    Notes: Xylella fastidiosa strains are responsible for several plant diseases and since such isolates display a broad host range and complex biological behavior, genomic comparisons employing microarray hybridizations may provide an effective method to compare them. Thus, we performed a thorough validation of this type of approach using two recently sequenced strains of this phytopathogen. By matching microarray hybridization results to direct sequence comparisons, we were able to establish precise cutoff ratios for common and exclusive sequences, allowing the identification of exclusive genes involved in important biological traits. This validation will enable the use of microarray-based comparisons across a wide variety of microorganisms

    Notes: A biofilm is a community of microorganisms attached to a solid surface. Cells within biofilms differ from planktonic cells, showing higher resistance to biocides, detergent, antibiotic treatments and host defense responses. Even though there are a number of gene expression studies in bacterial biofilm formation, limited information is available concerning plant pathogen. It was previously demonstrated that the plant pathogen Xylella fastidiosa could grow as a biofilm, a possibly important factor for its pathogenicity. In this study we utilized analysis of microarrays to specifically identify genes expressed in X. fastidiosa cells growing in a biofilm, when compared to planktonic cells. About half of the differentially expressed genes encode hypothetical proteins, reflecting the large number of ORFs with unknown functions in bacterial genomes. However, under the biofilm condition we observed an increase in the expression of some housekeeping genes responsible for metabolic functions. We also found a large number of genes from the pXF51 plasmid being differentially expressed. Some of the overexpressed genes in the biofilm condition encode proteins involved in attachment to surfaces. Other genes possibly confer advantages to the bacterium in the environment that it colonizes. This study demonstrates that the gene expression in the biofilm growth condition of the plant pathogen X. fastidiosa is quite similar to other characterized systems.

    Notes: Xylella fastidiosa is the causal agent of citrus variegated chlorosis and Pierce's disease which are the major threat to the citrus and wine industries. The most accepted hypothesis for Xf diseases affirms that it is a vascular occlusion caused by bacterial biofilm, embedded in an extracellular translucent matrix that was deduced to be the exopolysaccharide fastidian. Fourier transform infrared spectroscopy analysis demonstrated that virulent cells which form biofilm on glass have low fastidian content similar to the weak virulent ones. This indicates that high amounts of fastidian are not necessary for adhesion. In this paper we propose a kinetic model for X. fastidiosa adhesion, biofilm formation, and virulence based on electrostatic attraction between bacterial surface proteins and xylem walls. Fastidian is involved in final biofilm formation and cation sequestration in dilute sap.

    Notes: Abstract Using a simplified form of the bremsstrahlung cross-section, we obtain an analytic expression for the intensity of electron-beam-produced hard X-ray emission with depth in solar flares. The results show that ‘footpoint’ emission is more likely than previously thought, and we discuss these results in the light of recent observations.

    Notes: Abstract We have qualitatively analyzed, in the H and K lines spectral region, 31 flares covering part of umbrae or penumbrae of sunspots. A strong narrowing of the emission lines has been observed over the umbrae, and the lines are, in general, much weaker than in common flares suggesting that the optical thickness is quite low in these parts. We have calculated the Stark broadening of the Hɛ line from the general theory, and it has been applied to obtain the electron density in 9 flare spectra. In all cases it has been found that n e 〉 1013 cm−3. Goldberg's method has been applied to find the kinetic temperature from the H and K lines of Ca ii, and from the ratio between the central intensities of the lines we have calculated the optical thickness in the K line. Much evidence supports the assumption that the flare emission is highly diluted The term ‘dilution’ has been used with the same meaning as in Švestka's (1972) paper, i.e., the case when the integrated flare area is not completely covered by the flare elements. in the cases considered, and we propose a two-component model for the calcium emission lines.

    Notes: Abstract We have studied three pairs of prominences, two of them clearly showing the effect of interaction by a dynamical plasma bridge. The other one was a superimposed optical pair which also showed, after being spectroscopically studied, the possible effect of interaction. We have calculated the atom density in the prominence and bridges, and applied the Goldberg-Unno method for the determination of Doppler-widths of the calcium H and K lines.

    Notes: Abstract We analyze Ca ii K-line profiles of one flare and EUV continuum observations of two other flares in order to infer values for the temperature enhancements (over active region values) produced in the upper photosphere around and above the temperature minimum region. The results, obtained through a partial redistribution calculation of the Ca ii K-line profiles and an LTE approach to the continuum observations, show that the flare temperature minimum is depressed some two scale heights below its preflare level, and that substantial temperature enhancements are produced even at this depth. Estimates for the energy release in these photospheric layers are given, and are found to be comparable with that released in chromospheric Hα and Lα emission. We then turn our attention to the investigation of possible heating mechanisms which might be responsible for the observed enhancements. Bombardment by both electrons and protons, and irradiation by soft X-rays, are each considered and found to be largely ineffective, due to the large attenuation of flux by photospheric depths, unless new ideas on the precise nature of these mechanisms are invoked, particularly if the same mechanism is also to explain the observed chromospheric emissions. We therefore conclude that it is most likely that some other mechanism must be advocated in order to explain the observed heating. Possibilities for this are (a) heating by EUV radiation, (b) proton beams with low dispersion energy spectra centered around 10–20 MeV, and (c) localized heating at temperature minimum levels.

    Notes: Abstract A study is made of several ATM flares, to investigate the effect of soft X-ray heating at chromospheric levels. It is shown that the amount of energy released in Lyα and Lyman continuum emissions, and their spatial and temporal behavior, in compact flares correspond to what is expected from model calculations. An additional source of heating, which could be heat conduction, has to be postulated to explain ribbons of enhanced Lα and C ii λ1336 Å emission in large flares. As expected, the effect of X-ray heating is more important in small compact flares than in large ones.
