    Description: After the outbreak of various viral diseases, specifically white spot syndrome (WSS) in farmed Litopenaeus vannamei in the past few years, this non-native shrimp species in Iran requires a generator-builder with specific a pathogen-free approach. This Research was done by the Iran Shrimp Research Center (ISRC) to access accurate information regarding specific pathogen-free shrimp production and avoid using shrimp broodstock production through foreign investors. This research was part of the molecular viral study. For this trial, selected shrimps in farms were sampled and screened for main viruses (OIE list). Shrimps without infection were brought to the quarantine system for one month and at the end of the quarantine they were screened again. In addition, in winter spending, before and after spawning, it’s offspring in F1 and F2 were also screened for viral pathogens by IQ2000 kit at the Iran Shrimp Research Center lab and Iranian veterinary organization lab. All shrimp products such as fresh and consumed food were tested for OIE list at all stages during project performance and the result of tests were negative. Sequenced and molecular tests with specific primers were used to determine the presence of infected samples carrying the virus in any of the products, such as shrimp and consumed food, the infected samples. The result of all the tests during last three years, shows that all the shrimp and their food products were pathogen-free and safe to use.

    Description: At present, the aquaculture industry to provide proper instructions in the field of health management, including production of Specific Pathogen Free shrimp (SPF), require sensitive and reliable methods for the detection and identification of pathogenic microorganisms. Molecular methods which used in the detection of microorganisms have a high discriminatory power in the taxonomy and in relation to libraries in the world. On the other hand, the accurate identification of microorganisms, providing the genetic data bank of shrimp pathogens and maintenance of these strains is the step to promote further research on the mechanisms of pathogenesis of pathogens, diagnosis, treatment, prevention of disease, identify indigenous production kits, diagnosis re emerging and emerging diseases and their origin. Therefore, in this project, by using ribotyping technique, native isolated pathogenic bacteria and fungi were identified and recorded in the gene bank database center. During sampling of shrimp and water of Specific Pathogen Free shrimp center, 40 bacterial strains were isolated, which 8 of them had the most frequency and identification based on 16S rDNA sequencing was performed. Bacteria identified are: Vibrio nigripulchritudo strain IS013(GenBank:KP843725), Vibrio brasiliensis strain IS014 (GenBank:KR186076), Vibrio rotiferianus strain IS015 (GenBank:KR186077), Vibrio azureus strain IS012 (GenBank:KJ018724.1), Vibrio owensii strain IS016 (GenBank:KR186078), Agarivorans gilvus strain IS017 (GenBank:KR186079), Vibrio brasiliensis IS018 (GenBank:KR186080) and Vibrio alginolyticus strain IS019 (GenBank:1817854), which were recorded in The World Bank genes. In this study fungal isolates were not detected.

    Description: This study is a part of the national and technology master plan entitled "Attain to technical knowledge of specific pathogen free shrimp production and cut off to dependence on foreign products". The goals of this work were to assess and analysis the risk factors of production of specific pathogen free Litopenaeus vannamei shrimp in its all stages (broodstocking, laravl production, shrimp farming, feeding and water quality), determining the pattern of outcomes monitoring, management and control of outcomes from April 2012 to September 2015 in Bandargah research station and Persian Gulf SPF Shrimp research station and their environment, belonging to Shrimp Research Center, located in Bushehr. Identification and screening of risk factors have been done based on Environment Aspect and Effect Analysis method, (EA)2, using a fine statement questionnaire and quantification of risks. The number of identified risk factors in Bandargah research station was 15 while it was 13 in Persian Gulf SPF Shrimp research station. The more number of risk factors in Bandargah station in respect to its number in Persian Gulf station is might due to lower level of biosecurity, physical structure and aging of Bandargah station and existence of two possible source of contamination, the Bushehr nuclear power plant pollutants and Bandargah fishing harbor, near to the station. On the basis of results, the maximum values of APN in Bandargah station was 42 while it was 30 in Persian Gulf station. These results revealed that the level of risk in Bandargah station is lower than that in Persian Gulf station. The most probable risks in both studied research stations are related to transmission of pathogens in shrimp feeding and water intake pollution.

