Zusammenfassung Die Anwendungsmöglichkeiten der Fluorimetrie zur Bestimmung der Aktivität von Monoaminoxydase (MAO) wurden untersucht. Von verschiedenen Substraten hat sich Kynuramin als das geeignetste erwiesen. Bei seiner oxydativen Desaminierung durch MAO bildet sich 4-Hydroxychinolin, das eine intensive Fluorescenz aufweist (Aktivierung 310 nm, Fluorescenz 390 nm). Die optimalen Bedingungen für diese Reaktion hängen vom pH, der Temperatur, der Art des Puffers und der Substratkonzentration ab. Es wurde eine relativ einfache und schnelle Methode zur Bestimmung der MAO-Aktivität entwickelt, die eine Messung in sehr kleinen Gewebeproben (0,1 mg Frischgewicht) erlaubt. Die Werte der MAO-Aktivität werden in Einheiten angegeben (1 Einheit=1 μM 4-Hydroxychinolin/g Frischgewicht/h). Die Spezifität der Reaktion wurde nachgewiesen: 1. durch die Übereinstimmung der charakteristischen Aktivierungs- und Emissionskurven des nach Einwirkung von MAO auf Kynuramin entstandenen Produktes mit den charakteristischen Kurven von 4-Hydroxychinolin und 2. durch die Möglichkeit der Unterbrechung der Reaktion durch spezifische Hemmstoffe für MAO. Die Anwendbarkeit der Methode wurde am Beispiel der Bestimmung der MAO-Aktivitätsverteilung im Nervensystem der Teleosten wie auch in Geweben verschiedener anderer Tiere demonstriert.

Abstract A detailed study of the application possibilities of fluorimetry in the determination of monoamine oxidase (MAO) activity is presented. Of the different substrates used, kynuramine was found to be the most suitable. Through the oxidative desamination of kynuramine by MAO 4-hydroxyquinoline is formed. The optimal conditions for this reaction depend upon the pH, temperature, the type of buffer and the concentration of the substrate. This study led to the development of a relative simple, fast and highly sensitive (0.1 mg tissue wet-weight) method for the determination of MAO activity. The specificity of the method is proved by the fact that, first, the characteristic shape of the emission curves of the desamination product is identical with that of 4-hydroxyquinoline and, second, that the desamination reaction can be blocked by specific inhibitors for MAO (Marplan). The method was used for the mapping of MAO activity in the nervous system of teleosts and for the determination of the enzyme activity in different tissues of toad, fish and rat.
