    Description: Safety and reliability of transgene integration in human genome continue to pose challenges for stem cell-based gene therapy. Here, we report a baculovirus-transcription activator-like effector nuclease system for AAVS1 locus-directed homologous recombination in human induced pluripotent stem cells (iPSCs). This viral system, when optimized in human U87 cells, provided a targeted integration efficiency of 95.21% in incorporating a Neo-eGFP cassette and was able to mediate integration of DNA insert up to 13.5 kb. In iPSCs, targeted integration with persistent transgene expression was achieved without compromising genomic stability. The modified iPSCs continued to express stem cell pluripotency markers and maintained the ability to differentiate into three germ lineages in derived embryoid bodies. Using a baculovirus-Cre/LoxP system in the iPSCs, the Neo-eGFP cassette at the AAVS1 locus could be replaced by a Hygro-mCherry cassette, demonstrating the feasibility of cassette exchange. Moreover, as assessed by measuring -H2AX expression levels, genome toxicity associated with chromosomal double-strand breaks was not detectable after transduction with moderate doses of baculoviral vectors expressing transcription activator-like effector nucleases. Given high targeted integration efficiency, flexibility in transgene exchange and low genome toxicity, our baculoviral transduction-based approach offers great potential and attractive option for precise genetic manipulation in human pluripotent stem cells.

    Description: Chronic myelogenous leukaemia (CML) can progress from a slow growing chronic phase to an aggressive blast crisis phase, but the molecular basis of this transition remains poorly understood. Here we have used mouse models of CML to show that disease progression is regulated by the Musashi-Numb signalling axis. Specifically, we find that the chronic phase is marked by high levels of Numb expression whereas the blast crisis phase has low levels of Numb expression, and that ectopic expression of Numb promotes differentiation and impairs advanced-phase disease in vivo. As a possible explanation for the decreased levels of Numb in the blast crisis phase, we show that NUP98-HOXA9, an oncogene associated with blast crisis CML, can trigger expression of the RNA-binding protein Musashi2 (Msi2), which in turn represses Numb. Notably, loss of Msi2 restores Numb expression and significantly impairs the development and propagation of blast crisis CML in vitro and in vivo. Finally we show that Msi2 expression is not only highly upregulated during human CML progression but is also an early indicator of poorer prognosis.     Description: Auxin plays important roles in many aspects of plant growth and development. It is known that auxin binds to auxin-binding protein ( Abp57 ) and activates the plasma membrane H + -ATPases. In order to elucidate the biological function of rice Abp57 in plants, overexpression transgenic arabidopsis lines were generated. We found overexpression transgenic arabidopsis harbouring rice Abp57 showed tolerance to osmotic stress. Phenotypic transgenic arabidopsis showed longer roots than the wildtype when grown on ½ MS media supplemented with various concentrations of NaCl or PEG-6000. Besides, transgenic arabidopsis has relatively lower water loss rate, higher chlorophyll content and higher biomass when growing under osmotic stress compared to the wildtype. These results suggested that Abp57 has important roles in giving tolerance to arabidopsis against drought and salt stresses.

    Notes: Abstract. The miscibility of polysulfone (PSf) and poly(ether sulfone) (PES) with three tertiary amide polymers has been studied. PES is miscible with poly(N-methyl-N-vinylacetamide) (PMVAc) and with poly(N,N-dimethylacrylamide) (PDMA) but not with poly(2-methyl-2-oxazoline) (PMOx). Miscible PES/PDMA blends show lower critical solution temperature behavior. However, PSf is immiscible with all the three tertiary amide polymers. Previous studies have shown that both PES and PSf are miscible with poly(N-vinyl-2-pyrrolidone). PES is also miscible with poly(2-ethyl-2-oxazoline) but PSf is not. Therefore, PES is more readily miscible with tertiary amide polymers as compared to PSf.

    Description: In this study, the 3D printing technique was utilized to manufacture the sandwich composites. Composite filament fabrication based 3D printer was used to print the face-sheet, and inkjet 3D printer was used to print the sandwich core structure. This work aims to study the compressive failure of the sandwich structure manufactured by using these two manufacturing techniques. Two different types of core structures were investigated with the same type of face-sheet configuration. The core structures were printed using photopolymer, while the face-sheet was made using nylon/glass. The out-of-plane compressive strength of the 3D printed sandwich composite structure has been examined in accordance with ASTM standards C365/C365-M and presented in this paper.