    Description: After severe mortality due to white spot disease incidence in cultured shrimps of Helleh grows out farms, disease spread to Delvar,Mond, Boeyrat and Bandar Rig sites in Bushehr province during 2005. Many crustaceans keep this virus in the body, act as a carrier, and can spread of the infection. In order to investigate status of contamination of wild shrimp and crab's population to white spot virus in Bushehr province waters (north of Persian Gulf), samples were collected and tested by Nested PCR for detection of WSV from estuaries and input and output channels water around the Helleh, Shif and Mond sites during 2007 and 2008. Total tested shrimps were 401 peaces, includes green tiger shrimp (Penaeus semisulcatus) 220 pieces, white shrimp (Metapenaeus affinis) 181 pieces, also total tested crabs were 292 pieces includes Ocypode rotundata 68 pieces and Portunus pelagicus 224 pieces. All the samples showed negative results of WSV contamination. However, it can never be sure that the population is free from WSV unless we collect a random sample of 300 pieces from each species and test them individually for WSV. Therefore, we will be confident up to 95% that the wild sources in that area have no prevalence of 1% or upper WSV contamination.

    Description: The first time the white spot disease (WSD) and IHHNV were reported in Iran was in 2004 and 2008, respectively in Bushehr Province. In Sistan and Balochestan province only the WSD was reported in 2008.The aim of this study was detecting these two viral diseases in these provinces, during December 2009 to April 2010. A total of 364 samples were collected according to suspected gross signs from hatcheries and shrimp farms in Bushehr and Sistan and Blochestan provinces (I.R. Iran) respectively, including larvae (72, 43 samples), post larvae (48, 37 samples), juveniles (57, 32 samples), sub adults (39, 22 samples) and broodstock (29, 13 samples) of Litopenaus vannamei . WSD was detected from juveniles (23 samples), sub adults (14 samples), and broodstock (14 samples), and IHHNV was also detected from juveniles (16 samples), sub adults (9 samples) and broodstock (5 samples) based on gross signs, PCR and histopathological changes from Bushehr province but from Sistan and Blochestan province only WSD was detected from juveniles (26 samples), sub adults (18 samples) and broodstock (7 samples). Histopathological observations of WSSV showed basophilic Cowdry type A inclusion bodies in all tissues such as gills, haematopoietic tissue, cuticle epithelium, lymphoid organ and connective tissue. However histologically, the hepatopancreas tissue showed vacuolization of B cells , without inclusion bodies, but histopatholgical changes caused IHHNV including eosinophilic Cowdry type A inclusion bodies which were ectodermal, mesodermal and rarely endodermal.

    Description: During the last decade, shrimp aquaculture has developed rapidly in many developing countries. Antibiotic resistance is a major problem in shrimp aquaculture. Seaweeds are considered as potent source of bioactive compounds that are able to produce biological activities. The aim of this study was to evaluate the antibacterial activity of ethanolic and chloroformic crude extracts of the brown algae, Sargassum latifolium derived from Persian Gulf waters. The extractions against shrimp selective pathogen bacteria including Vibrio alginolyticus, V. parahaemolyticus and V. harveyi were used by disk diffusion agar method. Results revealed that all mentioned bacteria were sensitive to both ethanolic and chloroformic crude extracts of S.latifolium. The minimum inhibitory concentration of chloroformic extract of S. latifolium against the bacteria was performed by tube dilution technique. MICs were 5, 10 and 10 mg ml-1 for V. alginolyticus, V. parahaemolyticus and V. harveyi, respectively while minimum bactericidal concentrations (MBC) were 5, 15 and 20 mg ml-1. This study recommended that S.latifolium isolated from Persian Gulf waters has antibacterial activity potential against shrimp pathogen bacteria.

