    Notes: Two Saccharomyces cerevisiae strains were employed to investigate the effects of medium enrichment on the expression and secretion of a recombinant protein. One was a stable autoselection strain with mutations in the ura3, fur1, and urid-k genes. The combination of these three mutations blocks both the pyrimidine nucleotide biosynthetic and salvage pathways and is lethal to the cells. Retention of the plasmid, which carries a URA3 gene, was essential for cell viability. Therefore, all media were selective, allowing cultivation of the strain in complex medium. The second strain was a nonautoselection (control) strain and is isogenic to the first except for the fur1 and urid-k mutations. The plasmid utilized contains the yeast invertase gene under the control of the MFα1 promoter and leader sequence. The expression and secretion of invertase for the autoselection strain were examined in batch culture for three media: a minimal medium (SD), a semidefined medium (SDC), and a rich complex medium (YPD). Biomass yields and invertase productivity (volumetric activity) increased with the complexity of the medium; total invertase volumetric activity in YPD was 100% higher than in SDC and 180% higher than in SD. Specific activity, however, was lowest in the SDC medium. Secretion efficiency was extremely high in all three media; for the majority of the culture, 80-90% of the invertase was secreted into the periplasmic space and/or culture medium. A glucose pulse at the end of batch culture in YPD facilitated the transport of residual cytoplasmic invertase. For the nonautoselection strain, invertase productivity did not improve as the medium was enriched from SDC to YPD, and plasmid stability in the complex YPD medium dropped from 54% to 34% during one batch fermentation. During long-term sequential batch culture in YPD, invertase activity decreased by 90% and the plasmid-containing fraction dropped from 56% to 8.8% over 44 generations of growth. The expression level for the autoselection strain, however, remained high and constant over this time period, and no reversion at the fur1 or urid-k locus was observed. © 1993 John Wiley & Sons, Inc.

    Notes: The chromosomal δ sequences of the yeast Saccharomyces cerevisiae were employed as recombination sites to integrate the bacterial neor gene and the yeast SUC2 gene into the yeast genome. A dominate selection method employing the aminoglycoside antibiotic G418 was used. Transformation efficiencies and growth behaviors of the transformants were studied. Transformants were obtained with more than 40 integrations; the majority of insertions were tandem with a maximum of three different insertion sites utilized at one time. After 70-100 generations of growth in nonselective medium, the high copy number SUC2-neor integrants were found to be unstable; only minor instability was observed for the neor and low copy number SUC2-neor integrants. © 1996 John Wiley & Sons, Inc.

    Description: Inertial navigation devices include star sensor, GPS, and gyroscope. Optical fiber and laser gyroscopes provide high accuracy, and their manufacturing costs are also high. Magnetic suspension rotor gyroscope improves the accuracy and reduces the production cost of the device because of the influence of thermodynamic coupling. Therefore, the precision of the gyroscope is reduced and drift rate is increased. In this study, the rotor of liquid floated gyroscope, particularly the dished rotor gyroscope, was placed under a thermal field, which improved the measurement accuracy of the gyroscope. A dynamic theory of the rotor of liquid floated gyroscope was proposed, and the thermal field of the rotor was simulated. The maximum stress was in x, 1.4; y, 8.43; min 97.23; and max 154.34. This stress occurred at the border of the dished rotor at a high-speed rotation. The secondary flow reached 5549 r/min, and the generated heat increased. Meanwhile, the high-speed rotation of the rotor was volatile, and the dished rotor movement was unstable. Thus, nanomaterials must be added to reduce the thermal coupling fluctuations in the dished rotor and improve the accuracy of the measurement error and drift rate.

    Description: Large-scale coastal reclamation has caused significant changes in Spartina alterniflora (S. alterniflora) distribution in coastal regions of China. However, few studies have focused on estimation of the wetland vegetation biomass, especially of S. alterniflora, in coastal regions using LiDAR and hyperspectral data. In this study, the applicability of LiDAR and hypersectral data for estimating S. alterniflora biomass and mapping its distribution in coastal regions of China was explored to attempt problems of wetland vegetation biomass estimation caused by different vegetation types and different canopy height. Results showed that the highest correlation coefficient with S. alterniflora biomass was vegetation canopy height (0.817), followed by Normalized Difference Vegetation Index (NDVI) (0.635), Atmospherically Resistant Vegetation Index (ARVI) (0.631), Visible Atmospherically Resistant Index (VARI) (0.599), and Ratio Vegetation Index (RVI) (0.520). A multivariate linear estimation model of S. alterniflora biomass using a variable backward elimination method was developed with R squared coefficient of 0.902 and the residual predictive deviation (RPD) of 2.62. The model accuracy of S. alterniflora biomass was higher than that of wetland vegetation for mixed vegetation types because it improved the estimation accuracy caused by differences in spectral features and canopy heights of different kinds of wetland vegetation. The result indicated that estimated S. alterniflora biomass was in agreement with the field survey result. Owing to its basis in the fusion of LiDAR data and hyperspectral data, the proposed method provides an advantage for S. alterniflora mapping. The integration of high spatial resolution hyperspectral imagery and LiDAR data derived canopy height had significantly improved the accuracy of mapping S. alterniflora biomass.

A rapid and non-destructive method of early SSB detection is essential for rice-growth protection. In this study, hyperspectral imaging combined with chemometrics was used to detect early SSB infestation in rice and identify the degree of infestation (DI). Visible/near-infrared hyperspectral images (in the spectral range of 380 nm to 1030 nm) were taken of the healthy rice plants and infested rice plants by SSB for 2, 4, 6, 8 and 10 days. A total of 17 characteristic wavelengths were selected from the spectral data extracted from the hyperspectral images by the successive projection algorithm (SPA). Principal component analysis (PCA) was applied to the hyperspectral images, and 16 textural features based on the gray-level co-occurrence matrix (GLCM) were extracted from the first two principal component (PC) images. A back-propagation neural network (BPNN) was used to establish infestation degree evaluation models based on full spectra, characteristic wavelengths, textural features and features fusion, respectively. BPNN models based on a fusion of characteristic wavelengths and textural features achieved the best performance, with classification accuracy of calibration and prediction sets over 95%. The accuracy of each infestation degree was satisfactory, and the accuracy of rice samples infested for 2 days was slightly low. In all, this study indicated the feasibility of hyperspectral imaging techniques to detect early SSB infestation and identify degrees of infestation.
