    Description: Iron overload is a major life-threatening complication of thalassemia major and other iron-loading anemias treated by regular blood transfusions. Although the clinical manifestations of iron overload may be prevented by desferrioxamine, the only iron-chelating drug in routine clinical use, this treatment requires subcutaneous infusion of desferrioxamine for 12 hours each day. New orally effective iron chelators are urgently needed, and pyridoxal isonicotinoyl hydrazone (PIH), which was first recognized as an effective iron chelator in vitro and subsequently in vivo, shows promise for the treatment of iron overload. More recently, over 40 analogs of PIH were synthesized, and some of them proved to be very potent in mobilizing 59Fe in vitro from 59Fe-labeled cells. In this study, we show that PIH analogs such as pyridoxal benzoyl hydrazone, pyridoxal p-methoxybenzoyl hydrazone (PMBH), pyridoxal m-fluorobenzoyl hydrazone (PFBH), and pyridoxal-2-thiophenecarboxyl hydrazone, compounds previously shown to mobilize iron from cells in vitro, are also effective in vivo. All of these chelators significantly enhanced biliary excretion of iron (measured by atomic absorption spectrophotometry) following their intraperitoneal (IP) and/or oral administration to rats. The most effective was PFBH, which increased iron concentration in the bile about 150-fold, as compared with basal biliary iron concentration, within 1 hour following a single IP dose of 0.2 mmol/kg body weight. In contrast, desferrioxamine increased the biliary iron concentration only 20-fold to 30-fold under the same conditions. Moreover, while control rats excreted ≈ 0.8 μg Fe in 2 hours, treatment with PFBH, PMBH, and desferrioxamine resulted in cumulative excretions of 87, 59, and 22 μg Fe, respectively, in the same period of time. Interestingly, PMBH was also quite effective following gastric administration, resulting in a 6-hour cumulative value of 34 μg Fe. These compounds are nontoxic and are inexpensive and easy to make. Their further evaluation as candidate drugs for the treatment of iron overload is warranted.

    Notes: Abstract Housedusts and garden soils were sampled in 14 houses in the vicinity of a secondary Pb smelter and analysed for concentrations of Pb, Zn, Cu, Cd, As, and Hg. Sixty-one topsoil samples were also taken from a 2 km2 grid covering the smelter grounds and surrounding residential areas and analysed for concentrations of Pb, Zn, Cd and Cu. Contour maps generated from the grid data indicate significant contamination in the area (maximum Pb concentration 58 500 μg g-1), particularly down-wind of the smelter grounds. A geometric mean Pb concentration of 2225 μg g-1 was recorded in garden soil and similarly elevated levels were recorded for Zn, Cd, As and Sb. In housedusts, a geometric mean Pb concentration of 1668 μg g-1 was observed. Whilst housedust metal concentrations were generally elevated, compared to other urban or residential areas, there appears to be a large degree of attenuation of the metals between the exterior and interior environments of the homes studied. A significant correlation was not recorded between metal concentrations of garden soils and housedusts. There were significant correlations for: distance from the smelter against garden soil metal concentrations; garden soil metal concentrations against each other; housedust metal concentrations against each other; and house age against garden soil metal concentrations.

    Notes: Abstract Soils and housedusts were collected from three areas of Pribram,an historic metal mining and smelting town in the Czech Republic. The main objectives of the study were: (i) to assessthe influence of physico-chemical form, particle size, soilproperties and contaminant source on Pb bioavailability andexposure risk; (ii) compare the Pb bioavailability data obtainedfrom the mining and smelting areas and assess whether anydifferences observed could be attributed to the factors thoughtto exert an influence. Lead concentrations were highest in thesmelter area. Mining area garden soils also contained elevatedPb concentrations. Solubility of housedust Pb in 0.12 M HCl (asurrogate for stomach acid) was similar in all study areas andwas similar to values reported in the literature. However, 0.12M HCl solubility of garden soil Pb was low in the mining areacompared to the other study areas and compared to other urbanareas. Blood Pb concentrations were also relatively low in themining area compared to the other study areas and the reducedsoil Pb solubility observed in this area was suspected as aninfluencing factor. However, exposure pathways may also beimportant in explaining the differences observed.

Die Adsorption von Schwermetallen aus Lösungen niedriger Konzentrationen wurde für beide polymeren Thiole untersucht. Die Affinität steigt bei Experimenten in vitro in der Reihe Pb 〈 Ag 〈 Hg an. Bei Experimenten in vivo wurde die Resorption des Quecksilbers im Darmtrakt von Ratten durch das Methacrylatpolymere verhindert, wenn Quecksilberchlorid oder Methylquecksilberchlorid eingesetzt wurde, nicht aber im Falle von Phenylquecksilberchlorid.
    Notes: Poly(vinyltoluene-α-thiol) and poly(1-methacryloyloxyethane-2-thiol) were prepared by the polymeranalogous hydrolysis of the respective polymeric isothiuronium salts. The sorption of heavy metals from low-concentrated solutions was studied for both thiol polymers. Affinity increases in the series Pb 〈 Ag 〈 Hg in experiments in vitro. In vivo the mercury resorption in the gastrointestinal tract of rats was partly blocked by the methacrylate derivative if mercuric chloride or methylmercurichloride was administered (in the case of phenylmercurichloride the polymeric thiol was ineffective).
