    Notes: Abstract  Intrageneric phylogeny among ten representative Ceanothus species was investigated using DNA sequences of the chloroplast encoded ndhF and rbcL genes. Parsimony analysis of the ndhF sequences identified two main clades corresponding to two subgenera Ceanothus and Cerastes. The phylogenetic results suggest that three monophyletic clades within the subgenus Ceanothus can be delimited on the basis of (1) evergreen or (2) deciduous leaves and (3) thorn presence within the evergreen clade. The estimated divergence time based on rbcL sequences suggests that the two subgenera diverged 18–39 million years ago whereas species within each subgenus diverged more recently. Taken together, the results support the division of Ceanothus into two monophyletic subgenera and are consistent with the postulated recent divergence of many species within each subgenus.

    Notes: Abstract  In the genus Pinus the internal transcribed spacers (ITS1 and ITS2) and the 5.8s region of the nuclear ribosomal DNA are approximately 3000 bp in length. ITS1 is considerably longer than ITS2 and partial sequences of ITS1 indicate that this region is evolving rapidly and exhibits intraspecific variation. The ITS2 and 5.8s regions are relatively conserved. We surveyed restriction fragment length variability of PCR-amplified fragments (PCR-RFLP) of the ITS region in four populations (86 individuals) of Pinus rzedowskii, a pine endemic to western Michoacán, Mexico. Five of the restriction endonucleases assayed revealed variation, with a total of 13 variants, most of which were length mutations of 300–900 bp. A moderate degree of population differentiation was detected. The average diversity (Shannon’s index) of ITS fragment size patterns was 1.19, with 34% of the variation due to differences among populations and 66% due to differences among individuals within populations. The same individuals were assayed for nine polymorphic isozymes, which gave diversity measures similar to those of each restriction endonuclease.

    Notes: Abstract Datisca (Datiscaceae) is a ditypic genus with an intercontinentally disjunct distribution. Chloroplast DNA restriction site data was obtained from 23 populations and four 10–20 year old herbarium specimens ofD. glomerata and three populations ofD. cannabina from throughout their geographic ranges in western North America and southwest-central Asia, respectively. InD. glomerata, plastome diversity is partitioned geographically. All populations from southern California have a common plastome, while most populations north of this region share a relatively divergent plastome (0.49% sequence divergence). Likewise, these plastomes are highly divergent (0.87% mean sequence divergence) from those found inD. cannabina. Biogeographic processes dating to the Pleistocene and Late Miocene may be responsible for these intra- and interspecific patterns of chloroplast DNA divergence.

    Notes: Trifolium incarnatum L.) is a cool-season annual forage legume. Few morphological traits are recorded to distinguish genotypes and very little is known about its genetic diversity. The objective of this research was to classify and characterize the genetic diversity in the USDA-ARS National Plant Germplasm System (NPGS) accessions and USA and European Economic Community (EEC) cultivars. Descriptions of EEC cultivars and their breeding histories are also presented. A secondary objective was to determine if T. incarnatum var. molinerii (Balb. ex Hornem.) Ser. should be considered a distinct species. Genomic DNA from leaves of 37 taxa was examined in 1993 by the polymerase chain reaction (PCR) using 29 random amplified polymorphic DNA (RAPD) markers. Plants grown in the greenhouse were also observed for inflorescence color and relative flowering time. Cluster analysis of pairwise means of RAPD maker distances and cultivar pedigree histories revealed similar genetic backgrounds among most cultivars. Flower color was associated with RAPD marker grouping (P ≤ 0.02) but flowering time was not (P ≤ 0.31). Both red vs. non-red inflorescence color and early- vs. late-flowering traits could be predicted by specific RAPD bands (95 and 93% accuracy, respectively). Analysis of internal transcribed spacer region sequences (ITS) from ribosomal DNA (rDNA) showed diverse crimson clover taxon to be more similar to one another than to var. molinerii (0.4 and 0.9% base pair differences, respectively; P ≤ 0.001). Because of the limited diversity within the current NPGS collection, additional accessions should be collected, morphologically evaluated, and genetically analyzed to broaden the genetic base of the collection. Crimson clover var. molinerii is a sister taxa of more typical crimson clover accessions but exhibits enough ITS diversity to be classified as another species.
