    Notes: Abstract. A digoxigenin-labelled DNA probe was used for in situ detection of the Penaeus monodon-type baculovirus (MBV) derived from cloned MBV polyhedrin genome in cultured Penaeus monodon Fabricius. First, the specificity of the probe against MBV DNA with dot blot hybridization analysis was verified. This probe indicated that cloned MBV polyhedrin fragment can be used as an MBV-specific probe. This was then used to microscopically examine sections of MBV-infected tissues for a blue-purple precipitate indicative of a positive reaction for MBV. MBV-positive cells were located only in the epithelium of the hepatopancreatic tubules and of the midgut. Furthermore, comparison of the susceptibility to MBV infection among several life-stages of the shrimp showed that the MBV genome was found in the zoea, mysis, post-larva, and adult stages, whereas MBV DNA was not detected in either eggs or nauplii. The results were quantified from in situ hybridization with an image analyser to compare the degree of cell infection among groups of cultured P. monodon collected from various farms in Taiwan.

    Notes: Abstract. Penaeus monodon-type baculovirus (MBV) was isolated and purified from the hepatopancreases of MBV-infected Penaeus monodon Fabricius. MBV DNA was extracted and used as a template in a polymerase chain reaction (PCR). The primers were chosen from conserved regions of the polyhedrin gene of Autographa californica nuclear polyhedrosis virus (AcNPV). One DNA fragment (674 base pairs) was amplified after PCR. There was a 65% homology between the predicted amino acid sequence of this PCR product with that of the polyhedrin polypeptide of AcNPV. Nucleotide sequence analysis indicated that the amplified DNA is the open reading frame of the MBV polyhedrin gene. This 674 bp DNA fragment was subsequently used as a probe in a dot blot analysis. The probe was able to hybridize with the DNA extracted from the purified MBV and from the MBV-infected P. monodon, but not from the MBV uninfected P. monodon.

    Notes: Since 1987, farmers in southern Taiwan have reported significant disease-related mortalities in freshwater prawns. Most mortalities have occurred during the winter, and usually almost all the adult prawns die within 4–5 days after clinical signs appear. Histopathological studies show that cells in hepatopancreatic ducts and tubules are vacuolized and degenerated. Large numbers of membrane-bound yeast aggregates are observed in the affected tissues. Two hundred and seventeen yeast isolates were obtained from diseased prawns, pond water and sediments from six areas. DNA restriction fragment length polymorphism (RFLP) was used to analyse the yeast genomes and to categorize them into five groups. Conventional biochemical and metabolic methods were then used to identify the yeasts of each group. The results showed that two strains of Candida sake (I and II), Pichia anomala, Endomyces fibuliger and Candida famata were present. In addition, the RAPD (random amplified polymorphic DNA) method was used to determine their genomic similarities. Two strains (I and II) of C. sake were the most similar (72%). C. sake I appears to be the primary causative agent of disease, based on frequencies of occurrence of the yeasts found in the diseased prawns.

    Notes: The morphogenesis of a highly organized cellular membrane structure, a membranous labyrinth (ML), in the cells of penaeid shrimp infected with Penaeus monodon baculovirus (MBV) is described. The ML possibly originates from dilated Golgi apparatus, endoplasmic reticulum (ER) or the outer nuclear membrane. The ML apparently grows by proliferation of the cellular membranes of these systems. In addition, its degree of development was correlated with stages of MBV reproduction.

    Notes: Abstract Recently, two distinct cDNA clones encoding the catalytic subunit of the vacuolar H+-ATPase (V-ATPase) were isolated from the allotetraploid cotton species Gossypium hirsutum L. cv ‘Acala SJ-2’ (Wilkins 1992, 1993). Differences in the nucleotide sequence of these clones were used as molecular markers to explore the organization and structure of the V-ATPase catalytic subunit genes in the A and D genomes of diploid and allotetraploid cotton species. Nucleotide sequencing of polymerase chain reaction (PCR) products amplified from G. arboreum (A2, 2n=26), G. raimondii (D5, 2n=26), and G. hirsutum cv ‘Acala SJ-2’ [(AD)1, 2n=4x=52] revealed a V-ATPase catalytic subunit organization more complex than indicated hitherto in any species, including higher plants. In the genus Gossypium, the V-ATPase catalytic subunit genes are organized as a superfamily comprising two diverse but closely related multigene families, designated as vat69A and vat69B, present in both diploid and allotetraploid species. As expected, each vat69 subfamily is correspondingly more complex in the allotetraploid species due to the presence of both A and D alloalleles. Because of this, about one-half of the complex organization of V-ATPase catalytic subunit genes predates polyploidization and speciation of New World tetraploid species. Comparison of plant and fungal V-ATPase catalytic subunit gene structure indicates that introns accrued in the plant homologs following the bifurcation of plant and fungi but prior to the gene duplication event that gave rise to the vat69A and vat69B genes approximately 45 million years ago. The structural complexity of plant V-ATPase catalytic subunit genes is highly conserved, indicating the presence of at least ten introns dispersed throughout the coding region.

    Description: Paralarval and juvenile cephalopods collected in plankton samples on 21 western North Atlantic cruises were identified and enumerated. The 3731 specimens were assigned to 44 generic and specific taxa. This paper describes their spatial and temporal distributions and their developmental morphology. The smallest paralarvae recognized for anumber of species are identified and illustrated. The two most abundant and most frequently collected taxa were identifiable to species based on known systematic charactersof young, as well as on distribution of the adults. These were the neritic squids Loligo pealeii and Illex illecebrosus collected north of Cape Hatteras, both valuablefishery resources. Other abundant taxa included two morphotypes of ommastrephids, at least five species of enoploteuthids, two species of onychoteuthids, and unidentified octopods. Most taxa were distributed widely both in time and in space, although some seasonal andmesoscale-spatial patterns were indicated. The taxa that appeared to have distinct seasonal distribution included most of the neritic species and, surprisingly, the young of the bathypelagic cranchiids. In eight seasonal cruises over the continental shelf of the middle U.S. Atlantic states,neritic taxa demonstrated approximately the same seasonal patterns during two consecutive years. Interannual differences in the oceanic taxa collected on the shelfwere extreme. The highest abundance and diversity of planktonic cephalopods in the oceanic samples were consistently found in the vicinity of the Gulf Stream.Only eight of the oceanic taxa appeared to have limited areal distributions, compared with twelve taxa that were found throughout the western North Atlantic regionssampled in this study. Many taxa, however, were not collected frequently enough to describe seasonal or spatial patterns. Comparisons with published accounts of other cephalopod surveys indicate both strengths and weaknesses in various sampling techniques for capturing the youngof oceanic cephalopods. Enoploteuthids were abundant both in our study and in other studies using midwater trawls in several areas of the North Atlantic. Thus, this family probably is adequately sampled over its developmental range. In contrast, octopoteuthids and chtenopterygiids are rare in collections made by small to medium-sized midwater trawls but are comparatively common in plankton samples. Forfamilies that are relatively common in plankton samples, paralarval abundance, derived similarly to the familiar ichthyoplankton surveys of fisheries science, may be the most reliable method of gathering data on distribution and abundance.