    Description: White Spot Virus (WSV) is currently the most serious viral pathogen of shrimp worldwide it causes mortality up to 100% within 7-10 days in commercial shrimp farms. Infected Indian white shrimp Fenneropenaeus indicus samples were collected from Guatr shrimp site in Sistan and Baluchestan province in south of Iran and WSV infection was confirmed by Nested PCR. WSV was isolated from infected shrimp samples by centrifugation and filtration and multiplied in crayfish by intramuscular inoculation, the isolated virus was called WSV/IRN/1/2010. In order to determine the dilution resulting in 90-100% mortality in Penaeus semiculcatus, diluted virus stock in steps from 100 till 105 times in sterile PBS was injected intramuscularly to 14 shrimps in each group. Also the virus stock was diluted in steps from 1/2 till 1/32 times in sterile PBS and injected intramuscularly in Astacus leptodactylus crayfish. Therefore the LD50 of live virus stock in Astacus leptodactylus and Penaeus semiculcatus crayfish were calculated by the Karber method 10 3.29 /ml and 10 5.35 /ml, respectively.

    Description: This study was conducted to evaluate the possibility of replacing Z. multiflora instead of antibiotics and chemicals in shrimp aquaculture industry to protect the environment and prevention of microbial resistance. In vitro MIC of Z. multiflora essential oil against Candida albicans and Fusarium solani obtained 0.004% and 0.008% respectively, also MFC of Z. multiflora essential oil against C.albicans and F.solani was 0.004% and 0.04%, respectively. The results showed that 0.0002% dosage of Z. multiflora essential oil against total count of fungi in sea water (salinity 39±1ppt) can decrease growth of fungi after 6 h and 0.0004%, 0.0008% and 0.0016% dosages can decrease the total count of fungi in sea water. But very low dosage of Z. multiflora essential oil in shrimp Litopenaeus vannamei leads to mortality. Lethal dose of Z. multiflora essential oil in shrimp postlarvae, juvenile (2±0.2g) and adults (10-12 g) was 0.00050%, 0.00045% and 0.00035%, respectively. The results indicated that Z. multiflora have a significant anti-fungal effect but is toxic for L. vannamei. We recommend that Z. multiflora essential oil can be used in non-living environment, such as disinfecting tanks and equipments.

    Description: Nowadays, white spot virus disease is serious threat for breeding and culture industry of shrimp. In this study was increasing resistance of shrimp against white spot virus by using modern methods such as shrimp vaccination with inactivated viruses and recombinant proteins. The aim of this study were determine the growth and survival rates vaccinated and non-vaccinated of Litopenaeus vannamei (5 to 15 and 12 to 26 day), that were challenged with white spot virus in 40 and 60 day rearing. This study consisted two separate groups were vaccinated and non-vaccinated with 11 treatments experimental and each of replicate was stocked 1000 pieces by post larva of 5 to15 and 12 to 26 day. After vaccination, two groups of post larvae exposed to the white spot virus at 40 and 60 day, one groups no exposure to the virus. Samplings were randomly of shrimp in 40, 60 and 80 days 10 pieces each of treatment experimental and measured mean of weight and length. Also, number of deaths was recorded at morning and evening daily and calculated survival rate at the end of study. The results showed growth rate of post larvae vaccinated (5 to15 day) which exposed to white spot virus at 40 and 60 was significantly lower than non-vaccinated of post larval, while the growth rate of post larval exposed to virus in 60 day was significantly increased. On the other hand, growth rate of post larval vaccination (12 to 26 day) exposed to virus in 60 day compared with post larval vaccination (5 to 15 and 12 to 26) exposed to virus in 40 and 60 days was significantly increased. Hence, growth rate was significantly increased in post larval vaccinated (5 to 15 and 12 to 26) which non-exposed to virus. Although the survival rate was post larval vaccinated (12 to 26 days) exposed to virus Post larvae in 60 day higher than post larval were exposed to virus in 40 and 60, but no significant differences were observed. However, relative mortality of post larval vaccination in 12 to 26 day compared with post larval vaccination in 5 to 15 days exposed to virus were significantly lower. Considering growth and survival index was improved of post larval vaccination can be concluded that the optimum age for vaccination against white spot virus of L.vannamei was 12 to 26 day.
