistant diploids were classified into two groups, i.e., one group showed relatively high transmission rates of resistance with an average of 25.4 % and the other extremely low with an average of 1.2 % in their backcrossed and s el fed progenies., Meiotic chromosome behavior in a resistant diploid group with high transmission rates was considerably normal as compared to that in a resistant diploid group with low transmission rates. Chromatid bridges and acertric fragments were detected in 93 % of resistant diploids and in 46 % of susceptible diploids. Two different sized fragments occurred in resistant diploids, while only a smaller fragment was present in susceptible diploids. A frequency of sporocytes with bridges-fragments was 17.4% at anaphase I and 13.9 % at anaphase II in resistant diploids, while in susceptible diploids a frequency was 2.9 % and 5.3 % at the respective stages. These results suggest that at least two paracentric inversions are present in resistant diploids, one of which is linked to nernatode resistance and may be responsible for the low transmission rate of resistance.

    Notes: Abstract Objective: We investigated whether interindividual differences in the pharmacokinetic disposition of lansoprazole are attributed to the genetic polymorphism of CYP2C19 which occurred by two mutations, CYP2C19m1 and CYP2C19m2, in 20 Japanese subjects. Methods: Polymerase chain reaction (PCR) restriction fragment length polymorphism procedures were used to detect the CYP2C19m1 mutation in exon 5 and the CYP2C19m2 mutation in exon 4 using SmaI and BamHI, respectively. Results: Ten subjects were homozygous (wt/wt subjects) for the wt allele in both exon 5 and exon 4, four subjects were heterozygous (wt/m1) for the CYP2C19m1 mutation, and two subjects were heterozygous (wt/m2) for the CYP2C19m2. The remaining four subjects had both mutated alleles in CYP2C19 genes, i.e., two were homozygous (m1/m1) for the defect in exon 5 and two were heterozygous (m1/m2) for the two defects in exons 5 and 4. The subjects in group 1 (wt/wt, wt/m1 and wt/m2) were the extensive metabolizers (EMs) for 5-hydroxylation of lansoprazole and were in the range of hydroxylation indexes from 3.83 to 19.8, whereas the subjects in group 2 (m1/m1 and m1/m2) were the poor metabolizers (PMs) and the indexes were in the range of 38.5 to 47.6. In group 2, AUC, t1/2 and CL/f of lansoprazole were significantly greater, longer, and lower, respectively, than those in group 1. Conclusion: The hydroxylation of lansoprazole to 5-hydroxylansoprazole was apparently impaired in the subjects with the genetic defects of CYP2C19 (m1/m1 or m1/m2).

    Notes: Summary The pharmacokinetics of tosufloxacin enantiomers after oral administration of racemic tosufloxacin were examined in healthy volunteers. Only small differences were observed in time to peak concentration (2.6±0.3 [mean ± SEM] h for (+)-tosufloxacin vs 2.4±0.2 h for (−)-tosufloxacin), elimination half-life (3.61±0.24 h vs 3.49±0.23 h), and area under the curve (2.78±0.19 h·μg/ml vs 2.87±0.19 h·μg/ml); however, peak concentration (0.40±0.03 μg/ml vs 0.44±0.03 μg/ml), renal clearance (226±10 ml/min vs 202±10 ml/min), and urinary recovery (35.4±2.2% vs 32.4±1.9%) differed significantly between enantiomers.

    Notes: Summary Alloplasmic common wheat (Triticum aestivum L. cultivais ‘Penjamo 62’ and ‘Siete Cerros 66’) with cytoplasms of wheatgrass (Agropyron trichophorum and Ag. glaucum) showed two aberrant phenotypes, i.e., gross reduction in plant vigor and male sterility. Plant vigor and male fertility were restored by cytoplasm-specific telocentric chromosomes (telosomes). Studies on carbon assimilation and consumption and on oxygen evolution and uptake showed that maximum rates of apparent photosynthesis were significantly lower in the alloplasmic lines than in their corresponding euplasmic lines and that the telosomes restored a normal level of photosynthesis. The decreased apparent photosynthetic rates in the alloplasmic lines were shown to be not due to decreased rates of true photosynthesis but to increased rates of dark respiration in the green leaves. In contrast, dark respiration in the roots was significantly low in the alloplasmic lines. The alloplasmic lines also showed decreased rates of respiratory consumption of new photosynthates. These results suggest that growth depression and male sterility in the alloplasmic lines are related to aberrant mitochondrial function, which is compensated for by cytoplasm-specific telosomes.

    Notes: Summary In vitro culture of inflorescence tissue of a Triticum crassum (6x) x Hordeum vulgare cv. ‘Bomi’ (2x) intergeneric hybrid resulted in the proliferation of totipotent callus from which plants were regenerated. Regeneration was also achieved from immature inflorescence callus of T. crassum but not from H. vulgare. T. crassum x H. vulgare regenerates had a somatic chromosome number of 28, identical to that of the original hybrid. Four chimeric plants with a partially doubled chromosome number were obtained by in vitro colchicine treatment of hybrid callus prior to induction of plant regeneration. All T. crassum regenerates had 35 chromosomes rather than the expected number of 42. Meiotic analysis of a 35-chromosome plant revealed an extremely abnormal meiosis which might be attributable to a complete disturbance in meiotic control system(s) including that of meiotic pairing.

    Notes: Abstract  To develop a transposon tagging system in wheat (Triticum aestivum L.), transgenic wheat lines containing a transposase gene of the maize Activator (Ac) element were produced and characterized. The Ac transposase gene under the control of a cauliflower mosaic virus 35S promoter was introduced into cultured wheat embryos by particle bombardment. Several transgenic wheat plants expressing the transposase gene were independently recovered. Southern- and Northern-blot analyses of their progeny showed that the expression of the Ac transposase gene was stably inherited, and three fixed Ac lines were established. By RT-PCR analysis, products from fully spliced transcripts of the Ac element were confirmed. Cultured embryos isolated from the stable Ac lines were further bombarded with plasmids having a maize Dissociation (Ds) element located between a rice Act1 promoter and a β-glucuronidase (gus) gene, and transient gus expression was observed after the Ds excision. These findings suggest that the maize Ac transposase gene is precisely processed and an active transposase protein is synthesized in the transgenic Ac lines. The Ds element is trans-activated and excised in wheat cells by the action of the Ac transposase gene.

    Notes: Summary Based on successful results of an acetocarmine squash method for staining chromosomes of many plant species, the method was applied to chromosomes of sugar beet (Beta vulgaris L.). This simple method proved to be applicable to the relatively small chromosomes of sugar beet. Several known disadvantages of acetocarmine staining techniques are overcome by applying acid hydrolysis and maceration. Squashing the stained specimens in 45% acetic acid increases the contrast between stained chromosomes and destained cytoplasm. The advantages of this method are its simple procedure and applicability to both somatic and meiotic chromosomes. Uniform and high quality preparations make detailed analysis possible even with the relatively small chromosomes of sugar beet.
