    Description: Previously identified neural correlates of deception, such as the prefrontal, anterior cingulate, and parietal regions, have proven to be unreliable neural markers of deception, most likely because activity in these regions reflects executive processes that are not specific to deception. Herein, we report the first fMRI study that provides strong preliminary evidence that the neural activity associated with perception but not executive processes could offer a better marker of deception with regard to face familiarity. Using a face-recognition task, activity in the left precuneus during the perception of familiar faces accurately marked 11 of 13 subjects who lied about not knowing faces that were in fact familiar to them. This level of classification accuracy is much higher than the level predicted by chance and agrees with other findings by experts in lie detection. Scientific Reports 3 doi: 10.1038/srep01636

    Description: This study presents an innovative piezoelectric actuator, capable of long travel distance and fast speed. Main components of the actuator include a rod and two piezoelectric buzzers of disk geometry. Being subject to the piezoelectric force and Coulomb friction, the piezoelectric actuator can move rapidly. Equations of motion are derived based on piezoelectric properties and Newton’s law. Static and kinetic friction effects are incorporated. Stress and strain calculations and piezoelectric equations are included in derivation. Circular plate models with polar coordinates are used to account for piezoelectric properties of buzzers. Experimental results are compared with theoretical results. According to experimental results, the present actuator can move at the speed of 37.5 mm/sec and generate the force of 15.4 mN.

    Description: Transcription factors play a central role in cell development, differentiation and growth in biological systems due to their ability to regulate gene expression by binding to specific DNA sequences within the nucleus. The dysregulation of transcription factor signaling has been implicated in the pathogenesis of a number of cancers, developmental disorders, inflammation and autoimmunity. There is thus a high demand for convenient high-throughput methodologies able to detect sequence-specific DNA-binding proteins and monitor their DNA-binding activities. Traditional approaches for protein detection include gel mobility shift assays, DNA footprinting and enzyme-linked immunosorbent assays (ELISAs) which tend to be tedious, time-consuming, and may necessitate the use of radiographic labeling. By contrast, luminescence technologies offer the potential for rapid, sensitive and low-cost detection that are amenable to high-throughput and real-time analysis. The discoveries of molecular beacons and aptamers have spearheaded the development of new luminescent methodologies for the detection of proteins over the last decade. We survey here recent advances in the development of luminescent detection methods for DNA-binding proteins, including those based on molecular beacons, aptamer beacons, label-free techniques and exonuclease protection.

    Description: Cellular senescence has been recently shown to have an important role in opposing tumour initiation and promotion. Senescence induced by oncogenes or by loss of tumour suppressor genes is thought to critically depend on induction of the p19(Arf)-p53 pathway. The Skp2 E3-ubiquitin ligase can act as a proto-oncogene and its aberrant overexpression is frequently observed in human cancers. Here we show that although Skp2 inactivation on its own does not induce cellular senescence, aberrant proto-oncogenic signals as well as inactivation of tumour suppressor genes do trigger a potent, tumour-suppressive senescence response in mice and cells devoid of Skp2. Notably, Skp2 inactivation and oncogenic-stress-driven senescence neither elicit activation of the p19(Arf)-p53 pathway nor DNA damage, but instead depend on Atf4, p27 and p21. We further demonstrate that genetic Skp2 inactivation evokes cellular senescence even in oncogenic conditions in which the p19(Arf)-p53 response is impaired, whereas a Skp2-SCF complex inhibitor can trigger cellular senescence in p53/Pten-deficient cells and tumour regression in preclinical studies.     Description: Akt signaling plays a central role in many biological functions, such as cell proliferation and apoptosis. Because Akt (also known as protein kinase B) resides primarily in the cytosol, it is not known how these signaling molecules are recruited to the plasma membrane and subsequently activated by growth factor stimuli. We found that the protein kinase Akt undergoes lysine-63 chain ubiquitination, which is important for Akt membrane localization and phosphorylation. TRAF6 was found to be a direct E3 ligase for Akt and was essential for Akt ubiquitination, membrane recruitment, and phosphorylation upon growth-factor stimulation. The human cancer-associated Akt mutant displayed an increase in Akt ubiquitination, in turn contributing to the enhancement of Akt membrane localization and phosphorylation.
