    Description: Accelerated telomere loss has been proposed to be a factor leading to end-stage organ failure in chronic diseases of high cellular turnover such as liver cirrhosis. To test this hypothesis directly, telomerase-deficient mice, null for the essential telomerase RNA (mTR) gene, were subjected to genetic, surgical, and chemical ablation of the liver. Telomere dysfunction was associated with defects in liver regeneration and accelerated the development of liver cirrhosis in response to chronic liver injury. Adenoviral delivery of mTR into the livers of mTR(-/-) mice with short dysfunctional telomeres restored telomerase activity and telomere function, alleviated cirrhotic pathology, and improved liver function.     Description: Herbivore effects on leaf litter can have a strong impact on ecosystem nutrient cycling. Although such effects are well described for insect herbivory, research on the impacts of browsing by mammalian herbivores on leaf litter dynamics and nutrient cycling has been more limited, particularly at the level of the individual plant. Clipping treatments (66% shoot removal twice, plus unclipped) were applied to analyse the effect of browsing on the phenology (start date and pattern of leaf shedding) and leaf litter quality (nitrogen (N), soluble sugars, starch and total non-structural carbohydrate concentrations, plus C : N ratios) of Betula pubescens Ehrh. and Quercus petraea [Matt.] Liebl. saplings. Clipping decreased leaf litter biomass and delayed leaf senescence and shedding, but did not change the phenological timing of litterfall between senescence and shedding. The quality of leaf litter of both species was increased by simulated browsing, through an increase in N and carbohydrate concentrations (mainly soluble sugars) and a decreased C : N ratio. This is the first evidence we are aware of that browsing may cause changes in leaf shedding phenology, delaying the process without altering its pattern. Our results also indicate that simulated browsing increases the quality of leaf litter. However, the potential positive effect of browsing on N cycling through litter quality may be offset by its negative impact on the amount of N shed per tree.

    Description: Given their heterogeneity and lack of defining markers, it is surprising that multipotent mesenchymal stem/stromal cells (MSCs) have attracted so much translational attention, especially as increasing evidence points to their predominant effect being not by donor differentiation but via paracrine mediators and exosomes. Achieving long-term MSC donor chimerism for treatment of chronic disease remains a challenge, requiring enhanced MSC homing/engraftment properties and manipulation of niches to direct MSC behaviour. Meanwhile advances in nanoparticle technology are furthering the development of MSCs as vehicles for targeted drug delivery. For treatment of acute injuries, systemic cell-free exosome delivery may ultimately displace current emphasis on empiric donor-cell transplantation for anti-inflammatory, immunomodulatory and repair-promoting effects. Exploration of potential clinical sources of MSCs has led to increased utilisation of perinatal MSCs in allogeneic clinical trials, reflecting their ease of collection and developmentally advantageous properties. Three major paradigms are envisaged for the exploitation of mesenchymal stem cells (MSC) as therapeutic agents. Each paradigm utilises different properties ascribed to MSC regardless of their ontological source. These include low immunogenicity, differentiation potential, rich production of paracrine factors and being amenable to modification to act as therapeutic-carriers.

    Description: We discuss a portable edge illumination x-ray phase contrast imaging system based on compact piezoelectric motors, which enables its transportation to different environments, e.g., hosting different x-ray source technologies. The analysis of images of standard samples reveals an angular sensitivity of 270 ± 6 nrad, which compares well with the 260 ± 10 nrad reported for previous systems based on stepper motors, demonstrating that system portability can be achieved without affecting phase sensitivity. The results can also be considered a test of the performance of the piezoelectric motors, and as such could be of interest to researchers planning their use in other imaging systems.

    Notes: Nuclear magnetic resonance (NMR) spectroscopy and magnetic resonance imaging (MRI) were used to analyse changes in the physical state of water in wheat crowns during cold acclimation and during the freezing/thawing cycle. Spectroscopically measured average spin-spin relaxation times (T2) decreased during cold acclimation and increased when plants were grown at normal temperature. Spin-spin relaxation images whose contrast is proportional to T2, times were calculated allowing association of water relaxation with regions of tissue in spin-echo images during acclimation and freezing. Images taken during freezing revealed nonuniform freezing of tissue in crowns and roots. Acclimated and non-acclimated wheat crowns were imaged during freezing and after thawing. Spin-echo image signal intensity and T2 times decreased dramatically between -4°C and -8°C as a result of a decrease in water mobility during freezing. Images collected during thawing were diffuse with less structure and relaxation times were longer, consistent with water redistribution in tissue after membrane damage.
