    Notes: Graft copolymerization of methyl methacrylate and acrylamide onto ovalbumin was carried out with redox system with ceric ion. Initiation of grafting occurred very rapidly, and the number of grafting sites reached a maximum after about 2 min. regardless of initial ceric ion concentration. The grafted polymer chains were separated by selective hydrolysis of the protein backbone with hydrochloric acid or protease in order to characterize the graft copolymers. The grafted polyacrylamide separated by the proteolytic digestion contained a carbohydrate residue at the end of the polymer molecule. The problem of grafting sites on ovalbumin is discussed.

    Notes: The attachment and growth of L-929 cells on films made of Bombyx mori silk proteins - fibroin and sericin and their mixtures - was studied by a cell culture method. Both cell attachment and growth were dependent on a minimum of around 90% sericin in the mixture. The results from electron micrography as well as from the DSC measurements supported the notion that the mixture of the two proteins fibroin and sericin has a phase-separated structure in the solid state. The observed minimum of sericin in the cell attachment and growth is thought to be a result of this phase-separated structure. Films of pure component proteins (i.e., 100% fibroin or sericin) exhibited as high a cell attachment and growth as collagen, a widely used mammalian cell culture substrate. However, a morphological study of the attached cells revealed that the cells attached to silk fibroin were extended and had a spindle shape, just like the cells attached to collagen, while the cells attached to the silk sericin had a different shape. It is concluded, therefore, that the attachment condition on silk fibroin is ideal for the viability, growth and function of the cells. © 1995 John Wiley & Sons, Inc.

    Notes: A new method was developed for evaluating kinetically the antithrombogenicity of materials. The amount of thrombus formed was measured gravimetrically at an appropriate interval of time after calcium chloride was added to ACD blood in contact with a test material. Graphic curves for thrombus formation on glass, Silastic, Hydron, and treated natural tissues were obtained and compared. There was good correlation between our in vivo tests and in vivo observations.

    Notes: Poly(hydroxyethyl methacrylate) (PHEMA) was implanted subcutaneously in rats, hamsters, and guinea pigs for up to 24, 23, and 13 months, respectively. The tissue response and tumorigenesis caused by PHEMA were compared among the three species of the animals. Thick fibrous capsule formation and tumor production were noted only in rats, while no tumorigenesis and thin fibrous capsule formation were observed in hamsters and guinea pigs.These results support our previous hypothesis that animals producing a thick and avascular capsule tend to develop tumors at the implant site. PHEMA retrieved was characterized by scanning electron microscopy and x-ray energy dispersive analysis. Calcium and phosphorus were the main elements detected on the surface of PHEMA implants. PHEMA is suggested to be not suitable for long-term implant material.

    Notes: Random copolypeptides and block copolypeptides were synthesized, and an interaction between these polypeptide membranes and the cells was studied by a cell culture method (cell line, Ca. 9.22). In random copolypeptides composed of γ-methyl L-glutamate and γ-benzyl L-glutamate, cell attachment and cell growth depended on the monomer composition, and showed a maximum at around 70 mole % of benzyl glutamate. Block copolypeptide composed of L-methionine and oxyethylene exhibited low cell attachment and cell growth even at 10 mole % of oxyethylene content, compared to L-methionine homopolymer. ESCA study of the membrane suggested this result to be due to concentration of the poly(oxyethylene) block chain of the polymer on the surface of the membrane. Block copolypeptide composed of N5-(3-hydroxypropyl) L-glutamine and L-leucine exhibited low cell attachment and cell growth, while the corresponding random copolypeptide exhibited high cell attachment and cell growth. This difference is attributable to the microheterophase structure with the hydrophilic domains embedded in the hydrophobic matrix in the block copolypeptide membrane.

Die erhaltenen Pfropfcopolymeren wurden durch oxydativen Abbau des Polyvinylalkoholanteils charakterisiert. Es liegen hauptsächlich Blockcopolymere vor.
    Notes: Graft copolymerization of methyl methacrylate onto poly(vinyl alcohol) was carried out by the ceric ion method. The graft copolymers were separated from the reaction products by acetylation of the poly(vinyl alcohol) component. The graft copolymers obtained were characterized by oxidative degradation of the poly(vinyl alcohol) backbone, and were found to be predominantly block-type copolymers.

    Description / Table of Contents: Eine kinetische Untersuchung der Reaktion zwischen Epoxydgruppen und Aminen wurde mit Acrylnitril-Glycidylacrylatcopolymeren und Modellverbindungen durchgeführt. Bei 70°C wurden Di-n-butylamin und Diäthanolamin mit Dimethylformamidlösungen der Epoxydverbindungen umgesetzt. Bei der Reaktion der Modellverbindungen und der Copolymeren, die kleine Mengen an Glycidylacrylat enthielten, wurde eine Kinetik 2. Ordnung beobachtet. Bei den Copolymeren war in den meisten Fällen die Reaktion jedoch sehr komplex, und es war unmöglich, sie quantitativ zu behandeln. Ein höherer Nitrilgehalt der Modellverbindung oder der Copolymeren verursacht eine Steigerung der Reaktionsgeschwindigkeit. Ein Nachbargruppeneffekt von Nitril- und Hydroxylgruppen auf die Reaktivität der Epoxydgruppen der Copolymeren wurde beobachtet.
    Notes: A kinetic approach to the reaction between epoxy groups and amines has been made with acrylonitrile-glycidyl acrylate copolymers and model compounds. The reaction has been carried out at 70°C. in a dimethylformamide solution by using di-n-butylamide and diethanolamine. Second-order kinetics has been observed in the reaction of model compounds and the copolymer containing a small amount of glycidyl acrylate. Whereas, in most cases of copolymers, the reaction has been very complex and it has been impossible to treat it quantitatively. An increase in nitrile content of the model compounds and copolymers caused an increase in the rate of reaction. It has been observed a neighboringgroup effect of nitrile and hydroxyl group on the reactivity of the epoxy group of the copolymers.

    Notes: Styrene was polymerized in ceric salt-alcohol redox systems and hydroxyl endgroups on the polystyrene obtained were characterized quantitatively by the spectrophotometric determination of the polymer treated with p-phenylazophenyl isocyanate. A series of polymerization of acrylamide initiated in redox systems of ceric salt and various alcohols was studied as a model system for graft copolymerization onto cellulose. From studies of the rate of polymerization it was shown that the grafting was likely to occur at hemiacetal and 1,2-glycol units. The cellulose-styrene graft copolymer was isolated from the reaction products by a selective extraction technique and characterized by acetolysis of the cellulose backbone. The graft copolymers obtained were found to be a mixture of block and graft copolymers.

    Notes: The synthesis of five different polymers containing sugar residues on side chain is described. 1-O-Methacryloyltetra-O-acetyl-D-glucose, 1-O-methacryloyltetra-O-acetyl-D-galactose, and 6-O-methacryloyltetra-O-acetyl-D-glucose were prepared and polymerized. The polymethacrylates obtained were converted into water-soluble polymers by removing the acetyl protective group with sodium methoxide. 6-O-Methacryloyldiisopropylidene-D-galactose was also prepared and polymerized. The isopropylidene protective group was removed by acid hydrolysis. Poly (N-methacryloylglucosamine) was prepared directly by the polymerization of N-methacryloyl-D-glucosamine without the use of any protective group.
