    Description: A new finite fault inversion strategy is developed to explore the uncertainty range for the energy based average coseismic stress drop ( $\overline {{\rm{\Delta }}{\tau _E}} $ ) of large earthquakes. For a given earthquake, we conduct a modified finite fault inversion to find a solution that not only matches seismic and geodetic data but also has a $\overline {{\rm{\Delta }}{\tau _E}} $ matching a specified value. We do the inversions for a wide range of stress drops. These results produce a trade-off curve between the misfit to the observations and $\overline {{\rm{\Delta }}{\tau _E}} $ , which allows one to define the range of $\overline {{\rm{\Delta }}{\tau _E}} $ that will produce an acceptable misfit. The study of the 2014 Rat Islands M w 7.9 earthquake reveals an unexpected result: when using only teleseismic waveforms as data, the lower bound of $\overline {{\rm{\Delta }}{\tau _E}} $ (5–10 MPa) for this earthquake is successfully constrained. However, the same data set exhibits no sensitivity to its upper bound of $\overline {{\rm{\Delta }}{\tau _E}} $ because there is limited resolution to the fine scale roughness of fault slip. Given that the spatial resolution of all seismic or geodetic data is limited, we can speculate that the upper bound of $\overline {{\rm{\Delta }}{\tau _E}} $ cannot be constrained with them. This has consequences for the earthquake energy budget. Failing to constrain the upper bound of $\overline {{\rm{\Delta }}{\tau _E}} $ leads to the conclusions that (1) the seismic radiation efficiency determined from the inverted model might be significantly overestimated and (2) the upper bound of the average fracture energy E G cannot be constrained by seismic or geodetic data. Thus, caution must be taken when investigating the characteristics of large earthquakes using the energy budget approach. Finally, searching for the lower bound of $\overline {{\rm{\Delta }}{\tau _E}} $ can be used as an energy-based smoothing scheme during finite fault inversions.

    Description: We use interferometric synthetic aperture radar, Global Positioning System, and teleseismic data to constrain the relative location of coseismic slip from 11 earthquakes on the subduction interface in northern Chile (23-25 S) between the years 1993 and 2000. We invert body wave waveforms and geodetic data both jointly and separately for the four largest earthquakes during this time period (1993 Mw 6.8; 1995 Mw 8.1; 1996 Mw 6.7; 1998 Mw 7.1). While the location of slip in the teleseismic-only, geodetic-only, and joint slip inversions is similar for the small earthquakes, there are differences for the 1995 Mw 8.1 event, probably related to nonuniqueness of models that fit the teleseismic data. There is a consistent mislocation of the Harvard centroid moment tensor locations of many of the 6 〈 Mw 〈 8 earthquakes by 30-50 km toward the trench. For all models, the teleseismic data are better able to resolve fine details of the earthquake slip distribution. The 1995 earthquake did not rupture to the maximum depth of the seismogenic zone (as defined by the other earthquakes). In addition to the above events, we use only teleseismic data to determine the rupture characteristics of four other Mw 〉 6 earthquakes, as well as three Mw 〉 7 events from the 1980s. All of these earthquakes appear to rupture different portions of the fault interface and do not rerupture a limited number of asperities.

    Description: 〈p〉The Cdc48 adenosine triphosphatase (ATPase) (p97 or valosin-containing protein in mammals) and its cofactor Ufd1/Npl4 extract polyubiquitinated proteins from membranes or macromolecular complexes for subsequent degradation by the proteasome. How Cdc48 processes its diverse and often well-folded substrates is unclear. Here, we report cryo–electron microscopy structures of the Cdc48 ATPase in complex with Ufd1/Npl4 and polyubiquitinated substrate. The structures show that the Cdc48 complex initiates substrate processing by unfolding a ubiquitin molecule. The unfolded ubiquitin molecule binds to Npl4 and projects its N-terminal segment through both hexameric ATPase rings. Pore loops of the second ring form a staircase that acts as a conveyer belt to move the polypeptide through the central pore. Inducing the unfolding of ubiquitin allows the Cdc48 ATPase complex to process a broad range of substrates.〈/p〉

    Description: 〈p〉Meiotic resumption-coupled degradation of maternal transcripts occurs during oocyte maturation in the absence of mRNA transcription. The CCR4–NOT complex has been identified as the main eukaryotic mRNA deadenylase. 〈i〉In vivo〈/i〉 functional and mechanistic information regarding its multiple subunits remains insufficient. 〈i〉Cnot6l〈/i〉, one of four genes encoding CCR4–NOT catalytic subunits, is preferentially expressed in mouse oocytes. Genetic deletion of 〈i〉Cnot6l〈/i〉 impaired deadenylation and degradation of a subset of maternal mRNAs during oocyte maturation. Overtranslation of these undegraded mRNAs caused microtubule–chromosome organization defects, which led to activation of spindle assembly checkpoint and meiotic cell cycle arrest at prometaphase. Consequently, 〈i〉Cnot6l〈/i〉〈sup〉–/–〈/sup〉 female mice were severely subfertile. The function of CNOT6L in maturing oocytes is mediated by RNA-binding protein ZFP36L2, not maternal-to-zygotic transition licensing factor BTG4, which interacts with catalytic subunits CNOT7 and CNOT8 of CCR4–NOT. Thus, recruitment of different adaptors by different catalytic subunits ensures stage-specific degradation of maternal mRNAs by CCR4–NOT. This study provides the first direct genetic evidence that CCR4–NOT-dependent and particularly CNOT6L-dependent decay of selective maternal mRNAs is a prerequisite for meiotic maturation of oocytes.〈/p〉

    Description: Motivation: In recent years, genome-scale metabolic models (GEMs) have played important roles in areas like systems biology and bioinformatics. However, because of the complexity of gene–reaction associations, GEMs often have limitations in gene level analysis and related applications. Hence, the existing methods were mainly focused on applications and analysis of reactions and metabolites. Results: Here, we propose a framework named logic transformation of model (LTM) that is able to simplify the gene–reaction associations and enables integration with other developed methods for gene level applications. We show that the transformed GEMs have increased reaction and metabolite number as well as degree of freedom in flux balance analysis, but the gene–reaction associations and the main features of flux distributions remain constant. In addition, we develop two methods, OptGeneKnock and FastGeneSL by combining LTM with previously developed reaction-based methods. We show that the FastGeneSL outperforms exhaustive search. Finally, we demonstrate the use of the developed methods in two different case studies. We could design fast genetic intervention strategies for targeted overproduction of biochemicals and identify double and triple synthetic lethal gene sets for inhibition of hepatocellular carcinoma tumor growth through the use of OptGeneKnock and FastGeneSL, respectively.     Description: This paper aims at exploring a fuzzy-based adaptive dynamic surface control (DSC) for a class of non-linear systems in strict-feedback form. Incorporating DSC technique into a fuzzy logic system based minimal-learning parameter algorithm and by introducing an initialization technique based on a surface error modification, it is shown that the design procedure and the computational burden can be greatly reduced and the L performance of system tracking error can be achieved. Besides, some restrictions on controlled systems in stability analysis of previous works are also removed. Simulation results are presented to demonstrate the efficiency of the proposed scheme.
