    Notes: Activity of some of the main enzymes involved in protein digestion and absorption (acid and alkaline proteases, leucine-aminopeptidase, acid and alkaline phosphatases) as well as those of amylase and lipase, was assessed during larval development of white sea bream Diplodus sargus. All enzyme activity was detected at the moment of mouth opening. The variations observed in the activity profiles of the digestive enzymes were correlated either to developmental events, such as the functional start of the stomach (22 days after hatching), or to changes in the nature of the diet. The early and noticeable development of digestive enzyme activities was linked to a high survival after weaning.

    Notes: Abstract Ten types of plant baits were tested in the laboratory to assess their capacity to detect pathogenic Pythium species. These were orange tree leaves, tomato leaves, pepper leaves, geranium leaves, Bermuda grass leaves, pine needles, immature carnation petals, hemp-seed cotyledons, pepper and cucumber fruits. The Pythium spp. tested were P. aphanidermatum, P. irregulare and Pythium ‘group F’ from hydroponic market garden crops in the Poniente region of Almería (south-east Spain). The test consisted of observing the velocity at which five baits were colonized and the day of colonization of the first bait. Results indicated that the slowest baits to be infected were immature carnation petals and pine needles. These two, together with Bermuda grass leaves, were also the baits infected in lowest number, such that practically no further infection was produced in the baits after the fifth day of contact with the inoculated water. The other plant baits tested were equally suitable for detection of Pythium spp. over the first two days, although only orange leaves and hemp-seed cotyledons were infected on the first day.

    Notes: Abstract Adult Cosmopepla bimaculata discharge a volatile secretion from paired ventral metathoracic glands (MTG) when disturbed. Collected volatiles were similar in both sexes and consisted of n-tridecane (67%), (E)-2-decenal (12%), (E)-2-decenyl acetate (12%), (E)-2-hexenal (3%), hexyl acetate (2%), n-dodecane (2%), a tridecene isomer (1%), and n-undecane, n-tetradecane, and n-pentadecane (all 〈1%). In addition, undisturbed males produced a novel insect compound, (E)-8-heneicosene, whose function is unknown. The MTG secretion emerges as an enlarging droplet, which is held in place by a cuticular projection and a pleural scent area consisting of specialized rough cuticle surrounding the gland opening. Insects can selectively discharge from either the right or left gland or both glands simultaneously, can control the amount of fluid ejected, and can resorb the ejected secretion droplet back into the gland reservoir. In feeding trials, killdeer (Charadrius vociferous), starlings (Sturnus vulgaris), robins (Turdus migratorius), and anole lizards (Anolis carolinensis) rejected or demonstrated aversion to feeding on the bugs. Furthermore, bugs that lacked the secretion were more susceptible to predation than bugs with secretion, suggesting that the secretion functions in defense against predators.

    Notes: Summary The plasma membranes of polarized epithelial cells are divided into two distinct domains separated by tight junctions and characterized by distinct polypeptide compositions. Study of the cellular mechanisms involved in generating this anisotropy has recently been facilitated through the introduction of a surface labeling technique employing NHS-biotin (29). Incorporation of the biotin marker allows proteins present at one or the other cell surface domain to be identified and isolated. This methodology is applicable to investigations of both the steady state and the dynamic aspects of epithelial polarity. We discuss protocols through which this technique can be applied to various experimental situations. Methodologic details and special considerations are discussed. The utility of surface biotinylation for studies of epithelial polarity depends on this technique's capacity to report quantitatively on the surface distributions of plasmalemmal proteins. We find that under certain circumstances, surface biotinylation is extremely inefficient. Furthermore, in some cases the efficiency of surface biotinylation is dramatically different at the two plasmalemmal domains of epithelial cells grown on permeable filter supports. Thus, surface biotinylation does not always provide an accurate picture of plasma membrane protein distributions. These situations are discussed and methods to assess the efficiency and accuracy of surface biotinylation are proposed.
