    Description: Hearing loss is the most widespread sensory disorder, with an incidence of congenital genetic deafness of 1 in 1600 children. For many ethnic populations, the most prevalent form of genetic deafness is caused by recessive mutations in the gene gap junction protein, beta 2, 26 kDa ( GJB2 ), which is also known as connexin 26 ( Cx26 ). Despite this knowledge, existing treatment strategies do not completely recover speech perception. Here we used a gene delivery system to rescue hearing in a mouse model of Gjb2 deletion. Mice lacking Cx26 are characterized by profound deafness from birth and improper development of cochlear cells. Cochlear delivery of Gjb2 using an adeno-associated virus significantly improved the auditory responses and development of the cochlear structure. Using gene replacement to restore hearing in a new mouse model of Gjb2- related deafness may lead to the development of therapies for human hereditary deafness.

    Description: 〈span〉〈div〉Abstract〈/div〉In Japan, folklore says that uncommon appearances of deep‐sea fish are an earthquake precursor. If this folklore is proved to be true, the appearance of deep‐sea fish could be useful information for disaster mitigation. However, a statistical survey has not been conducted on this subject because a database of such information had yet to be compiled. In Japanese domestic local newspapers, such appearances have often been reported because rare appearances might attract readers. The authors constructed a database of reports from newspapers, academic articles, and the marine museum. In this study, fish species generally implicated in earthquakes, such as oarfish and slender ribbonfish, were the focus. Although the catalog used might not include all of the events of deep‐sea fish appearances around Japan because of a lack of whole coverage observation, the earthquake occurrence rate after deep‐sea fish appearances can be evaluated. Thus, the usefulness of the deep‐sea fish appearance information for disaster mitigation was evaluated. From this investigation, the spatiotemporal relationship between deep‐sea fish appearances and earthquakes was hardly found. Hence, this Japanese folklore is deemed to be a superstition attributed to the illusory correlation between the two events.〈/span〉

    Description: Ebola virus is a highly pathogenic filovirus causing severe hemorrhagic fever with high mortality rates. It assembles heterogenous, filamentous, enveloped virus particles containing a negative-sense, single-stranded RNA genome packaged within a helical nucleocapsid (NC). We have used cryo-electron microscopy and tomography to visualize Ebola virus particles, as well as Ebola virus-like particles, in three dimensions in a near-native state. The NC within the virion forms a left-handed helix with an inner nucleoprotein layer decorated with protruding arms composed of VP24 and VP35. A comparison with the closely related Marburg virus shows that the N-terminal region of nucleoprotein defines the inner diameter of the Ebola virus NC, whereas the RNA genome defines its length. Binding of the nucleoprotein to RNA can assemble a loosely coiled NC-like structure; the loose coil can be condensed by binding of the viral matrix protein VP40 to the C terminus of the nucleoprotein, and rigidified by binding of VP24 and VP35 to alternate copies of the nucleoprotein. Four proteins (NP, VP24, VP35, and VP40) are necessary and sufficient to mediate assembly of an NC with structure, symmetry, variability, and flexibility indistinguishable from that in Ebola virus particles released from infected cells. Together these data provide a structural and architectural description of Ebola virus and define the roles of viral proteins in its structure and assembly.

    Description: In the visual system, the establishment of the anteroposterior and dorsoventral axes in the retina and tectum during development is important for topographic retinotectal projection. We identified chick Ventroptin, an antagonist of bone morphogenetic protein 4 (BMP-4), which is mainly expressed in the ventral retina, not only with a ventral high-dorsal low gradient but also with a nasal high-temporal low gradient at later stages. Misexpression of Ventroptin altered expression patterns of several topographic genes in the retina and projection of the retinal axons to the tectum along both axes.     Description: Semaphorins and their receptor plexins constitute a pleiotropic cell-signalling system that is used in a wide variety of biological processes, and both protein families have been implicated in numerous human diseases. The binding of soluble or membrane-anchored semaphorins to the membrane-distal region of the plexin ectodomain activates plexin's intrinsic GTPase-activating protein (GAP) at the cytoplasmic region, ultimately modulating cellular adhesion behaviour. However, the structural mechanism underlying the receptor activation remains largely unknown. Here we report the crystal structures of the semaphorin 6A (Sema6A) receptor-binding fragment and the plexin A2 (PlxnA2) ligand-binding fragment in both their pre-signalling (that is, before binding) and signalling (after complex formation) states. Before binding, the Sema6A ectodomain was in the expected 'face-to-face' homodimer arrangement, similar to that adopted by Sema3A and Sema4D, whereas PlxnA2 was in an unexpected 'head-on' homodimer arrangement. In contrast, the structure of the Sema6A-PlxnA2 signalling complex revealed a 2:2 heterotetramer in which the two PlxnA2 monomers dissociated from one another and docked onto the top face of the Sema6A homodimer using the same interface as the head-on homodimer, indicating that plexins undergo 'partner exchange'. Cell-based activity measurements using mutant ligands/receptors confirmed that the Sema6A face-to-face dimer arrangement is physiologically relevant and is maintained throughout signalling events.     Description: Krev-1 is an anti-oncogene that was originally identified by its ability to induce morphologic reversion of ras-transformed cells that continue to express the ras gene. The Krev-1-encoded protein is structurally related to Ras proteins. The biological activities of a series of ras-Krev-1 chimeras were studied to test the hypothesis that Krev-1 may directly interfere with a ras function. The ras-specific and Krev-1-specific amino acids immediately surrounding residues 32 to 44, which are identical between the two proteins, determined whether the protein induced cellular transformation or suppressed ras transformation.     Description: 〈p〉Tephra beds are considered to be potential failure planes for submarine landslides. Here, we report on an example of a coarse-ash/lapilli-tuff bed influencing translational slides. The studied mass-transport deposit (MTD) is intercalated in the Pleistocene forearc basin fill exposed in east-central Japan. This MTD consists of stacked siltstone blocks resulting from repeated imbricate thrusts branching from the décollement. The basal slide plane is located immediately below a pumice-rich coarse ash/lapilli-tuff bed. The material comprising the slip zone is injected into the overlying coarse-ash/lapilli-tuff bed, suggesting an upwards escape of excess porewater that resulted from elevated pore pressure. To explain this mode of occurrence, we propose that the detachment preferentially occurred at the top and base of the coarse-ash-tuff-rich interval which appears to have been stronger relative to the adjacent silt-dominated interval. The pumiceous coarse-ash and lapilli-tuff bed behaved as a rigid plate on top of the high-pore-pressure slip zone, which sustained the translational slide on the gentle continental slope. Therefore, in translational submarine landslides, the preferential formation of a slide plane is caused by differing frictional resistances in the layered sediments.〈/p〉
